Influence of Oxidation Inhibitor on Carbon-Carbon Composites: 6. Studies on Friction and Wear Properties of Carbon-Carbon Composites (산화억제제 첨가에 의한 탄소/탄소 복합재료의 물성에 관한 연구 : 6. 탄소/탄소 복합재료의 마찰 및 마모특성)

  • The friction and wear properties of carbon-carbon composites made with different weight percent of $MoSi_2$ as an oxidation inhibitor were investigated using a constant speed wear test apparatus in an oxidation environment. The results indicated the carbon-carbon composites undergoing an abrupt transition of friction coefficient, from low-friction behavior(${\mu}$=0.15~0.2) during normal wear regime to the high-friction behavior(${\mu}$=0.5~0.6) during dusting wear regime at the frictional temperature range of 150~180${\circ}C$. The existence of temperature-dependent friction and wear regimes implied that the performance of specimen made with carbon-carbon composites was markedly affected by the thermal properties of the composites. The carbon-carbon composites filled with MoSi2 exhibited two times lower coefficient of friction and wear rate in comparison with the composites without $MoSi_2$. Especially, the composites containing 4wt% $MoSi_2$ filler showed a significantly improved activation energy for wear due to the reduction of both the porosity and powdery debris film formation on sliding surface when compared to those without $MoSi_2$.

  • High temperature corrosion behavior of AISI-type 316L stainless steel for the MCFC(molten carbonate fuel cell) bipolar application was studied by immersion test and penetration attack method in anode environment ($650^{\circ}C$, $Li_2CO_3/K_2CO_3=62/38$ mol%, $H_2/CO_2=80/20$ vol%) without or with different $CaCO_3$ content. Not only immersion test method but also morphological observation of samples in the carbonate melts are adopted as experimental methods. With aid of the morphological observation of cross section of samples immersed in a carbonate melt was possible to obtain penetration attack. The concentration effect of $CaCO_3$ inhibitor was investigated in order to verify the optimum concentration for practical application in MCFC stack operation. The corrosion rate in the presence of $CaCO_3$ was proven to be decreased as a function of $CaCO_3$ concentration. The corrosion rate in the presence of $CaCO_3$ was measured with a value of 6.9 mpy which is 2.4 times lower than that of inhibitor-free electrolyte. The cross section microscopy revealed that the internal penetration by oxidation in molten carbonate is very severe. In this case, the attack was occurred not only dissolution loss in the electrolyte by corrosion reaction but also weight gain through oxide layer by internal penetration.

Impact of glucose and pyruvate on adenosine triphosphate production and sperm motility in goats

  • Objective: This study evaluates goat sperm motility in response to metabolic substrates and various inhibitors, aiming to assess the relative contribution of glycolysis and mitochondrial oxidation for sperm movement and adenosine triphosphate (ATP) production. Methods: In the present study, two main metabolic substrates; 0 to 0.5 mM glucose and 0 to 30 mM pyruvate were used to evaluate their contribution to sperm movements of goats. Using a 3-chloro-1,2-propanediol (3-MCPD), a specific inhibitor for glycolysis, and carbonyl cyanide 3-chlorophenylhydrazone as an inhibitor for oxidative phosphorylation, cellular mechanisms into ATP-generating pathways in relation to sperm movements and ATP production were observed. Data were analysed using one-way analysis of variance for multiple comparisons. Results: Sperm motility analysis showed that either glucose or pyruvate supported sperm movement during 0 to 30 min incubation. However, the supporting effects were abolished by the addition of a glycolysis inhibitor or mitochondrial uncoupler, concomitant with a significant decrease in ATP production. Although oxidative phosphorylation produces larger ATP concentrations than those from glycolysis, sperm progressivity in relation to these two metabolic pathways is comparable. Conclusion: Based on the present study, we suggest that goat sperm use glucose and pyruvate to generate cellular energy through glycolysis and mitochondrial respiration pathways to maintain sperm movement.

