Optimization of Conditions for Isolating and Cultivating Bacillus sp. Se-103 with a Mesophilic Feather-Degrading Activity (중온성 우모 분해균 Bacillus sp. SE-103의 분리 및 배양 조건 최적화)

  • This study was carried out to investigate the possibility to utilize feather meal by bacterial strains. A bacterial strain SE-103 producing keratinolytic enzyme was isolated from the soil of the poultry slaughterhouses. It was identified as Bacillus sp. by judging from its morphological and physiological characteristics. Subsequently the optimal culture conditions for the production of keratinolytic protease by Bacillus sp. SE-103 were investigated. The composition of optimal medium was 3.0% glucose, 0.4% urea, 0.2% $NaNO_3$, and 0.15% KCl. In addition, optimal initial pH and temperature were 6.0 and $35^{\circ}C$, respectively.

  • Xenorhabdus nematophilus Kor-Al was cultured at flask and 5L jar fermentor at $28^{\circ}C$, 5% YS media condition. Antibiotic activity for X. nematophilus Kor-Al was experimented by paper disk method. As the result, antibiotic activity was growth associated form during culture time of X. nematophilus Kor-Al at flask. The maximum production and antibiotic activity were obtained at stationary period of cell growth. The optimum conditions of cell growth and antibiotic production at 5L jar fermentor were 400rpm agitation and 50% DO conditions.

  • As flocculent strains are likely to have considerable potential for internal cell recycle, kinetic studies on glucose medium with flocculent Saccharomyces uvarum were carried out in batch and continuous culture. Using a mathematical model, the kinetic parameters at each temperature and pH were estimated in order to establish optimal conditions. It was found that an overall optimum temperature for growth and ethanol production in the range 33-35$^{\circ}C$ was desirable. With regard to the effect of pH, ethanol production by S. uvarum was found to be relatively insensitive to pH value between 4 and 6, with an optimum pH of around 5. At these optimal conditions a maximum ethanol productivity of 12 g/$\ell$/h was determined using semi-batch process together with 5. uvarum.

  • The effects of steeping time of glutinous rice and of beating time of the cooked rice on characteristics of Ganjung was examined. and the optimum conditions of traditional Gangjung preparation were determined. Moisture absorption of glutinous rice was the maximum at 2 hours of steeping in water and did not increase much after then. pH of the steep water was the lowest at 7 days of steeping and increased slightly thereafter. Reducing sugar content of the steep water increased as the steepting time was extended. Expansion ratio of Gangiung was the greatest with 5 days of steepting, and tended to decrease with longer beating time. Optimum conditions of Gangjung preparation was 7 days of steeping glutinous rice and 4 minutes of beating (40 times/min) the cooked rice.

Optimization of the Manufacturing Process for Mandarin Dry Chip Using Response Surface Methodology (RSM) (반응표면분석법을 이용한 감귤건조칩 제조조건 최적화)

  • The purpose of this study was to optimize the mandarin dry chip manufacturing using a response surface methodology. The experiment was designed based on a CCD (Central Composite Design), and the independent variables were the drying temperature ($X_1$, $50-90^{\circ}C$), drying time ($X_2$, 12-36 hours), and microwave pretreat time ($X_3$, 0-4 minutes). The results of appearance ($Y_5$), color ($Y_6$), taste ($Y_8$) and overall acceptance ($Y_{10}$) were fitted to the response surface methodology model ($R^2=0.86$, 0.88, 0.89, and 0.84, respectively). Increasing the drying temperature and microwave treatment time were negatively evaluated for consumer acceptance. On the other hand, a high value of consumer acceptance was evaluated when the drying time was more than 24 hr. Therefore, the optimal conditions of $X_1$, $X_2$, and $X_3$ were $52.989^{\circ}C$, 24 hr, and 1 min, respectively. Under these optimal conditions, the predicted values of $Y_5$, $Y_6$, $Y_8$, and $Y_{10}$ were 5.066, 5.338, 5.063, and 5.339, respectively.

Optimizing Production Conditions for Germinated Brown Rice Cookies Prepared with Onion Powder (양파가루 첨가 발아현미쿠키의 제조조건 최적화)

  • The principal objective of this study was to determine the optimal mixing conditions for three amounts of onion powder, sugar, and butter to prepare onion powder cookies. The experimental design was based on the central composite design methodology of response surface, which included 16 experimental points including two replicates for onion powder, sugar, and butter. The mechanical and sensory properties of the cookies were measured, and these values were applied to the mathematical models. A canonical form and perturbation plot showed the influence of each ingredient on the mixed final product. The results of the spread ratio did not show significant results, but hardness increased with increasing quantities of onion powder and sugar but decreased with butter (p<0.01). The color lightness "L" value increased with increasing quantities of sugar and butter but decreased with added onion powder. In contrast, the redness color "a" value increased with increasing quantities of onion powder and sugar. Sugar did not affect the yellowness color "b" value, but the color b value increased with increasing onion powder and sugar. The results of a sensory evaluation using the predicted model showed significant values for flavor (p<0.01), texture (p<0.05), taste (p<0.05), and overall quality (p<0.01). As a result, the optimum formulation by numerical and graphical methods was calculated as 12.58 g onion powder, 35 g sugar, and 52.38 g butter.

