  • Crystal structure of two inclusion compounds Cd(pn)Ni(CN)4·0.25G (G=toluene and p-xylene, pn=1,2-diaminopropane) have been determined from single-crystal X-ray diffraction data; toluene clathrate: monoclinic P21/n, a=13.838(6), b=26.893(5), c= 7.543(5) Å, γ=90.92(3)°, Z=4, R=0.0616; p-xylene clathrate: monoclinic P21/n, a=13.895(2), b=26.900(3), c=7.613(1) Å, γ=91.06(1)°, Z=4, R=0.0486. The host structures determined for toluene- and pxylene-guest clathrates are substantially identical to the U-type structures observed for the straight chain aliphatic-guest clathrates. However, the alternating arrangement of occupied channels with the guest molecules and vacant channels appears in the host structure. The non-centrosymmetric toluene molecules are distributed about the inversion center to give an image like p-xylene molecule. The guests, toluene and p-xylene, prefer the U-type channel, favoring the interaction between the π-electrons of the aromatic ring and the pn-amino groups to hold the aromatic ring vertical to the cyanometallate meshes.

크실렌 노출로 인한 요중 메틸마뇨산 배설에 미치는 유전자 다형성 연구 (A Study on Polymorphism Affecting Excretion of Urinary Methylhippuric Acid due to Xylene Exposure)

  • Objectives : The purpose of this study was to investigate the effect of genetic polymorphism of cytochrome P450 2E1 (CYP2E1) and aldehyde dehydrogenase 2 (ALDH2) on the xylene metabolism. Methods : Among 247 workers, 116 were occupationally exposed to xylene and 131 were not. Workers exposed to xylene had different work such as spray, touch-up, mix & assist, and pre-treat. Questionnaire variables were age, sex, use of personal protective equipment, smoking, previous night's drinking and work duration. The urinary methylhippuric acid was measured in the urine collected in the afternoon and corrected by urinary creatinine concentration. The genotypes of CYP2E1 and ALDH2 were investigated by using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) methods with DNA extracted from venous blood. Results : 1. The urinary concentrations of o-, m-, and p-methylhippuric acid and total methylhippuric acid in the exposed group were significantly higher than those in the non-exposed group (p<0.001). 2. In multiple regression analysis, the urinary methylhippuric acid concentration was significantly influenced by exposure grade (Job-exposure matrixes), smoking, drug use and kind of protective equipment (p<0.1). 3. Genetic polymorphism of CYP2E1 and ALDH2 did not affect urinary methylhippuric acid level in the exposed group (p>0.05). Conclusions : Exposure grade, smoking, drug use and kind of protective equipment affected urinary methylhippuric acid level, whereas genetic polymorphism of CYP2E1 and ALDH2 did not. However, further investigation for the effect of genetic polymorphism on the metabolism of xylene with a larger sample size is needed.

인삼백합탕(人蔘百合湯) 및 인삼백합탕가미방(人蔘百合湯加味方)의 효능(效能)에 관(關)한 부험적(實驗的) 연구(硏究) (A study on the effects of Insambaekhabtang and Insambaekhabtanggamibang on the Experimental lung damage and the hemostasis)

  • The experimental studies were done to research the clinical effects of lnsambaekhabtang and Insambaekhabtanggamibang on the xylene-poisoned-lung damage and hypoprothrombinemia induced by warfarin injection in rats and mice. The results are as follows: 1. As to the hemostatic time in mice, lnsambaekhabtang was recongnized no significance but Insambaekhabtanggamibang was recognized significance( P<0.02 ). 2. As to the prothrombine time in rats. lnsambaekhabtang and lnsambaekhabtanggamibang Were recognized significance ( P<0.05, P<0.05 ). 3. As to the recalcification in rats, Insambaekhabtang and lnsambaekhabtanggamibang Were recognized significance ( P<0.05, P<0.02). 4. As to the lung TBA values in Xylene intoxicated rats, Insambaekhabtang was recognized significance but Insambaekhabtanggamibang was no recognized significance ( P<0.05). 5. As to the oxygen consumption in Xylene intoxicated rats, Insambaekhabtang and lnsambaekhabtanggamibang were no recognized significance. According to the above findings, it is suggested that the solid extract of Insambaekhabtang and Insambaekhabtanggamibang revealed effects on the xylene-poisoned-Iung damage and hypoprothrombinemia induced by warfarin injection in rats and mice.

