Preparation of Calcium Peroxide Originated from Oyster Shell Powder and Oxygen Releasing Ability (패각 분말기반 과산화칼슘 제조와 산소 유리 특성)

  • Bioremediation in situ is heavily dependent on the oxygenic environment which would privide the dwelling microorganism with sufficient oxygen. The situation could be easily resolved with supply of an Oxygen Releasing Compound (ORC). In this paper we prepared that sort of material out of oyster shell powder (mostly calcium carbonate) that prevails every shore areas of the country. We used two different oxidizing methods in the first step of the whole manufacturing process-conventional heating in a furnace and an ultrasound generator to obtain calcium oxide. Then that calcium oxide was further oxidized into calcium peroxide which may release oxygen under a moisturized condition. The oxygen releasing experiments were run to test the performance of our products, and to determine the gas kinetics during the experiments. Interestingly, calcium peroxide derived from ultrasound treatment was much more energy-effective as ORC than that from furnace heating although the heat derived process was better than that of ultrasound in terms of oxygen content and its releasing rate. We also found that most of the data collected from the gas releasing experiments fairly supported an ordinary $1^{st}$ order kinetics to oxygen concentration, which shaped a sharp discharge of oxygen at the very early moment of each test.

  • We studied the release characteristics of the marine sediment which could facilitate sea eutrophication through some lab-scale simulation experiments. Environmental indicators such as pH, ORP(oxidation reduction potential), nitrogens, and phosphates were measured in order to calculate the corresponding release rates. $CaO_2$, an oxygen releasing compound was used to determine how it would effect on the natural process of sedimental release. COD, ammonia nitrogen, phosphorous compounds were less released under the oxic environment caused by $CaO_2$. This basic data will help developing methodology for reducing marine eutrophication which may be initiated by the sedimental release.

  • Sediment is a useful natural source but deteriorated continually by anthropogenic and industrial sources. Therefore, it is imperative to search a suitable method for improving or restoring sediment quality. Sediment has been tested to identify the effects of some external agents on a polluted area for 28 days. Chemical analysis and total viable counts (TVC) test have been conducted for 4 days interval to assess their performance. The analyses of chemical oxygen demand (COD), acid volatile sulfide (AVS), total phosphorous (T-P), total nitrogen (T-N) indicate that the chemical agents was more efficient to improve sediment quality whereas the microbial agent was more efficient for nutrient releasing from sediment. Oxygen releasing property of the chemical agent was thought to be providing with more congenial environment for the higher growth of the bacterial community than the direct application of microbial agents.

Oxidative Modification of Cytochrome c by Tetrahydropapaveroline, an Isoquinoline-Derived Neurotoxin

  • Tetrahyropapaveroline (THP) is compound derived from dopamine metabolism and is capable of causing dopaminergic neurodegenerative disorder, such as Parkinson's disease (PD). The aim of this study was to evaluate the potential of THP to cause oxidative damage on the structure of cytochrome c (cyt c). Our data showed that THP led to protein aggregation and the formation of carbonyl compound in protein aggregates. THP also induced the release of iron from cyt c. Reactive oxygen species (ROS) scavengers and iron specific chelator inhibited the THP-mediated cyt c modification and carbonyl compound formation. The results of this study show that ROS may play a critical role in THP-induced cyt c modification and iron releasing of cyt c. When cyt c that has been exposed to THP was subsequently analyzed by amino acid analysis, lysine, histidine and methionine residues were particularly sensitive. It is suggested that oxidative damage of cyt c by THP might induce the increase of iron content in cells and subsequently led to the deleterious condition. This mechanism is associated with the deterioration of organs under neurodegenerative disorder such as PD.

