Inhibitory Substance on the Snake Venoms Produced by Penicillium sp. (사독의 조해물질에 관한 연구)

  • One strain of Penicillium sp. (175-66-B), isolated from soil, was able to produce a substance that has a strong inibition activity against the Agkistrodon and Trimeresurus venoms. In this experiment, the chemical and biological properties of the sample were investigated. As an inhibitory substance, it was effective to the proteinase, hemorrhagic and lethal factors of Agkistrodon and Trimeresurus venoms, and also effective to several fractions of the proteinases and hemorrhagic factors of Agkistrodon halys blomhoffi venom. Moreover, in the addition of prednisotone, it was more effective for the cure of the mouse envenomated with the venom amount of two fold of MLD$_{100}$. This substance was very stable to the acid, alkali and heat. Its melting point was high enough to sublime at 222$^{\circ}C$ without any decomposition. This sample was easily dissolved only in hot water, but not in several organic solvents except for a little dissolution in elate. It did not have the chelating activity. It had very strong specificity to the snake venoms. but its activity was depressed by the addition of zinc or cupric salts. This sample had no acute toxicity to the mouse. Its chemical formula was $C_{16}$ $H_{12}$$N_2$ $O_{10}$ with the molecular weight of about 392. It has two epoxy groups and four carboxyl radicals, but amino, nitrite and nitrate radicals, unsaturated bonds and aromatic ring were not detected. Study on the Mobility of Cd Ion Using Extraction by Solutions in Stem Wood of Pinus densiflora L. (용매추출(溶媒抽出)에 의한 소나무(Pinus densiflora L.) 수간부(樹幹部)에서의 Cd 이동성(移動性)에 관(關한) 연구(硏究))

  • In order to understand the chemical binding forms of Cd in terms of its binding strength in wood, the radial distribution of it through dendroanalysis and its different chemical fractions in trunk wood of mature red pine(Pinus densiflora L.) trees grown in a relatively unpolluted area, in Kangwon-Province, were examined. Total Cd concentration in stem wood was determined by dry digestion and $H_2O$ and 0.01 M solutions of citric acid, malic acid, $CaCl_2$, $MgCl_2$, $Na_2EDTA$ were used as extracting solutions for Cd. Extracting efficiencies of applied solutions were expressed as % of Cd concentration extracted by $Na_2EDTA$ which extracted all Cd instead of Cd total concentration. Total concentration of Cd decreased from the innermost rings near pith towards the the outermost rings near cambium. Especially through the transition zone this tendency was observed clearly. $H_2O$ was the least effective extractant of Cd, then extracting efficiency increased in the order $MgCl_2$ < $CaCl_2$ $Na_2EDTA$ (extracted all Cd). According to the radial position, extracting efficiency of Cd was ranging from 7 to 30% by $H_2O$ and even from 60 to 95% by citric acid. The Cd extracting efficiencies of $H_2O$ and organic acids increased continuously from the stem center to outermost annual rings, whereas salts showed somewhat different tendency, suddenly decreasing near the outermost ring. The results are discussed with regard to the different chemical binding forms of Cd and the mobility of Cd in wood and comparable with ones in previous studies on Pinus sylvestris and Quercus patraea. According to the results, usefulness of radial distribution pattern of Cd in pine tree trunks for retrosective biomonitoring is questioned.

Preparation of Organophilic MMT Modified with Various Aromatic Amines and Characterization of Polyimide Nanocomposite Films (다양한 구조의 방향족 아민으로 개질된 친유기성 MMT의 제조와 이를 이용한 폴리이미드 나노복합필름의 특성)

  • In this work, we have prepared organophilic MMT having thermal stability by ion exchange reaction of various aromatic ammonium salts with MMT containing sodium ion. The organic modifiers having alkyl side chains and amine functional group were successfully synthesized by effectively introducing the surfaces of MMT via ion exchange reaction to form organophilic MMTs with a view to improve the reactivity and thermal stability. The WAXD patterns of organophilic MMT showed the more increased gallery spacing by $3.3{\AA}$ than that of the pristine MMT and also the onset of initial decomposition of organophilic MMT was $275^{\circ}C$ as determined by a thermogravimetric analysis. The polyimide (PI) nanocomposite films based on poly(amic acid) and organophilic MMT were prepared by a solution blending followed by cyclodehydration reaction. We have investigated the dispersity of organophilic MMTs in PI matrix by using WAXD and the effect of the organophilic MMT content on the mechanical properties of PI nanocomposite films was studied.

