  • This paper focuses on disassembly sequencing, which is the problem of determining the optimum disassembly level and the corresponding disassembly sequence for a product at its end-of-life with the objective of maximizing the overall profit. In particular, sequence-dependent operation times, which frequently occur in practice due to tool-changeover, part reorientation, etc, are considered in the parallel disassembly environment. To represent the problem, a modified directed graph of assembly states is suggested as an extension of the existing extended process graph. Based on the directed graph, the problem is transformed into the shortest path problem and formulated as a linear programming model that can be solved straightforwardly with standard techniques. A case study on a photocopier was done and the results are reported.

Mornitoring and Identification of Human Astrovirus from Groundwater in Korea Based on Highly Sensitive RT-nested PCR Primer Sets

  • Human Astrovirus (HuAstV) is an important gastrointestinal pathogen that is frequently reported worldwide. Monitoring of contaminated groundwater has been suggested since HuAstV is transmitted through the fecal-oral route. This study developed a test method based on conventional reverse transcription (RT)-nested polymerase chain reaction (PCR) that involves SL® non-specific reaction inhibitor for unknown non-specific amplification taking place in the groundwater environment. An optimal method for detecting HuAstV in groundwater sample through analysis and comparison against conventionally reported method was also suggested. The developed method enabled the production of nested PCR amplicon of 630 nt, which is a sufficient length for similarity analysis based on sequencing and genotyping. Amplicons suspected to be HuAstV were amplified in two out of the twenty groundwater samples collected in Korea, presenting 99.77% and 99.73% similarity against HuAstV 1 strain lhar/2011/kor (JN887820.1) in sequencing, respectively. These amplicons were identified as HuAstV 1.

연속 회분식 반응기에서 최적 질소 제거를 위한 최적 궤적 찾기와 재최적화 (Optimal Trajectory Finding and re-optimization of SBR for Nitrogen Removal)

  • 본 연구는 생물학적 폐수 처리 공정인 연속 회분식 반응기(sequencing batch reactor, SBR)에서 질소 제거 최적화를 위해 활성 오니 공정모델(activated sludge model, ASM No.1, ASM1)과 반복 동적 프로그래밍(iterative dynamic programming, IDP)을 이용하여 SBR의 처리 기준을 만족하면서 최적 운전 조건을 탐색하고 하는 것을 목적으로 하였다. 연속 회분식 반응기의 최적화를 위해 에너지 최소화와 최소 회분 시간이 질소 처리의 농도 그래프의 면적과 비례하는 점을 이용하여 이를 고려한 새로운 performance index를 제안하였다. 회분 시간과 에너지에 대항하는 면적에 적절한 비중(weight)을 줌으로써 최소 회분 시간과 최소 에너지 문제를 동시에 고려하였다. SBR에서 IDP를 이용한 최적 운전서 최적 용존 산소 농도의 설정치가 전체 회분 시간과 전체 에너지 비용에 동시에 영향을 미침을 알 수 있었고 최적 운전시 기존의 운전 방법과 같은 유기물과 질소 제거가 가능하고 동시에 전체 비용을 20%까지 줄일 수 있었다. 더 나아가 공정이상으로 실제 공정이 모델과 다른 모델링 에러에 의해 잘못된 모사의 경우에도 IDP를 이용하여 다시 재최적화할 수 있음을 보였다.

Enhanced Carboxymethylcellulase Production by a Newly Isolated Marine Bacterium, Cellulophaga lytica LBH-14, Using Rice Bran

  • The aim of this work was to establish the optimal conditions for production of carboxymethylcellulase (CMCase) by a newly isolated marine bacterium using response surface methodology (RSM). A microorganism producing CMCase, isolated from seawater, was identified as Cellulophaga lytica based 16S rDNA sequencing and the neighborjoining method. The optimal conditions of rice bran, ammonium chloride, and initial pH of the medium for cell growth were 100.0 g/l, 5.00 g/l, and 7.0, respectively, whereas those for production of CMCase were 79.9 g/l, 8.52 g/l, and 6.1. The optimal concentrations of $K_2HPO_4$, NaCl, $MgSO_4{\cdot}7H_2O$, and $(NH_4)_2SO_4$ for cell growth were 6.25, 0.62, 0.28, and 0.42 g/l, respectively, whereas those for production of CMCase were 3.72, 0.54, 0.70, and 0.34 g/l. The optimal temperature for cell growth and the CMCase production by C. lytica LBH-14 were $35^{\circ}C$ and $25^{\circ}C$, respectively. The maximal production of CMCase under optimized condition for 3 days was 110.8 U/ml, which was 5.3 times higher than that before optimization. In this study, rice bran and ammonium chloride were developed as carbon and nitrogen sources for the production of CMCase by C. lytica LBH-14. The time for production of CMCase by a newly isolated marine bacterium with submerged fermentations reduced to 3 days, which resulted in enhanced productivity of CMCase and a decrease in its production cost.

