Release of Cytochrome c from Isolated Mitochondria by Etoposide

  • The efficacy of chemotherapeutic agents on tumor cells has been shown to be modulated by tumor suppressor gene p53 and its target genes such as Bcl-2 family members (Bax, Noxa, and PUMA). However, various chemotherapeutic agents can induce cell death in tumor cells that do not express the functional p53, suggesting that some chemotherapeutic agents may induce cell death in a p53-independent pathway. Here we showed that etoposide can induce the similar degree of cell death in p53-deficient HCT 116 cells, whereas 5'-FU-mediated cell death is strongly dependent on the existence of functional p53 in HCT 116 cells. Further, we provide the evidence that etoposide can induce the cytochrome c release from isolated mitochondria, and etoposide-induced cytochrome c release is not accompanied with the large amplitude swelling of mitochondria. These data suggest that etoposide can directly induce the mitochondrial dysfunction irrespective of p53 status, and it may, at least in part, account for the p53-independent pathway in cell death induced by chemotherapeutic agents.

Detection of p53 Common Intron Polymorphisms in Patients with Gastritis Lesions from Iran

  • Background: p53 alterations have been implicated in the development of many cancers, such as gastric cancer, but there is no evidence of p53 intron alterations in gastritis lesions. The aim of this study was to investigate the p53 intron alterations in gastritis along with p53 and mismatch repair protein expression and microsatellite status. Materials and Methods: PCR-sequencing was conducted for introns 2-7 on DNA extracted from 97 paired samples of gastritis lesions and normal adjacent tissue. Abnormal accumulation of p53 and mismatch repair proteins was investigated using immunohistochemistry. In addition, microsatellite status was evaluated with reference to five mononucleotide markers. Results: Gastritis cases included 41 males and 56 females in the age range of 15-83 years, 87.6% being H.pylori positive. IVS2+38, IVS3ins16 and IVS7+72 were the most polymorphic sites. Their minor allele frequency values were as follows: 0.38, 0.21 and 0.06, respectively. Samples with GG genotype at IVS2+38 and CT at IVS7+72 had no insertion. Moreover, most of the stable samples (91.9 %) had a G allele at IVS2+38. All of the samples were IHC negative for p53 protein, microsatellite stable and expressed mismatch repair proteins. p53 alterations were prominent in the H. Pylori+ group, but without statistical significance. Conclusions: According to our results, some p53 polymorphisms such as IVS2+38, IVS3ins16 and IVS7+72, because of their correlations together or with microsatellite status may contribute to gastritis development. However, so far effects on p53 expression and function remain unclear. Therefore, a comprehensive survey is needed to delineate their biological significance.

Effects of Microbial Phytase Supplementation on Growth Performance and Nutrient Excretion in Growing Pigs (육성돈에 있어 성장 및 영양소 배설에 대한 미생물성 Phytase의 첨가 효과)

  • For Exp, 1, eighty [(DurocYorkshire)Landrace) pigs(38.63$\pm$0.68kg average initial BW) were used to evaluate the effects microbial phytase supplementation on the growth performance and nutrient digestibility in growing pigs. There were ten pigs per pen and four pens per treatment. Treatments were 1) CON(com-SBM based diet), 2) Low-P(low-P diet+500 FTU/kg microbial phytase). For overall period, ADG, ADFI and gain/feed were significantly improved(P<0.05) by supplementation of Low-P diet. Also, pigs fed Low-P diet increased N, crude ash, Ca and P digestibilities compared to pigs fed CON diet(P<0.05). For Exp. 2, sox [(DurocYorkshire) Landrace) pigs(32.7kg average initial BW) were used to evaluate the effects microbial phytase supplementation on the nutrient excretion in growing pigs. Experimental treatments were as described for Exp. 1. pigs fed Low-P diet decreased N(8.70 vs 6.43g/d), crude ash(38.13 vs. 25.33 g/d), Ca(6.03 vs. 3.93g/d) and P(4.53 vs. 2.77g/d) excretion compared to pigs fed CON diet(P<0.05). In conclusion, microbial phytase supplementation in low P diets improved growth and nutrient digestibility.

