Effect of Gonadotropin Administration on Blood Ovarian Steroid Hormone Level in Rabbit (성선자극 호르몬 투여가 토끼의 혈중 난소 스테로이드호르몬 수준에 미치는 영향)

  • The present study was carried out to investigate the effect of gonadotropin administration on blood ovarian steroid hormone in angora rabbit. Mature angora rabbits were primed for superovulation with PMSG 100IU. Eighty hours later, the rabbit were induced to ovulate with HCG 100IU. In exp 1, blood progesterone and estradiol of superovulated does were measured by radiommunoassay. Blood progesterone concentration at 93, 99, 102 and 114 hours after HCG injection were 12.9$\pm$0.5, 34.8$\pm$5.1, 12.2$\pm$2.7 and 43.4$\pm$5.8ng/ml, respectively. Mean progesterone concentration of blood collected at 99 and 114 hours after HCG injection(p<0.05). However, mean blood estradiol concentration was not changed. In exp 2, superovulated does were unilaterally ovariectomized at 96 hours after HCG injection. Blood progesterone concentration was tend to be decreased after ovariectomy. Nosignificant changes in blood estradiol concentration was observed after ovariectomy. In exp 3, superovulated does were bilaterally ovariectomized at 96 hours after HCG injection Ovariectomized does were treated with progesterone. Blood progesterone level in the rabbits treated, twice daily, with 5mg progesterone after ovariectomy was similiar to that in the superovulated intact rabbits. Blood estradiol concentration of the rabbits after bilateral ovariectomy was beyond detection range. Blood progesterone concentration was significantly decreased to 7.6$\pm$3.0ng/ml wi thin 3 hours after ovriectomy(p<0.05). However, that value was increased to 34.8$\pm$8.2ng/ml by 5 mg progesterone treatment and this elevated level was significatnly decreased to 7.3$\pm$2.4ng/ml at 12 hours after progesterone administration(p<0.05).

Effect of Ovariectomy and Calcium Intake on Femur and Lumbar Compositions and Bone Related Factors in Adult Rats (성숙한 쥐의 대퇴골과 요추의 조성과 골격관련 인자에 대한 난소절제와 칼슘섭취 효과)

  • This study explored the effect of calcium levels and/or ovariectomy on bone composition and its related factors using the female Sprague-Dawley rats which achieved peak bone mass in normal(0.5%) calcium intake during growth period. The rats were randomly divided into six groups and fed 0.1%, 0.5% and 1.5% calcium diets for 8 weeks after ovariectomized and sham operation. The results indicated that body weight gain was higher in ovariectomized groups than sham groups, regardless of dietary calcium levels and food intakes. Serum Ca and P concentrations were normal level regardless of dietary calcium levels and operation. Estrogen concentration was decreased in ovariectomized rat groups. Serum alkaline phosphatase activity and urinary hydroxyproline were increased in ovariectomized groups. When rats were fed normal Ca diet during growing period, weight, length and breaking force of femur were not significantly different in all groups but increased the same level. Generally, lipid contents in lumbar and femur were lower in low calcium groups and this effect was more pronounced in femur. In case of femur, the significant decrease in Ca contents of bone was observed in a relatively short period of feeding low Ca diet, even though it appeared th desirable peak bone mass had achieved through the growing period by supplementing the adequate amount of calcium. The marked decrease of estrogen levels after ovariectomy did not seem to influence greatly bone parameters measured except bone wet weight. Therefore, this study indicated that lower intake of Ca might be more important determinating factor against osteoporosis than postmenopausal state stimulated by ovariectomy in female rats. With normal or high intake of Ca it appears possible to prevent bone loss in postmenopausal period, and this might apply only in case of achieving peak bone mass in the growing with the adequate intake of calcium. A histochemical study of argentaffin endocrine cells in the gastrointestinal tract of ovariectomized rats

  • The regional distributions and frequencies of argentaffin endocrine cells in gastrointestinal (GI) tract of osteoporotic Sprague-Dawley rat induced by ovariectomy were studied by Masson-Hamperl silver stain. The experimental animals were divided into two groups, one is non-ovariectomized group (Sham) and the other is ovariectomized group (OVX). Samples were collected from each part of GI tract (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) at 10th week after ovariectomy or sham operation. Argentaffin cells were detected throughout the entire GI tract with various frequencies regardless of ovariectomy except for the rectum of OVX in which no cells were detected. Most of these argentaffin cells in the mucosa of GI tract were generally spherical or spindle in shape (open type cell) while cells showing round in shape (close type cell) were rarely found in gland regions. Significant decrease of argentaffin cells was detected in OVX compared to that of Sham except for the fundus and jejunum. However, in the fundus and jejunum, argentaffin cells in OVX showed similar frequency compared to that of Sham. In conclusion, the endocrine cells are the anatomical units responsible for the production of gut hormones that regulate gut motility and digestion including absorption, and a change in their density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion. Ovariectomy induced severe quantitative changes of GI argentaffin endocrine cell density, and the abnormality in density of GI endocrine cells may contribute to the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis.