Isolation and Culture of Protoplasts from Embryonic Cotyledons of Pinus densiflora S. et Z. (소나무 자엽(子葉)을 이용(利用)한 원형질체(原形質體) 나출(裸出) 및 배양(培養))

  • Protoplasts were isolated from cotyledons of germinating Pinus densiflora S. et Z. seeds. The seeds were germinated for nine days, and excised embryonic cotyledons about 3-4mm long were incubated with Cellulase Onozuka R-10 and Macerozyme. After 8 hours of incubation, large number of viable protoplasts were isolated. Isolated protoplasts were cultured in a medium containing basal salts of $B_5$ medium, vitamines, amino acids, organic acid, sugars, and growth hormones. The first evidence of protoplast budding was observed after twelve hours in culture, and it suggested that high potential of the embryonic cotyledons for rapid cell division affected the early budding, rather than effect of culture medium was shown in twelve hours. The three- to four-cell stage was reached after three to four days of culture. Most cell divisions were achieved by additional buddings rather than equal binary cell division. No further cell division was observed beyond the four-cell stage. Protoplasts isolated from fully expanded cotyledons (germinated for 17 to 24 days) seldom initiated or failed to initiate cell division.

Ice Melting Capacity Evaluation of Applicable Materials of De-icing Fluid for High Speed Railway Rolling Stock (고속철도차량용 제빙액으로의 적용가능물질에 대한 융빙성능 평가)

  • In winter season, the snow and ice accretion on the bottom of the high speed railway rolling stock and boogie part has fallen at a high speed from the ballast section (gravel section for the transmission of the rolling stock load received by sleepers and fixing sleepers), causing the gravel to be scattered, thereby damaging the railway rolling stock structures and facilities. In order to solve these problems, the gravel scattering prevention net, manual de-icing, and movable hot air machine were used, but their efficiency was low. For the more efficient de-icing than ever before, an optimum material for de-icing fluid for high speed railway rolling stock was developed by evaluating the ice melting capacity, kinematic viscosity, evaporation of the material used as a chemical de-icing fluid. Four kinds of organic acid salts (sodium formate, sodium acetate, potassium formate and potassium acetate) and two different alcohols (propylene glycol, glycerol) were used as evaluation materials. Potassium formate, potassium acetate, and propylene glycol had similar ice melting capacities in the indoor test, but the propylene glycol showed the best ice melting capacity in spraying the system simulation test. This is because the kinematic viscosity of propylene glycol was 2.989029 St, which is higher than those of other materials therefore, it could stay longer on the ice and de-icing. In addition, potassium formate and potassium acetate were difficult to be used since the crystals precipitated and adversely affected the appearance of the rolling stock. The propylene glycol is the most optimum as an de-icing fluid for the high speed railway rolling stock.

  • Organic and inorganic characteristics including bacterial cell number, enzyme activity, nutrients, and heavy metals have been monitored in twelve acrylic experimental tanks for two weeks to estimate and compare self-purification capacities in two Korean wet-land environments, tidal flat and rice field, which are possibly different with the environments in other countries because of their own climatic conditions. FW tanks, filled with rice field soils and fresh water, consist of FW1&2 (with paddy), FW3&4 (without paddy), and FW5&6 (newly reclaimed, without paddy). SW tanks, filled with tidal flat sediments and salt water, are SW1&2 (with anoxic silty mud), SW3&4 (anoxic mud), and SW5&6 (suboxic mud). Contaminated solution, which is formulated with the salts of Cu, Cd, As, Cr, Pb, Hg, and glucose+glutamic acid, was spiked into the supernatent waters in the tanks. Nitrate concentrations in supernatent waters as well as bacterial cell numbers and enzyme activities of soils in the FW tanks (except FW5&6) are clearly higher than those in the SW tanks. Phosphate concentrations in the SW1 tank increase highly with time compared to those in the other SW tanks. Removal rates of Cu, Cd, and As in supematent waters of the FW5&6 tanks are most slow in the FW tanks, while the rates in SW1&2 are most fast in the SW tanks. The rate for Pb in the SW1&2 tanks is most fast in the SW tanks, and the rate for Hg in the FW5&6 tanks is most slow in the FW tanks. Cr concentrations decrease generally with time in the FW tanks. In the SW tanks, however, the Cr concentrations decrease rapidly at first, then increase, and then remain nearly constant. These results imply that labile organic materials are depleted in the FW5&6 tanks compared to the FW1&2 and FW3&4 tanks. Removal of Cu, Cd, As from the supernatent waters as well as slow removal rates of the elements (including Hg) are likely due to the combining of the elements with organic ligands on the suspended particles and subsequent removal to the bottom sediments. Fast removal rates of the metal ions (Cu, Cd, As) and rapid increase of phosphate concentrations in the SW1&2 tanks are possibly due to the relatively porous anoxic sediments in the SW1&2 tanks compared to those in the SW3&4 tanks, efficient supply of phosphate and hydrogen sulfide ions in pore wates to the upper water body, complexing of the metal ions with the sulfide ions, and subsequent removal to the bottom sediments. Organic materials on the particles and sulfide ions from the pore waters are the major factors constraining the behaviors of organic/inorganic elements in the supernatent waters of the experimental tanks. This study needs more consideration on more diverse organic and inorganic elements and experimental conditions such as tidal action, temperature variation, activities of benthic animals, etc.