  • Acid mine drainage occurs when sulfide minerals are exposed to an oxidizing environment. The objective of this study was to inhibit the oxidation of pyrite by applying various coating agent such as $KH_2PO_4$, MgO and $KMnO_4$ over its surface as an oxidation inhibitors. Experiments were conducted for 8 days to test the feasibility of oxidation inhibitors. The optimal condition of coating agent for standard pyrite and IK mine was the combination of 0.01M $KH_2PO_4$, 0.01M NaOAc and 0.01M NaClO. Otherwise, for YD mine the combination of 0.01M $KMnO_4$, 0.01M NaOAc and 0.01M NaClO. The $SO_4^{2-}$ reduction efficiency of pyrite, IK and YD mine samples was 70, 92 and 84%, respectively. For 8 days, no significant increase of $SO_4^{2-}$ from pyrite sample coated with inhibitor was observed. The pH of solution remains in between 4 to 6 for the reaction conditions.

  • In this work, the effect of molybdenum disilicide(MoSi$_2$) content on the impact properties of carbon-carbon composites(C/C composites) was investigated in the presence of MoSi$_2$. The content of MoSi$_2$ was varied in 0, 4, 12 and 20 wt% on the basis of resin matrix for anti-oxidation properties of the composites under high temperature. As a result, the composites made with MoSi$_2$ resulted in an increase of interfacial adhesion between fibers and matrix, which could improve the impact properties of the composites. Especially, 12 wt% Mosi$_2$ composites showed the highest impact properties in the present system. This was probably due to the existence of brittle-to-ductile transition(BDT) properties of MoSi$_2$ in the vicinity of 90$0^{\circ}C$, resulting from increasing the interfacial adhesion force among fibers, filler, and matrix in the composites.

Screening of Inhibitor of Thyroid Peroxidase, an Oxidative Coupling Enzyme from Natural Products (생약으로 산화적 결합 효소인 갑상선 peroxidase의 저해제 검색)

  • Thyroid peroxidase is a biochemical target protein for the antithyroid drugs. Ethanol extracts from one hundred and thirty seven natural products were screened for the inhibition of thyroid peroxidase activity. Thyroid peroxidase was purified from porcine thyroids, and the inhibition of peroxidase activity was evaluated using guaiacol oxidation (C-C coupling) assay. Twenty one natural products expressed a remarkable inhibition (>50%) of peroxidase activity at $330{\mu\textrm{g}}$ solid weight/m. The 50% inhibitory concentration ($IC_{50}$) of 70% ethanol extract from six potent natural products ranged from 3.1 to $31.2{\;}{\mu\textrm{g}}$ solid weight/m, in contrast to the range ($0.33~0.54{\;}{\mu\textrm{g}}/ml$) of $IC_{50}$ values fro catechin and epigallocatechin gallate as positive controls. Noteworthy, the extract of Camellia taliensis showed irreversible inhibition of the enzyme. It is suggested that extract from some natural products such as Camellia taliensis, Rheum undulatum or Euphorbia perinensis, exhibiting a potent inhibition of peroxidase activity, may be developed as sources of potent antithyroid agents.

Antiplatelet and antithrombotic activities of purpurogallin in vitro and in vivo

  • Enzymatic oxidation of pyrogallol was efficiently transformed to an oxidative product, purpurogallin (PPG). Here, the anticoagulant activities of PPG were examined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT), and the activities of thrombin and activated factor X (FXa). And, the effects of PPG on expression of plasminogen activator inhibitor type 1 (PAI-1) and tissue-type plasminogen activator (t-PA) were evaluated in tumor necrosis factor (TNF)-${\alpha}$ activated human umbilical vein endothelial cells (HUVECs). Treatment with PPG resulted in prolonged aPTT and PT and inhibition of the activities of thrombin and FXa, as well as inhibited production of thrombin and FXa in HUVECs. In addition, PPG inhibited thrombin-catalyzed fibrin polymerization and platelet aggregation. PPG also elicited anticoagulant effects in mice. In addition, treatment with PPG resulted in significant reduction of the PAI-1 to t-PA ratio. Collectively, PPG possesses antithrombotic activities and offers a basis for development of a novel anticoagulant.

