Objectives: The purpose of this study is to evaluate the optimum conditions for oral mucosal irritation testing using the buccal pouch of hamsters. Methods: Test materials were applied to the buccal pouch of seven-week old male Syrian hamsters (SLC, Japan) four times at one-hour intervals and macroscopic changes were examined at 24 hours after final treatment. After sacrifice, the buccal pouches were removed and prepared for histopathological evaluation. In order to set the exposure time, we performed exposure tests of 5, 12, 18 and 23 minutes using sodium lauryl sulfate (SLS) 1% and set the treatment volume from the test results at 2, 3, or 4 ml treatment using SLS 1%, Triton X-100 1% and ethanol. After setting the experimental conditions, seven groups of materials [sodium lauryl sulfate (SLS) (1%), Triton X-100 (1%), hydrogen peroxide (3%), ethanol (100%), chlorhexidine (0.2%, 2%), phosphate buffer saline (PBS)] were assessed. Results: Experimental conditions of material exposure time were fixed as 18 minutes from the exposure tests of 5, 12, 18 or 23 min using sodium lauryl sulfate (SLS) 1%. Treated volume was set as 4 ml per each pouch from the test results of 2, 3, or 4 ml treatments using SLS 1%, Triton X-100 1% and ethanol. The results in terms of irritation degree were in the order of sodium lauryl sulfate (SLS) (1%) > Triton X-100 (1%) ${\fallingdotseq}$ hydrogen peroxide (3%) > ethanol (100%) ${\fallingdotseq}$ chlorhexidine (0.2%, 2%) > phosphate buffer saline (PBS). Conclusion: From this study, suitable conditions for hamster mucosal irritation testing were suggested and this method was verified through materials commonly used on oral mucosal membranes.
The author studied the histopathologic changes according to a single or a split dose and the time after irradiation on the acinar cells of rat parotid gland. 99 Sprague Dawley rats, weighing about l20gm, were divided into control and 3 experimental groups. In experimental groups, GroupⅠ and Ⅱ were delivered a single dose of l5Gy, 18Gy and Group Ⅲ and Ⅳ were delivered two equal split doses of 9Gy, 10.5Gy for a 4 hours interval, respectively. The experimental groups were delivered by a cobalt-60 teletherapy unit with a dose rate of 222cGy/min, source-skin distance of 50㎝, depth of l㎝ and a field size of l2×5㎝. The animals were sacrificed at 1, 2, 3, 6, 12 hours, 1, 3, 7 days after irradiation and examined by light and electron microscopy. The results were as follows: 1. As the radiation dose increased and the acinar cells delivered a single dose exposure were more damaged, and the change of acinar cells appeared faster than those of a split dose exposure. 2. The histopathologic change of acinar cells appeared at 1 hour after irradiation. The recovery from damaged acinar cells appeared at 1 day after irradiation and there was a tendency that the recovery from damage of a split dose exposure was somewhat later than that of a single dose exposure. 3. Light microscope showed atrophic change of acinar cells and nucleus, degeneration and vesicle formation of cytoplasm, widening of intercellular space and interlobular space. 4. Electron microscope showed loss of nuclear membrane, degeneration of nucleus and nucleoli, clumping of cytoplasm, widening and degeneration of rough endoplasmic reticulum, loss of cristae of mitochondria, lysosome, autophagosome and lipid droplet. 5. Electron microscopically, the change of rough endoplasmic reticulum was the most prominent and this appeared at 1 hour after irradiation as early changes of acinar cells. The nuclear change appeared at 2 hours after irradiation and the loss of cristae of mitochondria was observed at 2 hours after irradiation in all experimental groups.
The purpose of this study was to evaluate the effect of scatter radiation to dental x-ray film with long time-exposure in the different structures of the tooth, by using pinhole camera. For this study, pinhole camera, skull with tooth, and pocket dosimeter were used. The radiation with 70 and 90kVp and exposure time (minimum: 2.5 min., maximum 10 hrs.) was projected to the film in the pinhole camera. And density of the obtained x-ray film was measured with densitometer. In the intra-oral film taking, the amount of exposure of the scatter radiation affecting the thyroid gland area was measured with the dosimeter at the thyroid gland. The density of radiographs was compared in radiation projected with or without the metal cone of dental machine. The effect of the back scatter radiation to the film was also evaluated when the lead foil was removed. The obtained results were as follows: 1. A pinhole camera was a valuable device for locating the source of x-ray. 2. The scatter radiation affected the dental x-ray film when the radiation source was exposed. more than 5 hours'. In that case, the density of the scatter radiation could be observed visually. 3. The scatter radiation caused by short exposure of dental radiation didn't affect the diagnostic quality of the dental x-ray film. 4. The differences of densities between the tooth and the soft tissue according to exposure time showed 0.16 in 5 hours' exposure & 0.17 in 10 hours' exposure at 70 kVp & 0.12 in 5 hours' exposure & 0.13 in 10 hours' exposure at the 90kVp. 5. The differences of densities between the tooth and the soft tissue according to kVp showed no difference between 5 hours' exposure of tooth at 70 kVp and soft tissue at 90 kVp, but showed 0.05 high density in tooth when 10 hours' exposure at 90 kVp. 6. No difference of density was on radiographs taken with or without dental machine cone. 7. Back scatter radiation was recorded image of radiographs for only 3 min. 8. The amounts of the scatter radiation exposed to the thyroid gland in intraoral film taking were 1.12 mr in upper anterior, 0.55 mr in upper posterior, 2.75 mr in lower anterior, and 1.92 mr in lower posterior teeth.