Evaluation of the Potential of Wood Preservatives Formulated with Okara (두부비지를 이용한 목재 방부제의 사용가능성 평가)

  • The use of CCA as a wood preservative was recently inhibited due to its environmental pollution and human harmfulness. Instead of CCA, copper azole (CuAz) and alkaline copper quaternary (ACQ) have been used as alternative wood preservatives, but the price of the preservatives is much more expensive than that of CCA. As a substitute for high-priced CuAz and ACQ, environmentally friendly wood preservatives were formulated with okara, which is an organic waste from the production of tofu. Prior to formulating the preservatives, okara was hydrolyzed by three levels of sulfuric acid concentration (1, 2.5 and 5%) to easily penetrate the effective components of the preservatives into wood blocks. Final preservative solutions were formulated with the hydrolyzed okara and metal salts, such as copper sulfate, copper chloride and borax. The preservatives were treated into wood blocks by vacuum-pressure method to measure the treatability of the preservatives, and the treated wood blocks were placed in hot water for three days to measure the leachability of the preservatives. The effective components of the preservatives might be successfully penetrated into wood blocks through the uses of hydrolyzed okara and ammonia water. However, the leached amount of effective components was increased as the concentration of acid used for the hydrolysis of okara increased. The treatability and leachability of the preservatives were not affected by hydrolysis temperature but negatively affected by the addition of borax. Based on the results above, the optimal conditions for formulating okara-based wood preservatives cost-effectively and environmentally might be 1% acid hydrolysis of okara and the use of $CuCl_2$ as a metal salt. In addition, the treatability and leachability of okara-based wood preservatives were superior or no differences comparing with those of CuAz. Therefore, it is concluded that okara-based wood preservatives might have a potential to be used as an environmentally friendly wood preservative.

Chemical characteristics of wet precipitation in urban and mountainous sites of Jeju Island

  • Wet precipitation samples were collected in Jeju City and Mt. Halla-1100 site (a site at an altitude of 1100 m on Mt. Halla) during 2011-2013, and their major ionic species were analyzed to examine the chemical composition and characteristics. A comparison of ion balance, electric conductivity, and acid fraction of precipitation revealed correlation coefficients in the range of r = 0.950~0.991, thereby implying the high quality of analytical data. Volume-weighted mean pH and electric conductivity corresponded to 4.86 and 25.5 µS/cm for Jeju City, and 4.98 and 15.1 µS/cm for Mt. Halla-1100 site, respectively. Ionic strengths of the wet precipitation in Jeju City and Mt. Halla-1100 site corresponded 0.3 ± 0.5 and 0.2 ± 0.2 mM, respectively, thereby indicating that more than 30 % of total precipitation was within a pure precipitation criteria. The precipitation with a pH range of 4.5 - 5.0 corresponded to 40.8 % in Jeju City, while the precipitation with a pH range of 5.0 - 5.5 corresponded to 56.9 % in Mt. Halla-1100 site, thereby indicating slightly more weak acidity than that in Jeju city. The volume-weighted mean concentration (µeq/L) of ionic species was in the order of Na+ > Cl- > nss-SO42- > NO3- > Mg2+ > NH4+ > H+ > nss-Ca2+ > PO43- > K+ > CH3COO- > HCOO- > NO2- > F- > HCO3- > CH3SO3- at Jeju City area, while it corresponded to Na+ > Cl- > nss-SO42- > NO3- > NH4+ > H+ > Mg2+ > nss-Ca2+ > PO43- > CH3COO- > K+ > HCOO- > NO2- > F- > HCO3- > CH3SO3- at Mt. Halla-1100 site. The compositions of sea salts (Na+, Cl-, Mg2+) and secondary pollutants (NH4+, nss-SO42-, NO3-) corresponded to 66.1 % and 21.8 %, respectively, in Jeju City and, 49.9 % and 31.5 %, respectively, in Mt. Halla-1100 site. The acidity contributions in Jeju City and Mt. Halla-1100 site by inorganic acids, i.e., sulfuric acid and nitric acid, corresponded to 93.9 % and 91.4 %, respectively, and the acidity contributions by organic acids corresponded to 6.1 % and 8.6 %, respectively. The neutralization factors in Jeju City and Mt. Halla1100 site by ammonia corresponded to 29.8 % and 30.1 %, respectively, whereas the neutralization factors by calcium carbonate corresponded to 20.5 % and 25.2 %, respectively. From the clustered back trajectory analysis, the concentrations of most ionic components were higher when the airflow pathways were moved from the continent to Jeju area.

