Studies on Microbial Penicillin Amidase (II) Characteristics and the Reactor Performance of Whole Cell Immobilized Penicillin Amidase of Escherichia coli (미생물 페니실린 아미다제에 관한 연구 (II) E. coli의 균체 고정화 페니실린 아미다제의 특성 및 반응조에 관한 연구)

  • Whole cell penicillin amidase of Escherichia coli was immobilized by entrapment in gelatin followed by extrusion and crosslinking with glutaraldehyde. The immobilized engyme preparation demonstrated the recovery yield of activity up to 70% and good stability during storage and operation. The half life of activity decay during the operation was estimated to be about 50 days. The optimum pH and temperature for both of immobilized and soluble enzyme are 8.5 and 5$0^{\circ}C$, respectively. No significant change was demonstrated in the effect of pH and temperature, but the increase in heat stability at high temperature was observed in the case of the immobilized enzyme. It was found that the plug flow reactor could be operated favorably since the pH drop along the column path due to tile reaction product was minimized by employing substrate solution with moderate buffer strength. The optimal condition of reactor operation was discussed with regard to the effect of substrate concentration and the residence time on the conversion efficiency and productivity.

Detection of fluorescence from soils contaminated with monoaromatic hydrocarbons (유류 오염 토양에서의 단일방향족 탄화수소 농도 측정을 위한 자외선 형광 분석에 관한 연구)

  • In order to determine the contamination of the aromatic hydrocarbons in soil, a fiber-optic sensing technique with fluorescence detector has been proposed. Previous researches have shown that the optimal condition for detecting benzene, toluene, ethylbenzene, xylene (BTEX) was 260 nm /290 nm (excitation/emission wavelength). However, broader fluorescence spectra of BTEX-polluted soil sample ranging from 300 nm to 600 nm were observed. Additionally, the intensity of fluorescence increased with increasing BTEX concentration, which was conspicuous in the fine-particle soil, The overall results indicated that the suggested technique could be useful for in-situ monitoring system for subsurface oil-storage tank.

Optical information storage using diffraction properties of volume hologram in Fe-LiNbO$_3$ crystal (Fe-LiNbO$_3$결정에서 부피형 홀로그램의 회절특성을 이용한 광정보 저장)

  • In this paper, we experiment the characteristics of coupling coefficient, gain, diffraction efficiency and dependence of time determined by TWM(Two-Wave Mixing), using Fe-LiNbO$_3$ crystal(doped with 0.015Wt.%). From these results, we proposed to apply for optical memory application. The highest coupling angle of 14。 and maximum coupling coefficient of 6.9$cm^{-1}$ / are obtained at 514.5nm wavelength. Also, maximum diffraction efficiency is 54.13% when intensity ratio and writing beam incident angle are 0.1 and 14o, respectively. After fixing process, diffraction efficiency is 21.4%. As an example, we demonstrated the writing and reconstruct optical data using spatial light modualtor and angular multiplexing in most optimal condition.

A Comparison Study on Flow-Friction Characteristic of Polymer Solution and Surfactant as Drag Reduction Additive (고분자물질 및 계면활성제의 유동마찰 저감 특성 비교 연구)

  • The drag reduction(DR) for Betaine+Amin and Xantan Gum as kinds of surfactant and Polyacrylamide as kinds of polymer solution according to the fluid velocity, temperature and surfactant concentration were compared experimentally. For this study, two kinds of experimental apparatus for short time and long time measurement were established. Each experimental appratus was equipped with hot water storage tanks, pumps, testing pipe network, flowmeter, two pressure gauges and data logging system was built for them. Results showed that Betaine+Amin and Xanthan Gum as kinds of surfactant had appeared optimal DR around 200-500 ppm and their DR tended to be decreased when flow velocity increased but Polyacrylamide as kinds of polymer solution showed the opposite trend to be increased when flow velocity increased. The both of them showed above 40% DR in the case of better condition by the short term measurement. But Polyacrylamide as kinds of polymer solution showed more degradation than Betaine+Amin and Xanthan Gum as kinds of surfactant by the long term measurement. As a result, Betaine+Amin and Xanthan Gum as kinds of surfactant showed better materials to use to the district heating system.