대학생들의 혈액 내 휘발성 유기화합물 노출에 영향을 미치는 요인 (Determinant Factors for Blood VOCs Exposures in College Students)

  • Objectives: The objective of this study was to identify determine factors for blood VOCs levels in college students. Methods: Venous blood samples were collected from healthy 29 male and 25 female volunteers. The samples were analyzed with solid-phase microextraction gas chromatography and mass spectrometry. Results: Blood concentrations of benzene, toluene, ethylbenzene, m-,p-xylene, o-xylene were higher in smoking group than in non-smoking group. Geometric means of smoking group and non-smoking group were 0.45 and $0.36{\mu}g/L$ in benzene, 0.82 and $0.47{\mu}g/L$ in toluene, 0.44 and $0.41{\mu}g/L$ in ethylbenzene, 0.49 and $0.39{\mu}g/L$ in m-,p-xylene and 0.47 and $0.43{\mu}g/L$ in o-xylene, respectively. Gender, alcohol drinking, coffee, commuting time (30 minutes) by a car and adjacency between home and road were not associated with increasing blood VOCs concentrations of college students. Conclusion: Smoking habit was significantly associated with blood concentrations of benzene, toluene, ethylbenzene, o-,m-,p-xylene in college students.

  • In order to develop the methods for exposure assessment and find susceptibility markers for the workers who are exposed to low doses of toluene, xylene and other chemical in petroleum industries, we investigated the application of P-450 expression in human lymphocytes utilizing mouse monoclonal anti-rat CYP2B1/2, the levels of toluene and xylene in air and their metabolite levels in urine with the levels of expressed CYP2B1/2 proteins. The general characteristics such as age, smoking and drinking habit were no significant difference between the control and exposed workers, but the working durations and working hours were significantly different. Workers in exposed group were exposed to the mean of 2.1 ppm (range, 0.00-4.54) of toluene and 0.3 ppm (rang, 0.00-1.23) of xylene. The mean concentration of urinary hippuric acid was low and less than 1/5 of the biological exposure index recommended by the Ministry of Employment and Labor Korea. Methyl hippuric acid in urine was not detected in control and exposed workers. Also, there were no significant differences in the levels of the urinary metabolites between the control and exposed group. When chemiluminescence dot blottings were carried out utilizing mouse monoclonal antibody against CYP2B1/2, the strong density dots corresponding to a mouse monoclonal antibody was observed in the human lymphocytes from the exposed workers. These results suggested that the chemiluminescence dot blot assay for CYP of lymphocytes should be valuable for identifying CYP expression as biomarkers in the workers exposed to toluene and xylene.

Isolation and Characterization of a Rhodococcus Species Strain Able to Grow on ortho- and para-Xylene

  • Rhodococcus sp. strain YU6 was isolated from soil for the ability to grow on o-xylene as the sole carbon and energy source. Unlike most other o-xylene-degrading bacteria, YU6 is able to grow on p-xylene. Numerous growth substrate range experiments, in addition to the ring-cleavage enzyme assay data, suggest that YU6 initially metabolizes 0- and p-xylene by direct aromatic ring oxidation. This leads to the formation of dimethylcatechols, which was further degraded largely through meta-cleavage path-way. The gene encoding meta-cleavage dioxygenase enzyme was PCR cloned from genomic YU6 DNA using previously known gene sequence data from the o-xylene-degrading Rhodococcus sp. strain DK17. Subsequent sequencing of the 918-bp PCR product revealed a $98\%$ identity to the gene, encoding meth-ylcatechol 2,3-dioxygenase from DK17. PFGE analysis followed by Southern hybridization with the catechol 2,3-dioxygenase gene demonstrated that the gene is located on an approximately 560-kb megaplasmid, designated pJY J1

폴리메틸벤젠과 할로겐사이의 전하이동착물생성에 관한 열역학적 연구 (제1보) (The Thermodynamics of the Formation of Polymethylbenzene-Halogens Charge Transfer Complexes (Ⅰ))

