Development of Remote Monitoring System for groundwater purifier apparatus for community wells (마을 공동 우물용 지하수 정수 장치의 원격 모니터링 시스템 개발)

  • Recently, the pollution of groundwater has become serious. In particular, the contamination of groundwater near livestock farms is becoming increasingly severe and it is difficult to drink with drinking water. In this paper, a groundwater purifier apparatus that can be installed in a community well was designed. The designed groundwater purifier apparatus enables a RO membrane filter and UV sterilization to remove pollutants, such as heavy metals, bacteria, and organic compounds. In addition, electrical conductivity, pressure, and flow sensors were added for remote monitoring. Remote monitoring of the system can determine the level of fouling and contamination of RO membrane filters through pressure and flow sensor data, and can record changes in the contamination and condition of groundwater through the electrical conductivity of the feed water. The designed groundwater purifier apparatus was installed at a farmhouse and remote monitoring. The result after 15 days of operating a groundwater purifier apparatus and analyzing the monitoring data revealed an average permeate water flow rate of 2.67L/min and an average water pressure of 7.09kgf/㎠, indicating that the RO Membrane filtered without fouling and clogging. The average electrical conductivity was 796.6 S/㎠ of the feed water and 55.6 S/㎠ of permeate water, which is similar to that of general tap water. Through this, it was confirmed that no pollutant occurred in the surroundings. Therefore, the designed groundwater purifier apparatus can confirm the replacement time of the RO membrane filter in advance through remote monitoring, and check the pollution state of the groundwater.

  • Solutions to water pollution, global warming, and climate change have been currently discussed. Pressure retarded osmosis (PRO) using a difference in salt concentration between two fluids is proposed to meet the demand for clean water and produce eco-friendly energy. Although PRO has been researched continuously, it has not been commercialized yet due to limitations such as lack of technology and the high price of membranes. Meanwhile, the membrane is one of the most significant parts of the PRO engine and salinity gradient power (SGP) technology. Research continues to technologically develop graphene oxide membranes and nanocomposite membranes used in salinity gradient power generation. Studies on efficient membranes, solvents, and solutes are active to enable high energy efficiency of the osmotic heat engine even at low temperatures of waste. Studies have been conducted on reducing internal concentration polarization and increasing power density by using membranes with balanced permeability and selectivity. In this review, dealing with these studies, we discuss the types of PRO membranes, theoretical modeling of technologies through efficient membranes, and other technologies to develop the process efficiency.

Effect of Salt Level in Water on Feed Intake and Growth Rate of Red and Fallow Weaner Deer

  • Under a typical Mediterranean environment in southern Australia, the evaporation rate increases significantly in hot summers, resulting in highly saline drinking water for grazing animals. Also in the cropping areas, dryland salinity is a problem. Grazing animals under these environments can ingest excessive amount of salt from feed, drinking water and soil, which can lead to a reduction in growth rate. To understand the impact of high salt intake on grazing deer, two experiments were conducted to assess the effect of salt levels in drinking water on feed intake and growth rate of red and fallow weaner deer. The results revealed that fallow deer did not show any abnormal behaviour or sickness when salt level in drinking water was increased from 0% to 2.5%. Feed intake was not affected until the salt content in water exceeded 1.5%. Body weight gain was not affected by 1.2% salt in drinking water, but was reduced as salt content in water increased. Compared with deer on fresh water, the feed intake of red deer on saline water was 11-13% lower when salt level in drinking water was 0.4-0.8%. An increase in salt level in water up to 1% resulted in about a 30% reduction in feed intake (p<0.01). Body weight gain was significantly (p=0.004) reduced when salt level reached 1.2%. The deer on 1% salt tended to have a higher (p=0.052) osmotic pressure in serum. The concentration of P, K, Mg and S in serum was affected when salt level in water was over 1.0%. The results suggested that the salt level in drinking water should be lower than 1.2% for fallow weaner deer and 0.8% for red weaner deer to avoid any reduction in feed intake. Deer farmers need to regularly test the salt levels in drinking water on their farms to ensure that the salt intake of grazing deer is not over the levels that deer can tolerate.

