Copper(II) Binding Mechanisms with Water Soluble Organic Fractions Extracted from Sewage Sludge Amended Soils (구리(II) 이온과 Sewage Sludge를 시용(施用)한 토양(土壤)에서 추출(抽出)한 수용성유기물(水溶性有機物)과의 착화합물(錯化合物) 형성방법(形成方法))

  • Cu(II) binding mechanisms with water soluble organic fractions (WSOF) extracted from an agricultural soil (W), a soil treated with sludge for 6 years ($WS_6$), a sludge-soil mixture incubated for one week ($WS_1$), and sewage sludge (SS) were studied by electron spin resonance (ESR) spectroscopy and potentiometric titrations. Cu(II)-WSOF complexes produced $g_{11}$ values which were larger than $g_{\perp}$ values, indicating that the coordination of Cu(II) complex was an elongated octahedron. At liquid $N_2$ temperature (77K), the Cu(II)-W complex showed an anisotropic ESR spectrum while the Cu(II)-SS complex showed an isotropic spectrum. These spectral results suggest that the oxygen donor ligands of W may form relatively strong bonds with $Cu^{2+}$ due to extensive chelation while ligands of SS may form little or no chelate bonds with $Cu^{2+}$. The ESR spectra of Cu(II)-SS complex also suggest that each of four in-plane ligands (e.g., $COO^-$, $H_2O$, $Cl^-$, etc.) may act independently as monodentate ligands. Oxygen donor ligands such as aromatic carboxyl groups were probably the major Cu(II) binding sites in W. Sulfonate, aliphatic carboxyl group, and N-containing ligands were probably the major binding sites in SS at pH 5. The Cu(II) complexation with N-containing groups increased as sludge was added to the soil. Much higher (6x) pyridine concentrations were required to displace W from Cu(II)-W complex as compared to the Cu(II)-SS complex.

Influence of Solution pH on Pyrene Binding to Sorption-Fractionated and Kaolinite-Bound Humic Substance

  • Changes in pyrene binding by dissolved and kaolinite-associated humic substances (HS) due to HS adsorptive fractionation processes were examined using purified Aldrich humic acid (PAHA) at different pH (4, 7 and 9). Irrespective of solution pH, molecular weight (MW) fractionation occurred upon adsorption of PAHA onto kaolinite, resulting in the deviation of residual PAHA MW from the original MW prior to sorption. Variation in $K_{OC}$ by bulk PAHA was observed at different pH due to relative contributions of partitioning and size exclusion effects (i.e., specific interactions). For all pH conditions investigated, carbon-normalized pyrene binding coefficients for nonadsorbed, residual fractions $(K_{OC}(res))$ were different from the original dissolved PAHA $K_{OC}$ value $(K_{OC}(orig))$ prior to contact with the kaolinite suspensions. Positive correlations between pyrene $(K_{OC}(res))$ and weight-average molecular weight $(MW_W)$ for residual PAHA fractions were observed for pH 7 and 9. However, such a positive correlation was not found at pH 4 due to the absence of the dramatic fractionation observed for high pH conditions (i.e., exclusive fractionation with respect to higher MW), suggesting that actual MW distribution pattern is more important for sorption-fractionated HS than the composite MW value. For adsorbed PAHA, conformational changes of PAHA upon adsorption seem to be important for the extent of pyrene binding. At relatively high pH (7 and 9), lower extent of pyrene binding was observed for adsorbed PAHA versus nonadsorbed PAHA. The conformation effects were more pronounced at higher pH.

Functional Properties of Hydrolysates Prepared from Agar Treated with Organic Acids (한천 유기산 가수분해물의 생리활성)

