굴의 질병에 관한 연구 1, 병리 조직학적 연구 (STUDIES ON THE OYSTER DISEASES 1. Pathogenetic Investigation)

  • The present paper deals with mortality and pathogenetic investigation of the oysters Crassostrea gigas cultured by tile coventional bamboo and hanging method in Kimhae and Koje-Do in 1969. The results of the investigation may be summarized as follows: 1. Mortality of the oysters by the bamboo method in Kimhae was $9.5\%$ in June, $10.2\%$ In July, $12.3\%$ in August and $12.1\%$ in September, respectively, 2. Mortality of the oysters cultured by the hanging method in Koje-Do was $2.4\%$ in June, $2.9\%$ in July, $30.7\%$ in October, $37.3\%$ in November and $30.0\%$ in December, respectively 3. The diseased oysters had severe inflammation, necrosis and multiple abscess in the epithelia of stomach, mid-gut, digestive tubules, blood vessels and gonads, mucous membrane and surrounding tissue. 4. From August gram negative bacteria were found in the nodules of connective tissue and multiple abscess of the diseased oysters. Particularly the connective tissue of the diseased oysters contained more bacteria than epithelia. 5. Since the bacteria are less abundant in the region of digenerated tissue, mortality of the oysters is not caused only by the infectious bacteria but seems that is also caused by other environmental factors such as extreme temperatures and salinities.

다환방향족탄화수소 (PAHs) 에 노출된 굴, Crassostrea gigas의 생리 반응 변화 (The change of the physiological response of the Crassostrea gigas exposed to PAHs)

  • PAHs (Polycyclic Aromatic Hydrocarbons: PAHs) is the hydrophobic inorganic material composed of carbon and hydrogen that is easily adsorbed biological organisms in the ocean. Bivalves is the indicator of environment monitoring because of reflect growth, physiological response of bivalve followed their habitat environment. The aim of research is understand the change of oysters (Crassostrea gigas) physiological response under exposed PAHs concentration for control, 1, 10 and $100{\mu}g/L$. We investigated induced immune change response for oyster hemocyte and effect of tissue RNA/DNA ratio for mantle, gill and adductor muscle individually. As a result of experiment change of immune response the oyster hemocyte when exposed PAHs showed that viability and adhesion is no significant difference (ANOVA test, p < 0.05). However phagocytosis decreased under the over $10{\mu}g/L$ of PAHs concentration and ROS increased with the increase of PAHs concentration. The change of RNA/DNA ratio is R/D ratio decreased with the increase of PAH concentration in adductor muscle. However gill and mantle showed no change of R/D ratio with PAHs concentration. The oysters when exposed inorganic pollutant that decreased of physiological condition and damaged protein synthesis of adductor muscle.

양식환경이 통영 북만의 참굴, Crassostrea gigas에 기생하는 난소기생충, Marteilioides chungmuensis 감염에 미치는 영향 (Influence of Environmental Factors on the Prevalence of the Ovarian Parasite, Marteilioides chungmuensis, in Crassostrea gigas, Cultured in Pukman Bay, Tongyeong)

  • Occurrence and prevalence of Marteilioides chungmueasis have been reported in several waters around Tongyeong but no report has been made for Pukman bay. Therefore, we investigated that the prevalence and infection intensities in Pukman Bay at the inside and the outside areas which are hydrographically divided by tidal current. Furthermore, various environmental parameters were investigated in order to elucidate effective parameter for parasitic infection. Infection rates of Marteilioides chungmuensis in adult oysters were ranged 3.3-20.0% at the inside area during September 2002 through January 2003, and 3.3-30.9% at the outside area during August 2002 through January 2003. External manifestation of infected oyster consisted of abnormal egg masses with nodular appearance in the soft tissue. Histopathological symptoms included massive hemocytic infiltration within or around the follicle wall and atrophic epithelium of digestive diverticula. For the environmental parameters, comparative study made differences between two side of the Bay during the infection period: inside > outside for SS while inside < outside for chlorophyll-a. A positive relationship was observed between chlorophyll-a and infection period, which might indicate the difference in food availability between two areas. The prevalence of ovarian parasite Marteilioides chungmuensis, therefore, was highly associated with food availability. Pearson's correlation analysis was made between environmental parameters and infection prevalence. Significance was observed in water temperature (p < 0.05), suspended solids (p < 0.01) and chlorophyll-a (p < 0.05). A principle component analysis showed that infection of the ovarian parasite, Marteilioides chungmuensis, exhibited effects of seasonality (component I = 55.2%) and chemical/physical environmental factors (component II = 24.4%). These results clearly indicate that the infection of ovarian parasite, M. chungmuensis in the Pacific oyster Crassostrea gigas is closely associated with seasonality and food availability.