  • Taurine is one of the most abundant free ${\beta}$-amino acids in the human body that accounts for 0.1% of the human body weight. It has a sulfonic acid group in place of the more common carboxylic acid group. Mollusks and meat are the major dietary source of taurine, and mother's milks also include high levels of this amino acid. The leukocytes, heart, muscle, retina, kidney, bone, and brain contain more taurine than other organs. Furthermore, taurine can be synthesized in the brain and liver from cysteine. There are no side effects of excessive taurine intake in humans; however, in case of taurine deficiency, retinal abnormalities, reduced plasma taurine concentration, and other abnormalities may occur. Taurine enters the cell via a cell membrane receptor. It is excreted in the urine (approximately 95%) and feces (approximately 5%). Taurine has a number of features and functions, including conjugation with bile acid, reduction of blood cholesterol and triglyceride levels, promotion of neuron cell differentiation and growth, antioxidant effects, maintenance of cell membrane stability, retinal development, energy generation, depressant effects, regulation of calcium level, muscle contraction and relaxation, bone formation, anti-inflammatory effects, anti-cancer and anti-atherogenic effects, and osmotic pressure control. However, the properties, functions, and effects of taurine require further studies in future.

The effect of high potassium adaptation on hybridoma cell growth

  • Hybridoma cells were adapted in media containing up to 80 mM $K^+$. The adapted cells obtained tolerance at high osmotic pressure and low pH. The adapted cells showed a maximum viable cell density of $1.1{\times}10^6$ cells/ml when a batch culture was progressed in a nutrient-fortified medium with Erlenmeyer flask at 450 $mOsm/kgH_2O,$ as compared to $4.8{\times}10^5$ cells/ml for non-adapted cells grown under the same conditions. The adapted cells also showed a maximum viable cell density of $7.8{\times}10^5$ cells/ml with the same method at initial pH 7.0 as compared to $5.3{\times}10^5$ cells/ml for non-adapted cells. Adaptation of animal cells at high $K^+$ levels may therefore lead to an improvement of their performance at limited conditions.

Analysis of Students' Responses on the Items of Chemistry II in the College Scholastics Ability Test (대학수학능력시험 화학II 문항에 대한 학생들의 응답 분석)

  • In this study, the students' responses on the items of chemistry II in the College Scholastics Ability Test (CSAT) implemented for the past 3 years since 1999 was investigated. The number of applicants and mean score of chemistry II, average percent correct by contents and inquiry process, and the items with high and/or low percent correct are analysed and discussed. The percentage of applicants for natural science track is reduced each year. However, more than 25% of natural science track applied to chemistry II, which ranked second to biology II. The mean score of chemistry II was increased in 2001 along with those of other subjects. There is no difference in average percent correct either by contents or inquiry process. Students performed generally poor at solving test items relating to 'colligative properties of solution' such as 'boiling point elevation' or 'osmotic pressure', and very well at solving test items relating to 'reaction rates'. Educational implications are discussed.

  • A cyst is a pathologic lesion characterized by a cavity filled with fluid, celluar products, air, or a combination of these. Dentigerous cysts were formed around the crown of unerupted teeth. The reduced enamel epithelium persists around the crown after it has formed. Proliferation of the epithelium in a fluid-filled sac may be induced by osmotic pressure. In the first decade the most frequent location is the premolar site. In each subsequent decade the largest number of cysts are in the mandibular third molar site, with the second most frequent site being the maxillary canine. The treatment of odontogenic cyst can be mostly classified into three types of cyst enucleation, marsupialization and decompression. We should consider age of patient, anatomic structure, location and size for choosing a treatment method. Advantage of cyst enucleation is fast healing, but a injury of a surrounding structure is highly. Marsupialization is conservative treatment that can reduce the damage of a adjacent structure, but it is only limited at superficial lesion. Decompression also is conservative treatment, but it has the difficulty of the oral hygiene and the troublesome of the lavage. We present the possibility that reduces the defect of decompression and cures the lesion efficiently. We report a male patient with the dentigerous cyst developed at left mandibular third molar in this study. We used the decompression for a treatment and created special appliance to treat the lesion efficiently. We report a case of the cyst treatment that is association with efficiency of decompression appliance.