Effect of oyster shell powder on nitrogen releases from contaminated marine sediment

  • Nitrogen flux release from organically enriched sediments into overlying water, which may have significantly influence on water quality and increasing continuous eutrophication. The purpose of this study is to evaluate the remediation efficiency of oyster shell powder and its treated product into organically enriched sediment in terms of nitrogen flux, organic matter, chlorophyll-a, pH and dissolved oxygen (DO). The TOSP was mainly composed of CaO2. The application of TOSP into the sediment has increased the pH, DO and significantly decreased the concentrations of NH4+-N and T-N compared to other basins. On the other hand, nitrate was enriched with the addition of treated oyster powder, an oxygen releasing compound on both phases. Furthermore, chlorophyll-a was found to be increasing with time in the control basin meanwhile it dropped drastically with the addition of TOSP, which implied on the repression of algal growth owing to blockage of nitrogen source migrating from the sediment. This study has shown that the TOSP was effective to improve sediment-water quality, diminish eutrophication and control harmful algae blooms in a marine environment. Therefore, it is a good reference as an effective environmental remediation agent.

Correlation of Releases of Nutrient Salts in Sediment with Vicinal Oxic Conditions (퇴적물의 영양염류 용출과 호기적 조건과의 상관성 분석)

  • The aim of this paper is to correlate the release characteristics of marine and lake sediment with their vicinal oxic conditions. We performed lab-scale simulation experiments using field sediment and water in order to compare the release concentrations and the release rates one another. To provide a few different kinds of oxic environments we used natural air flow and some oxygen releasing compounds such as $CaO_2$ and $MgO_2$. In case of phosphates, in each oxic condition, removal of phosphorus via biological activity and that via salt precipitation with the metal ions lowered the release rates. The behavior of the nitrogen-origin salts seemed to greatly depend on the typical biological actions - growth of biomass, nitrification, and partial denitrification. Generally speaking, the control of releases of $NH_3$-N, $PO_4$-P, T-N and T-P was successful under the oxic conditions meanwhile COD, nitrates and nitrites were difficult to reduce the releases into the bulk water because of the considerable microbial oxidation. Based on typical diffusive mass transfer kinetics the changes of concentrations of the nutrients were computed for qualitative and quantitative comparisons.

Enviro-Chemical Changes in Shoreline Sediment by MgO2 for Enhancement of Indigenous Microbial Activity (토착균주 활성화를 위한 산소발생제 MgO2 투입에 의한 연안저질의 이화학적 성분 변화 관찰)

  • Rapid industrialization has brought Nam-Hae area serious environmental problems associated with released oil and other hydrocarbons. In this work, in order to enhance the quality of the shoreline sediment we made enviro-chemical analyses of its substances, TPHs and microbial growth after treating with oxygen releasing compound(ORC) such as $MgO_2$. Total organic compound(TOC) was reduced from 33.45% to 25.1~31.08% meanwhile COD decreased from 27.5~28.9mg/$g{\cdot}dry$ to 19.9~26.1mg/$g{\cdot}dry$ for input of 2~10% $MgO_2$ in 20days. For 10% $MgO_2$ input, TP and TN were reduced by 13.3% and 18.8%, respectively. Most of all TPH was decomposed by max. 42.4% in 21days, and the total viable count of microbes was found to be exponentially increased by 75.9%.

Development of Immobilized Naphthoquinone for Effective Algicidal Activity under Various Environmental Conditions and It's Ecological Changing Monitoring (다양한 환경에서의 효율적 녹조 저감을 위한 Naphthquinone 물질의 담체화 기술 개발 및 이에 따른 생태계 변화 모니터링)

