Mechanism of DNA Cleavage Induced by Fe2+ Autoxidation

  • This work investigated the difference between $Fe^{2+}$ autoxidation-induced and Fenton-type cleavage of pBR322 plasmid DNA. $^{\cdot}OH$ generation reactions in the absence and presence of $H_2O_2$ under various conditions were also investigated. Although both the $Fe^{2+}$ autoxidation and Fenton-type reactions showed DNA cleavage and $^{\cdot}OH$ generation, there were significant differences in their efficiencies and reaction rates. The rate and efficiency of the cleavage reaction were higher in the absence of 1.0 mM of $H_2O_2$ than in its presence in 20 mM phosphate buffer. In contrast, the $^{\cdot}OH$ generation reaction was more prominent in the presence of $H_2O_2$ and showed a pH-independent, fast initial reaction rate, but the rate was decreased in the absence of $H_2O_2$ at across the entire tested pH range. Studies using radical scavengers on DNA cleavage and $^{\cdot}OH$ generation reactions in both the absence and presence of $H_2O_2$ confirmed that both reactions spontaneously involved the active oxygen species $^{\cdot}OH$, ${O_2}^{\cdot-}$, $^1O_2$ and $H_2O_2$, indicating that a similar process may participate in both reactions. Based on the above observations, a new mechanism for the $Fe^{2+}$ autoxidation-induced DNA cleavage reaction is proposed.

Photophysical Efficiency Factors of Singlet Oxygen Generation from Core-modified Trithiasapphyrin Derivatives

  • The photophysical properties and the singlet oxygen generation efficiencies of meso-tetraphenyl-trithiasapphyrin $(S_3TPS)$ and meso-tetmkis(p-methoxy phenyl)-trithiasapphy rin ((p-MeO)-$S_3TPS$) have been investigated, utilizing steady-state and time-resolved spectroscopic methods to elucidate the possibility of their use as photosensitizers for photodynamic therapy (PDT). The observed photophysical properties were compared with those of other porphyrin-like photosensitizers in geometrical and electronic structural aspects, such as extended ${\pi}$ conjugation, structural distortion, and internal heavy atoms. The steady-state electronic absorption and fluorescence spectra were both red-shifted due to the extended ${\pi}$-conjugation. The fluorescence quantum yields were measured as very small. Even though intersystem crossing rates were expected to increase due to the increment of spin orbital coupling, the triplet quantum yields were measured as less than 0.15. Such characteristics can be ascribed to the more enhanced internal conversion rates compared with the intersystem crossing rates. Furthermore, the triplet state lifetimes were shortened to -1.0 ${\mu}s$ as expected. Therefore, the singlet oxygen quantum yields were estimated to be near zero due to the fast triplet state decay rates and the inefficient energy transfer to the oxygen molecule as well as the low triplet quantum yields. The low efficiencies of energy transfer to the oxygen molecule can be attributed to the lower oxidation potential and/or the energetically low lying triplet state. Such photophysical factors should be carefully evaluated as potential photosensitizers that have extended ${\pi}$-conjugation and heavy core atoms synthesized for red-shifted absorption and high triplet state quantum yields.

MS-5, a Naphthalene Derivative, Induces the Apoptosis of an Ovarian Cancer Cell CAOV-3 by Interfering with the Reactive Oxygen Species Generation

  • Reactive oxygen species (ROS) are widely generated in biological processes such as normal metabolism and response to xenobiotic exposure. While ROS can be beneficial or harmful to cells and tissues, generation of ROS by diverse anti-cancer drugs or phytochemicals plays an important role in the induction of apoptosis. We recently identified a derivative of naphthalene, MS-5, that induces apoptosis of an ovarian cell, CAOV-3. Interestingly, MS-5 induced apoptosis by down-regulating the ROS. Cell viability was evaluated by water-soluble tetrazolium salt (WST-1) assay. Apoptosis was evaluated by flow cytometry analysis. Intracellular ROS ($H_2O_2$), mitochondrial superoxide, mitochondrial membrane potential (MMP) and effect on cycle were determined by flow cytometry. Protein expression was assessed by western blotting. The level of ATP was measured using ATP Colorimetric/Fluorometric Assay kit. MS-5 inhibited growth of ovarian cancer cell lines, CAOV-3, in a concentration- and time-dependent manner. MS-5 also induced G1 cell cycle arrest in CAOV-3 cells, while MS-5 decreased intracellular ROS generation. In addition, cells treated with MS-5 showed the decrease in MMP and ATP production. In this study, we found that treatment with MS-5 in CAOV-3 cells induced apoptosis but decreased ROS level. We suspect that MS-5 might interfere with the minimum requirements of ROS for survival. These perturbations appear to be concentration-dependent, suggesting that MS-5 may induce apoptosis by interfering with ROS generation. We propose that MS-5 may be a potent therapeutic agent for inducing apoptosis in ovarian cancer cell through regulation of ROS.

