We examined the effects of unripe black raspberry water extract (UBR-W) on lipid metabolism and oxidative stress in mice. C57BL/6J mice were divided into 4 groups: those administered a control diet (CTL), high-fat diet (HFD), UBR-W and simvastatin for 12 weeks. In the HFD group, LDL cholesterol were significantly higher than in the CTL group. However, the UBR-W treated group showed dose-dependent reduction of plasma LDL levels. Hepatic total lipid, TC, and malondialdehyde were significantly increased in hyperlipidemic mice. However, supplementation with either UBR-W or simvastatin effectively reduced these lipid profiles and lipid peroxidation. UBR-W increased mRNA expression of the LDL receptor, sterol regulatory element binding protein 2 (SREBP2), 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase and ATP-binding cassette transporter A1 (ABCA1) compared to that observed in the HFD group. In addition, UBR-W and simvastatin showed significantly reduced oxidized LDL uptake by the scavenger receptor CD36. These results suggest that UBR-W is useful for treatment and prevention of hyperlipidemia and lipid peroxidation.
This study investigated the effects of Angelica keiskei Koidzumi and turmeric extract supplementation(ATE) on blood lipids, antioxidant and inflammatory markers in 35 hypercholesterolemic Korean adults with high blood cholesterol levels (serum total cholesterol$\geq200mg/d{\ell}$ or LDL-cholesterol$\geq130mg/d{\ell}$). They received ATE(n=21, 14 females and 7 males) or placebo(control group, n=14, 11 females and 3 males) for 4 weeks. There was no significant change in serum total cholesterol, LDL-cholesterol and HDL-cholesterol levels after ATE supplementation in the both groups. However, the LDLcholesterol: HDL-cholesterol ratio(LPH) was significantly decreased and both serum prostagrandin E2(PGE2) levels were significantly decreased in those receiving ATE. No significant changes were evident in interleukin(IL)-$1\beta$, IL-6, IL-8, 8-isoprostane, malondialehyde, total antioxidant capacity and oxidized-LDL. These results suggest that complex extract of Angelica keiske and turmeric has the potential to decrease cardiovascular risk by reducing LPH and inflammatory mediator $PGE_2$ in hypercholesterolemic adults.
Apolipoprotein B-100 (apo B-100), the main protein component that makes up LDL (Low density lipoprotein), consists of 4,536 amino acids and serves to combine with the LDL receptor. The oxidized LDL peptides by malondialdehyde (MDA) or acetylation in vivo were act as immunoglobulin (Ig) antigens and peptide groups were classified into 7 peptide groups with subsequent 20 amino acids (P1-P302). The biomimetic peptide P99 (KGTYG LSCQR DPNTG RLNGE) out of B-group peptides carrying the highest value of IgM antigens were selected for structural studies that may provide antigen specificity. Circular Dichroism (CD) spectra were measured for peptide secondary structure in the range of 190-260 nm. Experimental results show that P99 has pseudo α-helice and random coil structure. Homonuclear (COSY, TOCSY, NOESY) 2D-NMR experiments were carried out for NMR signal assignments and structure determination for P99. On the basis of these completely assigned NMR spectra and proton distance information, distance geometry (DG) and molecular dynamic (MD) were carried out to determine the structures of P99. The proposed structure was selected by comparisons between experimental NOE spectra and back-calculated 2D NOE results from determined structure showing acceptable agreement. The total Root-Mean-Square-Deviation (RMSD) value of P99 obtained upon superposition of all atoms were in the set range. The solution state P99 has mixed structure of pseudo α-helix and β-turn(Gln[9] to Thr[13]). These NMR results are well consistent with secondary structure from experimental results of circular dichroism. Structural studies based on NMR may contribute to the prevent oxidation studies of atherosclerosis and observed conformational characteristics of apo B-100 in LDL using monoclonal antibodies.

