We investigated the branchial osmoregulatory response of black sea bream Acanthopagrus schlegelii to short-term (3-48 h) exposure to a hyposaline environment (5 psu). Gill $Na^+/K^+$-ATPase (NKA) activity was decreased after 3 h in fish transferred to 5 psu compared to salt water-acclimated (control) fish, but the level of activity returned to that observed in the control fish at 6 h after transfer. NKA activity increased significantly at 24 h after transfer, but it returned to the level observed in the control fish at 48 h after transfer. Immunohistochemical staining revealed that gill NKA was localized to chloride cells. The number of chloride cells tended to change in parallel with NKA activity. Substantial decreases in plasma $Na^+$, $Cl^-$, and osmolality were observed after 12 h of exposure to 5 psu; however, these parameters began to recover to the values detected in the controls at 24 h after transfer. In conclusion, our results suggest that black sea bream are able to adjust their osmoregulatory mechanisms to shift from hypo- to hyperosmoregulation within 6 h of exposure to a hypoosmotic environment.
Growth hormone (GH) is known as one of the main osmoregulators in euryhaline teleosts during seawater (SW) adaptation. Many of the physiological actions of GH are mediated through insulin-like growth factor-I (IGF-I), and the GH/IGF-I axis is associated with osmoregulation of fish during SW acclimation. However, little information is available on the response of fish IGF-II to hyperosmotic stress. Here we present the first cloned IGF-I and IGF-II cDNAs of marine medaka, Oryzias dancena, and an analysis of the molecular characteristics of the genes. The marine medaka IGF-I cDNA is 1,340 bp long with a 257-bp 5' untranslated region (UTR), a 528 bp 3' UTR, and a 555-bp open reading frame (ORF) encoding a propeptide of 184 amino acid (aa) residues. The full-length marine medaka IGF-II cDNA consists of a 639 bp ORF encoding 212 aa, a 109 bp 5' UTR, and a 416 bp 3' UTR. Homology comparison of the deduced aa sequences with other IGF-Is and IGF-IIs showed that these genes in marine medaka shared high structural homology with orthologs from other teleost as well as mammalian species, suggesting high conservation of IGFs throughout vertebrates. The IGF-I mRNA level increased following transfer of marine medaka from freshwater (FW) to SW, and the expression level was higher than that of the control group, which was maintained in FW. This significantly elevated IGF-I level was maintained throughout the experiment (14 days), suggesting that in marine medaka, IGF-I is deeply involved in the adaptation to abrupt salinity change. In contrast to IGF-I, the increased level of marine medaka IGF-II mRNA was only maintained for a short period, and quickly returned a level similar to that of the control group, suggesting that marine medaka IGF-II might be a gene that responds to acute stress or one that produces a supplemental protein to assist with the osmoregulatory function of IGF-I during an early phase of salinity change.
Rice is one of the world's staple crops and is a major source of carbohydrate. Rice is exported from several countries, providing a major source of income. There are many documents reporting that rice is a salt-sensitive crop in its developmental stages. The objective of this investigation is to evaluate the effective salt-tolerance defense mechanisms in aromatic rice varieties. Pathumthani 1(PT1), Jasmine(KDML105), and Homjan(HJ) aromatic rice varieties were chosen as plant materials. Rice seedlings photoautotrophically grown in-vitro were treated with 0, 85, 171, 256, 342, and 427 mM NaCl in the media. Data, including sodium ion$(Na^+)$ and potassium ion$(K^+)$ accumulation, osmolarity, chlorophyll pigment concentration, and the fresh and dry weights of seedlings were collected after salt-treatment for 5 days. $Na^+$ in salt-stressed seedlings gradually accumulated, while $K^+$ decreased, especially in the 342-427 mM NaCl salt treatments. The $Na^+$ accumulation in both salt-stressed root and leaf tissues was positively related to osmolarity, leading to chlorophyll degradation. In the case of the different rice varieties, the results showed that the HJ variety was identified as being salt-tolerant, maintaining root and shoot osmolarities as well as pigment stabilization when exposed to salt stress or $Na^+$ enrichment in the cells. On the other hand, PT1 and KDML105 varieties were classified as salt-sensitive, determined by chlorophyll degradation using Hierarchical cluster analysis. In conclusion, the HJ-salt tolerant variety should be further utilized as a parental line or genetic resource in breeding programs because of the osmoregulation defensive response to salt-stress.
