Effect of Silkworm Powder on Oxygen radicals and Their Scavenger Enzymes in Brain membranes of SD Rats (뇌조직의 활성산소 및 그 제거효소에 미치는 누에분말의 영향)

  • This study was designed to investigate the effect of silkworm powder on oxygen radicals and their scavenger enzymes in brain membrances of SD rats. Hydroxyl radical (OH) levels resulted in a considerable decreases in brain mitochondria fraction. Superoxide radical (O$_2$) levels were a slightly decreased in brain cytosol fraction. Lipid peroxide (LPO) and Oxidized protein (OP) levels were significantly decreased in brain mitochondria and microsomes fraction. Mn-superoxide dismutase (SOD) activity was remarkably increased in the mitochondria fraction. Cu and Zn-SOD activities were effectively increased in brain cytosol fraction. GSHPx activity was considerably increased in brain cytosol fraction. These results suggest that anti-aging effect of silkworm plays an effective role in attenuating an oxidative stress and increasing a scravenger enzyme activity in brain membranes.

  • Chemotaxonomy and numerical identification were carried out for an isolate strain No.8001 producing an antibiotic that generates oxygen radical. The genus of strain No.8001 was decided as Streptomyces sp. from chemotaxonomic data. Thirty-nine taxonomic unit characters were tested and the data were analyzed numerically using the TAXON program. The isolate was best matched to Streptomyces citreofluorescens in the major cluster 1B of Streptomyces. Therefore, it was concluded that the isolate was identified to be a member of Streptomyces citreofluorescens.

  • We investigated the active oxygen scavenging activity and active compound from Humulus japonicus. The ethyl acetate and butanol fraction exhibited strong scavenging effects on hydroxyl radical. Furthermore active compound was isolated and purified using Amberlite XAD-2 column chromatography and preparative HPLC from ethyl acetate fracton, and major compound was identified as a luteolin-7-O-$\beta$-D-glucoside by the MS, UV, 1H-NMR and 13C-NMR analyses. Luteolin-7-O-$\beta$-D-glucoside exhibited strong scavenging effect on active oxygens such as superoxide radical, hydroxyl radical and hydrogen peroxide.

  • Maltol(2-methyl-3-hydroxy-r-pyrone), a component known to be present in Korean Ginseng root showed an antioxidant action but its potency as an antioxidant was low; about 1150th that of other antioxidants such as p-phenylenediamine , BHA and BHT. However, maltol was able to protect the oxidation adamants in biological systems such as adriamycin-induced membrane damage in isolated cardiomyocytes, parquet-induced toxicities in isolated hepatocytes and repercussion injury in isolated hearts. The antioxidant action of maltol was also shown to be effective in vivo. The antioxidant action of this compound was probably due to the removal of hydroxyl radicals. In view of the roles of oxygen radical in various pathological processes, Korean Ginseng root, which contains several antioxidants including maltol, is expected to have beneficial efforts on the oxygen radical-involved processes.   • For a long time, there is much interest in the superoxide anion radical as one of reactive oxygen species (ROS) not only in the basic research field of chemistry and physics but also in the life science (or biotechnology). Recently, it is becoming ever more vital since the toxic property of nanomaterials as well as advanced oxidation processes (AOP) frequently employed for controlling pollutants are connected with the formation of superoxide anion radicals. Despite many researches on superoxide anion radical, the quantitative information of its presence and its detailed reaction mechanism in aqueous environments remains largely unclear, causing the controversy and confusion. In this review paper, we attempted to summarize the physicochemical property, mechanisms, and applications of superoxide anion radical. In addition, we briefly incorporated the important application of superoxide anion radical in AOP, nanomaterials, and life science (or biotechnology).

  • ln an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen See radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde(MDA) level and activities of catalase and superoxide dismutase in the heart of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260gm were used. Experimental groups consist of Control group (=breathing with air), HBO group(=exposed to hyperbaric oxygen(HBO, 3ATA, 100%) after air breath), CO group(=exposed to CO(3,970 ppm) after air breath), CO-Air group(=exposed to CO after air breath followed by air breath) and CO-HBO group(=exposed to CO after ai. breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of Co-induced cardiotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the heart of the rats exposed to carbon monoxide.

Effect of EGF against Oxygen Radical-Induced Neurotoxicity in Cultured Spinal Dorsal Root Ganglion Neurons of Mouse (산소자유기에 의해 저해된 배양 척수감각 신경절 세포에 대한 상피세포성장인자의 영향)

  • In order to elucidate the cytotoxic effect of oxygen radicals on cultured spinal dorsal root ganglion(DRG) neurons derived from mouse. the neurotoxic effect of oxygen radicals w as examined after cultured DRG neurons were exposed to xanthine oxidase(XO) and hypoxanthine(HX)-oxygen radical generating system. In addition. neuroprotective effect of epidermal growth factor(EGF) against oxidant-induced neurotoxicity was also evaluated in these cultures. The results were, as follows: 1. Lethal concentration 50(LC$_{50}$) was 35mU/ml XO and 0.1mM HX in cultured DRG neurons. 2. Oxygen radicals induced the morphological changes such as the decrease of cell number and loss of neurites in these cultures. 3. EGF increased the cell viability and neurofilament in neurons damaged by oxygen radicals. From above the results, it is suggested that oxygen radicals have a cytotoxic effect on cultured DRG neurons of neonatal mouse and selective neurotrophic factors such as EGF are, effective, in blocking the neurotoxicity induced by oxygen radicals in cultured spinal DRG neurons.