수확 전 염화칼슘 및 알카리환원수 처리가 느타리버섯 저장에 미치는 영향 (Effect of Preharvest Treatment of $CaCl_2$ and Alkaline-Reduced Water on the Quality of Oyster Mushroom during Storage)

  • This experiment was focused to determine the effect of preharvest treatment of alkaine-reduced water (ARW) and $CaCl_2$ on the storage of oyster mushroom. ARW was prepared through electrolysis of water with $CaCl_2$ (0.1%) as salts and $CaCl_2$ (0.1%) was dissolved in deionized water. Mushrooms were harvested after two sprays of each solutions and quality factors including firmness and browning was determined during storage. The severity of browning was more greater when stored 20 days. Unlike $CaCl_2$, ARW containing 139 ug/L calcium was positive on maintaining freshness in terms of delay of firmness loss and respiration drop, keeping soluble carbohydrate content such as trehalose, and inhibiting the occurrence of browning. Browning, however, seemed to be more influenced by the developmental stage of mushroom at harvest. Even no significant difference was found, the level of ${\beta}$-glucan was low in ARW treatment compared to that of control. Further research including application and preparation methods of ARW is required to clarify the potential of ARW on keeping freshness of oyster mushroom through inhibiting tissue browning.

  • The unwanted artificial oil-spill has severely contaminated the coastal environment in the world. Level of contamination has so far been monitered by various indicator species including mussel, oysters, flounder, and cockle. In this study, we decided to use the oyster as a model organism to observe the morphological changes being exposed to the artificial oil-spill in the coastal areas in Taean, Korea. The oysters were collected from four local sites (Sindu-ri, Uiwang-ri, Jonghyeon-dong, Ansan and Uihang-ri) exposed to various levels of pollution after an oil spill in Taean. Microscopic analysis of the hepatopancreatic microstructure in the digestive gland from the collected oysters show that the swelling, whorl, and destruction phenomenon of the nuclear membrane, a well-known microstructure induced by heavy metal exposure, was observed. Nuclear body (Nb), another typical characteristic of contamination or infection were also observed in some samples. Necrosis was observed in tissue samples collected from the area with a high degree of oil pollution. In addition, parasite-like particles (virus, perkinsus) were observed in most samples. Taken together, these results suggest that oil contamination in the oyster habitats influences the cytopathological changes in Crassostrea gigas.

Application of the rpoS Gene for Species-Specific Detection of Vibrio vulnificus by Real-Time PCR

  • Vibrio vulnificus is a causative agent of serious diseases in humans, resulting from the contact of wound with seawater or consumption of raw seafood. Several studies aimed at detecting V. vulnificus have targeted vvh as a representative virulence toxin gene belonging to the bacterium. In this study, we targeted the rpoS gene, a general stress regulator, to detect V. vulnificus. PCR specificity was identified by amplification of 8 V. vulnificus templates and by the loss of a PCR product with 36 non-V. vulnificus strains. The PCR assay had the 273-bp fragment and the sensitivity of 10 pg DNA from V. vulnificus. SYBR Green I-based real-time PCR assay targeting the rpoS gene showed a melting temperature of approximately $84^{\circ}C$ for the V. vulnificus strains. The minimum level of detection by real-time PCR was 2 pg of purified genomic DNA, or $10^3$ V. vulnificus cells from pure cultured broth and $10^3$ cells in 1 g of oyster tissue homogenates. These data indicate that real-time PCR is a sensitive, species-specific, and rapid method for detecting this bacterium, using the rpoS gene in pure cultures and in infected oyster tissues.

천연 추출물 첨가에 의한 생굴(Crassostrea gigas)의 식품학적 품질 유지 (Extending Raw Pacific Oyster Crassostrea gigas Shelf-life by Addition of the Natural Food Additives)

  • Oysters are a highly consumed seafood throughout Korea, but they have a short shelf life because they support rapid microbial growth due to their of high moisture content and fragile muscle tissue. We examined natural food additives including lactic acid bacteria fermentation powder, rosemary extract, and lemon juice for their ability to preserve raw oyster Crassostrea gigas quality. Samples were stored at 4℃, and microbiological and physicochemical analyses were conducted. Among the natural additives tested, lemon juice was the most effective. Lemon juice was thus applied at different concentrations (50-300 ppm) to quantitatively assess its effect on total viable cell count, pH, glycogen, soluble protein, and turbidity. 200 ppm was confirmed to be optimal, and is projected to extend shelf life by 2 days compared to the control group.

환경가스조절 저장방법을 이용한 느타리버섯과 표고버섯의 유통기간 연장 (Modified Atmosphere Storage for Extending Shelf Life of Oyster Mushroom and Shiitake)

  • 버섯의 유통기간을 연장하기 위한 방법으로 환경가스조절(modified atmosphere, MA) 저장의 효과를 조사하기 위하여 표고버섯과 느타리버섯을 두께가 $50\;{\mu}m$인 폴리에틸렌 필름으로 포장한 후 $0.5^{\circ}C$에서 저장하였다. 재래식 골판지 상자에 4kg씩 포장된 대조구 버섯은 수분증발, 외관수축, 변색 및 균사발생으로 인하여 유통기간이 $5{\sim}6$일에 불과하였다. MA 저장구의 버섯은 저장 4주 후에도 중량감소가 $1.6{\sim}1.7%$로 낮아 갓 안쪽 주름의 변형이 없었고 갓이 퍼지지도 않았으며 변색도 거의 없었는데 이는 포장내 습도가 포화상태로 유지되고 포장내 산소가 $1.0{\sim}1.3%$, 이산화탄소가 10% 이상으로 유지되어 버섯의 호흡이 억제되었기 때문으로 생각된다. MA 저장 느타리버섯은 조직연화가 품질을 결정하는 가장 중요한 요인이었으며 이를 기준으로 판단한 유통기간은 15일 정도였으며 MA 저장 표고버섯은 갓의 황변이 품질저하의 가장 중요한 요인이었으며 이를 기준으로 판단한 동고의 유통기간은 15일, 화고의 유통기간은 30일 이상이었다.