Purpose: To compare the film density of Insight dental X-ray film (Eastman Kodak Co., Rochester, NY, USA) with that of Ektaspeed Plus film (Eastman Kodak) under manual and automatic processing conditions. Materials and Methods : Insight and Ektaspeed Plus films were exposed at three different exposure conditions with an aluminum step wedge on the films under the three different exposure times. The exposed films were processed by both manual and automatic ways. The Base plus fog density and the optical density made by exposing step wedge were calculated using a digital densitometer (model 07-443, Victoreen Inc, Cleveland, Ohio, USA). The optical densities of the Insight and Ektaspeed film versus thickness of alumimun wedge at the same exposure time were plotted on the graphs. Statistical analyses were applied for comparing the optical densities of the two films. Results: The film density of both Insight films and Ektaspeed Plus films under automatic processing condition was significantly higher over the manual processing. The film density of Insight films was significantly higher than that of Ektaspeed Plus films on both automatic and manual processing conditions. Conclusion: The radiation exposure time can be reduced when using Insight over Ektaspeed Plus film. To take the full advantage of reducing exposure time, Insight film should be processed automatically.
Recent studies indicate that reactive oxygen species (ROS) are critically involved in persistent pain primarily through spinal mechanisms, and that mitochondria are the main source of ROS in the spinal dorsal horn. To investigate whether mitochondrial ROS can induce changes in membrane excitability on spinal substantia gelatonosa (SG) neurons, we examined the effects of mitochondrial electron transport complex (ETC) substrates and inhibitors on the membrane potential of SG neurons in spinal slices. Application of ETC inhibitors, rotenone or antimycin A, resulted in a slowly developing and slight membrane depolarization in SG neurons. Also, application of both malate, a complex I substrate, and succinate, a complex II substrate, caused reversible membrane depolarization and enhanced firing activity. Changes in membrane potential after malate exposure were more prominent than succinate exposure. When slices were pretreated with ROS scavengers such as phenyl-N-tert-buthylnitrone (PBN), catalase and 4- hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPOL), malate-induced depolarization was significantly decreased. Intracellular calcium above $100{\mu}M$ increased malateinduced depolarization, witch was suppressed by cyclosporin A, a mitochondrial permeability transition (MPT) inhibitor. These results suggest that enhanced production of spinal mitochondrial ROS can induce nociception through central sensitization.
The purposes of this study were clinical comparison and evaluation between digital radiography and conventional radiography for the detection of artificial interproximal caries. Four freshly extracted, unrestored posterior teeth were obtained and caries was simulated by drilling semicircled defects with variable size at the interproximal surface of each tooth. The experiments were performed with IBM-PC/32 bit-DX compatible, video camera(VM-S8200, Hitachi Co., Japan), and color monitor(Multisync 3D, NEC, Japan). Sylvia Image Capture Board for the ADC(analog to digital converter) was used, and spatial resolution was 512 × 480 with 256 gray levels. The obtained results were as follows: 1. At the condition of under exposure, the radiographs were superior to the digital images in readability. Also, as the size of the artificial lesion was increased, readability of the radiographs was elevated. 2. The digital images were superior to the radiographs in readability especially under over exsposure. 3. As the exposure time and size of lesion was increased, the gray level of region of interest of the digital image was decreased proportionally. 4. As the F-value of average gray level of region of interest at individual exposure time and size of lesion, gray level of the all lesion sizes was decreased in comparison with that of the normal enamel.