Color Enhancement by Oxygen Torch in Blue Sapphires (블루사파이어와 루비의 고온산소 화염처리에 의한 색향상)

  • We enhanced the color of blue sapphires and rubies successfully by using a oxygen-propane torch flame annealing, which had not been employed so far. We confirmed that about 1 mm-thick de-coloring of the corundum samples were available with 60 minutes flame annealing through eye evaluation, color coordination characterization, and methylene iodide immersion observation. We also suggest that the color centers such as $[Fe_{Al}^{\cdot}]$ may transform into transparent $[Fe_{Al}^{x}],\;[Cr_{A1}^{x}]$ sites with $[V_o^']$ generation at the elevated temperature in oxygen-rich atmosphere by diffusion mechanism. Our results implied that the longer diffusion time and the higher oxygen partial pressure might lead to the better de-coloring enhancement in corundum gem stones.

Preparation of Photosynthesis Nanofiber Composite Membrane by Using Chlorophyll and Polymer Nanofiber (식물 엽록소와 고분자 나노섬유를 이용한 광합성 나노섬유복합막의 제조)

  • In this study, chlorophylls were been extracted from common local plants, deposited on polypropylene (PP) substrate using various approaches, and the oxygen generation effect of the chlorophylls were investigated. The loading of chlorophylls on the substrates was achieved by dipping and spraying methods, where the spraying coating showed overall better results regarding oxygen generation from the combustion experiments in the closed vessel or in the isolated vacuum oven cell than those of dip coating. In addition, a composite substrate was prepared by nylon6/6 nanofiber on the PP substrate, and it exhibited an increase in the activation of chlorophylls. In the case of samples containing titanium dioxide ($TiO_2$), the reaching time of oxygen concentration from 16% to 21% and the combustion test using a candle for a sample with 50% chlorophylls showed similar results to those of a sample without $TiO_2$. As such, combining a spray coating and $TiO_2$ incorporation into gas separation membrane systems are expected to be useful to understand the fundamentals of material properties for their applications as oxygen generation membranes and air filtration systems.

Superoxide Generation by Blood Monocyte and Pulmonary Alveolar Macrophage in Patients with Pulmonary Tuberculosis (폐결핵환자의 폐포대식세포 및 말초혈액내 단구세포에서 분비하는 과산화음이온의 비교 관찰)

  • Background: Mycobacterium tuberculosis is a facultative intracellular pathogen which persists and multiplies within macrophage. Competent cell mediated immunity by cooperation of both T lymphocyte and macrophage of the host is required to kill the Mycobacterium tuberculosis. But a precise understanding of the pathogenesis of tuberculosis infection in pulmonary alveolar macrophage has not been achived. Research on the macrophage's basic microbicidal mechanism has elucidated the importance of oxygen-dependent or oxygen-independent components. Oxygen dependent processing begins with the reduction of oxygen by NADPH oxidase and generation of superoxide. In this study, the oxidative metabolic status of blood monocyte and pulmonary alveolar macrophage in patients with active pulmonary tuberculosis was accessed and compared with that of healthy control subjects to know whether there was a basic difference in superoxide generation by mononuclear cells between two groups. Methods: Pulmonary alveolar macrophage was purified after performing BAL(bronchoalveolar lavage) through the bronchi of infected lesion by plastic adhesion method. Blood monocyte was purified by Ficoll-Hypaque method. Superoxide generation by blood monocyte and pulmonary alveolar macrophage was measured by ferricytochrome-C reduction method after either stimulated with PMA(phorbol myristate acerate) or non-stimulated states. We also measured the effect of pulmonary tuberculosis patient's serum on superoxide generation by monocyte. Results: 1) Generation of superoxide by alveolar macrophage obtained from patients with pulmonary tuberculosis was little higher than those of controls, and PMA enhanced the generation of 2) Generation of superoxide by blood monocyte obtained from patients with pulmonary tuberculosis was little higher than those of control(p>0.05), and PMA more enhanced the generation of superoxide in patientswith pulmonary tuberculosis than those in controls(p<0.02). 3) Patient's serum enhanced the generation of superoxide by blood monocyte obtained from patients with pulmonary tuberculosis and controls, but not in the case of PMA stimulated blood monocyte. Conclusion: The present study suggest that the phenomenon of M.tuberculosis escape the microbicidal action of macrophage was not result of suppressed superoxide generation by blood monocyte and pulmonary alveolar macrophage, rather there might be a factor to stimulate the generation of superoxide by blood monocyte in pulmonary tuberculosis patient serum, but the comparision with effect of control's serum on superoxide generation needs further elucidation.