  • This study was concerning various physiological activities of agar hydrolysates. All agar hydrolysates showed strong antimicrobial activity against Bacillus subtilis and Bacillus cereus. Also, the agar hydrolysates prepared at the temperature of $110^{\circ}C$ or $120^{\circ}C$ showed antimicrobial activity against St. aureus and E. coli. Among the agar hydrolysates, several hydrolysates treated with citrate or malate at $110^{\circ}C$ or $120^{\circ}C$ conditions showed tyrosinase activity inhibition, and their inhibition rates of tyrosinase activity were about 80%. Some tested samples treated with 0.5% organic acid at $100^{\circ}C$ or $110^{\circ}C$ inhibited the growth of cancer cell. Two agar hydrolysates prepared with 0.5% citrate and lactate at $110^{\circ}C$ for $180^{\circ}C$ min had relatively high cancer cell growth inhibition among the tested samples. The agar hydrolysates treated with citrate and lactate at $110^{\circ}C$ for 180 min obtained the main peaks of six and seven from Sephadex G-15 column chromatography. Among the main peaks, the cancer cell growth inhibition of C-3 and L-3 fractions were higher than that of other fractions.

On the Identification of Strains and the Cultural Characteristics of Flammulina velutipes in Korea (한국산 팽나무 버섯의 계통류별과 그 배양적 특성에 관한 연구)

  • In order to investigate morphological and cultural characteristics of strains in species, Flammulina velutipes, the author collected isolates of Flammultina velutipes at 49 locations in Korea and cultivated these isolates on the various kinds of solid media. After investigating the cultural characteristics, appeared on the various media, he obtained the following results : 1. The variation of colors in the fruit bodies is connected with the variation of climatic environments(composite effects between mean temperature in January and number of rain days of 1mm and over precipitation). The author, therfore, can find out the trend that brown type is distributed in the midland climatic region and yellow type in the southern climatic regoin. 2. Two types can be classified into several strains respectively : the strain of abundant or insufficient productivity, and strain of selectivity or non-selectivity of media. 3. According to the results of mutual comparison of soluble mycelial proteins by disc electrophoresis using polyacrylamide gels, each type has special common protein fractions(brown type : band located at 26..5mm position from surface of gel, yellow type : band located at 24.5mm position from surface of gel), and each strain has special protein fractions too. Therefore this phenomenon seems to support the results obtained by the above-stated morphological and cultural studies. 4. In the adaptability of strains to the temperature, every strain has the nature of growing in lower temperature(the optimum temperature of 20.deg.C to 25.deg.C) except that YI-1 strain has the optimum temperatue of $25^{\circ}C$-26^{\circ}C. And mycelial growth of every strain is discontinued at $35^{\circ}C.$ 5. In the adaptability of strains to the H-ion concentration, every strain has wide adaptable range of H-ion concentration, and has optimum range of pH 5.5 to 6.6 in mycelial growth excepting YA01, BI-2 and YI-1. 6. In the utility of carbon sources, the mycelial growth of every strain is very poor on the media containing xylose(average diameter of mycelial growth : 18mm), and most strains utilize favorably sucrose(39mm), maltose(37mm) and dextrose(35mm) in mycelial growth. In the utility of nitrogen sources, every strain utilizes favorably organic nitrogens(36 mm)more than inorganic nitrogens (25 mm), and utilizes fully peptone nad asparagine in organic nitrogens. Especially BA-1, BIK-2 and YA-1 strains grow vigorously on each media containing various carbon and nitrogen sources. 7. The characteristic tests of the number of days required for mycelial growth, the number of days requried for sprout of young bodies, the length of stipe and the number of fruit bodies formation seem to be useful methods in the early selection of the strain of the abundant productivity.

Suppressive Effects of Lees from Sweet Potato Soju on LPS-induced Inflammatory Responses in RAW 264.7 Cells (고구마 소주 주박에 의한 RAW 264.7 세포주에서 lipopolysaccharide로 유도된 염증 반응의 억제 효과)

  • In the current study, the ethanol extracts and their subsequent organic solvent fractions from lees of sweet potato soju were prepared and the prepared samples were designated as from KSD-E8-1 to KSD-E8-5. Their effects on cell viability and nitric oxide (NO) production in mouse macrophage RAW 264.7 cells were investigated. The results showed that the ethyl acetate fraction (KSD-E8-3) of lees extracts from sweet potato soju significantly decreased nitric oxide (NO) production in LPS-activated RAW 264.7 cells, whereas they did not affect cell viabilities. The fraction KSD-E8-3 reduced the expression of pro-inflammatory genes such as COX-2, iNOS and TNF-alpha and also decreased protein expression of iNOS in a dose dependent manner, which were detected with RT-PCR and Western blot analysis, respectively. In addition, we detected the expression of mitogen-activated protein kinases (MAPKs) such as p38, JNK, and ERK1/2 and their phosphorylated forms. The results indicated that the treatment of the fraction KSD-E8-3 did inhibit phosphorylation of p38, JNK, and ERK1/2 MAPKs, indicating that the fraction KSD-E8-3 regulates LPS-induced inflammatory response via suppressing MAPK signaling pathway. Overall, these results may contribute to understand the molecular mechanism of anti-inflammatory effects by the ethyl acetate fraction of lees extracts from sweet potato soju.

