환경 독성 유기인 화합물 분해를 위하여 재조합 대장균에서 세포내 간극으로 분비된 Organophosphorus Hydrolase의 생산 (Production of Periplasmic Space-Secreted Organophosphorus Hydrolase from Recombinant Escherichia coli for Degradation of Environmental Toxic Organophosphate Compounds)

  • 본 연구에서는 유기인 화합물인 Paraoxon의 분해를 위하여 재조합 대장균에서 세포내 간극(periplasmic space)으로 분비되는 organophosphorus hydrolase(OPH)의 생산에 대하여 고찰하였다. OPH 생산의 향상을 위하여 성장 배지에 첨가되는 최적의 조건은 1.0 mM isopropyl-${\beta}$-D-thiogalactopytanoside (IPTG), 0.25 mM $Co^{2+}$ 및 0.1 mM ethylenediamine tetraacetate (EDTA) 이었다. 이 조건에서 최대OPH 생산은 $174Unit/L{\cdot}OD$를 나타내었다. 또한 1 mM의 Paraoxon은 OPH에 의하여 완전히 분해되었다. 이러한 연구 결과는 토양 및 수계에 잔류하는 환경독성 유기인 화합물을 제거하는 bioremediation의 수수단으로 활용될 수 있음을 보여주었다.

아산만과 경기만의 유기인계 잔류농약 분포 (Distribution of organophosphorus pesticides in Asan and Kyeonggi Bay, Korea)

  • 1999년 하계에 아산만과 경기만 해역의 유기인계 잔류농약 분포를 조사하였다. 표층해수 중 IBP는 모든 해역에서 광범위하게 분포하였다 아산만의 표층해수 중 IBP 농도 범위는 7월과 9월에 각각 10.3-1030.1(m=339.1), ND-319.1(m=74.8)ng/L였고 경기만 표층해수 중 IBP 농도는 7월과 9월에 각각 ND-76.2(m=29.1), ND-270.3(m=46.6)ng/L였다. Diazinon, DDVP, Ethoprophos와 Chlorpyrifos는 다소 낮은 농도를 보였지만 모든 해역에서 검출되었다. 다른 유기인계 농약(Disulfoton, rarathion Methyl, Fenchlorfos, Prothiofos, EDDP)은 일반적으로 검출한계 미만이었다. 퇴적물 중에서는 Chlorpyrifos가 다근 유기인계 농약보다 널리 분포하였다. 유기인계 농약의 시공간적 분포는 자 지역에 사용되어지는 농약의 사용량과 종류에 따라 달라진다고 보여진다.

  • Organophosphorus pesticides (OPPs) contents in the sewage sludge derived from the residential and industrial areas were determined in order to characterize contemporary organic contamination levels as a part of the ongoing development of the environmentally sound sewage sludge management strategy in Korea. OPPs were extracted from freeze-dried sludges in a ultrasonic extractor. The extracts were cleaned-up by florisil column and subsequently fed into gas chromatograph/nitrogen phosphorus detector (GC/NPD) for determining OPP contents. Diazinon, dimethoate, disulfoton, EPN, malathion, methyl parathion, parathion, phorate and sulfotep were present in the appreciable amount in the domestic sewage sludges. 제주도 연안 해양환경 중 유기린계 농약의 잔류 (Residue of Organophosphorus Pesticides in the Coastal Environment on the Cheju Island)

  • To study the residue of organophosphorus pesticides in the ocean environment on the Cheju island, EPN[O-ethyl-O-4-nitrophenyl phenylphosphonothioate] and monocrotophos [Dimethyl-1-methyl-2-methyl carba-moylvinylphosphate] of organophosphorus pesticides are surveyed on coastal environment of Cheju city and Wimi in May and October 1996. The qualified limit detection of EPN and monocrotophos are 0.005ng/mL and 0.006ng/mL in the water by GC-FPD, respectively. The qualified limit detection of EPN and monocrotophos are 0.010ng/g and 0.012ng/g in the solid such as sediment and sea organism by GC-FPD, respectively. EPN and monocrotophos are not detected in seawater and sedimnet. Moreover EPN and monocrotophos are neither detected in seaweed cava (Ecklonia cava), Agar (Gelidium amansii), turban sell(Batillus cornutus) and sea urchin(Anthocidaris Crassispina). EPN, monocrotophos used in the farm on the Cheju island are not residued in the coastal environment in the Cheju island.

