Strain Response Analysis of RC Beams Strengthened with Optical Fiber-embedded CFRP Sheet (광섬유 매립 CFRP 쉬트로 보강한 RC 보의 변형률 응답 분석)

  • This paper reports the results of an experimental study using the BOTDR sensor to detect the unbonded location of attached CFRP sheet for structural rehabilitation. A specimens with the unattached CFRP sheet were fabricated for this study, on which BOTDR sensor was attached with a nylon net. During the flexural test of the specimens, the strain of the CFRP sheet was measured using the BOTDR sensor and electric resistance gauges. From the results, it was confirmed that the strain distribution obtained through the BOTDR sensor can be effectively used to visualize and detect the unbonded position of the CFRP sheet. In addition, In addition, the strain measured by the BOTDR sensor was found to be more effective in analyzing the overall structure behavior than the electric resistance strain gauge. The development of a BOTDR sensor with a measuring longth of less than 100 mm will enable accurate detection of the local unbonded position of the CFRP sheet.

A Prediction of the Behavior in Normally Consolidated Clay with Application of Isotropic Single Hardening Constitutive Model (등방단일경화구성모델에 의한 정규압밀점토의 거동 예측)

  • The results of a series of triaxial compression tests on remolded normally consolidated clay are compared with the predictions .by the isotropic single -hardening constitutive model, which incorporates eleven parameters. The parameters can be determined from undrained triaxial compression tests on isotropically consolidated specimens of remolded clay. The model with the determined parameters is applied to predict the stress-strain and pore pressure behaviors for untrained triaxial compresion tests on anisotropically consolidated specimens. Also the model is utilized to predict the stress strain and voltmetric strain behavior for drained triaxial compression tests on both isotropic and anisotropic specimens. The predicted response agrees well with the measured behavior for undrained triaxial compression tests on not only isotropically but also anisotroically but also anisotropically consolidated specimens. The initial volumetric strain is, however, predicted to be less than the measured value from drained triaxial compression tests, while the predicted volumetric strain close to failure is greater than the measured value. Nevertheless, it may be stated generally that overall acceptable predictions are produced. Therefore, the results of this study indicate that the applicability of the model on prediction of the behavior of normally consolidated clay is achieved sufficiently.

Safety Assessment of Potential Lactic Acid Bacteria Bifidobacterium longum SPM1205 Isolated from Healthy Koreans

  • The safety assessment of Bifidobacterium longum SPM1205 isolated from healthy Koreans and this strain's inhibitory effects on fecal harmful enzymes of intestinal microflora were investigated. The overall safety of this strain was investigated during a feeding trial. Groups of SD rats were orally administered a test strain or commercial reference strain B. longum $1\times10^9\;CFU/kg$ body weight/day for four weeks. Throughout this time, their feed intake, water intake and live body weight were monitored. Fecal samples were periodically collected to test harmful enzyme activities of intestinal microflora. At the end of the four-week observation period, samples of blood, liver, spleen, kidney, and gut tissues were collected to determine for hematological parameters and histological differences. The results obtained in this experiment demonstrated that four weeks of consumption of this Bifidobacterium strain had no adverse effects on rat's general health status, blood biochemical parameters or histology. Therefore, it is likely to be safe for human use. Fecal harmful enzymes such as $\beta-glucosidase,\;\beta-glucuronidase$, tryptophanase and urease, were effectively inhibited during the administration of the B. longum SPM1205. These results suggested that this B. longum SPM 1205 could be used for humans as a probiotic strain.

Reduction of Acetate and Lactate Contributed to Enhancement of a Recombinant Protein Production in E. coli BL21

  • Acetate and lactate in growth media are detrimental to the production of Thermus maltogenic amylase (ThMA), a heterologous protein, as well as to the growth of recombinant Escherichia coli. Only 50 mM of acetate or 10 mM of lactate reduced 90% of specific ThMA activity. In this study, mutant E. coli strains blocked in the ackA-pta or ackA-pta and ldh pathways were created, characterized, and assessed for their culture performace in 300 L-scale fermentation. The ackApta and ldh double-mutant strain formed significantly less lactate and acetate, and produced a concomitant increase in the excretion of pyruvate (17.8 mM) under anaerobic conditions. The ackA-pta mutant strain accumulated significant acetate but had an approximately 2-fold increase in the formation of lactate. The ackA-pta and ldh double-mutant strain had superior overall performance in large-scale culture under suboptimal conditions, giving 67% higher cell density and 66% higher ThMA activity compared with those of the control strain. The doublemutant strain also achieved a 179% improvement in volumetric ThMA production.

Improvement of FK506 Production in the High-Yielding Strain Streptomyces sp. RM7011 by Engineering the Supply of Allylmalonyl-CoA Through a Combination of Genetic and Chemical Approach

