Objectives : The water extract of Samul-tang (SMT) has traditionally been used for treatment of ischemic heart and brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of SMT rescues cells from these damages. Methods: This study was designed to investigate the protective mechanisms of SMT on oxidative stress-induced toxicity in H9c2 cardiomyoblast cells. Treatment with $H_2O_2$ markedly induced death of H9c2 cardiomyoblast cells in a dose-dependent manner. Results: The characteristics of H20z-induced death of H9c2 showed apparent apoptotic features such as DNA fragmentation and morphological change. However, SMT significantly reduced both H202-induced cell death and morphological change. The decrease of Bc-2 expression by High were inhibited by SMT. In addition, the increase of Bax expression was also inhibited by SMT. The cotreatment of SMT and $H_2O_2$ in H9c2 cells also induced the phosphorylation of ERK in a time-dependent manner. Moreover, PD98059, a specific inhibitor of ERK1/2 attenuated the protective effects of SMT on $H_2O_2-induced$ toxicity in H9c2 cardiomyoblast cells. These results suggest that both ERK1/2 signaling pathways play important roles in the protective effects of SMT on $H_2O_2-induced$ apoptotic death of H9c2 cells. Also, the expression profile of proteins in $H_2O_2$ cardiomyoblast cells were screened by using two-dimensional (2-D) gel electrophoresis. Among 300 spots resolved in 2-D gels, the comparison of control versus apoptosis cells revealed that signal intensity of 17 spots increased and 11 spots decreased. Conclusions: Taken together, this study suggests that the protectiw effects of the water extract of SMT against oxidative damages may be mediated by the modulation of Bc1-2 and Bax expression via the regulation of the ERK signaling pathway.

The purpose of this study is to examine the toxic effects caused by xanthine oxidase/hypoxanthine (XO/HX) and the effects of herbal extracts such as Mangeum-tang (萬金湯: MGT) and Gamimangeum-tang (加味萬金湯: GMGT) on the treatment of the toxic effects. The results of these experiments were XO/HX, an oxygen radical-generating system, decreased the survival rates of the cultured cells on XTT assay, the amount of DNA syntheses, and the amount of neurofilaments, and increased c-fos positive cells, MGT and GMGT have the efficacy of increasing the survival rates of the cultured cells by increasing the amount of neurofilaments and DNA synthesis and decreasing the c-fos positive cells damaged by XO/HX, From the above results, it is suggested that MGT and GMGT have marked efficacy as a treatment for the damages caused by the XO/HX-mediated oxidative stress. And MGT and GMGT are thought to have certain pharmacological effects.
Ascorbic acid has been reported to extend replicative life span of human embryonic fibroblast (HEF). Since the detailed molecular mechanism of this phenomenon has not been investigated, we attempted to elucidate. Continuous treatment of HEF cells with ascorbic acid at ($200{\mu}M$) from 40 population doubling (PD) increased maximum PD numbers by 18% and lowered $SA-{\beta}-gal$ positive staining, an aging marker, by 2.3 folds, indicating that ascorbic acid extends replicative life span of HEF cells. Ascorbic acid treatment lowered DCFH by about 7 folds and Rho123 by about 70%, suggesting that ascorbic acid dramatically decreased ROS formation. Ascorbic acid also increased aconitase activity, a marker of mitochondrial aging, by 41%, indicating that ascorbic acid treatment restores age-related decline of mitochondrial function. Cell cycle analysis by flow cytometry revealed that ascorbic acid treatment decreased G1 population up to 12%. Further western blot analysis showed that ascorbic acid treatment decreased levels of p53, phospho-p53 at ser 15, and p21, indicating that ascorbic acid relieved senescence-related G1 arrest. Analysis of AP (apurinic/apyrimidinic) sites showed that ascorbic acid treatment decreased AP site formation by 35%. We also tested the effect of hydrogen peroxide treatment, as an additional oxidative stress. Continuous treatment of $20{\mu}M$ of hydrogen peroxide from PD 40 of HEF cells resulted in premature senescence due to increased ROS level, and increased AP sites. Taken together, the results suggest that ascorbic acid extends replicative life span of HEF cells by reducing mitochondrial and DNA damages through lowering cellular ROS.