Impact of Career Identity and Career Decision-Making Type on Career Preparation Behavior among Nursing Students (간호대학생의 진로정체감과 진로의사결정유형이 진로준비행동에 미치는 영향)

  • This study identified the impact of career identity and career decision-making type on career preparation behavior in nursing students. The participants were 198 nursing students enrolled in the 3rd and 4th grades located in J province, and data were collected from June 14, 2021 to 27, 2021, using an online questionnaire. Data were analyzed with descriptive statistics, independent t-test, one-way ANOVA, Pearson's correlation, and hierarchical regression using SPSS/WIN 23.0 program. As a result, the mean score of career identity, rational type, intuitive type, dependent type, and career preparation behavior were 2.79(range 1~4), 3.78(range 1~5), 3.38(range 1~5), 3.01(range 1~5), and 3.51(range 1~5), respectively. Career preparation behavior was significantly correlated with career identity(r=.40, p<.001), rational type(r=.50, p<.001), intuitive type(r=.22, p=.002), and dependent type(r=-.20, p=.004). Factors influencing career preparation behavior were career identity(β=.23, p=.001), rational type(β=.31, p<.001), intuitive type(β=.27, p<.001), and dependent type(β=-.20, p=.002). These variables accounted for 24.6%p of the variance in career preparation behavior(F=11.93, p<.001). These research results suggest that it is necessary to establish a career identity and develop and apply a program that includes career guidance according to career decision-making type to improve the career preparation behavior of nursing students.

The Effect of Foot Cooling on Body Temperature (발바닥 부위 쿨링이 심부 체온에 미치는 효과)

  • In this study, We investigated the effect of foot cooling on the reduction of body temperature after hard exercise at the high temperature of $40^{\circ}C$. We performed a total of 30 subjects, and the subjects performed treadmill exercise for 30 minutes. We produced the cooling device to cool the foot using Peltier module. After the end of the exercise, We performed normal recovery method and cooling recovery method(one foot, both feet) for 1 hour on the same indoor environmental conditions and confirmed the change of body temperature of subjects. The results of deep body temperature measurement showed average $38.78{\pm}0.22^{\circ}C$ to $38.54{\pm}0.15^{\circ}C$ when the normal recovery method was performed. Cooling recovery method on one foot showed average $38.69{\pm}0.14^{\circ}C$ to average $38.06{\pm}0.17^{\circ}C$ and Cooling recovery method on both feet showed average $38.69{\pm}0.15^{\circ}C$ to average $37.84{\pm}0.21^{\circ}C$. There was a significant difference between the normal recovery method and the one foot cooling recovery method(p < .05), there was a significant difference between the normal recovery method and the both feet cooling recovery method(p < .05) and there was a significant difference between the one foot cooling recovery method and the both feet cooling recovery method(p < .05). Body temperature showed the lowest decrease rate when the normal recovery method was performed, and body temperature showed the highest decrease rate when the both feet cooling recovery method was performed. Therefore, recovery of cooling on the foot after hard exercise have decreased body temperature, delay fatigue in the body, and will be contributed to improvement of athlete performance.

Feeding broiler chicks with Schizosaccharomyces pombe-expressed phytase-containing diet improves growth performance, phosphorus digestibility, toe ash, and footpad lesions

  • Objective: The objective of this study was to evaluate the effects of dietary supplementation of Schizosaccharomyces pombe (S. pombe) -expressed phytase on growth performance, apparent ileal digestibility, organ indexes, meat quality, toe ash, and footpad lesions score in broiler chicks. Methods: A total of 390 one-day-old broiler chicks were randomly assigned to 5 groups based on the initial body weight (42.15±0.17 g), there were 6 replicate cages per treatment and 13 birds (mixed sex) per cage. The experimental period was 45 days, including 4 periods (starter, days 1 to 10; grower, days 11 to 24; finisher 1, days 25 to 38; finisher 2, days 39 to 45). Dietary treatments were based on a corn-soybean meal-basal diet and supplemented with 500, 750, 1,000, and 1,500 FTU/kg S. pombe-expressed phytase. One phytase unit (FTU) was defined as the amount of enzyme that catalyzes the release of one micromole phosphate from phytate per minute at 37℃ and pH 5.5. Results: The inclusion of increasing levels of phytase in the diet linearly increased the body weight gain during days 1 to 10 (p = 0.001), 25 to 38 (p = 0.016), 39 to 45 (p = 0.018), and 1 to 45 (p = 0.004), feed intake during days 25 to 38 (p = 0.032), feed conversion ratio during days 1 to 10 (p = 0.001), 39 to 45 (p = 0.038), and 1 to 45 (p = 0.012), carcass weight (p = 0.035), toe ash (p<0.001), and apparent ileal phosphorus digestibility (p = 0.049). However, the footpad lesions score (p = 0.040) decreased linearly with the increase in phytase levels in the diet. Conclusion: Dietary supplementation of S. pombe-expressed phytase was beneficial to the growth performance, toe ash, apparent ileal phosphorus digestibility, and footpad lesions of broiler chicks in a dose-dependent manner.