Aureobasidium pullulans produced high concentration of polyols extracellularly in the media of sucrose, glucose and mannose as sole carbon source. Mannitol was the main polyol produced during the late exponential and stationary phases of growth together with small quantities of glycerol. Sucrose and glucose were rather rapidly metabolized to mannitol among carbon sources examined where the initial glucose concentration showed no difference in the amount of mannitol. In contrast 20%(w/v) of sucrose was the most appropriate concentration tested. However, the yield of mannitol based on substrate used($Y_{p/s}$) was independent on the initial concentration, and the mean value of mannitol yield in 10% glucose and sucrose media was 0.144 and 0.188, respectively. Mannitol production was reduced in response to an elevated water stress imposed by salts within the range from 0.25 to IM of NaCl or KCl as stress solutes. However, glycerol contents and its ratio to mannitol were increased at the conditions of high salinity. Based on the results, extracellular mannitol produced by A. pullulans probably resulted partly from osmoregulation(in case of glycerol) and mainly from, as known to occur in most of fungi, enzymatic reduction of the corresponding hexoses through phosphate pathway.
This study was carried out to establish a proper cultivation site and diagnose the drought-tolerance of Aster scaber and Synurus deltoides leaves by using Pressure-volume curves. In order to measure pressure-volume (P-V) curves, Aster scaber and Synurus deltoides were cut off above ground part and the tip of the cutting were placed in water, which was covered with a plastic bag. Samples were kept overnight (about 12 hours) in darkness at room temperature (20~25$^{\circ}C$) to achieve maximal turgor (full saturation). The pressure in the chamber was gradually increased from 0.3MPa to 1.8MPa by nitrogen gas. After measured, leaf samples were dried at 80$^{\circ}C$ for 48 hours and dry weight of each samples were determined. The result of the original bulk osmotic potential at maximum turgor ${\Psi}^{sat}_o$ sat was lower -0.8 MPa in Aster scaber leaves than -0.7 MPa Synurus deltoides leaves. Also the osmotic potential at incipient plasmolysis ${\Psi}^{tlp}_o$ in Aster scaber leave was -0.9 MPa. In contrast, the value of maximum bulk modulus of elasticity $E_{max}$ of Aster scaber leaves were approximately two folds higher than that of Synurus deltoides leaves. The values of the relative water content at incipient plasmolysis $RWC^{tlp}$ are all above 90% showing that the function of osmoregulation is somewhat better, and Vo/DW, Vt/DW, Ns/DW of Synurus deltoides leaves were approximately 1~2 times higher than that of Aster scaber leaves. Thus, responses to water relations of Aster scaber and Synurus deltoides such as ${\Psi}^{sat}_o$, ${\Psi}^{tlp}_o$, $E_{max}$, ${\Psi}_{P,max}$, $RWC^{tl}$ were shown that the Aster scaber leaves was slightly higher drought-tolerance than Synurus deltoides leaves. However, in both of Aster scaber and Synurus deltoides, occurring incipient plasmolysis at the high water content, have a relatively lower drought-tolerance property indicating that growth of these plants are cultivated appropriate in high moisture soil sites.
This study was conducted to investigated effects of water relation of mulching and trickle irrigation on the external and internal fruit quality in Satsuma mandarin grafted on trifoliate orange rootstock in a orchard assigned to randomly three groups; whole period of Tyvex mulching (TM), Tyvex mulching with trickle irrigation once a week from October 22 to harvesting season (WM) and non-mulching treatment (NM). The average soil moisture content in the TM was lower than the WM during the time of trickle irrigation from Oct. 21 to Nov. 28. The leaf water potential was at the level of ${\Psi}max$ of -1.5 to -2.5 MPa during whole period of Tyvex mulching treatment but gradually increased at the point of supplement of water. The water and osmotic potential in juice vesicle was decreased by drought but increased again in response to the supply of water in WM. The total soluble solids (TSS) in fruit juice was increased by drought stress, but diminished in response to supply of water after drought. The content of titratible acidity was increased by drought stress but gradually decreased due to supplement of water after drought, reached it at the level of 1%. It was suggested that the accumulation of the total soluble solids compensates the degree of active osmoregulation and the decrease in content of acidity accounts for the fast respiration and water uptake resulted of the water after drought.
This study was conducted to compare the physiological conditions of black porgy (Acanthopagrus schlegeli) when reared in freshwater (0 psu) and seawater (35 psu) during 90 days. In terms of stress response, there was no significant difference in cortisol levels of the fish reared either freshwater or seawater until 60 days. Although cortisol level of fish reared in freshwater(12.6$\pm$5.0 ng/ml) was significantly higher than in seawater (4.5$\pm$2.9 ng/ml) at the end of experiment, these values were stable levels as compared with that of non-stressed fish. No significant differences in plasma osmolalities were recognized between the fish reared in freshwater (346.7$\pm$4.6$\~$356.5$\pm$2.1 mOsm/kg) and seawater (350.0$\pm$2.0$\~$357.0$\pm$22.6 mOsm/kg). Normal structure of gill lamellae without histological damage or cell necrosis has been observed in the fish reared in freshwater. In connection with growth and survival rate, total length and body weight of fish reared in freshwater were slightly longer and higher than those of fish reared in seawater. Survival rate of black porgy reared in freshwater was slightly higher than that of fish reared in seawater, but there was no significant difference. The results suggest that black porgy is reared with normal growth rate in freshwater without stress.