Experimental results show that P143 has β-sheet in Ile[1]-Asn[9] and α-helice in Gln[16]-Tyr[20] structure. Homonuclear 2D-NMR (COSY, TOCSY, NOESY) experiments were carried out for NMR signal assignments and structure determination for P143. Both metabolic syndrome (MetS) and elevated LDL cholesterol (LDL-C) increase the risk for cardiovascular disease (CVD). We hypothesized that low HDL cholesterol (HDL-C) would further increase CVD risk in women having both conditions. To assess this, we recruited 89 women with MetS (25-72 y) and LDL-C ${\geq}$ 2.6 mmol/L. To determine whether plasma HDL-C concentrations were associated with dietary components, circulating atherogenic particles, and other risk factors for CVD, we divided the subjects into two groups: high HDL-C (H-HDL) (${\geq}$ 1.3 mmol/L, n=32) and low HDL-C (L-HDL) (< 1.3 mmol/L, n=57). Plasma lipids, insulin, adiponectin, apolipoproteins, oxidized LDL, Lipoprotein(a), and lipoprotein size and subfractions were measured, and 3-d dietary records were used to assess macronutrient intake. Women with L-HDL had higher sugar intake and glycemic load (P< 0.05), higher plasma insulin (P< 0.01), lower adiponectin (P< 0.05), and higher numbers of atherogenic lipoproteins such as large VLDL (P < 0.01) and small LDL (P<0.001) than the H-HDL group. Women with L-HDL also had larger VLDL and both smaller LDL and HDL particle diameters (P<0.001). HDL-C was positively correlated with LDL size (r=0.691, P<0.0001) and HDL size (r=0.606, P<0.001), and inversely correlated with VLDL size (r=-0.327, P<0.01). We concluded that L-HDL could be used as a marker for increased numbers of circulating atherogenic lipoproteins as well as increased insulin resistance in women who are already at risk for CVD.
1. Objectives This study aimed to investigate the anti-oxidative effect of Nocyongdaebo-tang (NYD) decoction in oxidized rats induced by AAPH. 2. Methods AAPH (2.2-azobis-2 aminodinopropane-hydrochloride) was injected intraperitoneally to induce oxidization and NYD was orally administered to the rats. To observe the anti-oxidative effect of NYD, we performed blood chemistry analysis, histological analysis, and evaluated the levels of SOD, catalase, glutathione, NO and MDA in liver. 3. Results & Conclusions 1) Serum Albumin level was increased significantly in the NYD group as compared with the control group and the saline group. 2) Serum LDL-cholesterol level was reduced significantly in the NYD group as compared with the control group. 3) GPT level in liver was reduced significantly in the NYD group as compared with the control group. 4) SOD, catalase activity and glutathione levels were increased significantly in the NYD group as compared with the control group and the saline group. 5) The levels of NO and MDA were reduced significantly in the NYD group as compared with the control group. 6) The degenerative change of liver tissue of NYD group was decreased in comparison with the control group. In conclusion, NYD decoction is considered to have an anti-oxidative effect in rats.
This study was designed to investigate the effects of silkworm powder (SWP) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (SWP-200and SWP-400 groups)added 200 and 400mg/kg BW/day for 6 weeks. Triglyceride (TG)levels were significantly inhibited (10% and 25%) in SWP-200 and SWP-400 groups, but there were no singificant differences in total, LDL-and HDL-cholesterol levels in both SWP-200 and SWP-400 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases (about 20%) in both SWP-200 and SWP-400 groups compared with control group. Superoxide radical (O2.-)and hydrogen peroxide formations resulted in a significant decreases in SWP-400 group compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (SOD) and catalase (CAT) activiteies were remarkably increased (10∼25% and 40∼50%)in SWP-200 and SWP-400groups. Glutathione peroxidase(GSHPX) activities were significantly increased (about 10%) in SWP-400 groups compared with control group. These results suggest that anti-aging effect of silkworm powder (SWP) may play a pivotal role in attenuation a various chronic degenerative diseases age-related changes.
