In this study, cattle microsatellite markers recommended for diversity studies of cattle by the EU AIRE 2066 Concerted Action Group were used to study the genetic diversity of 105 Thai swamp buffalo which were randomly selected from eight different research stations of the Department of Livestock Development, Thailand. Of 34 primer pairs, 16 were successfully amplified while the rest showed non-specific amplification. The lowest number of alleles was two while the highest was nine, with an average of 4.7 alleles per locus. The average unbiased heterozygosity for all eight populations was 0.5233, with a low of 0.4772 (Samui) and a high of 0.5616 (Burirum). The genetic distance ranged from 0.0574 to 0.2575. Populations from Lopburi and Burirum showed the closest relationship, whereas Srisagat and Samui were the most divergent. The results generated with the primers recommended by the EU AIRE 2066 Concerted Action Group are at a slight variance from our previous study, possibly as a result of the number of specific amplification products obtained, suggesting that cattle markers may not be optimal for studies of the genetic diversity of the Thai swamp buffalo.
The present study describes the gene cloning, overexpression and characterization of a novel nitrilase from hyperthermophilic bacterium Thermotoga maritima MSB8. The nitrilase gene consisted of 804 base pairs, encoding a protein of 268 amino acid residues with a molecular mass of 30.07 kDa after SDS-PAGE analysis. The optimal temperature and pH of the purified enzyme were 45℃ and 7.5, respectively. The enzyme demonstrated good temperature tolerance, with 40% residual activity after 60 min of heat treatment at 75℃. The kinetic constants Vmax and Km of this nitrilase toward 3-cyanopyridine were 3.12 μmol/min/mg and 7.63 mM, respectively. Furthermore, this novel nitrilase exhibited a broad spectrum toward the hydrolysis of the aliphatic nitriles among the tested substrates, and particularly was specific to aliphatic dinitriles like succinonitrile, which was distinguished from most nitrilases ever reported. The catalytic efficiency kcat/Km was 0.44 /mM/s toward succinonitrile. This distinct characteristic might enable this nitrilase to be a potential candidate for industrial applications for biosynthesis of carboxylic acid.

Overlay routing has emerged as a promising approach to improve reliability and efficiency of the Internet. The key to overlay routing is the placement and maintenance of the overlay infrastructure, especially, the selection and placement of key relay nodes. Spurred by the observation that a few relay nodes with high betweenness centrality can provide more optimal routes for a large number of node pairs, we propose a resilient routing overlay network construction method by introducing Super-Relay nodes. In detail, we present the K-Minimum Spanning Tree with Super-Relay nodes algorithm (SR-KMST), in which we focus on the selection and connection of Super-Relay nodes to optimize the routing quality in a resilient and scalable manner. For the simultaneous path failures between the default physical path and the overlay backup path, we also address the selection of recovery path. The objective is to select a proper one-hop recovery path with minimum cost in path probing and measurement. Simulations based on a real ISP network and a synthetic Internet topology show that our approach can provide high-quality overlay routing service, while achieving good robustness.

This paper proposes a two-phase genetic algorithm (GA) to solve the problem of obtaining an optimum configuration of a paired single row assembly line. We pair two single-row assembly lines due to the shared usage of several workstations, thus obtaining an optimum configuration by considering the material flow of the two rows simultaneously. The problem deals with assigning workstations to a sequence and selecting the best arrangement by looking at the length and width for each workstation. This can be considered as an enhancement of the single row facility layout problem (SRFLP), or the so-called paired SRFLP (PSRFLP). The objective of this PSRFLP is to find an optimal configuration that seeks to minimize the distance traveled by the material handler and even the use of the material handler itself if this is possible. Real-world applications of such a problem can be found for apparel, shoe, and other manual assembly lines. This research produces the schematic representation solution using the heuristic approach. The crossover and mutation will be utilized using the schematic representation solution to obtain the neighborhood solutions. The first phase of the GA result is recorded to get the best pair. Based on these best matched pairs, the second-phase GA can commence.
The chitinase-producing strain SC081 was isolated from Korean traditional soy sauce and identified as Bacillus atrophaeus based on a phylogenetic analysis of the 16S rDNA sequence and a phenotypic analysis. A gene encoding chitinase from B. atrophaeus SC081 was cloned in Escherichia coli and was named SCChi-1 (GQ360078). The SCChi-1 nucleotide sequences were composed of 1788 base pairs and 596 amino acids, which were 92.6, 89.6, 89.3, and 78.9% identical to those of Bacillus subtilis (ABG57262), Bacillus pumilus (ABI15082), Bacillus amyloliquefaciens (ABO15008), and Bacillus licheniformis (ACF40833), respectively. A recombinant SCChi-1 containing a hexahistidine tag at the amino-terminus was constructed, overexpressed, and purified in E. coli to characterize SCChi-1. $H_6SCChi$-1 revealed a hydrolytic band on zymograms containing 0.1% glycol chitin and showed the highest lytic activity on colloidal chitin and acidic chitosan. The optimal temperature and pH for chitinolytic activity were $50^{\circ}C$ and pH 8.0, respectively.