The Effect of Hybrid Reburning on NOx Reduction in Oxygen-Enriched LPG Flame (산소부화 LPG 화염에서 혼합형 재연소 방법에 의한 NOx 저감 효과)

  • In order to enhance combustion efficiency, oxygen-enriched combustion is used by increasing the oxygen ratio in the oxidizer. However, since the flame temperature increases, NOx formation in the furnace seriously increases for low oxygen enrichment ratio. In this case, reburning is a useful technology for reducing nitric oxide. In this research, experimental studies have been conducted to evaluate the hybrid effects of reburning/selective non-catalytic reaction (SNCR) and reburning/air staging on NOx formation and also to examine heat transfer characteristics in various oxygen-enriched LPG flames. Experiments were performed in flames stabilized by a co-flow swirl burner, which were mounted at the bottom of the furnace. Tests were conducted using LPG gas as main fuel and also as reburn fuel. The paper reported data on flue gas emissions, temperature distribution in furnace and various heat fluxes at the wall for a wide range of experimental conditions. Overall temperature in the furnace, heat fluxes to the wall and NOx generation were observed to increase by low level oxygen-enriched combustion, but due to its hybrid effects of reburning, SNCR and Air staging, NOx concentration in the exhaust have decreased considerably.

Generation of Reactive Oxygen Species by Nonenzymatic Reaction of Menadione with Protein Thiols in Plasma (Menadione과 Plasma내의 Protein Thiol의 비효소적인 화학반응에 의한 활성산소 생성)

  • Quinones have been reported to undergo nonenzymatic reaction with thiols to generate reactive oxygens. It is therefore possible that the nonenzymatic reaction of quinones with thiols in plasma could lead to potentJared cellular toxicity or disease. When 1 mM menadione was added in plasma under pH 11.2, 7.4 and 5.0, the increase in oxygen consumption rate was the order of pH 11.2 > pH 7.4 > pH 5.0. In addition, oxygen consumption rates under plasma anticoagulated with trisodium citrate solution (pH 7.85) was significantly higher than those with acid-citrate-dextrose solution (pH 6.87). SOD and catalase reduced the rate of oxygen consumption induced by menadione in plasma. Taken together, these results suggest that the menadione-induced increased oxygen consumption was due to nonenzymatic reaction of menadione with thiols in the plasma. The presence of plasma has an additive effect on the increased oxygen consumption rates induced by the menadione treatments on our model tissue, platelets, as compared between washed platelet (WP) and platelet rich plasma (PRP). Cytotoxicity, as determined by LDH release, are well correlated with the oxygen consumption rates observed in each system and strongly suggest that menadione-induced cytotoxicity can be increased with the presence of blood plasma.

Development of Three-dimensional Thermo-fluid Numerical Model for Steam Drum of a Basic Oxygen Furnace (순산소 전로의 증기드럼 내의 3차원 열 유동 해석모델 개발)

  • The efficient steam drum should be required to reduce carbon oxide emissions and heat recovery in oxygen converter hood system. However, steam generation is limited to the time of the oxygen blowing period, which is intermittent or cyclical in operation of steel-making process. Thus, steam drum should be optimized for an effective steam generation during the oxygen blowing portion of the converter cycle. In this study, a three-dimensional computational fluid dynamics (CFD) model has been developed to describe the impacts of changing various operating conditions and geometric shape on thermo-fluid characteristics and performance of the steam drum. This model encompasses not only fluid flow and heat transfer but also evaporation and condensation at the interfacial surface in the steam drum by using VOF (Volume of Fluid) method. To validate the prediction performance of this model, comparison of the steam flow rate between numerical and experimental result has been performed, resulting in the accuracy of the relative error by less than 3.2%.

Protective Effect of Acanthopanax senticosus Extract on Alloxan-induced β-cell Damage

  • The protective effect of Acanthopanax senticosus (AS) extract on alloxan-induced pancreatic β-cell damage was investigated in HIT T-15 cells, a Syrian hamster pancreatic β-cell line. Alloxan caused the pancreatic β-cell damage through the generation of reactive oxygen free radicals, increased DNA fragmentation, and decreased cellular NAD/sup +/ levels. The β-cell damage was significantly prevented by the pretreatment with water soluble extract of AS roots. These results suggest that the protective effect of AS extract, on alloxan-induced β-cell damage, is primarily due to the inhibition of the generation of reactive oxygen free radical species (ROS) by alloxan.