Free Radical Scavenging Effect of Seed and Skin Extracts of Black Olympia Grape (Vitis labruscana L.) (거봉 포도 종자 및 과피 추출물의 항산화 활성)

  • Optimal extraction condition and free radical scavenging effect of ethanol extracts of Olympia grape (Vitis labruscana L.) seeds and skins were determined. Extraction yield of grape seeds increased with increasing extraction temperature increased, but was not affected by ethanol concentration in grape skins. Highest extraction yields obtained with 100% ethanol concentration at $78^{\circ}C$ were 36 and 49.5% in seeds and skins, respectively. Ethanol extract of grape seeds obtained at $78^{\circ}C$ showed stronger $RC_{50}\;(409.7\;{\mu}g/mL)$ than that of grape skins. Among organic solvent fractions, ethylacetate fraction obtained from ethanol extract of grape seed showed the strongest $RC_{50}\;(136.7\;{\mu}g/mL)$. Overall, ethanol extracts of grape seeds and their organic fractions showed significantly stronger free radical scavenging effect than those of skin extracts.

Anti-adipogenic, Anti-inflammatory, and Anti-proliferative Activities of Extracts from Lees and Nuruk (주박과 누룩의 추출물에 의한 지방세포형성억제, 항염증 및 항성장 활성)

  • This study examined extracts from five different kinds of lees and nuruk and their organic solvent fractions in terms of several biological functions, such as anti-adipogenic, anti-inflammatory, and anti-proliferative activities. The anti-adipogenic activity was investigated by treating mouse pre-adipocyte 3T3-L1 cells with one extract (YE) and four organic solvent fractions (YAc, PAc, RAc, and WPAc) during adipogenesis. Among the treated samples, the ethyl acetate fraction of W-Ju lees (WPAc) showed the strongest anti-adipogenic effect, which was confirmed with oil red O staining and down-regulation of pro-adipogenic genes such as PPAR-gamma and SCD-1. Treatment with WPAc also reduced the expression of PPAR-gamma in a time-dependent manner. The effects of five different extracts were examined on nitric oxide (NO) production in mouse RAW 264.7 cells to determine anti-inflammatory activity. The ethyl acetate fraction of B-Ju lees (PAc) significantly decreased NO production in LPS-stimulated RAW 264.7 cells and it also inhibited NO production in a dose-dependent manner. The PAc fraction also dramatically decreased the viability of human colorectal cancer HCT116 cells in a dose-dependent manner. In addition, PAc increased the expression of NAG-1 and ATF3 genes in a dose dependent manner. Overall, these results indicate that lees and nuruk have several biological functions, including anti-adipogenic, anti-inflammatory, and anti-proliferative activities.

Fractionation of DOC and its Correlation to AOX(FP) in the Advanced ater Treatment Process (고도정수처리 공정에서 DOC 분획 특성 및 AOX(FP)와의 관계)