  • This study was used homomixer as one of the manufacturing method to establish optimal manufacturing condition with magnesium chloride emulsion as coagulant. The more rotational speed of homomixer was fast, the more particle size of magnesium chloride emulsion was minute. The particle size was distributed between 1 and $5{\mu}m$. The more minute particle size of emulsion had an effect on increasing viscosity of emulsion and delay of oil phase separation during storage period, so the quality of magnesium chloride emulsion had correlation with dispersed phase particle size. After all the experiments, when manufacturing magnesium chloride emulsion, it used more than 10,000 rpm of rotational speed of homomixer, it showed the best result as coagulant according to the state of texture and the water separation ratio of soybean curd.

  • In this study, optimization was performed to improve the conventional liquefaction process of offshore plants, such as a LNG-FPSO(Liquefied Natural Gas-Floating, Production, Storage, and Offloading unit) by maximizing the energy efficiency of the process. The major equipments of the liquefaction process are compressors, expanders, and heat exchangers. These are connected by stream which has some thermodynamic properties, such as the temperature, pressure, enthalpy or specific volume, and entropy. For this, a process design problem for the liquefaction process of offshore plants was mathematically formulated as an optimization problem. The minimization of the total energy requirement of the liquefaction process was used as an objective function. Governing equations and other equations derived from thermodynamic laws acted as constraints. To solve this problem, the sequential quadratic programming(SQP) method was used. To evaluate the proposed method in this study, it was applied to the natural gas liquefaction process of the LNG-FPSO. The result showed that the proposed method could present the improved liquefaction process minimizing the total energy requirement as compared to conventional process.

  • Sustainable energy supplies without the recharging and replacement of the charge storage device have become increasingly important. Among various energy harvesters, the triboelectric nanogenerator (TENG) has attracted considerable attention due to its high instantaneous output power, broad selection of available materials, eco-friendly and inexpensive fabrication process, and various working modes customized for target applications. In this study, the amount of voltage and current generated was measured by applying the PSD profile random vibration test of the electronic vibration tester and ISTA 3A according to the time of Anodized Aluminum Oxide (AAO) pore widening of the manufactured TENG device Teflon and AAO. The discharge and charging tests of the integrated module during the random simulated transport environment and the recognition distance of RFID were measured while agricultural products (onion) were loaded into the returnable folding plastic box. As a result, it was found that AAO alumina etching processing time to maximize TENG performance was optimal at 31 min in terms of voltage and current generation, and the integrated module applied with the TENG module showed a charging effect even during the continuous use of RFID, so the voltage was kept constant without discharge. In addition, the RFID recognition distance of the integrated module was measured as a maximum of 1.4 m. Therefore, it was found that the surface condition of AAO, a TENG element, has a great influence on the power generation of the integrated module, and due to the characteristics of TENG, the power generation increases as the surface dries, so it is judged that the power generation can be increased if the surface drying treatment (ozone treatment, etc.) of AAO is applied in the future.

Preservation of Simmental bull sperm at 0℃ in Tris dilution: effect of dilution ratio and long-distance transport