Purpose : To measure dose-width product (DWP) values used for dental panoramic radiography in Anyang city, Korea. Materials and Methods : Thirty-six panoramic dental radiographic sets (17 analogue panoramic sets and 19 digital panoramic sets) in 36 dental clinics in Anyang city were included in the study. Each patient's panoramic exposure parameters were simulated and the panoramic radiation doses were measured at the secondary collimator using a Mult-O-Meter (Unfors Instruments, Billdal, Sweden) at each dental clinic during 2006. The third quartile DWP was determined from 310 surface dose measurements on adult. Results : The third quartile DWP for adult panoramic radiograph was 106.7 mGy mm. For analogue and digital panoramic radiograph, 3/4 DWP were 116.8 mGy mm and 72 mGy mm respectively. The overall third quartile DWP of panoramic radiography was 106.7 mGy mm. Conclusion : The measured 3/4 DWPs were higher than the 3/4 DWP of 65 mGy mm recommended by NRPB. Dentists who are operating above the reference dose should lower their panoramic exposure doses below the recommended reference value by changing the exposure parameters and/or their panoramic equipments.
Purpose: This study aimed to provide comparative measurements of the effective dose from direct and indirect digital panoramic units according to phantoms and exposure parameters. Materials and Methods: Dose measurements were carried out using a head phantom representing an average man (175 cm tall, 73.5 kg male) and a limbless whole body phantom representing an average woman (155 cm tall, 50 kg female). Lithium fluoride thermoluminescent dosimeter (TLD) chips were used for the dosimeter. Two direct and 2 indirect digital panoramic units were evaluated in this study. Effective doses were derived using 2007 International Commission on Radiological Protection (ICRP) recommendations. Results: The effective doses of the 4 digital panoramic units ranged between $8.9{\mu}Sv$ and $37.8{\mu}Sv$. By using the head phantom, the effective doses from the direct digital panoramic units ($37.8{\mu}Sv$, $27.6{\mu}Sv$) were higher than those from the indirect units ($8.9{\mu}Sv$, $15.9{\mu}Sv$). The same panoramic unit showed the difference in effective doses according to the gender of the phantom, numbers and locations of TLDs, and kVp. Conclusion: To reasonably assess the radiation risk from various dental radiographic units, the effective doses should be obtained with the same numbers and locations of TLDs, and with standard hospital exposure. After that, it is necessary to survey the effective doses from various dental radiographic units according to the gender with the corresponding phantom.

Purpose: This study assessed the intensity of artifacts produced by 2 metal posts, 2 cements, and different exposure parameters using 2 cone-beam computed tomography (CBCT) units. Materials and Methods: The sample was composed of 20 single-rooted premolars, divided into 4 groups: Ni-Cr/zinc phosphate, Ni-Cr/resin cement, Ag-Pd/zinc phosphate, and Ag-Pd/resin cement. Samples were scanned before and after post insertion and cementation using a CS9000 3D scanner with 4 exposure parameters (85/90 kV and 6.3/10 mA) and an i-CAT scanner with 120 kV and 5 mA. The presence of artifacts was assessed subjectively by 2 observers and objectively by a trained observer using ImageJ software. The Mann-Whitney, Wilcoxon, weighted kappa, and chi-square tests were used to assess data at a 95% confidence level(α<0.05). Results: In the subjective analyses, AgPd presented more hypodense and hyperdense lines than NiCr (P<0.05), and more hypodense halos were found using i-CAT (P<0.05) than using CS9000 3D. More hypodense halos, hypodense lines, and hyperdense lines were observed at 10 mA than at 6.3 mA (P<0.05). More hypodense halos were observed at 85 kV than at 90 kV (P<0.05). CS9000 3D presented more hypodense and hyperdense lines than i-CAT (P<0.05). In the objective analyses, AgPd presented higher percentages of hyperdense and hypodense artifacts than NiCr (P<0.05). Zinc phosphate cement presented higher hyperdense artifact percentages on CS9000 3D scans(P<0.05). CS9000 3D presented higher artifact percentages than i-CAT(P<0.05). Conclusion: High-atomic-number alloys, higher tube current, and lower tube voltage may increase the artifacts present in CBCT images.
The comparative detectability of the artificial defects among conventional radiographs, digital images and digital subtraction images was evaluated. The artificial defects were made within spogy bone of 24 unilateral mandibles of adult dogs. The results were as follows: 1. With normal exposure time, the detectability of digital subtraction radiographs was 90.3% which was statistically significant superior to those of conventional radiographs(78.0%) and digital images(75.9%) (p<0.05). 2. With half-exposure time, the detectability of conventional radiographs, digital images and digital subtraction radiographs was 68.4%, 67.3% and 69.9% respectively. There was no statistical significant difference among the detectability of these methods(p>0.05). 3. All radiographic images with normal exposure time showed statistically significant superior detectability to those with half-exposure time(p<0.05). 4. The detectability of digital subtraction radiographs was not linearly related to the standard deviation of the grey levels of reference line(p<0.05).