  • Paper electrophoresis of aluminium ion was studied using a closed horizontal type apparatus. The optimal conditions of voltage, pH of electrolytic supporting solution and migration time were determined for the electrophoretic migration of the ion on the paper strip saturated by Michaelis buffer solution. The mobility of the ion and the isoelectric point of $Al(NO_3)_3$ solution were $(1.004{\pm}0.0020){\times}10^{-4}\;cm^2\;sec^{-1}\;volt^{-1}$ at pH = 2.38 and pH = 3.23 respectively.

Direct Interaction between Ras Homolog Enriched in Brain and FK506 Binding Protein 38 in Cashmere Goat Fetal Fibroblast Cells

  • Ras homolog enriched in brain (Rheb) and FK506 binding protein 38 (FKBP38) are two important regulatory proteins in the mammalian target of rapamycin (mTOR) pathway. There are contradictory data on the interaction between Rheb and FKBP38 in human cells, but this association has not been examined in cashmere goat cells. To investigate the interaction between Rheb and FKBP38, we overexpressed goat Rheb and FKBP38 in goat fetal fibroblasts, extracted whole proteins, and performed coimmunoprecipitation to detect them by western blot. We found Rheb binds directly to FKBP38. Then, we constructed bait vectors (pGBKT7-Rheb/FKBP38) and prey vectors (pGADT7-Rheb/FKBP38), and examined their interaction by yeast two-hybrid assay. Their direct interaction was observed, regardless of which plasmid served as the prey or bait vector. These results indicate that the 2 proteins interact directly in vivo. Novel evidence is presented on the mTOR signal pathway in Cashmere goat cells.

Paclitaxel Stimulates Cyclooxygenase-2 Expression via MAP Kinase Pathway in Rabbit Articular Chondrocytes

  • Paclitaxel, an antimicrotubule agent, binds to beta-tubulin in the microtubule and stabilizes the polymer, thereby repressing dynamic instability. Here, we have demonstrated that microtubule cytoskeletal architecture involved in regulation of the COX-2 expression in chondrocyte treated with paclitaxel. Paclitaxel enhanced COX-2 expression and prostaglandin E2 production, as indicated by the Western blot analysis, reverse transcriptase PCR(RT-PCR) and immunofluorescence staining, and $PGE_2$ assay, respectively. In our previous data have shown that paclitaxel treatment stimulated activation of ERK-1/2 and p38 kinase(Im et al., 2009). SB203580, an inhibitor of p38 kinase, blocked the induction of COX-2 expression by paclitaxel. Also PD98059, an inhibitor of ERK-1/2 kinase was blocked the induced COX-2 expression. These results indicate that activation of ERK-1/2 and p38 kinase is required for COX-2 expression induced by paclitaxel in rabbit articular chondrocytes.

The Regulation of Osteoclastogenesis by L-Type Channel Agonist (L-형 칼슘 이온통로에 의한 파골세포 분화의 조절)

  • We investigated the role of L-type $Ca^{2+}$ channel in receptor activator of nuclear factor-kappaB ligand (RANKL)-induced osteoclast formation. BayK 8644, a L-type $Ca^{2+}$ channel agonist, was shown to increase the RANKLinduced osteoclastogenesis and actin ring formation in mouse bone marrow-dereived macrophage (BMM) culture system. BayK 8644 stimulated RANKL-induced extracellular signal-regulated kinase (ERK) and p38 MAP kinase (MAPK) activation, which leads to increased nuclear factor of activated T cells (NFAT)c1 expression. Taken together, these data indicate that L-type $Ca^{2+}$ channel regulates osteoclast formation possibly through ERK- and p38-mediated NFATc1 expression.