  • As a part of dissolved organic matter, dissolved organic carbon (DOC) or biodegradable DOC (BDOC) fraction in particular is one of important issues in water treatment. Due to role as a nutrient source for bacteria, BDOC, therefore, may cause regrowth problems in water distribution system. The main objectives of this study were to investigate the possibility to minimize the concentration of BDOC in advance water treatment process. DOC in water is fractionized into four fractions such as AnBDOC (adsorbable and non-biodegradable DOC) which possesses adsorption properties but no biodegradation ability; nABDOC (biodegradable and non-adsorbable DOC) which has biodegradation properties but no adsorption ability; ABDOC (adsorbable and biodegradable DOC) which has adsorption properties and biodegradable characteristic; and non-removal DOC (nAnBDOC) which do not have either adsorbability or biodegradability. BAC process was effective for adsorbable DOC (AnBDOC+ABDOC) removal. However, in some cases, the removal ratio of adsorbable DOC was not sufficient. BDOC removal rate is very low or irremovable. Thus, for the control of residual DOC, it is necessary to change the operation condition by BAC process. From the analysis results of DOC fractions, water treatment processes appeared to be effective because it could grasp a remarkable amount of biodegradable, adsorbable and non-removal DOC. The concentration of AOX in non-prechlorination process was reduced from 7.1 ${\mu}g$/L to 0.51 ${\mu}g$/L in BAC process followed by ozonation.

Vertical Flow Zeolite-Filled Reed Bed with Intermittent Feeding for Sewage Treatment (수직 흐름 제올라이트 갈대 여과상에 의한 생활하수 처리)

  • A sewage was treated using a vertical flow zeolite-filled reed bed. The sewage from the student dormitory of Changwon National University was fed into the reed bed for 10 minutes every 6 hours at the hydraulic load of $314L/m^3{\cdot}$day. The filtering height of the reed bed was 100 em and the zeolite mixture was filled in the reed bed. The mixture consisted of the same volume of two types of zeolite: 0.5$\sim$1 mm and 1$\sim$3 mm in diameter. Annual average removal efficiency was 88 89.9%, $COD_{Cr}$ 86.1 %, $COD_{Mn}$ 81.2%, T-N 34.0%, $NH_4^+$-N 97.3% and T-P 34.6%. T-N of effluent was mostly $NO_3^-$-N and the concentration of $NO_2^-$-N in effluent was lower than 0.1 mg/L. All removal efficiencies did not show a remarkable seasonal change. The ranking of phosphorous fractions fixed to the zeolite in column test was Ca-P > Fe-P > reductant soluble Fe-P > occluded P > saloid P > AI-P at all depths of the filter. All phosphorous fractions except for AI-P reduced at deeper filter layer, while their content ratios increased at deeper filter layer. Organic matter content was the highest at the highest layer (0$\sim$5 cm from the top of the filter) and only small differences were observed at the deeper filter layer than 5 em from the top. Organic matter content increased at all depths of the filter with the operating time.

Ultimate Anaerobic Biodegradability and Multiple Decay Rate Coefficients of Organic Wastes (유기성 폐자원의 최종생분해도 및 다중 분해속도 평가)

  • Anaerobic mesophilic batch test of several organic wastes were carried out by a graphical statistic analysis (GSA) to evaluate their ultimate biodegradability and two distinctive decay rates ($k_1$ and $k_2$) with their corresponding degradable substrate fractions ($S_1$ and $S_2$). Each 3 L batch reactor was operated for more than 100 days at the substrate to inoculum ratio (S/I) of 0.5 as an initial total volatile solids (TVS) mass basis. Their Ultimate biodegradabilities were obtained respectively as follow; 69% swine waste, 45% dairy cow manure, 66% slaughterhouse waste, 79% food waste, 87% food waste leachate, 68% primary sludge and 39% waste activated sludge. The readily biodegradable fraction of 89% ($S_1$) of Swine Waste BVS ($S_o$) degraded with in the initial 31 days with $k_1$ of $0.116day^{-1}$, where as the rest 11% slowly biodegradable fraction ($S_2$) of BVS degraded for more than 100 days with the long term batch reaction rates ($k_2$) of $0.004day^{-1}$. For the Food Waste and Waste Activated Sludge, their readily biodegradable portions ($S_1$) appeared 89% and 80%, which degrades with $k_1$ of $0.195day^{-1}$ and $0.054day^{-1}$ for an initial 15 days and 28 days, respectively. Their corresponding long term batch reaction rates ($k_2$) were $0.003day^{-1}$ and $0.002day^{-1}$. Results from other organic wastes are addressed in this paper. The theoretical hydraulic retention times (HRTs) of anaerobic digesters treating organic wastes are easily determined by the analysis of multiple decay rate coefficients ($k_1$ and $k_2$) and their corresponding biodegradable substrate fractions ($S_1$ and $S_2$).
