Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.
Acetobacter sp. were isolated from persimmon vinegar. We studied about conditions of Aloe vinegar fermentation by an isolated strain and inhibitory effect against lipase activity. Strains DS-118 was strictly aerobic, motile, gram negative, non-spore-forming and short rod shaped. It reacted positively in catalase test, was oxidase test negative, was ureas negative, was produced acetic acid from alcohol. On the basis of these results, it was identified as a strain of Acetobacter sp. In the preparation of Aloe vinegar, optimun initial alcohol concentration, acidity, and fermentation temperature were 10%, $3{\sim}4%$ and $25^{\circ}C$, respectively. The major organic acid in Aloe vinegar was acetic acid (12%), but malic acid and ${\delta}-galactronic$ acid were also present in trace. The Aloe vinegar(acidity : 12%) inhibited lipase activity and it's $IC_{50}$ was 43%.
PMS(Pavement Management System) has been developed and/or operated for the freeway and national highway systems in Korea. There is, however, no municipal PMS that is necessary in many cities for efficient management of their roadways. In this study, a municipal Prototype PMS is developed. The Prototype PMS is designed to be operated on PC base. It uses GIS interface for user friendliness that applies color graphics, text and distress pictures to display pavement conditions and rehabilitation information for both current and historical situations. The computer program of the prototype PMS consists of four major functions(inquiry, analysis, data management, and print). Especially, prioritization and M&R(Maintenance and Rehabilitation) alternative selection procedure is designed to be flexible so that the users can modify the decision criteria whenever it is necessary. Database items of the Prototype PMS is carefully selected for the objectives and size of municipal PMS. Reference point method is basically used for the location referencing in the Database. The unit section consists of each loom segment of the road for effective management.
The application of micro-simulation model has been extended farther with improvement of computer performance and development of complicated model. To make a micro-simulation model accurately replicate field traffic conditions, model calibration is very crucial. Studies on calibration of micro-simulation model have not been enough while lots of studies on calibration of macro-simulation model have been continued in our country. This paper presents an auto-calibration of parameter values in micro-simulation model(VISSIM) using genetic algorithm. RMSE(Root Mean Square Error) of collected volume on the urban expressway versus simulated volume is set as MOP(measure of performance) and objective function of optimization is set as to minimize the RMSE. Applying to urban expressway(Nae-bu circular) as a case study, it shows that RMSE of optimized parameter values decrease 60.4%($19.3{\longrightarrow}7.6$) compared to default parameter values and the proposed auto-calibration system is very effective.
The land use-transportation models typically have complicated model structure that is good for empirical execution but bad for theoretical probe. This complexity makes it very difficult to derive the first-order conditions for system optimization in tractable forms. Yu and Rhee (2011) and Rhee (2012) show how to simplify the derivative of the model's objective function with respect to policy variables in the computable general equilibrium model of land use and transportation. However, the travel demand in their model was fixed. This drawback fundamentally limits the applicability of their methodology in the planning field. We relax this restriction. Once this is done, we can employ the methodology developed in analyzing the impacts of various types of policy instruments in the models where land market is treated endogenously and transportation network is embedded.
The experimental design and response surface methodologies haven been applied to the investigation of the chemical coagulation of livestock wastewater. The chemical coagulation reactions were mathematically described as a function of parameters raping mixing (rpm) of chemical coagulation ($X_1$), slow mixing (rpm) of chemical coagulation ($X_2$), $FeCl_3 $ concentration (mg/L) ($X_3$) and pH ($X_4$) being modeled by use of the central composite design. Empirical models were developed to describe relationship between the experimental variables and response. Statistical analysis indicates that three factors ($X_1$: raping mixing (rpm), $X_2$: slow mixing (rpm), $X_3$: $FeCl_3 $ concentration (mg/L) on the linear term (main effect), slow mixing (rpm) (${X_2}^2$) on the non-linear term (quadratic), and two factors ($X_1-X_3$, $X_2-X_3$) on the non-linear term (cross-product) had significant effects, respectively. The estimated ridge of maximum responses and optimal conditions for CODcr using canonical analysis was 87.44% ($X_1$: 229 rpm, $X_2$: 51 rpm, $X_3$: 877 mg/L, $X_4$: 4.3). To confirm this optimum condition, three additional experiments were performed and the mean CODcr removal (%) and concentration (mg/L) with a standard deviation of $87{\pm}1.2%$ ($576{\pm}34ppm$) were obtained.