  • Objective: This study aimed to assess the impact of the dilution ratio of Tris diluent, storage at 0℃, and long-distance transportation on the spermatozoa of Simmental cattle. It also validated the feasibility of the regional distribution of fresh semen. Methods: In experiment 1, semen was diluted at four dilution ratios (1:6, 1:9, 1:12, and 1:15) to determine the optimal dilution ratio of Tris diluent. In experiment 2, we assessed sperm viability, progressive motility (objectively assessed by computer-assisted sperm analyzer), and acrosome intactness in Tris dilutions kept at constant 0℃ for 1, 3, 6, 9, and 12 days. We compared them to Tianshan livestock dilutions (Commercial diluent). In experiment 3, semen was diluted using Tris diluent, and sperm quality was measured before and after long-distance transport. Artificial insemination of 177 Simmental heifers compared to 156 using Tianshan Livestock dilution. Results: The outcomes demonstrated that 1:9 was the ideal Tris diluent dilution ratio. The sperm viability, Progressive Motility, and acrosome integrity of both Tris and Tianshan dilutions preserved at 0℃ gradually decreased over time. sperm viability was above 50% for both dilutions on d 9, with a flat rate of decline. The decrease in acrosome integrity rate was faster for Tianshan livestock dilutions than for Tris dilutions when stored at 0℃ for 1 to 6 days. There was no significant difference (p>0.05) in sperm viability between semen preserved in Tris diluent after long-distance transportation and semen preserved in resting condition. The conception rates for Tris dilution and Tianshan livestock dilution were 49.15% and 46.15% respectively, with no significant difference (p>0.05). Conclusion: This shows that Tris diluent is a good long-term protectant. It has been observed that fresh semen can be successfully preserved for long-distance transport when stored under 0℃ conditions. Additionally, it is feasible to distribute semen regionally.

The 'Consequence Analysis' of Variables Affecting the Extent of Damage Caused by Butane Vapor Cloud Explosions (부탄가스 증기운폭발의 피해범위에 영향을 미치는 변수에 관한 고찰)

  • This paper presents a 'consequence analysis' for vapor cloud explosions caused by heavy gas leakages from commercially used storage tanks at petrochemical plants. Particularly, this paper emphasizes on evaluating the results of various vapor cloud explosion accidents from Butane storage tanks. Also this paper analyses the impact of variables on the accidents in order to acquire the optimum conditions for variables. $SuperChems^{TM}$ Professional Edition was applied to analyse the impact (If atmospheric and other variables in the situation where vapor cloud continuously disperses from the ground level. Under the assumption that practical operating conditions are selected as a standard condition, and Butane leaks from the storage tank for 15 minutes, the results show that the maximum distance of LFL (Lower Flammable Limit) was 52 meters and overpressure by the vapor cloud explosion was 1 psi at 128.2 meters. It is observed that the impact of the variables on accidental Butane storage tank leakage mainly varied upon atmospheric stability, wind velocity, pipe line size, visible length, etc., and changes in the simulation result occurred as the variables varied. The maximum distance of the LFL (Lower Flammable Limit) increased as the visible length became shorter, the size of the leak became larger, the wind velocity was decreased, and the climatic conditions became more stable. Thus, by analysing the variables that influence the simulation results of explosions of Butane storage tanks containing heavy gases, I am presenting the most appropriate method for 'consequence analysis' and the selection of standards for suitable values of variables, to obtain the most optimal conditions for the best results.

  • In SS branch of Korea District Heating Corporation, Combined Heat & Power power plant with 99MW capacity and 98Gcal / h capacity is operated as a district electricity business. In this region, it is difficult to operate the generator due to the problem of surplus heat treatment between June and September due to the economic recession and the decrease in demand, so it is urgent to develop an economical energy new business model. In this study, we will develop an optimized operation model by introducing a renewable energy hybrid system based on actual operation data of this site. In particular, among renewable energy sources, fuel cell (Fuel Cell) power generation which can generate heat and electricity at the same time with limited location constraints, photovoltaic power generation which is representative renewable energy, ESS (Energy Storage System). HOMER (Hybrid Optimization of Multiple Energy Resources) program was used to select the optimal model. As a result of the economic analysis, 99MW CHP combined cycle power generation is the most economical in terms of net present cost (NPC), but 99MW CHP in terms of carbon emission trading and renewable energy certificate And 5MW fuel cells, and 521kW of solar power to supply electricity and heat than the supply of electricity and heat by 99MW CHP cogeneration power, it was shown that it is economically up to 247.5 billion won. we confirmed the results of the improvement of the zone electricity business condition by introducing the fuel cell and the renewable energy hybrid system as the optimization process model.