Obesity is generally caused by quantitative changes in adipocyte differentiation and fat metabolism. Only a few studies have been determined the effect of red beans extract on obesity and plasma cholesterol concentration. We have been studied the functional activities of red-bean extracts including anti-oxidative effect against DNA and cell damages. Histological study including micro CT analysis showed that the accumulation of fat in hepatocytes and intestines was significantly decreased in red bean extract treated group. In addition, plasma cholesterol and triglyceride levels were decreased in blood samples. In addition, it was confirmed that the red bean extract inhibited the expression of $PPAR{\gamma}$, Fabp4 and RETN genes, which regulate total adipocyte differentiation and lipid metabolism. Red bean extract inhibits the expressions of transcription factors associated with adipocyte differentiation in a dose-dependent manner, thereby inhibiting fat accumulation and decreasing blood lipid levels in obese mice induced by high fat diet.

The purpose of this study is to examine the toxic effects caused by xanthine oxidase/hypoxanthine(XO/HX) and the effects of herbal extracts such as JHYJ and GJHYJ on the treatment of the toxic effects. For this purpose, experiments with the cultured hippocampal cells from new born mice were done. The results of these experiments were as follows. 1. XO/HX, a oxygen radical-generating system, decreased the survival rates of the cultured cells on XTT assay and INT assay, the amount of DNA syntheses, and the amount of neurofilaments, and increased the lipid peroxidation. 2. JHYJ and GJHYJ have the efficacy of increasing the survival rates of the cultured cells. 3. JHYJ and GJHYJ have the efficacy of increasing the amount of neurofilaments and of decreasing the lipid peroxidation. 4. JHYJ and GJHYJ have the efficacy of increasing the amount of DNA syntheses. From the above results, it is suggested that Jihwangyeumja and Gamijihwangyeumja have marked efficacy as a treatment for the damages caused by the XO/HX-mediated oxidative stress. And Jihwangyeumja and Gamijihwangyeumja are thought to have certain pharmacological effects. Further dinical study of this pharmacological effects of Jihwangyeumja and Gamijihwangyeumja should be complemented.
To evaluate the antimutagenic activity of the specialty rices, a giant embryonic rice and a pigmented rice, we measured the inhibitory effect on the chemically induced mutagenesis in E. coli and V79 cultured cell system, as well as on DNA strand scission induced by oxidative damages in vitro. When the inhibitory activity to mitomycin C-induced mutagenesis using SOS chromotest in E. coli cell was measured, the activities decreased in the following order: germinated pigmented rice (40.4%) > germinated giant embryonic rice (37.1%) > pigmented rice (35.5%) > germinated brown rice (15.7%) > giant embryonic rice (14.0%) > brown rice (0.8%). The activities for inhibiting mitomycin C-induced DNA strand scission decreased in the order of pigmented rice > giant embryonic rice > germinated pigmented rice > germinated brown rice > brown rice > germinated giant embryonic rice. We also determined antimutagenic activities of the specialty rices using the suppressing effect on 6-TG resistant colony formation by 4-NQO in V79 cells as a mutagenicity index. The order of antimutagenicity was germinated giant embryonic rice (53.2%) > pigmented rice (40.0%) > brown rice (21.2%) > germinated brown rice (14.4%) > giant embryonic rice (0.23%); in contrast, germinated pigmented rice showed promoting effect on 4-NQO-induced mutagenesis.

The water extract of Hwansodan has been traditionally used for treatment of ischemic brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of Hwansodan rescues cells from neurodegenerative disease. PC12 pheochromocytoma cells have been used extensively as a model for studying the cellular and molecular mechanisms of neuronal cell damages. Under deprivation of growth factor and ischemic injury, PC12 cells spontaneously undergoes apoptotic cell death. Serum and glucose deprivation markedly decreased the viability of PC12 cells, which was characterized with apparent apoptotic features such as membrane blebbing as well as fragmentation of genomic DNA and nuclei. However, the aqueous extract of Hwansodan significantly reduced serum and glucose deprivation-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Pretreatment of Hwansodan also ingibited the activation of caspase-3, in turn, degradation of ICAD/DFF45 was completely abolished in serum and glucose deprivated cells. Furthermore, pretreatment of Hwansodan obviously increased heme oxygenase 1 (HO-1) expression in PC12 cells. Taken together, the data suggest that the protective effects of Hwansodan against serum and glucose deprivation induced oxidative injuries may be achieved through the scavenging of reactive oxygene species accompanying with HO-1 induction.