Studies on the Amylase Production by Bacteria (세균(細菌)에 의(依)한 Amylase생산(生産)에 관한 연구(硏究))

  • 1. Isolation and identification of amylase-producing bacteria. The powerful strain A-12 and S-8 were respectively isolated from air and soil after screening a large number of amylase-producing bacteria. Their bacterial characteristics have been investigated and it has been found that all characteristics of strain A-12 and S-8 are similar to Bac. subtilis of Bergey's manual except for the acid formation from a few carbohydrates and the citrate utilization, i.e., the strain A-12 shows negative in the citrate utilization, and the acid formation from arabinose and xylose, S-8 shows negative in the acid formation from xylose. 2. Amylase production by Liquid cultures with solid materials. Several conditions for amylase production by strain A-12 in stationary cultures have been studied. The results obtained are as follows. (1) The optimum conditions are:temperature $35^{\circ}C$, initial pH 6.5 to 7.0 and incubation time 3 to 4 days. (2) The amylase production is not affected by the preservation period of the stock cultures. (3) Among the various solid material, the defatted soy bean is found to be the best for t1e amylase production. However, the alkali treatment of the defatted soy bean gives no effect contrary to the cage of defatted rape seed. The addition of soluble starch to the alkali extract of defatted soy bean shows the increased amylase production. (4) Up to 1% addition of ethanol to carbon dificient media gives the improved amylase production, whereas the above effect is not found in the case of carbon rich media. (5) The amylase production can be increased 2.5 times when 10% of defatted soy bean is admixed to cheaply available wheat bran. (6) The excellent effect is found for amylase production when 20% of wheat bran is admixed to defatted dry milk which is a poor medium. The activity is found to be $D^{40^{\circ}}_{30'}$ 7,000(L.S.V. 1,800) in 10% medium. (7) No significant effect is observed due to the addition of various inorganic salts. 3. Amylase production by solid cultures. Several conditions for amylase production by strain A-12 in wheat bran cultures have been studied and the results obtained are as follows. (1) The optimum conditions: are temperature $33^{\circ}C$, incubation lime 2 days, water content added 150 to 175% and the thickness of the medium 1.5cm, The activity is found to be $D^{40^{\circ}}_{30'}$ 36,000(L.S.V. 15,000) (2) No significant effect is found in the case of the additions of various organic and inorganic substances.

A Study on the Physico-Chemical Characteristics of Acid Sulfate Soil in Kimhae Plain (김해평야(金海平野)에 분포(分布)된 특이산성토(特異酸性土)(답)(沓)의 이화학적성질(理化學的性質)에 관(關)한 조사연구(調査硏究))

  • The study on physico-chemical characteristics of the acid sulfate soil present in Kimhae plain was carried out with 28 surface and subsoils from lower and higher produtive area and two representative profile samples from the areas reclaimed a few decades ago and around 10 years ago respectively. 1. There are no differences in soil texture between lower and higher productive soils being mostly silty clay loam and silty clay. 2. Very significant differences in pH, degree of base saturation and extractable aluminium content are observed; lower pH, lower degree of base saturation and higher aluminium in the lower productive soils and subsoils. The pH and degree of base saturation of these soils are extremely low whereas aluminium content is very high compared to ordinary paddy soil. 3. Cation exchange capacity of these soils are slightly higher than ordinary paddy soils. In higher productive soils, exchangeable calcium and magnesium are of same order, whereas in lower productive soils magnesium content is appreciably higher than calcium. 4. Though the soil is derived from marine and estuarine sediment, the soluble salt content is not high. There are only few lower productive surface soils and subsoils having Ec values of the saturation extracts higher than 4 mmhos but lower than 9 mmhos/cm. 5. Organic matter content of these soils is a bit higher compared to ordinary paddy soils, but, nitrogen content is comparatively low. C/N ratio of these soils is around 12. 6. Sulfur content is considerably higher but oxidizable sulfur is found to be very low. Total sulfur is generally high in subsoils and lower productive soils. 7. Active iron and available silica are slightly higher than ordinary paddy soils but easily reducible manganese is very low. Almost no differences are also observed between lower and higher productive soils. 8. Available phosphorus content is extremely low in particular, regardless of higher or lower productive soils. 9. The two representative profiles from the area of earlier reclamation and recent one show that samples from earlier reclaimed area contain less amount of free acids, sulfur compounds, toxic aluminium and soluble salts etc. than the other. This indicate greater leaching and possible addition of lime for a longer period of time. 10. From the results obtained, it can be concluded the higher productivity of group I soils is due to the greater leaching and neutralisation of acidity by liming materials, It can also be concluded that the productivity of both types can be increased by addition of liming materials and improvement of drainage facilities.