This study was conducted to determine the effect of feeding betaine on egg production, lipid metabolism and osmoregulation in laying hens. Three hundred and sixty layers were fed corn-soy basal diet containing 16% CP, 2800 kcal/kg ME from 18 to weeks. Hens were provided with betaine 0, 300, 500 or 1200 ppm. Egg production, feed intake, egg weight and feed conversion were examined in every four weeks. Liver and egg betaine. serum cholesterol and neutal fat were measured at the mid experiment. Egg production was not statistically different among treatments from 18 to 26 weeks of age, but significantly increased for the rest of period in hens fed more than 600ppm betaine(P<0.05). Betaine added treatments showed a tendency to increase feed intake. Feed conversion tended to improve from 26 to 33 weeks old in betaine supplemental groups. Egg qualify was not influenced by dietary supplemental betaine. Betaine supplementation seemed to increase in liver betaine. but not in egg betaine. Serum total cholesterol, triacylglycerol, abdominal fat content and ileal osmolality were not different. However, liver fat content was prone to decrease in betaine supplemental groups. The result of this experiment suggested that dietary supplemtal betaine can improve the egg production and decrease liver fat content.

Taurine (2-ethaneaminosulfonic acid) is one of the major intracellular ${\beta}$ -amino acids in mammals and is required for a number of biological processes including membrane stabilization, osmoregulation, antioxidation, detoxification, modulation of calcium flux and neurornodulation. The taurine transporter (TAUT) which contains 12 hydrophobic membrane-spanning domains has been cloned from dog kidney, rat brain, mouse brain, human thyroid, placenta and retina. In this study, The TAUT cDNA from the human intestinal epithelial cell, HT-29 was cloned and sequenced. Reverse-transcription polymerase chain reaction (RT-PCR) was performed to amplify partial cDNA encoding human intestinal TAUT. The coding region of the PCR product was 732 bp long. The primers were designed to encode highly conserved amino acid sequences near the transmembrane domains III (IPYFIFLF) and Ⅵ (KYKYNSYR) both in human and mouse. The TAUT cDNA amplified was ligated into the pGEX 4T-1 expression vector. The resulting sequence of human intestinal TAUT cDNA (Accession number of NCBI Genebank is AF346763) was identical to the sequences of the TAUTs previously determined in the human placenta and retina except 3 base pairs from that of the reported human thyroid. TAUT specific antibodies were generated to use them as biological tools in the studies of the biological role of TAUT. Peptides of 149-162 amino acid residue (14 amino acids) of the TAUT were synthesized. The synthetic peptide used in this study was LFQSFQKELPWAHC. This region was chosen not only to avoid putative glycosylation sites but also to exclude regions of known homology with GABA transporters in the extracellular hydrophilic domains. The synthetic peptide, TAUT-1 was conjugated with carrier protein, kehole lympet hemocyanin (KLH) to use as an antigen. When used for immunization on a rabbit to produce polyclonal antiserum, the conjugates elicited high -titered specific anti-TAUT-1 antibodies, which reacted well with the ovalbumin (OVA) conjugated peptides in ELISA. The KLH-conjugated peptide was also used as immunizing antigen in BALB/c mice to produce TAUT specific monoclonal antibodies. From the culture supernatant of the hybridoma, the specificity of anti-TAUT-1 monoclonal antibodies was confirmed by ELISA. Further applications of more tools in TAUT expression analysis will be performed such as western blotting and flow cytometry.
Two species of halophilic bacteria were isolated from five salted seafoods and identified by 16S rDNA sequencing homology. One was identified as Halomonas subglaciescola and other four strains were belong to Halomonas marina. The identity of all isolates with standard organisms was above $95\%.$ H. subglaciescola, H. marina IN, and H. marina SH-2 grew in salinity condition from $3%\;to\;18\%$ NaCl but growth of H. marina SQ and H. marina SH-l grew in salinity environment from $8\%\;to\;17\%.$ Maximum biomass of H. subglaciescola, H. marina IN, H. marina SQ, H. marina SH-1, and H. marina SH-2 growing in LB medium containing $15\%$ NaCl were about 3.2, 4.5, 4.5, 5.7, and 4.2, however the maximum biomass in LB medium containing $5\%$ NaCl were about 2.2, 1.1, 0.7, 0.2, and 2.4 as optical density at 660 nm, respectively. In scanning electron micrograph, unknown material (mucus) attached to outer membrane of all isolates was observed. When mucus isolated from halophilic bacterial cell was added to culture of E. coli, E. coli grew in medium containing $15\%$ NaCl.