  • o-, m- 및 p-크실렌계와 요오드사이에 형성되는 전하이동착물을 사염화탄소용액에서 자외선분광광도법을 사용하여 연구한 결과$ C_6H_4(CH_3)_2{\cdot}I_2 $형의 1:1 분자착물이 형성됨을 알았다. 이들 착물생성에 대한 흡수최대는 온도가 상승함에 따라 blue shift되므로 이를 고려하여 각 온도에서의 평형정수를 구했다. 이 값으로부터 이들 착물생성에 대한 $ {\Delta}$H, $ {\Delta}$G 및 $ {\Delta}$S의 열역학적 파라미터를 산출하였다. 이 결과와 전 연구결과를 종합하면 폴리메틸벤젠과 요오드와의 착물의 상대적 안정도가 다음 순서로 증가함을 알 수 있었다. Benzene < Toluene < o-Xylene < m-Xylene < p-Xylene 이 결과는 positive inductive effect에 의한 전자밀도증가와 steric hindrance effect에 의한 영향때문인 것으로 설명할 수 있다.

관능기화 메조포러스 실리카를 이용한 파라자일렌 흡착 특성 (Characteristics of p-Xylene Adsorption using Functionalized Mesoporous Silica)

  • 본 연구에서는 지하수 내의 BTEX 제거를 위한 흡착제로서 다양한 관능기화 메조포러스 실리카를 검토하였다. BTEX 물질로는 파라자일렌이 사용되었다. 관능기화 메조포러스 실리카는 공중합을 통해 MCM-41 기반 구조에 모노아민, 트리아민, 니트릴, 페닐, 옥틸기를 부착하여 합성되었다. 관능기화 메조포러스 실리카에서 파라자일렌 흡착 부위로 작용하는 부위는 Si-O-Si 결합, 계면활성제, 관능기인 것으로 사료된다. 관능기화 메조포러스 실리카의 자일렌 흡착능을 비교한 결과, 옥틸기를 부착한 경우에 MCM-41에 비해 자일렌 흡착능이 향상되었으며, 계면활성제로는 stearyltrimethylammonium chloride(STMACl)가 cetyltrimethylammonium bromide(CTMABr)에 비해 우수하였다. MCM-41을 기본 구조로, 옥틸기를 관능기로, STMACl을 계면활성제로 사용한 메조포러스 실리카는 $20^{\circ}C$에서 Langmuir model 상에서 최대 4.17 mmol/g의 최대 흡착량을 가질 것으로 평가되었으며, 이는 MCM-41에 비해 약 2.9배 높은 수치였다.

Highly sensitive xylene sensors using Fe2O3-ZnFe2O4 composite spheres

  • Pure ZnFe2O4 and Fe2O3-ZnFe2O4 hetero-composite spheres were prepared by ultrasonic spray pyrolysis of a solution containing Zn- and Fe-nitrates. Additionally, the sensing characteristics of these spheres in the presence of 5 ppm ethanol, benzene, p-xylene, toluene, and CO (within the temperature range of 275-350 ℃) were investigated. The Fe2O3-ZnFe2O4 hetero-composite sensor with a cation ratio of [Zn]:[Fe]=1:3 exhibited a high response (resistance ratio = 140.2) and selectivity (response to p-xylene/response to ethanol = 3.4) to 5 ppm p-xylene at 300 ℃, whereas the pure ZnFe2O4 sensor showed a comparatively lower gas response and selectivity. The reasons for the superior response and selectivity to p-xylene in Fe2O3-ZnFe2O4 hetero-composite sensor were discussed in relation to the electronic sensitization due to charge transfer at Fe2O3-ZnFe2O4 interface and Fe2O3-induced catalytic promotion of gas sensing reaction. The sensor can be used to monitor harmful volatile organic compounds and indoor air pollutants.

Formation of humus-bound residues in the course of BTX biodegradation in soil

  • To examine whether the xylene component of BTX (benzene, toluene, xylene) mixture is cometabolized and residues are produced in soil, $\^$14/C-labeled-0-xylene was added to sandy loam in combination with unlabeled benzene and toluene. After 4 weeks of incubation in a sealed system connected to an oxygen reservoir, 55.1% of the radiocarbon was converted to $\^$14/CO$\sub$2/, 3.0% was to 95.8% radiocarbon recovery. Biomass incorporation of o-xylene radiocarbon which was detected by fumigation/extraction was usually low (5.6%), but 32.1% radiocarbon became associated with soil humus. Most of the numus-bound radiocarbon was found in humin fraction. In addition to o-xylene, p-xylene and toluene also showed similar results. The evidence shows that some of their reactive methylcatechol biodegradation intermediates attach to the humic metrix in soil in preference to mineralization and biomass incorporation.