  • Bloom of small centric diatom Stephanodiscus is quite occasional in winter season in temperate freshwater ecosystems. It often leads to degradation of water quality and affects the quality of supplied drinking water. In a previous study, we have found that naphthoquinone (NQ) 4-6 derivate is an effective tool for efficient mitigation of natural S. hantzschii blooms. In the present research, polylactide (PLA) and agar foam were used as immobilized agent for NQ 4-6 to improve the efficiency of NQ 4-6 compound releasing process for its application under various field conditions. Mesocosm experiments at 10 ton scale suggested that the abundance of S. hantzschii was continuously increased in the control and upon treatment of the mesocosm with immobilized NQ 4-6 from PLA and agar foam. Their algicidal activities were 78.8% and 77.1%, respectively, on S. hantzschii after 10 days. In the mesocosm experiments, the dynamics of biotic (bacteria, HNFs, ciliates, zooplankton) and abiotic (water temperature, dissolved oxygen, pH, conductivity, nutrients) factors remained unaffected. They exhibited similar trends in the control and treatment groups. Therefore, the immobilized NQ 4-6 from PLA and agar foam has potential to be used as an alternative algicidal substance to effectively mitigate natural S. hantzschii blooms under various field conditions. In addition, it not only can be used to control S. hantzschii, but also is an effective technique. The immobilized NQ 4-6 showed stable controlled release in desired system.

A Field Research on Mud Flat Remediation by Biological Treatments (생물학적 처리에 따른 갯벌 복원을 위한 현장 적용성 연구)

  • A field test on mud flat remediation was carried out in order to observe the effects of the treatments such as microbial dose and an oxygen releasing compound like $CaO_2$. The size of each treatment site was $100m^2$ and the dosage was 3.6 kg per site. The 6 week monitoring showed that pH on two sites was below 7 and ORP increased from .178~-188 mV to .121~-142 mV. In Ignition loss and COD there were no significant changes. Meanwhile nitrogen and phosphorus concentrations changed: ammonia concentration decreased both on control and treatment sites. Nitrate nitrogen decreased more on combined treatment site than on single microbial treatment (11.3% vs. 7.3%) probably because the extra oxygen supplied by $CaO_2$ formed more oxic environment so that the facilitated nitrification might produce more nitrate but the nitrate would be much rapidly released into the water layer out of the sediment. That also explains the total nitrogen reduction(6.1%). Similarly, T-P and $PO_4-P$ reduced by 29% and 31.8%, respectively on combined treatment sites, resulting from the phosphorus release effect though the initial concentrations of the two factors were considerably high.

Inhibition of Nitric Oxide-induced Neuronal Apoptosis in PC12 Cells by Epigallocatechin Gallate

  • In the central nervous system, nitric oxide (NO) is associated with many pathological diseases such as brain ischemia, neurodegeneration and inflammation. The epigallocatechin gallate (EGCG), a major compound of green tea, is recognized as protective substance against neuronal diseases. This study is aimed to investigate the effect of EGCG on NO-induced cell death in PC12 cells. Administration of sodium nitroprusside (SNP), a NO donor, decreased cell viability in a dose- and time-dependent manner and induced genomic DNA fragmentation with cell shrinkage and chromatin condensation. EGCG diminished the decrement of cell viability and the formation of apoptotic morphologenic changes as well as DNA fragmentation by SNP. EGCG played as an antioxidant that attenuated the production of reactive oxygen species (ROS) by SNP. The cells treated with SNP showed downregulation of Bcl-2, but upregulation of Bax. EGCG ameliorated the altered expression of Bcl-2 and Bax by SNP. The release of cytochrome c from mitochondria into cytosol and expression of voltage -dependent anion channel (VDAC)1, a cytochrome c releasing channel in mitochondria, were increased in SNP-treated cells, whereas were attenuated by EGCG. The enhancement of caspase-9, preceding mitochondria-dependent pathway, caspase-8 and death receptor-dependent pathway, as well as caspase-3 activities were suppressed by EGCG. SNP upragulated Fas and Fas-L, which are death receptor assembly, whereas EGCG ameliorated the expression of Fas enhanced by SNP. These results demonstrated that EGCG has a protective effect against SNP-induced apoptosis in PC12 cells, through scavenging ROS and regulating the mitocondria- and death receptor-mediated signal pathway. The present study suggest that EGCG might be a natural neuroprotective substance.