This experimental design and response surface methodology (RSM) have been applied to the investigation of the electrocoagulation/flotation of dye wastewater. The electrocoagulation/flotation reactions were mathematically described as a function of parameters current (A), NaCl concentration (B), initial RhB concentration (C) and time (D) being modeled by use of the central composite design (CCD). The application of RSM using the CCD yielded the following regression equation, which is an empirical relationship between the RhB removal (%) and test variables in RhB removal (%) = $-300.42+129.21{\cdot}Current+46.99{\cdot}NaCl-0.11{\cdot}RhB-+43.71{\cdot}Time-5.67{\cdot}Current{\cdot}NaCl-3.18{\cdot}Current{\cdot}Time-2.41{\cdot}NaCl{\cdot}Time-19.79{\cdot}Current^2-2.27{\cdot}NaCl^2-1.59{\cdot}Time^2$. the model predictions agreed well with the experimentally observed result ($R^{2}=0.9728$). The estimated ridge of maximum response and optimal conditions for RhB removal (%) using canonical analysis was 99.4% (A: 1,77 A, NaCl concentration: 2.23 g/L, RhB concentration: 56.12 mg/L, Time: 9.98 min). To confirm this optimum condition, three additional experiments were performed and RhB removal (%) were within range of 86.87% (95% PI low)~111.93% (95% PI high) obtained.

Bioenergy production from lignocellulosic biomass and agriculture wastes have been attracted because of its sustainable and non-edible source. Especially, canola is considered as one of the best feedstock for renewable fuel production. Oil extracted canola and its agriculture residues are reuseable for bioethanol production. However, a pretreatment step is required before enzymatic hydrolysis to disrupt recalcitrant lignocellulosic matrix. To increase the sugar conversion, more efficient pretreatment process was necessary for removal of saccharification barriers such as lignin. Alkaline pretreatment makes the lignocellulose swollen through solvation and induces more porous structure for enzyme access. In our previous work, aqueous ammonia (1~20%) was utilized for alkaline reagent to increase the crystallinity of canola residues pretreatment. In this study, significant factors for efficient soaking in aqueous ammonia pretreatment on canola residues was optimized by using the response surface method (RSM). Based on the fundamental experiments, the real values of factors at the center (0) were determined as follows; $70^{\circ}C$ of temperature, 17.5% of ammonia concentration and 18 h of reaction time in the experiment design using central composition design (CCD). A statistical model predicted that the highest removal yield of lignin was 54% at the following optimized reaction conditions: $72.68^{\circ}C$ of temperature, 18.30% of ammonia concentration and 18.30 h of reaction time. Finally, maximum theoretical yields of soaking in aqueous ammonia pretreatment were 42.23% of glucose and 22.68% of xylose.
A fractional step finite difference model for the longitudinal dispersion of nonconservative pollutants is applied to the Nakdong River to simulate the phenol spill accident which occurred on March, 1971. Prior to the dispersion calculation, the flow conditions are simulated to provide inputs to the dispersion model. An unsteady flow model based on Preissmann's four-point scheme is used for this purpose. Sensitivities of the dispersion calculation to empirical equations for dispersion coefficient and to the first-order decay coefficient are analyzed. The time to peak concentration at a downstream location is significantly different depending on the formula for the dispersion coefficient. Although the decay coefficient does not affect the shape of the temporal concentration distribution, the concentration values depend on the decay coefficient verb significantly. An optimization technique is used to calibrate the dispersion model as well as the flow model. The time to the peak concentration is simulated for major positions of water intake along the Nakdong River.

The electrical loss of the photo-generated carriers is dominated by the recombination at the metal- semiconductor interface. In order to enhance the performance of the solar cells, many studies have been performed on the surface treatment with passivation layer like SiN, SiO2, Al2O3, and a-Si:H. In this work, Al2O3 thin films were investigated to reduce recombination at surface. The Al2O3 thin films have two advantages, such as good passivation properties and back surface field (BSF) effect at rear surface. It is usually deposited by atomic layer deposition (ALD) technique. However, ALD process is a very expensive process and it has rather low deposition rate. In this study, the ICP-assisted reactive magnetron sputtering method was used to deposit Al2O3 thin films. For optimization of the properties of the Al2O3 thin film, various fabrication conditions were controlled, such as ICP RF power, substrate bias voltage and deposition temperature, and argon to oxygen ratio. Chemical states and atomic concentration ratio were analyzed by x-ray photoelectron spectroscopy (XPS). In order to investigate the electrical properties, Al/(Al2O3 or SiO2,/Al2O3)/Si (MIS) devices were fabricated and characterized using the C-V measurement technique (HP 4284A). The detailed characteristics of the Al2O3 passivation thin films manufactured by ICP-assisted reactive magnetron sputtering technique will be shown and discussed.
