Study on the Manufacturing Technology of Mural Tomb in Goa-dong of Daegaya Period (대가야 고아동 고분벽화 제작기술에 관한 연구)

  • Rigorous analysis was performed to identify the structure and materials of the murals to study techniques used on mural tombs of ancient Daegaya era(6th century). The murals were painted by applying mortar on the walls and the ceiling after building a stone chamber and creating ground layers on mortar layers. Mud was applied on most of the mortar layers on four sides of the walls except the ceiling. Sand was not used in mortar but was made of materials with pure calcium substances. In addition, shells in irregular sizes with incomplete calcination were mixed; and the mortar's white powder was inferred as lime obtained by calcination of oyster shells. Kaolinite($Al_2Si_2O_5(OH)_4$) was used in the ground layer, Cinnabar(HgS) was used for red pigment, Malachite($Cu_2CO_3(OH)_2$) for green and Lead white($PbCO_3{\cdot}Pb(OH)_2$) for white. Mud plaster was applied on the mortar and was composed thinly and densely using clayey of particle size smaller than that of medium sand. It was assumed that the finishing was for repair after long time had passed since the mortar layer came off. Using lime made with oyster shells as mortar is unprecedented in ancient Korean mural tombs and its durability was very poor, suggesting that Gaya's mortar production technique was relatively behind compared to that of Koguryo's in the same era.

Characterizations of Shell and Mantle Edge Pigmentation of a Pacific Oyster, Crassostrea gigas, in Korean Peninsula

  • The objectives of this study were to investigate color patterns of shell and mantle edge pigmentation of a Pacific oyster, C. gigas, and to estimate variance components of the two colors. A sample of 240 F0 oysters was collected from six aquaculture farms in Tongyeong, Korea to measure shell color and mantle edge pigmentation. Among the F0s, male and female individuals with black (white) shell and black (white) mantle edge were selected and mated to generate three F1 full-sib black (white) cross families (N = 265). Two and four F2 cross families (N = 286) were also produced from black and white F1 selected individuals, respectively. Variance component estimates due to residuals and families within color were obtained using SAS PROC VARCOMP procedures to estimate heritability of shell and mantle edge pigmentation. In the F0 generation, about 29% (11%) had black (white) color for both shell and mantle edge. However, in the F1 and F2 black (white) cross families, 75% (67%) and 100% (100%) of oysters had black (white) shell colors, and 59% (23%) and 79% (55%) had black (white) mantle edge, respectively. Spearman correlation coefficients between shell and mantle edge color were 0.25, 0.74, and 0.92 in F0, F1, and F2 generations, respectively, indicating that, with generations of selection process, an individual with black (white) shell color is more likely to have black (white) mantle edge pigmentation. This suggests that shell color could be a good indicator trait for mantle edge pigmentation if selection of both the colors is implemented for a couple of generations. Estimates of heritability were 0.41 and 0.77 for shell color and 0.27 and 0.08 for mantle edge pigmentation in the F1 and F2 generations, respectively, indicating that, in general, significant proportions of phenotypic variations for the shell and mantle edge colors are explained by genetic variations between individuals. These results suggest that the two color traits are inheritable and correlated, enabling effective selection on shell and mantle edge color.

Application of Waste Resources for the Stabilization of Heavy Metals (Pb, Cu) in Firing Range Soils (폐자원을 이용한 사격장 토양내 중금속(Pb, Cu) 안정화 처리)

  • In this study, a heavy metal stabilization treatment using waste resource stabilizing agents was utilized on army firing range soil contaminated with Pb and Cu. Both calcined oyster shells (COS; 5% w/w) and waste cow bone (WCB; 3% w/w) were applied for a wet-curing duration of 28 days. Following the stabilization treatment, the process efficiency was evaluated by various extraction methods for Pb and Cu. Neutral and weak acid extraction methods, such as water soluble extraction and SPLP, did not show positive results for heavy metal stabilization with very low leachability. On the other hand, TCLP and 0.1 N HCl extraction showed that the stabilizing agents significantly reduced the amount of the heavy metals leached from the soil, which strongly supports that the treatment efficiency is positively evaluated in acidic leaching conditions. Specifically, in the 0.1 N HCl extraction, the reduction efficiencies of Pb and Cu leaching were 99.9% and 83.9%, respectively. From the sequential extraction results, a difference between Pb and Cu stabilization was observed, which supports that Pb stabilization is more effective due to the formation of insoluble Pb complexes. This study demonstrates that the application of waste resources for the stabilization of heavy metals is feasible.

