High Throughput-compatible Screening of Anti-oxidative Substances by Insect Extract Library (약용곤충추출물 라이브러리를 이용한 항산화 활성의 초고속 검색)

  • Oxidant stress is well-known for a pivotal parameter related to neuro-inflammatory diseases including Alzheimer's disease, Parkinson's disease, and ALS (Lou Gehrig's disease). In order to effectively screen for anti-inflammatory agents, we first established the infrastructure of high throughput screening for anti-oxidant agents from medicinal insect library extracted with water, methanol, ethanol, and dimethyl sulfoxide. By the screening system, we found that Tenodera angustipennis Saussure, Pyrocoela rupa Olivier and Papilio maackii Mntris had strong anti-oxidant activity. Moreover, Tenodera angustipennis Saussure and Tenodera aridifolia (Stoll) exhibited protection effects of cellular damage by treatment of an oxidant hydrogen peroxide. Together, the results suggest that some selected hits could be a potential agent against neuro-inflammation, although the in vivo studies should be clearly tested.

Heat stress resistance of Ilmi

  • Rice production and quality could be changed by temperature condition. Extremely high temperature event have been occurred more frequently for global warming. To increase rice quality against to global warming, breeding of heat tolerance rice cultivar is needed. Ilmi which is the one of Korean leading rice cultivar shows heat stress resistant character during ripening stage. Yield and quality (brown and milled rice) of Ilmi did not show significant different under high temperature condition compared to control condition. However, the main physiological characters for heat resistance of Ilmi have been not investigated yet. Therefore we try to investigate the heat tolerance characters of Ilmi. Two rice cultivars, Ilmi and Ilpum-which is heat susceptible rice cultivar, were cultivated under natural condition in wagnor pot until heading was appeared. After checking heading date, each material was cultivated under different temperature condition, heat($32/22^{\circ}C$) and control($26/16^{\circ}C$) condition. Anti-oxidant enzyme activity was checked during ripening stage in each material. Catalase and ascorbic peroxidase activity of leaf under heat stress condition were higher in Ilmi than Ilpum especially early ripening stage. Analyzing of stress resistance using $H_2O_2$, the flag leaf of Ilmi showed more green color than Ilpum with higher chlorophyll content than those of Ilpum. We also checked the amount of $H_2O_2$ content in young leaf of each material by treating high temperature. $H_2O_2$ content in each material was increased according to treatment time. However $H_2O_2$ content of young leaf in Ilmi was less than those in Ilpum. Also catalase and ascorbic peroxidase activity in leaf increased much faster in Ilmi than Ilpum. With those data, we confirmed that heat stress resistance of Ilmi is due to the higher anti-oxidant activity against to stress condition. We will investigate the heat tolerance characters of Ilmi more in further study to enhance the breeding effect of heat stress tolerance rice.

Effects of Hanbag Mushroom(Grifola frondosa) on Oxidative Stress in Diabetic Rats (당뇨유발 흰쥐에 있어서 산화적 스트레스에 대한 함박잎새버섯의 효과)

  • This research was carried out to investigate the effects of Hambag mushroom on the oxidative stress in diabetic rats, Sprague-Dawley. The diabetic rats induced by streptozotocin were fed with hambag mushroom-powder(G. frondosa) for 6 weeks. For the level of oxidative stress in liver and pancreas tissues, it was studied by measuring LPO (lipid oxide) level as an indicator of lipid peroxidation, XOD(xanthine oxidase) as one of important sources for free radicals and the levels of GSH and GST as anti-oxidant systems. Also, as an indicator of liver damaged by oxidative stress, the activities of serum ALT and AST were measured. It was observed that the levels of ALT, AST, LPO and XOD were higher by about two times in both tissues from diabetic rats than in those from control rats. This indicates that the oxidative stress induced by diabetes caused the tissues damages. However, these levels were decreased in the tissues from rats with hambag mushroom-powder. Futhermore, the activity of GST were higher in both tissues from diabetic rats fed with hambag mushroom-powder than in those from diabetic rats. Thus, it is considered that the hambag mushroom-powder decreases the level of oxidative stress by increasing activity of anti-oxidant system such as GSH and GST. It is suggested that the hambag mushroom-powder can be useful for preventing the tissues damaged by diabetes-induced oxidative stress.