Wireless mesh networks (WMNs) based on IEEE 802.11s have emerged as one of the prominent technologies in multi-hop communications. However, the deployment of WMNs suffers from serious interference problem which severely limits the system capacity. Using multiple radios for each mesh router over multiple channels, the interference can be reduced and improve system capacity. Nevertheless, interference cannot be completely eliminated due to the limited number of available channels. An effective approach to mitigate interference is to apply dynamic channel switching (DCS) in WMNs. Conventional DCS schemes trigger channel switching if interference is detected or exceeds a predefined threshold which might cause unnecessary channel switching and long protocol overheads. In this paper, a P-learning based dynamic switching algorithm known as learning automaton (LA)-based DCS algorithm is proposed. Initially, an optimal channel for communicating node pairs is determined through the learning process. Then, a novel switching metric is introduced in our LA-based DCS algorithm to avoid unnecessary initialization of channel switching. Hence, the proposed LA-based DCS algorithm enables each pair of communicating mesh nodes to communicate over the least loaded channels and consequently improve network performance.

In this paper, a microstripline-type linear amplitude equalizer is implemented to achieve the linearity of the slope of the amplitude over $6{\sim}18$ GHz with resistors inserted between their immediate pairs of a TX-line and a stub. The values of resistors and stubs are explored to have the optimal linear slope and return loss performance. Experiments reveal the slope of around 9 dB variation and return loss of less than 10 dB desired over $6{\sim}18$ GHz and validate the design methodology.
Conventional cantilevered piezoelectric energy harvesters (PEHs) are usually fabricated with continuous electrode configuration (CEC), which suffers from the electrical cancellation at higher vibration modes. Though previous research pointed out that the segmented electrode configuration (SEC) can address this issue, a comprehensive evaluation of the PEH with SEC has yet been reported. With the consideration of delivering power to a common load, the AC outputs from all segmented electrode pairs should be rectified to DC outputs separately. In such case, theoretical formulation for power estimation becomes challenging. This paper proposes a method based on equivalent circuit model (ECM) and circuit simulation to evaluate the performance of the PEH with SEC. First, the parameters of the multi-mode ECM are identified from theoretical analysis. The ECM is then established in SPICE software and validated by the theoretical model and finite element method (FEM) with resistive loads. Subsequently, the optimal performances with SEC and CEC are compared considering the practical DC interface circuit. A comprehensive evaluation of the advantageous performance with SEC is provided for the first time. The results demonstrate the feasibility of using SEC as a simple and effective means to improve the performance of a cantilevered PEH at a higher mode.
A tooth profile modification for loaded gears is used to avoid a tooth impact. Since a tooth profile error causes amplification of the cumbersome whine noise in automotive gear transmissions, an optimal quantity of tooth profile modifications must be obtained for good performance in the vibration sense. In this paper, a tooth profile modification curve considering profile manufacturing errors and elastic deformation of the gear tooth is formulated; in addition, transmission errors of the gear system with modified teeth are verified. The equivalent excitation due to transmission errors is formulated. For experimental evaluation of the transmission error, the transmission error for a simple gear system was measured by two rotational laser vibrometers. Finally, we perform a comparative analysis between the calculated and measured responses to the excitations due to the transmission error to verify the practicability of the application to automotive transmissions.
The effect of galleries on the earthquake behavior of dams should be investigated to obtain more realistic results. Therefore, a roller compacted concrete (RCC) dam with and without galleries are examined under ground motion effects. For this purpose, Cine RCC dam constructed in Aydın, Turkey, is selected in applications. The optimal mesh around galleries is investigated to obtain the most realistic results. Two-dimensional finite element models of Cine RCC dam with and without galleries are prepared by using ANSYS software. Empty and full reservoir conditions were taken into account in the time-history analyses. Hydrodynamic effect of the reservoir water was taken into account considering two-dimensional fluid finite elements based on the Lagrangian approach. It is examined that how principle stresses and displacements change by height and during earthquake. The dam-foundation-reservoir interaction was taken into consideration with contact-target element pairs. The displacements and principle stress components obtained from the linear analyses are compared each other for various cases of reservoir water and galleries. According to numerical analyses, the effect of galleries is clear on the response of RCC dam. Besides, hydrodynamic water effect obviously increases the principle stress components and horizontal displacements of the dam.