Lymphoid Lineage γδ T Cells Were Successfully Generated from Human Pluripotent Stem Cells via Hemogenic Endothelium

  • γδ T cells are a rare and unique prototype of T cells that share properties with natural killer cells in secondary lymphoid organs. Although many studies have revealed the function and importance of adult-derived γδ T cells in cancer biology and regenerative medicine, the low numbers of these cells hamper their application as therapeutic cell sources in the clinic. To solve this problem, pluripotent stem cell-derived γδ T cells are considered alternative cell sources; however, few studies have reported the generation of human pluripotent stem cell-derived γδ T cells. In the present study, we investigated whether lymphoid lineage γδ T cells were successfully generated from human pluripotent stem cells via hemogenic endothelium under defined culture conditions. Our results revealed that pluripotent stem cells successfully generated γδ T cells with an overall increase in transcriptional activity of lymphoid lineage genes and cytolytic factors, indicating the importance of the optimization of culture conditions in generating lymphoid lineage γδ T cells. We uncovered an initial step in differentiating γδ T cells that could be applied to basic and translational investigations in the field of cancer biology. Based on our result, we will develop an appropriate method to purify γδ T cells with functionality and it helpful for the study of basic mechanism of γδ T cells in pathophysiologic condition as well as clinic application.

Optimization of Anticoagulant Production from Phellinus linteus Mycelia (상황버섯(Phellinus linteus) 균사체로부터 항혈전 생산의 최적화)

  • To produce the functional food materials, 50 kinds of the mycelial extracts from edible mushroom were examined for anticoagulant activity and Phellinus linteus showed the highest activity through the activated partial thromboplastin test (aPTT). The maximum production of anticoagulant activity and the mycelial growth was observed in culture medium containing soluble starch 3.0%, peptone 0.1%, $MgSO_4{\cdot}7H_2O^{\circ}C$ 0.1%, $K_2HPO_4$ 0.1% and in the culture conditions controlled at initial pH 7.0, $30^{\circ}C$ and 150 rpm by the rotary shaker. In addition, the maximum production of mycelial dry weight was 7.5 mg/mL after 10 days under the optimal conditions, and anticoagulant activity was reached to 390 sec in 5 L-jar fermentor.

Optimization for Production of Phellinus linteus Mycelia with Anti-Complement Activity (항보체 활성을 갖는 Phellinus linteus 균사체 생산의 최적화)

  • To produce the functional food materials from edible mushrooms, hot-water extracts from 70 kinds of mushroom mycelia were examined for anti-complementary activity and Phellinus linteus showed the highest activity through the complement fIxation test. The maximum production of Phellinus linteus mycelia with anti-complementary activity was observed in culture medium containing soluble starch 3.0%, peptone 0.3%, yeast extract 0.4%, $MgSO_4{\cdot}7H_2O$ 0.1%, $K_2HPO_4$ 0.2% and in the culture conditions controlled at initial pH 7.0, $30^{\circ}C$ and 150 rpm by the rotary shaker. In addition, the maximum production of mycelial dry weight was 15 mg/mL after 18 days under the optimal conditions, and anti-complementary activity was reached to 88% in 5 L-jar fermenter.

  • Using recombinant Vaccinia virus(vSC8) that express ${\beta}$-galactosidase, a model heterologous protein, conditions for virus and protein production were investigated in tissue culture flask. As host animal cells HeLa and HeLa S3 were used. It was demonstrated that cells infected during the exponential growth phase gave higher protein yield than those infected during the stationary growth phase and calf serum concentration after virus infection did not significantly alter protein yield. Pretreatment of cell layer with hypotonic solution enhanced the virus infectivity. Optimum cell growth and recombinant protein production was achieved at $37^{\circ}C$. But, during 2 hours of virus infection period incubation temperature must be lowered to 20∼$30^{\circ}C$ for maximum recombinant protein yield. To enhance virus replication, the effects of adrenal glucocorticoid hormone (Dexamethasone) and silkworm hemolymph were evaluated. Only dexamethasone increased about 20% of ${\beta}$-galactosidase yield in HeLa S3 cells when added with 10-7∼10-5M concentration 24 hours before infection.