  • Most reactive oxygen species (ROS) are free radicals and implicated in the development of a number of disease processes including artherosclerosis, neurodegenerative disorders, aging and cancer. ROS are byproducts of a number of in vivo metabolic processes and are formed deliberately as part of nor-mal inflammatory response. On the other hand, ROS are generated either as by products of oxygen reduction during xenobiotic metabolism or are liberated as the result of the futile redox cycling of the chemical agents including several chemical carcinogens. A better understanding of the mechanisms of free radical toxicity may yield valuable clue to risks associated with chemical exposures that leading to the development of chronic diseases including cancer. The molecular biology of ROS-mediated alterations in gene expression, signal transduction and carcinognesis is one of the important subjects in free radical toxicology. Epidemiological studies suggest that high intake of vegetables and fruits are associated with the low incidence of human cancer. Many phytopolyphenols such as tea polyphenols, curcumin, resveratrol, apigenin, genistein and other flavonoids have been shown to be cancer chemopreventive agents. Most of these compounds are strong antioxidant and ROS scavengers in vitro and effective inducers of antioxidant enzymes such as superoxide dismutatse, catalase and glutathione peroxidase in vivo. Several cellular transducers namely receptor tyrosine kinase, protein kinase C, MAPK, PI3K, c-jun, c-fos, c-myc, NFkB, IkB kinase, iNOS, COX-2, Bcl-2, Bax, etc have been shown to be actively modulated by phyto-polyphenols. Recent development in free radical toxicology have provided strong basis for understanding the action mechanisms of cancer chemoprevention.

Effects of Nuruk or Wheat Bran Supplemented Diet on the Serum Levels of Cholesterol and Activities of Hepatic Oxygen Free Radical Metabolizing Enzymes in Rats (누룩 또는 밀기울 첨가식이로 성장시킨 흰쥐의 혈중 Cholesterol 및 간조직 유해산소 대사효소 활성 변동)

  • The effects of nuruk and wheat bran on cholesterol level in serum and activities of free radical metabolizing enzymes were investigated in rats. The rats were fed a diet containing nuruk or wheat bran for one month. Body weight and food intake were measured. Animals were sacrificed after one month. The increased food efficiency ratio throughout whole growth period was observed in the rats fed with either nuruk containing Aspergillus terreus or wheat bran compared with control group on normal diet. In the rats fed with nuruk, hepatic GSH content, glutathione S transferase activity, hepatic cytochrome P 450 content, and aniline hydroxylase activities were generally increased. In the rats fed with nuruk containing other fungi except Aspergillus terreus, xanthine oxidase activity was decreased. The decreased cholesterol level in serum was observed in rats fed with nuruk prepared from Aspergillus terreus and wheat bran. LDL cholesterol level was decreased in rats fed with nuruk prepared with other fungi such as Penicillium sp. and Rhizopus sp. But HDL cholesterol level was increased in all groups fed with nuruk from any fungi and wheat bran. These results suggested that nuruk or wheat bran supplemented diet might exert their effect by decreasing cholesterol level in serum and amount of oxygen free radical level.

A Study for Oxidants Generation on Oxygen-plasma Discharging Process Discharging System (산소-플라즈마 공정에서 산화제의 생성에 대한 연구)

  • This study carried out a laboratory scale plasma reactor about the characteristics of chemically oxidative species (${\cdot}OH$, $H_2O_2$ and $O_3$) produced in dielectric barrier discharge plasma. It was studied the influence of various parameters such as gas type, $1^{st}$ voltage, oxygen flow rate, electric conductivity and pH of solution for the generation of the oxidant. $H_2O_2$ and $O_3$.) $H_2O_2$ and $O_3$ was measured by direct assay using absorption spectrophotometry. OH radical was measured indirectly by measuring the degradation of the RNO (N-Dimethyl-4-nitrosoaniline, indicator of the generation of OH radical). The experimental results showed that the effect of influent gases on RNO degradation was ranked in the following order: oxygen > air >> argon. The optimum $1^{st}$ voltage for RNO degradation were 90 V. As the increased of $1^{st}$ voltage, generated $H_2O_2$ and $O_3$ concentration were increased. The intensity of the UV light emitted from oxygen-plasma discharge was lower than that of the sun light. The generated hydrogen peroxide concentration and ozone concentration was not high. Therefore it is suggested that the main mechanism of oxidation of the oxygen-plasma process is OH radical. The conductivity of the solution did not affected the generation of oxidative species. The higher pH, the lower $H_2O_2$ and $O_3$ generation were observed. However, RNO degradation was not varied with the change of the solution pH.