몇 종류의 물에서 pH 변화에 의한 유기인계 살충제의 분해과정에 관한 고찰 (A Study on the Degradation Process of Organophosphorus Insecticides depending on Variation of pH in Several Waters)

  • This study was to investigate time-dependent degradation process under various pH condition for organophosphorus(org-p) insecticides, namely Demeton-s-methyl, diazinon, Parathion, Phenthoate, and EPN in several waters. They were analysed by GC-FTD according to standard methods for the examination of water and wastewater. In pH=4, diazinon showed disappearance after 14 days in chromatogram. In pH=11, org-p insecticides were almost degradable after 7 days. In this condition, effect of pH on degradation process was greater than of light. In pH=7, org-p insecticides persisted residues after 112 days except Demeton-s-methyl. In BOD5 120, 250 ppm and domestic water, org-p insecticides showed also rapid degradation process.

새만금 해양환경에서 해수 중 유기인계 농약의 시공간적 분포 (Spatial and Temporal Distribution of Organophosphorus Pesticides in Seawater from Saemangeum Area)

  • Organophosphorus pesticides (OPs) were analyzed in surface water samples from the Saemangeum environment. IBP was the most abundant among 10 OPs analyzed, and accounted fur over 95% of total amount of OPs. Concentrations of IBP measured in Mangyeong River water and surface seawater ranged from 670 to 1100ng/l and from 45 to 1000 ng/l, respectively. In general, a decreasing trend of OP concentrations towards open sea was observed. The concentrations of IBP and salinity demonstrated a strong correlation $(r^2=0.96)$. This indicates that IBP showed conservative behavior in the study area, and seawater dilution is a major factor affecting distribution and variation of OPs in the Sammgem environment.

Development of an ELISA for the Organophosphorus Insecticide Chlorpyrifos

  • A selective enzyme-linked immunosorbent assay (ELISA) for the insecticide chlorpyrifos was developed. Four haptens for chlorpyrifos were synthesized and two of them were used as immunogens after coupling to keyhole limpet hemocyanin by two differe nt approaches. Rabbits were immunized with either of them and the sera were screened against 4 haptens coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigencoated ELISA was developed, which shows an I50 of 160 ppb with a detection limit of 10 ppb. An antibody-coated ELISA was also developed, which shows an $I_{50}$ of 20 ppb with a detection limit of 0.1 ppb. The antibodies showed negligible cross-reactivity with other organophosphorus pesticides except for insecticides chlorpyrifos-methyl and bromophos-ethyl, which makes these assays suitable for the selective detection of chlorpyrifos.

A Dipstick-Type Electrochemical Immunosensor for The Detection of The Organophosphorus Insecticide Fenthion

  • A dipstick-type immunochemical biosensor for the detection of the organophosphorus insecticide fenthion was developed using a screen-printed electrode system as an amperometric transducer with polyclonal antibodies against fenthion as a bioreceptor. The assay of the biosensor involved competition between the pesticide in the sample and pesticide-glucose oxidase conjugate for binding to the antibody immobilized on the membrane. This was followed by measurement of the activity of the bound enzyme by the supply of the enzyme substrate (glucose) and amperometric determination of the enzyme reaction product ($H_2O_2$). The activity of the bound enzyme was inversely proportional to the concentration of pesticide. The optimized sensor system showed a linear response against the logarithm of the pesticide concentration ranging from $10^{-2}$ to $10^3\;{\mu}g/L$.

  • A competitive enzyme-linked immunosorbent assay (ELISA) was developed for the quantitative detection of organophosphorus insecticide cyanophos. An analogue (hapten) of cyanophos was synthesized and was coupled to BSA to produce polyclonal antibodi es from rabbits. The antisera were screened against another hapten coupled to ovalbumin (OVA). Using the sera of highest specificity, an antigen-coated ELISA was developed, which showed an I50 of 310 ng/mL with the detection limit of 20 ng/mL. The antibodies showed negligible cross-reactivities with other organophosphorus pesticides except for parathion-methyl, which makes the assay suitable for the selective detection of cyanophos.

  • Monoclonal antibodies have been generated against a generic hapten, ο,ο-diethyl ο-(5-carboxy-2-fluorophenyl) phosphorothioate, for the determination of organophosphorus (OP) pesticides in a class-specific manner. In an indirect competitive enzyme-linked immunosorbent assay (ELISA) format, employing a heterologous coating antigen, these monoclonal antibodies showed desirable properties for use in the class-specific determination, i.e., broad specificity and high sensitivity. The IC50 values of four commonly used ο,ο-diethyl OP pesticides were fairly uniform ranging from 0.1 to 0.3 ㎛/mL. The IC50 values of three ο,ο-dimethyl derivatives were between 0.3 and 1.4 ㎛/mL. These values, together with the limits of detection (LOD), were better, in terms of the specificity and sensitivity, compared with the values obtained previously with polyclonal antibodies.