Antioxidant effect of Lonicera Caerulea on heat stress-treated male mice

  • Lonicera caerulea (Honey berry, HB) has been used in medical treatment in Russia, Japan, China and Korea. It has high level of vitamin C and polyphenolics. Polyphenolics can improve anti-inflammatory effect and prevent cancer, diabetes mellitus type 2. Also, Vitamin C is a representative anti-oxidant. however, it is still unknown what effect it will have on the oxidation stress of the reproductive system. In previous studies, ROS can be produced when it is exposed to heat stress and has negative effect on sperm's maturation, capacitation, hyperactivation, acrosome reaction and fusion of egg and sperm. Therefore, the purpose of this study is to investigate the antioxidant effects of L. Caerulea on the sperm and mice. At first, it conducted using ICR mouse (n = 20) for 4 weeks. There are four groups of mice (n = 5 per group). Also, L. Caerulea was taken by oral gavage. Group I (control) kept at 23℃-27℃ and administer D.W (0.5 mL/day), Likewise, Group II (HB) kept at room temperature but gave HB (250 mg/kg, 0.5 mL/day), Group III (HB + HS) received heat stress (40℃) using hyperthermia induction chamber and gave HB at same dose. and Group IV (HS) exposed heat stress only. Mainly, we showed degree of gene expression using Western blot in SOD, HSP 70, 17β-HSD and Real-time PCR. It can find correlation between intracellular activity like steroid hormone, apoptosis under ROS and antioxidant activity of L. Caerulea.

A Nudix Hydrolase Protein, Ysa1, Regulates Oxidative Stress Response and Antifungal Drug Susceptibility in Cryptococcus neoformans

  • A nucleoside diphosphate-linked moiety X (Nudix) hydrolase-like gene, YSA1, has been identified as one of the gromwell plant extract-responsive genes in Cryptococcus neoformans. Ysa1 is known to control intracellular concentrations of ADP-ribose or O-acetyl-ADP-ribose, and has diverse biological functions, including the response to oxidative stress in the ascomycete yeast, Saccharomyces cerevisiae. In this study, we characterized the role of YSA1 in the stress response and adaptation of the basidiomycete yeast, C. neoformans. We constructed three independent deletion mutants for YSA1, and analyzed their mutant phenotypes. We found that ysa1 mutants did not show increased sensitivity to reactive oxygen species-producing oxidative damage agents, such as hydrogen peroxide and menadione, but exhibited increased sensitivity to diamide, which is a thiol-specific oxidant. Ysa1 was dispensable for the response to most environmental stresses, such as genotoxic, osmotic, and endoplasmic reticulum stress. In conclusion, modulation of YSA1 may regulate the cellular response and adaptation of C. neoformans to certain oxidative stresses and contribute to the evolution of antifungal drug resistance.

  • Pulmonary system is a target organ and primary defense mechanism against environmental oxidants and polutants. Enzymatic and nonenzymatic antioxdant defense mechanisms undoubtedly protect the lung from oxidants even endogenous oxidative stress. In addition, new ways of augmenting pulmonary antioxidant defenses are developed, which can be used to support the intrinsic antioxidants. Therefore, improved understanding of antioxidant defense mechanisms will increase our knowledge of the cause and will suggest rational approaches for treating and preventing oxidant-induced lung injury. In this review, we discuss the formation and scavenging of free radicals, and the strategies for antioxidant defense of pulmonary system.

Effects of Vitamin C on Oxidative Stress Due to Anesthesia and Surgical Trauma in Dogs (개 마취와 수술 창상에 따른 산화스트레스에 대한 비타민 C의 효과)