Atherosclerosis is a cardiovascular disease in which plaque builds up inside of an artery and can lead to various complications such as myocardial infarction, stroke, and thrombosis. Recently, hypertriglyceridemia has attracted significant attention as contributors to development of atherosclerosis. However, molecular mechanism of its contribution to atherosclerosis is poorly understood. Here we proposed a potential link between triglyceride (TG) and atherosclerosis. TG treatment promoted downregulation of certain scavenger receptor, macrophage scavenger receptor-1 (MSR-1) in phorbol myristate acetate (PMA)-derived human macrophages. TG treatment caused reduction of MSR-1 mRNA expression in a time- and dose-dependent manner. Using chemical inhibitors, we found that inhibition of signaling pathways associated with PI3K and PLC enhances TG-induced reduction of MSR-1 expression in THP-1 macrophages implying that PI3K and PLC is implicated in the expression of MSR-1 in macrophages. Since MSR-1 is associated with uptake and clearance of atherogenic lipoprotein, oxidized low density lipoprotein (oxi-LDL), our data suggest that increase of oxi-LDL due to TG-mediated reduction of its receptor MSR-1 can promote atherosclerosis.

High blood cholesterol levels and oxidized cholesterol are risk factors for cardiovascular disease, which displays high annual incidence. Although studies on sericulture products, including pupae, silk protein, and blood lymph, as hypocholesterolemic substances have been reported, insufficient research in this field has been focused on silkworm larvae. Six larval extracts (Low temperature distilled water, LW; hot temperature distilled water, HW; and 30-100% ethanol, E30-E100) were prepared, and their effects on cholesterol metabolism were examined. LW most potently reduced the risk of cholesterol-related disorders. Polyphenols were highly represented in LW, corresponding with its increased antioxidant potency. The cholesterol biosynthesis enzyme, 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase (HMGCR) was strongly inhibited by LW. Hepatocytes over-expressed LDL receptor (LDLR) after LW stimulation, promoting cholesterol elimination from plasma. LW also increased ATP binding cassette transporter 1 (ABCA1) gene expression, upregulating HDL biogenesis. In conclusion, LW exhibited strong antioxidant activity, suppressed cholesterol biosynthesis, improved LDL uptake from plasma, and upregulated HDL biosynthesis. In aggregate, these activities could reduce blood cholesterol levels and prevent cardiovascular disease.
Objectives : The present study investigated effects of Artemisia Capillaris Thunberg ethanol extract(EtOH ext). on lowering lipid, anti-oxidation and concentration of plasma inflammatory mediators using rat fed on high oxidized fat. Methods : We divided fat sprague-dawley rats fed on high oxidized into 4 groups. They were normal group, feed with 100 mg/kg Artemisia Capillaris Thunberg group, feed with 200 mg/kg Artemisia Capillaris Thunberg group and feed with 300 mg/kg Artemisia capilaris Thunberg group. They were administered for 4 weeks. We measured concentration of plasma free fatty acid(FFA), plasma triglyceride, plasma total cholesterol, and plasma low density lipoprotein-cholesterol(LDL-cholesterol), plasma high density lipoprotein-cholesterol(HDL-cholesterol), concentration of liver total cholesterol and liver triglyceride (TG), concentration of plasma thiobarbituric acid reactive substance(TBARS) and liver thiobarbituric acid reactive substance(TBARS), glutathione peroxidase (GSH-Px) activity, superoxide dismutase(SOD) activity and catalase(CAT) activity, plasma nitric oxide(NO), ceruloplasmin and ${\alpha}-glycoprotein$. Results : 1. The Artemisia Capillaris Thunberg EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDL-cholesterol was increased in the Artemisia Capillaris Thunberg EtOH ext. groups. 2. Concentration of liver total cholesterol and liver TG showed a significantly decrement in all Artemisia Capillaris Thunberg EtOH ext. groups than that of control group. 3. The Artemisia Capillaris Thunberg EtOH ext. groups showed lower values in concentration of plasma TBARS and liver TBARS than that of control group. The values of GSH-Px activity, SOD activity and CAT activity were increased in the Artemisia Capillaris Thunberg EtOH ext. groups. 4. The values of plasma NO, ceruloplasmin and ${\alpha}-glycoprotein$ were decreased in Artemisia Capillaris Thunberg EtOH ext. groups. Conclusions : Based on the results in this study, the Artemisia Capillaris Thunberg EtOH ext. showed a positive effect in lowering lipid, anti-oxidation and decrement of plasma inflammatory mediators.