Bacterial Carotenoids를 생산하는 광합성세균 Erythrobacter longus SY-46의 분리 및 특성 (Isolation and Characteristics of Photosynthetic Bacterium, Erythrobacter longus SY-46 which Produces Bacterial Carotenoids)

  • The aerobic photosynthetic bacterium, which produces bacterial carotenoids was isolated and identified from coastal marine environments. This bacterium was identified by 16S rDNA sequencing and designated as Erythrobacter longus SY-46. E. longus SY-46 was Gram negative and rod shape, and the optimal culture conditions were $25^{\circ}C$, pH 7.0, and 3.0% NaCl concentration, respectively. The carbon and nitrogen sources required for the optimal growth were lactose and tryptone, respectively. Fatty acid compositions of E. longus SY-46 were $C_{18:1}$(78.32%), v-linolenic acid($C_{18:3n9.12.15c}:3.83%$), margaric acid($C_{17:0}$: 3.38%), palmitic acid($C_{16:0}$: 3.07%), and docosahexaenoic acid($C_{22:6n3}$: 2.21%). In addition, E. longus SY-46 showed the characteristic absorption peaks of bacterial carotenoids(in the region of 450 to 480 nm) and bacteriochlorophyll(770 to 772 nm). Major carotenoids of E. longus SY-46 were polyhydroxylated xanthophylls such as fucoxanthin and zeaxanthin.

  • Though multi-panel structures lined with a poroelastic material have been widely used to reduce sound transmission in various fields, most of the previous works to design them were conducted by repeated analyses or experiments based on initially given configurations or sequences. Therefore, it was difficult to obtain the optimal sequence of multi-panel structures lined with a poroelastic material yielding superior sound isolation capability. In this work, we propose a new design method to sequence a multi-panel structure lined with a poroelastic material having maximized sound transmission loss. Being formulated as a one-dimensional topology optimization problem for a given target frequency, the optimal sequencing of panel-poroelastic layers is systematically carried out in an iterative manner. In this method, a panel layer is expressed as a limiting case of a poroelastic layer to facilitate the optimization process. This means that main material properties of a poroelastic material are treated as Interpolated functions of design variables. The designed sequences of panel-poroelastic layers were shown to be significantly affected by the target frequencies; more panel layers were used at higher target frequencies. The sound transmission loss of the system was calculated by the transfer matrix derived from Biot's theory.

Purification and Characterization of an Antibacterial Substance from Aerococcus urinaeequi Strain HS36

  • A bacterial strain inhibiting the growth of Vibrio anguillarum, the causative agent of vibriosis, was isolated from fish intestines. The isolated strain HS36 was identified as Aerococcus urinaeequi based on the characteristics of the genus according to Bergey's Manual of Systematic Bacteriology and by 16S rRNA sequencing. The growth rate and antibacterial activity of strain HS36 in shaking culture were higher than those in static culture, while the optimal pH and temperature for antibacterial activity were 7.0 and 30℃, respectively. The active antibacterial substance was purified from a culture broth of A. urinaeequi HS36 by Sephadex G-75 gel chromatography, Sephadex G-25 gel chromatography, and reverse-phase high-performance liquid chromatography. Its molecular weight, as estimated by Tricine SDS-polyacrylamide gel electrophoresis, was approximately 1,000 Da. The antibacterial substance produced by strain HS36 was stable after incubation for 1 h at 100℃. Although its antibacterial activity was optimal at pH 6-8, activity was retained at a pH range from 2 to 11. The purified antibacterial substance was inactivated by proteinase K, papain, and β-amylase treatment. The newly purified antibacterial substance, classified as a class II bacteriocin, inhibited the growth of Klebsiella pneumoniae, Salmonella enterica, and Vibrio alginolyticus.

Isolation and Culture Properties of a Thermophilic Agarase-Producing Strain, Microbulbifer sp. SD-1

  • An agar-degrading enzyme-producing strain was isolated from seawater. The isolate was identified as Microbulbifer sp. SD-1 by 16S rRNA sequencing analysis. The optimal pH and temperature for growth were 6.0 and $30^{\circ}C$, respectively, and growth was possible at pH 9.0 and $60^{\circ}C$. The isolate required 5% NaCl for optimal growth and showed 45% growth activity without NaCl. Agar concentrations of 0-0.4% in the medium did not affect growth. Thin-layer chromatography analysis revealed that this strain could degrade agar into a monosaccharide and oligosaccharide, which may have industrial applications.

Common Due-Date Assignment and Scheduling with Sequence-Dependent Setup Times: a Case Study on a Paper Remanufacturing System

  • In this paper, we report a case study on the common due-date assignment and scheduling problem in a paper remanufacturing system that produces corrugated cardboards using collected waste papers for a given set of orders under the make-to-order (MTO) environment. Since the system produces corrugated cardboards in an integrated process and has sequence-dependent setups, the problem considered here can be regarded as common due-date assignment and sequencing on a single machine with sequence-dependent setup times. The objective is to minimize the sum of the penalties associated with due-date assignment, earliness, and tardiness. In the study, the earliness and tardiness penalties were obtained from inventory holding and backorder costs, respectively. To solve the problem, we adopted two types of algorithms: (a) branch and bound algorithm that gives the optimal solutions; and (b) heuristic algorithms. Computational experiments were done on the data generated from the case and the results show that both types of algorithms work well for the case data. In particular, the branch and bound algorithm gave the optimal solutions quickly. However, it is recommended to use the heuristic algorithms for large-sized instances, especially when the solution time is very critical.