  • The study evaluated the antioxidant effects of ascorbic acid (AA) against oxidative stress during laparotomy in dogs under general anesthesia using isoflurane. Twelve dogs were randomly assigned to either the ascorbic acid group (AAG) or the sham group (SG). AAG received intravenous dosage of 100 mg/animal of standard AA 10 minutes before anesthesia. Plasma levels of cortisol, glucose, total oxidant status (TOS), total antioxidant status (TAS), and oxidative stress index (OSI) were measured. Cortisol level increased significantly with time in both groups (p < 0.05). The change of glucose levels was not significantly different between both groups. TOS and OSI increased significantly with time in the SG (p < 0.05), whereas it did not significantly change with time in the AAG. The TOS and OSI of the AAG were significantly lower than in the SG (p < 0.05). However, TAS did not significantly change between both groups. The results suggest that the decrease of TOS in the AAG might be related to the conversion of antioxidants to oxidants. The decrease of OSI indicates that the decrease of reactive oxygen species (i.e., oxidative stress) produced at the site of injury is able to decrease surgical side effects of abnormal blood circulation, organ failure, and inflammation. Therefore, AA can be used to protect a surgical patient from oxidative stress in canine laparotomy.

The Protective Effect of Water Extract of Phellodendri Cortex in Chronic Reflux Esophagitis-induced Rats (만성 역류성 식도염 동물 모델에 대한 황백(黃柏) 물 추출물의 식도 점막 보호 효과)

  • Objective : Reflux esophagitis is a disease caused by the reflux of gastric acid and inflammation due to unstable gastroesophageal sphincter. The aim of the present study was to clarify the effect of Phellodendri Cortex (PC) on chronic reflux esophagitis (CRE) in rats. Methods : The anti-oxidant activity of PC was measured by total polyphenol, total flavonoid contents, 1, 1-diphenyl-2-picrylhydrazyl (DPPH), and 2, 2'-azinobis-3-ethyl-enzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity. A CRE was established surgically in SD rats. And then CRE rats were treated with water or PC 200 mg/kg body weight for 14 days. Besides, the anti-oxidant and inflammatory protein levels were evaluated using western blotting. Results : PC reduced esophagus tissues injury. The total polyphenol (36.05 ± 0.25 mg/g) and total flavonoid (72.90 ± 0.61 mg/g) of PC showed a high content. PC strongly reduced radical scavenging activities (DPPH IC50 43.58 ± 1.54 ㎍/㎖; ABTS IC50 36.75 ± 0.35 ㎍/㎖). Moreover, reactive oxygen species (ROS) and peroxynitrite (ONOO-) levels in serum, the protein expression of inducible nitric oxide synthases (iNOS) were significantly reduced. In addition, the protein expression of NADPH oxidases related to oxidative stress were significantly reduced in PC compared to CRE control. PC effectively reduced inflammatory factors including, TNF-α, and IL-6 via NF-κBp65 inactivation through the inhibition of p-IκBα and increased anti-oxidant enzyme such as HO-1, SOD, catalase, and GPx-1/2 via Nrf2 activation. Conclusions : Taken together, these results show that PC can alleviate the esophageal mucosal ulcer though the inhibition of NF-κB inflammatory and the enhancement of Nrf2 anti-oxidant pathway.

Cheogjogupye-Tang has Anti-oxidant Potential through the Activation of Nrf2 (청조구폐탕(淸燥救肺湯)의 Nrf2 매개 항산화 효능)

  • Transcription factor, Nrf2 was well known to protect cell from oxidative stress by up-regulating it's dependent anti-oxidative genes such as HO-1 and NQO1. Cheongjogupye-tang (CJGPT), a traditional herbal formula was originally recorded in 『EuiMunBeopRyul』, still having been used to treat pulmonary disease such as asthma and pulmonary inflammation, in Eastern Asian countries. However, the underlying therapeutic mechanisms remain elusive. The purpose of this study is to investigate the anti-inflammatory or anti-oxidative effects of CJGPT on the RAW 264.7 cells. To examine the anti-inflammatory or anti-oxidative effects of CJGPT, MTT assay, immunoblotting, RT-PCR and reporter gene assays were performed. Although CJGPT slightly suppressed the nuclear NF-κB expression, it did not decreased the expression of pro-inflammatory genes in LPS-stimulated RAW 264.7 cells. Moreover, it did not increased the transcriptional activity of NF-κB in reporter gene assay. However, CJGPT upregulated the nuclear expression of Nrf2, as well as increased the expression of Nrf2-dependent genes such as HO-1 and NQO1. In addition, CJGPT incresed the transcriptional activity of Nrf2. Taken together, our results showed that CJGPT exerts functions as an anti-oxidant mainly by activating Nrf2.