Hypertrophical Changes of the Corpus Allatum Caused by Ovariectomy in Blattella germanica (난소제거 바퀴에서 알라타체의 이상비대화에 관한 연구)

  • The present study is undertaken to investigate the hypertrophical changes of the corpus allatum (corpora allata, CA) after the ovariectomy in Blattella germanica. In particular, the ultrastructural aspects of the normal and ovariectomized conditions, and induced factors of the hypertrophic phenomenon are focused. Ultrastructure of the CA from an immediately emergent adult is similar to that of the last larval stage that has stopped secreting juvenile hormone. The CA is composed of undifferentiated cells, exhibiting an electron-lucent matrix, a few mitochondria and less smooth endoplasmic reticulum. The karyoplasm occupy most of the cytoplasm. Electron-dense materials are filled with the intercellular spaces and gap junctions are also found. Almost no ultrastructural changes have been noticed during seven days until the oviposition. However, considerable changes in structure have been detected soon after the oviposition. Mitochondria are increased dramatically in number and cristae, and changed to the filamentous form with a high electron density. In addition, Golgi complexes, microtubules, and polysomes are also increased. After an oviposition, the total volume of the CA are decreased again. The volume of the CA are increased continuously, hypertrophy, after the ovariectomy. Morphological aspects of the CA in an early stage after the removal are similar to the structure of the secondary egg maturation. Large and electron dense globules are observed in the ovariectomized CA cytoplasms and they are present in those cells for a long period of time. Yet such a hypertrophical phenomenon occur only in the specific cells. The hypertrophy are caused by hollowing the part of the CA cells and later filling such site with polysomes. In 42 days after the ovariectomy, the nuclear membranes disappear in the CA cells, thus, exhibiting the prokaryotic-like features. Some results of the current study will contribute to the establishment of the model that explain unusual changes accompanied by certain treatment in insects and/or further in animals.

THE HISTOLOGIC STUDY OF THE RAT TEMPOROMANDIBULAR JOINT CHANGE FOLLOWING OVARIECTOMY (자성백서의 난소절제시 악관절 변화에 관한 조직학적 연구)

  • This investigation studied the effect of ovariectomy on the temporomandibular joint (TMJ) of rat. Forty female Sprauge-Dawley strain white rats, each weighing about 200grams were used and divided into two groups. The one group was performed ovariectomy, and the other was performed sham operation. Both group were killed at the intervals of 3 days, 1, 2, 4, and 8 weeks postoperatively, paraffin section were stained with H&E, MT and histomorphometric study of the TMJ was performed in a synchronous manner with an sham operated control group The result were as follows: 1. At 4 weeks after ovariectomy, an adhesion between the inferior surface of the articular disc and the condyle was observed in the central portion. At 8 weeks after ovariectomy, a focal thickening of the bone (osteophyte) was observed in anterior portion and there was a marked decrease in the trabecular bone in the posterior region. 2. The thickness of cartilage zone was increased at 2 weeks postopratively in the central portion, and decreased at 4 weeks postopratively in the posterior portion. The thickness of transitional zone was decreased at 1 week postoperatively in the anterior and proliferating zone was decreased postoperatively. The thickness of articular zone was decreased at 3 days postopratively in the anterior portion, and decreased at 1 week postoperatively in the central portion. From the above finding, we could conclue that estrogen deficiency in rats causes alteration of TMJ in marked early stage.

Effect of Ovariectomy Induced Hyperphagia on the Intestinal Adaptation (난소절제에 의해 유발된 과식현상이 소장적응변화에 미치는 영향)

  • Ovariectomy induced hyperphagia was reported in various animal models. It was postulated that hyperphagia resulted in interstinal hypertrophy and hyperplasia. Therefore, this study was performed to compare the food intake and intestinal changes between ovariectomized rats(OVX) and sham operated rats(Sham). Results of this study showed that the food intake and body weight of OVX animal was significantly higher than those of Sham animal. This results confirmed the effect of ovarian hormone on the food intake regulation. Hyperphagia induced by ovariectomy influence the intestinal cell growth and showed hyperplasia determined by protein/DNA ratio. Maltase and alkaline phosphatase activity suggested that the intestinal cell was fully matured in body groups. There were no difference in weight of other organs, such as liver, heart and kindney between two groups.

  • The effect of Cervi Cornu Parvum aqua-acupuncture on serum levels of sex hormone was investigated in ovariectomized rats. Water extract of Cervi Cornu Parvum was daily injected for 30 days at points in rats corresponding to bilateral Taixi (Ki. 3) points of human and non-point at root of tail after ovariectomy, respectively. Serum levels of estradiol, testosterone, progesterone, LH and FSH were measured at 2 and 4 weeks after aqua-acupuncture treatment, respectively. Compared with ovariectomized rats, significant increase of estradiol and progesterone and significant decrease of FSH and LH were elicited at 2 and 4 weeks after aqua-acupuncture treatment. Aqua-acupuncture treatment at non-point did not produce any remarkable effect. These findings suggest aqua-acupuncture may restore dysfunction of hypothalamic-pituitary-ovarian axis, associated with a significant increase of serum estradiol level in ovariectomized rats.

  • The objective of this study was to compare the total surgical time, pain score and hemorrhage after ovariohysterectomy (OVH) and bilateral flank ovariectomy (BFOVE) with hemoclips in dogs. OVH was operated on six dogs and BFOVE was performed on six dogs. In BFOVE group, both ovarian pedicle hemostasis applied using hemoclips after removing ovaries. Total surgical time was recorded from starting incision of skin to closure last skin suture. Pain score was measured at 2, 6, 12, and 24 hours after surgery based on University of Melbourne Pain Scale (UMPS). Hemorrhage around clipping area, uterus, abdominal muscles, and skin were monitored after surgery. BFOVE technique could reduce total surgical time and pain after surgery. There was no hemorrhagic problems near ovarian pedicles following surgery. BFOVE with hemoclips could be useful and recommended for sterilization in dog.

  • This experiment study was carried out to prove the efficacy of Cheongpajeon(CPJ) extract in ovariectomized rats. 40 rats were divided into 4 groups, administrated saline after sham operation group(sham-op), administered saline after ovariectomy group(control), administered CPJ 1g/kg after ovariectomy group and administered Livial 0.042mg/kg after ovariectomy group(positive control). We examined the water extract of CPJ that is capable of affecting osteoblast proliferation using MG-63 and HOS-TE85. (omitted)