저장 온도와 포장재에 따른 느타리버섯의 MA 저장 중 품질변화 (Quality Changes in Oyster Mushrooms during Modified Atmosphere Storage as Affected by Temperatures and Packaging Materials)

  • 저장 온도와 포장재를 달리한 느타리버섯의 MA 저장 중 품질 변화를 알아보기 위해, 느타리버섯을 PE, EVA, 세라믹필름으로 각각 포장한 후 $0^{\circ}C,\;5^{\circ}C,\;10^{\circ}C$$20^{\circ}C$에 저장하면서 중량감소, 경도, 색변화, 필름 내부의 기체조성 및 조직 내 에탄을 함량을 조사하였다. 필름 처리한 버섯은 2kg 골판지 상자에 저장한 대조구 버섯의 중량이 급속히 감소한 것과 달리 필름의 종류에 관계없이 중량감소가 $0.8{\sim}1.3%$에 불과하였다. 경도는 대조구와 필름 처리구 모두 저장 초기에는 일시적으로 약간 증가하다가 시간이 경과함에 따라 감소하였으며 필름처리구가 대조구보다 높은 경도를 유지하였다. 버섯의 색은 저장 온도가 높을수록 황변이 많이 진행되긴 했으나 MA 저장에 의한 색변화 억제 효과가 나타남을 알 수 있었다. 필름 내부의 산소함량은 포장 하루만에 $1.2{\sim}1.4%$까지 급격히 낮아져 저장 기간 동안 안정적으로 유지됐으며, 이산화탄소 농도는 저장 1일까지는 비슷한 값을 보이다가 저장 2일 후부터 필름의 종류에 따라 뚜렷한 차이를 나타내어 세라믹 필름, PE, EVA 순으로 높게 나타났다. 저장 중 느타리버섯 조직의 에탄을 함량은 대조구에서는 초기치를 제외하고는 에탄올이 검출되지 않은 반면, MA 저장에서는 시간이 경과할수록, 저장 온도가 높을수록 에탄올 함량은 큰 폭으로 증가하였다. 부위별 에탄올 함량을 보면 기둥의 에탄올 함량이 갓보다 약간 높게 나타났으나 통계적인 차이는 보이지 않았다. MA 저장 버섯의 저장 수명은 $0^{\circ}C$$14{\sim}15$일, $5^{\circ}C$ 8일, $10^{\circ}C\;3{\sim}4$일, 그리고 $20^{\circ}C$ 2일 정도였다. 전체적인 실험결과를 볼 때 느타리버섯은 $0^{\circ}C$와 같은 저온 저장이 효과적이었으며, EVA필름 처리구가 세라믹필름이나 PE필름보다 효과적인 것으로 보인다.

굴 양식수역의 환경용량 산정 -II. 거제 · 한산만의 환경용량- (Estimating the Carrying Capacity of a Coastal Bay for Oyster Culture -II. The Carrying Capacity of Geoie-Hansan Bay-)

  • 과밀양식으로 생산성이 저하되고 있는 거제 $\cdot$ 한산만의 안정적이고 지속적인 굴 생산방안을 제시하기 위하여 생태계 모델을 이용하여 먹이 공급량을 추정하였고, 양식 굴의 여수율과 chloro-phyll $\alpha$ 농도로부터 먹이 요구량을 계산하여 양식 수용력을 산정한 결과는 다음과 같다. 수확 크기별 굴의 먹이 요구량은 습중량 4g의 알굴인 경우 1.40~4.82mgC/ind./day (평균 2.49mgC/ind./day), 습중량 7g의 알굴인 경우 1.96-6.77mgC/ind./day (평균 3.50mgC/ind./day) 범위로 나타나 알굴의 습중량이 증가할수록 먹이 요구량도 크게 나타났다. 월별로는 2월이 가장 작았고, 9월이 가장 크게 나타났다. 월별 먹이 공급량을 월별 먹이 요구량으로 나누어 수확 크기별 수용력을 산정한 결과 2월이 평균 6.10 ton/ha로 최저 수용력을 나타내었고, 4월이 14.91 ton/ha로 최대 수용력을 나타내었다. 거제 $\cdot$ 한산만의 알굴 생산량은 9ton/ha로 최대 수용력의 $60\%$ 수준이나 지속적 생산을 위해서는 임계 수용력인 2월의 6.1ton/ha와 통계자료에 의한 최대 생산량 5.5ton/ha를 감안하여 현재 시설량을 $32\%{\~}39\%$ 정도 줄여야 할 것으로 판단되었다.