Studies on the development of mushroom media for bottle culture in new Pleurotus ostreatus 'Miso' (신품종 '미소' 느타리버섯 병재배 배지개발)

  • This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.

Characteristics and breeding of a new cultivar Pleurotus eryngii var. ferulae, 'Beesan No.2' (아위느타리 신품종 '비산2호'의 육성 및 자실체 특성)

  • To develop a new cultivar of King oyster mushroom white variety (Pleurotus ferulae), GW10-68 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2798-24 derived from ASI 2798 and dikaryotic strain ASI 2803. The GW10-95(ASI 2803 ${\times}$ ASI 2798-24) was shown the best cultural characteristics, selected to be a new cultivar and designed as 'Beesan No.2'. The 'Beesan No.2' was formed incompatibility line distinctly in the confrontation growth of parental strains ASI 2803, ASI 2798 and Beesan No.1. Analysis of the genetic characteristics of the new cultivar 'Beesan No.2' showed a different DNA profile as that of the control strains, ASI 2803, ASI 2798 and 'Beesan No.1', when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about 123.3 g. And also the stipe was long. This new cultivar 'Beesan No.2' of Pleurotus ferulae was characterized white-like variety of King oyster mushroom in the color of stipe, that was long and low yield compared to that of other cultuvar 'Beesan No.1'. We therefore expect that this new strain will increase of export and substitute cultivar of King oyster mushroom.

Daily Variation of Particulate Organic Carbon in Wonmun Bay on the South Coast of Korea in Late Summer (늦여름 원문만 굴양식장 입자유기탄소의 일변동)

  • Daily variation of particulate organic carbon(POC) and some factors controlling its level were examined for a semi-enclosed bay(Wonmun Bay, south coast of Korea), in which a lot of suspended oyster culture farms existed, in September, 1992. Observations were made at hourly interval. In spite of the relatively short survey period, strong short-term variation of POC concentration could be observed. Concentrations of POC were the range of $58{\sim}582{\mu}g/l(average 272{\mu}g/l)$ and their variation pattern was similar to those of chlorophyll a with the range of $0.90{\sim}7.25{\mu}g/l(average 3.35{\mu}g/l)$. The low C/N ratios also suggested that marine microalgae was a major component of POC for Wonmun Bay. Primary production, average $1.97\;gC/m^2/day$, was the main source of POC beacuse the supply of POC via freshwater input and exchange with the outer part of the bay was little. Oyster population also excreted a small amount of POC. About $40\%$ of produced POC was decomposed heterotrophically. Another important cause for the fluctuation of observed POC was tidal cycle. Considerable POC, which amounted $37\%$ of produced POC, was lost from the bay due to flushing by tidal cycle. It was also calculated that about $16\%$ was transported onto the sediment. It seemed that a part of POC was consumed by oyster and other heterotrophs.