  • To show the effects of metallothionein (MT) on the activity of enzymes involved in the removal of reactive oxygen species, MT has been added to the assay systems of superoxide dismutase (SOD), catalase and peroxidase. We have used assay systems of SOD based on NADPH oxidation and nitrite formation from hydroxylammonium chloride as an assay of superoxide breakdown rate. The two assay systems showed different results at the high concentration of MT. MT showed the scavenging of superoxide in the SOD assay system in the presence and absence of SOD. MT added to the SOD assay system behaved as an activator of SOD, but apo-MT behaved as an inhibitor. When MT was added to the assay system in the presence of a fixed amount of SOD, the breakdown rate of superoxide increased. The effects of MT on the decomposition of hydrogen peroxide and the activity of catalase and peroxidase decomposing hydrogen peroxide were evaluated. MT decreased the activities of catalase and peroxidase. We have concluded that the function of MT as an antioxidant might effect the level of superoxide scavenging and not the level of hydrogen peroxide.

Inhibitory Effect against Angiotensin Converting Enzyme and Antioxidant Activity of Panax ginseng C. A. Meyer Extracts (인삼 추출물의 Angiotensin Converting Enzyme 저해 효과와 항산화 활성)

  • The study was performed for elucidating angiotensin converting enzyme (ACE) inhibitory activity and comparing antioxidative activity of Panax ginseng extracts prepared at different conditions. Total phenolic content, inhibitory activity on ACE and antioxidative effects were tested on 10 ethanolic extracts and correlation coefficient between total phenolic content and physiological activity was calculated. Yield and total phenolic content of 50% ethanolic extract prepared at $85^{\circ}C$ exhibited the highest value as 42.52% and 0.82%, respectively. Among the fractions obtained from 50% ethanolic extract prepared at room temperature, water fraction showed the highest value in yield as 72.08% and ethyl acetate fraction did in total phenolic content as 6.59%. In the test on ACE inhibitory activity, 50% ethanolic extract obtained at room temperature indicated the strongest effect of 93.8% which was higher than 85.2% of commercialized ACE inhibitor and solvent fractions showed potent inhibitory activity in order of hexane fraction, diethyl ether fraction, ethyl acetate fraction, butanol fraction and water fraction at concentration of $4000{\mu}g/ml$. 50% Ethanolic extract prepared at $85^{\circ}C$ had the most potent inhibition effect on human LDL oxidation as 78.2% at $200{\mu}g/ml$ and the other extracts also did above 60%. Diethyl ether fraction and ethyl acetate fraction showed strong inhibition activity $(34.38%{\sim}78.13%)$ on LDL oxidation at concentration of $10{\sim}200\;{\mu}g/ml$. From the statistical analysis via SAS program, correlation coefficient between total phenolic content and ACE inhibitory effect was 0.6353 at P<0.05. Conclusively, this report showed that the most efficient extraction condition for elevating inhibitory activity on ACE and LDL oxidation, phenolic content and yield from Panax ginseng was 50% ethanol extraction at room temperature or high temperature condition. And Panax ginseng would be used for preventing hypertension or atheroscrelosis for man via inhibitory action on ACE and LDL oxidation.

A Study on the Inhibiting Corrosion of Triethanolamine for the SCM440 Steel (CM440 강에 대한 트리에탄올아민의 부식억제에 관한 연구)

  • In this study, the current-voltage curves for the SCM440 steel by the addition of an organic corrosion inhibitor triethanolamine(TEA) was measured using the conventional three electrodes of cyclic voltammetry. As a result, the C-V characteristics of SCM440 steel were to be for an irreversible process due to the oxidation current from the cyclic voltammetry. Diffusion coefficient according to the twice increasing the concentration of TEA in the corrosion inhibitor from $2.5{\times}10^{-4}M$ to $5.0{\times}10^{-4}M$, the diffusion coefficient was found to be a good corrosion-inhibiting effect is reduced by 1.5 times, so for each $2.561{\times}10^{-6}cm^2s^{-1}$ to $1.707{\times}10^{-6}cm^2s^{-1}$. When according to the electrolyte concentration and the effect is to increase the electrolyte concentration to 1.0 N at 0.5 N, the diffusion coefficient is $2.56{\times}10^{-6}cm^2s^{-1}$ to $5.12{\times}10^{-6}cm^2s^{-1}$, each 2 times decrease in the use of the electrolyte of 1.0 N, because this was more appropriate.