Studies on Short Term Hardening Method of Tree Seedlings for Afforestation of Cut-Rock Slope (암반절개사면 녹화용 강건묘목의 속성육묘법에 관한 연구)

  • This study was carried out to develop a short term hardening method of tree seedlings of Rhus chinensis Mill., Evodia daniellii Hemsley and Parthenocissus tricuspidata(Sieb. et Zucc.) Planck for afforestation on a concave and a crack of cut-rock slope. The seedlings were grown in a cylinder shaped pot made of polyvinyl net with the soil media of peatmoss, vermiculite, clay, compost, fertilizer, and absorbant(40:25:19:15:1:0.1, v:v). They were cultivated in a greenhouse for four months and in field condition for two months. During the last three months of the growing period the seedlings were hardened by periodic desiccation and irrigation in 4 to 10 days interval. The hardened seedlings showed lower leaf water potential, higher leaf osmotic pressure, and lower T/R ratio than those before the hardening. The hardened seedlings survived well on the soil medium in the concave of cut-rock slope.

Transformation of an Alkalin Protease Overproducer, Vibrio metschnikovii Strain RH530, and Improvement of Plasmid Stability by the par Locus

  • Vibrio metschnikovii strain RH530 is a non-pathogenic, industrially-important alkaline protease producer which has been isolated from wastewater. In this paper, we report on the transformation of this strain by using the method of electroporation. A field strength of $7.5\;kVcm^{-1}$ and $25\;{\mu}F$, and using a 0.2-cm cuvette, appeared to be the optimal conditions for electroporation of the cells with the recombinant pSBCm plasmid carrying the vapK alkaline protease gene and the ColE1 replicon. Cells were subjected to osmotic shock in order to remove extracelluar DNase, and adding 200 mM of sucrose to electroporation buffer cells showed an increased transformation efficiency. Maximum efficiency of transformation was obtained at an early exponential growth phase. Using all of the conditions mentioned above, we routinely obtained a transformation efficiency of more than $10^4{({\mu}g\;plasmid\;DNA)}^{-1}$. The stability of the plasmid pSBCm in V. metschnikovii RH530 was 25% after 18h of growth (27 generations) in the medium without antibiotic selection. The insertion of the par locus to the pSBCm increased the stability of the plasmid up to 42% without selective pressure. The increase in plasmid stability was accompanied by the increase in the productivity of alkaline protease in the recombinant V. metschnikovii strain RH530. Determining optimal conditions for the transformation of the industrially-important, nonpathogenic Vibrio strain, and the improvement of plasmid stability by introducing the par locus into the high copy number plasmid vector, will allow the development of procedures involved in the genetic manipulation of this strain, particularly for its use in the production of industrial enzymes such as alkaline protease.

Effect of Sustained Release Recombinant Bovine Somatotropin on Semen Characteristics and Sexual Behavior in Dairy Young Bulls (소 성장호르몬 투여가 젖소 후보 종모우의 정액 성상 및 성 행동 양식에 미치는 영향)

  • This study was conducted to examine the effect of sustained release recombinant bovine somatotropin(SR-rBST) with different dosage(0, 0.03mg/kg body weight/14 day, 0.06mg/kg body weight/14 day, 0.09mg/kg body weight/14 day) on the economical traits of the semen characteristics and sexual behaviors and blood chemical values with sixteen Holstein dairy young bulls in Dairy Cattle Improvement Center of National Livestock Co-operatives Federation. Sensual testings of collected semens of Holstein young bulls were not different among treatments (p>0.05). Ejaculated semen volumes of control, SR-rBST 0.03mg, 0.06mg and 0.09mg groups were 5.6ml, 5.6ml, 6.0ml and 6.7m, respectively, but the result of SR-rBST 0.09mg group significantly increased semen volume(P<0.05). In sperm concentration, SR-rBST 0.06mg and 0.09mg groups significantly increased the total number of sperms than the other groups(P<0.05). In otherwise, SR-rBST treatments did not affect on pH, osmotic pressure, anti forzen rate, abnormality and motility of collected semen during whole experimental period. Reaction time(RT), sexual aggressiveness(SA) and tactile stimulation(TS) were not different among treatments. The libido scores(LS) of control, 0.03mg, 0.06mg and 0.09mg groups were 68.0, 80.5, 73.9 and 80.8, respectively, and LS were significantly improved by SR-rBST administration(P<0.05). The effect of SR-rBST on scrotal circumference measurement that is important factor to determine the ability of semen production, was not different among treatments. These studies indicates that SR-rBST treatments favourably affect on semen quality and sexual libido in Holstein young bull.