Analysis of nutritional contents and useful functional materials for finding breeding resources in Flammulina velutipes (팽이 기능성 육종소재 발굴을 위한 영양성분 및 유용 기능성 물질 분석)

  • Flammulina velutipes, known as winter mushroom in the family of Physalacriaceae, is the main edible and export mushroom with the third highest production after oyster and king oyster mushroom in Korea. However, as normal consumers regard F. velutipes as a simple subsidiary material, there is a limitation to increasing mushroom demand. In order to overcome the consumption limit and increase the differentiation of new varieties, it is necessary to breed varieties with enhanced functionality in consideration of consumer preferences. Therefore, the study was performed to analyze nutrient components and several useful functional substances with 26 genetic resources of F. velutipes. Analyses of inorganic compound(Ca, K, Mg) and 15 amino acids revealed that Strain 4148 had the highest content among the 26 strains. Beta-glucan, which increases immune activity and polyphenol, which exert antioxidant effects were higher in non-white strains than in white strains with a small number of exceptions. Among the five fatty acids, linoleic acid(an omega-6 fatty acid) and α-linolenic acid(an omega-3 fatty acid), were detected in six mushroom strains. α-linolenic acid, which was not found in five major mushrooms including oyster mushrooms, was identified in F. velutipes. The results of HPLC analysis showed that 'Auram' (Strain 4232) and 'Baekseung'(Strain 4230) had the highest content of the stabilizing neurotransmitter GABA(15.38 ㎍/ml and 20.56 ㎍/ml, respectively) among non-white and white strains, respectively. Our findings provide useful information for breeding F. velutipes to obtain strains with enhanced functionality.

  • We investigated the possibility of a bio-monitoring system for detecting hypoxic water in coastal area using shell valve movements of Pacific oyster (Crassostrea gigas), which showed most aquaculture production in Korea, with Hall element sensor. In filtrating water to confirm shell valve movement (SVM) under normal condition, it showed spikes which mean a relatively fast closing condition after opened condition of average 5~12 mm, and then the SVM showed back to opening condition slower than closing speed SVM numbers during light period were similar to that of dark period (p<0.05). When dissolved oxygen (DO) concentration was reduced from 7 mg l-1 to 3 mg l-1, SVM numbers were increasing with decreasing of DO, and showed abnormality SVMs as compare with normal condition. Moreover, in the condition of 2 mg l-1, Distance between light and left shell showed gradually decreased, and then we could not detected SVMs due to closed condition. Thus, if we quickly detect abnormal environmental variations as hypoxia water using bio-monitoring of SVM, it may be contribute to increased productivity by dramatically reducing damages in aquaculture.

  • 우리나라의 느타리버섯 재배는 70년대 후반 농촌진흥청에서 볏짚다발재배기술을 개발 보급하면서 느타리버섯 재배기술이 보편화하였으나 80년대 후반부터 점차 노동력을 절감 할 수 있는 폐면을 이용한 재배 방법으로 변화되었다. 그런데 이 폐면재배 방법은 해외에서 폐면을 대량 수입하여 사용하고 있으므로 외화낭비와 더불어 식물병균, 곤충, 잡초 등이 반입될 수도 있으며 재배기술면에서도 볏짚다발재배에 비하여 까다롭고 버섯병 발생이 쉬워 버섯재배에 실패하는 농가가 많은 실정이다. (중략)

In vitro Biological Control Against Trichoderma harzianum Using Antifungal Bacteria

  • Trichoderma harzianum is an aggressive causal agent of green mold disease on mushroom cultivation. Some bacterial strains isolated, from oyster mushroom compost in Wonju, were found to have in vitro antifungal activity against Trichoderma harzianum ATCC 6385, 6504, and our isolates Trichoderma spp. Y and G. Further in vitro antifungal studies on several strains of phytopathogenic fungi showed that all of 12 phytopathogenic fungal strains were significantly inhibited by the isolated antifungal bacteria in Petri dishes. Of these, KATB 99121 showed the broadest inhibiting effect and displayed as negative coagulase, negative sulfide production and rod shape. KATB 99121 was resistant to ampicillin, chlorampenicol, and kanamycin. Identification of isolates was determined by Biolog GN system, and KATB 99121 was identified as Photobacterium logei because of 96 probability, 0.65 similarity, and 4.97 disturbance. With electron microscopy, thin section of KATB 99121 strain revealed typical rod-like shaped cell (0.6-0.8${\mu}{\textrm}{m}$$\times$1.5-2.0${\mu}{\textrm}{m}$) with prokaryotic structure and organization.