  • FK506, a widely used immunosuppressant, is a 23-membered polyketide macrolide that is produced by several Streptomyces species. FK506 high-yielding strain Streptomyces sp. RM7011 was developed from the discovered Streptomyces sp. KCCM 11116P by random mutagenesis in our previous study. The results of transcript expression analysis showed that the transcription levels of tcsA, B, C, and D were increased in Streptomyces sp. RM7011 by 2.1-, 3.1-, 3.3-, and 4.1-fold, respectively, compared with Streptomyces sp. KCCM 11116P. The overexpression of tcsABCD g enes in Streptomyces sp. RM7011 gave rise to approximately 2.5-fold (238.1 μg/ml) increase in the level of FK506 production compared with that of Streptomyces sp. RM7011. When vinyl pentanoate was added into the culture broth of Streptomyces sp. RM7011, the level of FK506 production was approximately 2.2-fold (207.7 μg/ml) higher than that of the unsupplemented fermentation. Furthermore, supplementing the culture broth of Streptomyces sp. RM7011 expressing tcsABCD genes with vinyl pentanoate resulted in an additional 1.7-fold improvement in the FK506 titer (498.1 μg/ml) compared with that observed under non-supplemented condition. Overall, the level of FK506 production was increased approximately 5.2-fold by engineering the supply of allylmalonyl-CoA in the high-yielding strain Streptomyces sp. RM7011, using a combination of overexpressing tcsABCD genes and adding vinyl pentanoate, as compared with Streptomyces sp. RM7011 (95.3 μg/ml). Moreover, among the three precursors analyzed, pentanoate was the most effective precursor, supporting the highest titer of FK506 in the FK506 high-yielding strain Streptomyces sp. RM7011.

  • Traditional theories of the tensile failure of paper have assumed that uniform strain progresses throughout the sheet until an imperfection within the structure causes a catastrophic break. The resistance to tensile elongation is assumed to be elastic , at first, throughout the structure, followed by an overall plastic yield. However, linear image strain analysis (LISA) has demonstrated that the yield in tensile loading of paper is quite non-uniform throughout the structure, Traditional theories have failed to define the flaws that trigger catastrophic failure. It was assumed that a shive or perhaps a low basis weight area filled that role. Studies of the fracture mechanics of paper have typically utilized a well-defined flaw around which yield and failure could be examined . The flaw was a simple razor cut normal to the direction of tensile loading. Such testing is labeled mode I analysis. The included fla in the paper was always normal to the tensile loading direction, never at another orientation . However, shives or low basis weight zones are likely to be at random angular orientations in the sheet. The effects of angular flaws within the tensile test were examined. The strain energy density theory and experimental work demonstrate the change in crack propagation from mode I to mode IIas the initial flaw angle of crack propagation as a function of the initial flaw angle is predicted and experimentally demonstrated.

Residual seismic performance of steel bridges under earthquake sequence

  • A seismic damaged bridge may be hit again by a strong aftershock or another earthquake in a short interval before the repair work has been done. However, discussions about the impact of the unrepaired damages on the residual earthquake resistance of a steel bridge are very scarce at present. In this paper, nonlinear time-history analysis of a steel arch bridge was performed using multi-scale hybrid model. Two strong historical records of main shock-aftershock sequences were taken as the input ground motions during the dynamic analysis. The strain response, local deformation and the accumulation of plasticity of the bridge with and without unrepaired seismic damage were compared. Moreover, the effect of earthquake sequence on crack initiation caused by low-cycle fatigue of the steel bridge was investigated. The results show that seismic damage has little impact on the overall structural displacement response during the aftershock. The residual local deformation, strain response and the cumulative equivalent plastic strain are affected to some extent by the unrepaired damage. Low-cycle fatigue of the steel arch bridge is not induced by the earthquake sequences. Damage indexes of low-cycle fatigue predicted based on different theories are not exactly the same.

  • Currently the plates used for the various structures has a tendency to being slim to the degree of the sheet metal vis-a-vis the overall measurements of the structure, and the conditions of their usage and load have been getting diverse. As the structure material has been used more and more under the various conditions, the necessity of the strength analysis is to be required. While most of these methods are simply based on the strain gaging method; that is, directly attaching most of the material to the gage, using the non-contact method is still in the early stage. One of the non-contact methods is by the use of the laser that has high-level of the accuracy for the measurement, and this laser also has excellent characteristics on which many studies for its applications are focused throughout the many fields. Therefore, this dissertation is on the measurement of the strain caused by the characteristics of the TIG welded zone which is used with 3D ESPI system that is functionally modified through the laser ESPI system.

Effect of Porosity on Quality Index of Tensile Property of A356 Casting Alloys (A356합금의 품질지수에 미치는 미소기공율의 영향)

  • The dependence of the tensile properties on variations in the porosity of A356 aluminium alloys was investigated in terms of the quality index of the tensile properties based upon the ultimate tensile strength and elongation as well as the variation of the strength coefficient and strain-hardening exponent with regard to a T6 treatment. The test specimens were prepared by low-pressure die-casting and a subsequent T6 treatment, and the experimental results of a tensile test carried out at room temperature were compared to the theoretical description using a modified constitutive model. The nominal value of the quality index of A356 alloys increases gradually with a lapse of the ageing time upon a T6 treatment, despite the fact that this value is temporarily decreased during the initial stage of ageing from a solutionised condition. Additionally, the quality index depends practically upon the porosity variation with a power law relationship without regard to whether in solutionised or artificial aged conditions. The theoretical description indicates that the strength coefficient directly determines the nominal level of the quality index. Moreover, the overall dependence of the quality index on the porosity variation is remarkably weakened with an increase in the tensile strain, whereas the quality index depends sensitively upon the porosity variation with a low value of the strain-hardening exponent.

Complete chromosome and plasmid sequences of Staphylococcus aureus strain JDFM SA01, isolated from a milk filter in Korean dairy farm

  • Staphylococcus aureus is a significant pathogen that can source a variety of illness worldwide. In this announcement, we report here the complete genome sequence of S. aureus strain JDFM SA01, isolated from a milk filter collected from Korean dairy farm. The final complete genome assembly consists of one circular chromosome (2,748,925 bp) with an overall GC content of 32.9% and one circular plasmid sequence (24,655bp) with a GC content of 28.7%.
