The Effect of Human Recombinant Superoxide Dismutase Conjugated with Polyethylene Glycol on the Hepatic Toxicity of Acetaminophen (HrSOD-폴리에칠렌 접합체의 아세트아미노펜 간독성에 미치는 영향)

  • The covalent conjugation of human recombinant superoxide dismutase (hrSOD) with trichloros-triazine activated polyethylene glycol (PEG) 5000 formed soluble conjugates with molecular weight of 92KD, which retained $90{\sim}98%$ of original activity with a markedly prolonged plasma half-life of enzyme activity. The effect of hrSOD-PEG conjugates on acetaminophen (ACP)-induced hepatotoxicity was tested in male rats which were pretreated with 3-methylcholanthrene. HrSOD-PEG conjugates inhibited the hepatotoxicity produced by ACP, on the other hand, native hrSOD had no protective effect. The above results indicated that oxygen radicals might participate in the mechanism of the ACP-induced hepatotoxicity and that polymer conjugated-protein drugs with prolonged half-lives could be employed as an effective therapeutic agent.

  • Iron is an important element for brain oxygen transport, myelination, DNA synthesis and neurotransmission. However, excessive iron can generate reactive oxygen species and contribute neurotoxicity. Although brain iron deposition is the natural process with normal aging, excessive iron accumulation is also observed in various neurological disorders such as neurodegeneration with brain iron accumulation, Parkinson's disease, Alzheimer's disease, multiple sclerosis, Friedreich ataxia, and others. Magnetic resonance image (MRI) is a useful method for detecting iron deposits in the brain. It can be a powerful tool for diagnosis and monitoring, while furthering our understanding of the role of iron in the pathophysiology of a disease. In this review, we will introduce the mechanism of iron toxicity and the basics of several iron-related MRI techniques. Also, we will summarize the previous results concerning the clinical application of such MR imagings in various neurological disorders.

  • There is now increasing evidence that free radicals and active oxygen species are involved in a variety of pathological events. often associated with ageing. Free radical-mediated cell damage and free radical attack on polyunsaturated fatty acids result in the formation of lipid radicals. These lipid radicals react readily with molecular oxygen to produce peroxy radicals responsible for initiating lipid peroxidation. The peroxidation of cellular membrane lipid can lead to cell necrosis and considered to be implicated in a number of pathophysiological conditions as well as in the toxicity of many xenobiotics.   • In regard to $A\beta$ toxicity and AD, reactive oxygen species (ROS) are produced by macrophage families in response to $A\beta$ stimulation. In addition to this, neurotrophins (NTs) regulate the neuronal function as well as cell survival and the growth of various types of neurons in both the peripheral nervous system (PNS) and central nervous system (CNS). As high expressions of the ROS and NTs are a routine findings in neuronal cell damage, we wanted to investigate whether Rg3 can inhibit the production of ROS and NTs primary cell cultures. (omitted)

  • The peculiarities of the polyvinylpyrrolydone (PVP) interaction with transition metal ions of the first row in solution were studied. It was shown that PVP macromolecules due to their swelling conformation in organic solvents form the stable metal complexes. Metal ions were bond with oxygen and nitrogen atoms of PVP lactam rings. In water solution every metal ion interacts with one or two oxygen atoms out of 10-12 monomer units of the polymer. The additional contraction of PVP macromolecule coils in water have been found out by dissolving of the polymer metal complexes (PMC) synthesized in organic media. Toxicity, blood forming and immune stimulating activity and pharmaco-kinetic too of obtained polymers and their metal completes have been investigated. The factors and effects that responsible fur changing of PMC physical-chemical and biological properties have been estimated.

Evaluation and Comparison with Standard 48 hr Acute Bioassay and High Temperature Rapid Toxicity Test for Sewage Toxicity Test (하수의 독성평가를 위한 표준독성시험법과 온도증가 단기독성평가법의 비교 평가)

  • A new method, ToxTemp (TOXcity test based on TEMPerature control) using Ceridaphnia dubia was applied to evaluate the toxicity of insecticide materials and compared with the standard 48 hr acute bioassay. BPMC, diazinon and fenitrothion may cause the inhibition to the biological process in sewage treatment plant and need to detect toxicity within short contact time. The ToxTemp method showed sensitive detection with more shorter contact of 1-1.5 hr time than that of the standard 48 hr acute bioassay. To evaluate toxicity of real wastewater/sewage, the inhibition rate of nitrification and oxygen uptake rate (OUR) using activated sludge, the standard 48hr acute bioassay and ToxTemp method using C. dubia were compared, respectively. On the basis of the inhibition rate of nitrification, the OUR test showed the less sensitive results at the relatively strong toxic sewage. On the other hands, the standard 48hr acute bioassay and ToxTemp method using C. dubia represented the toxicity of each wastewater/sewage with high sensitivity. Even the slightly low (about 1.5%) sensitivity, the ToxTemp method showed the high applicability to the real site of sewage treatment plant.

  • Increasing evidence regarding free radical generating agents and inflammatory processes suggests that accumulation of reactive oxygen species can cause hepatotoxicity. A short-chain analog of lipid hydroperoxide, t-butyl hydroperoxide (t-BHP), can be metabolized to free radical intermediates by cytochrome P-450 in hepatocytes. which in turn can initiate lipid peroxidation, affect cell integrity and result in cell injury.   • Defect of the Cable is one of the most frequent cause of fire accident for years. It also take a big part of the fire load for train. In this study, we reviewed the standard code of other countries for cable fire test. Oxygen index, flame propagation test, smoke test and toxicity test codes were investigated. We also suggest the our national code for train cables.

  • In this study we reported the Smoke Density, the Toxicity, the Flame Spread Characteristics and the Oxygen Index of the FRP composite interior panels for raicars. Also investigated the test codes of the concerning items of other countries. We made some samples to the variance of resin types, surface finishing methods and panel shapes. It's not the really used ones but may help us effectively to guess the fire safety characteristics of railcars.

Because people want more environmental material than glass fiber, there are certain needs of finding safe and economical insulating material. We checked the flammability of some kinds insulators such as PE foam, neoprene rubber foam. melamine foam and glass fiber. You could find out the characteristics such as oxygen index. optical smoke density, toxicity index of that insulators.
