Optimal Medium Conditions for the Detection of Cellulolytic Activity in Ganoderma lucidum

  • To determine the optimal medium conditions for the detection of the cellulolytic activity in Ganoderma lucidum, we varied three media conditions: dye reagent, pH, and temperature. First, we evaluated the use of four dyes, Congo Red, Phenol Red, Remazol Brilliant Blue, and Trypan Blue. To observe the effect of pH on the chromogenic reaction, we also made and tested various media spanning acidic and alkaline pHs, ranging from 4.5 to 8.0. Furthermore, in order to research the effect of temperature on the clear zone and the fungus growing zone, we tested temperatures ranging from 15 to $35{^{\circ}C}$. On the whole, the best protocol called for Ganoderma lucidum transfer onto media containing Congo red with pH adjusted to 7.0, followed by incubation at $25{^{\circ}C}$ for 5 days. Our results will be useful to researchers who aim to study extracellular enzyme activity in Ganoderma lucidum.

생쥐 체외수정란의 초급속동결 및 이식에 관한 연구 I. pH, 삼수압 및 정자 전배양처리가 생쥐 체외수정율에 미치는 영향 (Studies on Transfer of In Vitro Fertilized Mouse Embryos Following Ultrarapid Freezing I. Effect of Treatment of pH, Osolality and Sperm Preincubation on In VitroFertilization Rate of Mouse Embryos)

  • These studies were carried out to investigate optimal physological conditions for in vitro fertilization (IVF) of mouse ova. The unfertilized ova were obtained by superovulation from ICR mice of 4 to 6 weeks old. Tyrode's 280 solution was used as basal media, and pH and osmolality of basal media were adjusted with the supplementation of sodium bicarbonate and sodium chloride, respectively. The optimal pH, and osmolality of culture media and the optimum period of sperm preincubation were examined in fertilization in vitro of mouse ova and the subsequent culture in vitro of embryos. The pH range of media examined was designed from 6.5 to 7.5 with 0.2 interval and the range of osmolality from 250 to 370 mOsm with 20 interval, and the period of sperm preincubation examined was 30, 60, 120, and 180 minutes. The ova developed to 2-cell embryosafter 26hrs. of incubation with preincubated sperm were evaluated as in vitro fertilized ones. The results obtained were summarized as follows: 1. The percentage of in vitro fertilized ova was highest (64.7%) in media of pH 7.1 and lowest (38.0%) in pH 6.7. No significant difference in % fertilized ova was found from the media of pH 7.1 to 7.5. Compared with the result from pH 7.1 medium, the pollyspermy was increased signifciantly (p<0.05) in the media of pH over 7.5 and below 6.9;, and the % degenerated ova was significantly (p<0.05) increased in the media of pH below 6.9. 2. The percentage of in vitro fertilized ova was highest (69.4%) in media of osmolality 330 mOsm and lowest (47.9%) in osmolality 250 mOsm. No significant difference in % fertilized ova was found from the media of osmolality 310 to 350 mOsm. Compared with the result from osmolality 330 mOsm in medium, the polyspermy aws increased significantly(p<0.05) in the media of osmolality over 350 mosmol and blow 290 mOsm, and the % degenerated ova was significantly (P<0.05) increased in the media of osmolality below 290 mOsm. 3. The percentate of in vitro fertlilized ova was highest (62.7%) in media of period sperm preincubation 180 min. and lowest (40.4%) in sperm preincubation 30 minutes. No significant difference in % fertilized ova was found from the media of sperm preincubation 120 to 180 minutes. Compared with the result from sperm preincubation 180 minutes in medium, the polyspermy was low differ no significantly(P<0.05) in the media of period sperm preincubation, and the % degenerated ova was signifciantly(P<0.05) increased in the media of sperm presincubation below 60 minutes.

The Optimal Culture Conditions Affecting the Mycelial Growth and Formation of Paecilomyces fumosoroseus

  • The fruiting body of Paecilomyces fumosoroseus was collected at Mt. Mani, Ganghwa Island, Korea in September, 2001. This study was carried out to obtain the basic informations for the mycelial growth and fruiting body production of P. fumosoroseus in artificial media. The optimal conditions for the mycelial growth were obtained at $25^{\circ}C$ and in the range of pH $6{\sim}9$, respectively. P. fumosoroseus showed the favorable growth on Hamada medium. The carbon and nitrogen source favorable for mycelial growth were dextrin and histidine, respectively. Optimum C/N ratio suitable for optimal growth of P. fumosoroseus was observed on the culture media adjusted to the ratio of 40:1. The mycelial growth of P. fumosoroseus was optimal on corn meal agar supplemented with 30% of silkworm pupae. The most favorable fruiting body formation of P. fumosoroseus was obtained in the medium containing unpolished rice supplemented with 20%(w/w) silk worm pupae at $25^{\circ}C$ under 100 lux.

느타리버섯 압축배지 재료에 대한 최적 배합 연구 (The optimum material mixture rate of the pressing media in Pleurotus ostreatus)

  • 느타리버섯 균사 배양에 배지의 최적 혼합비를 구하기 위해 주재료인 면실피, 톱밥, 볏짚과 부재료인 제올라이트, 옥수수강, 탈지강, 백솜, 이분, 왕겨, 피이트를 사용하였다. 주재료인 면실피 톱밥 : 볏짚의 최적 혼합비는 6: 3: 1 (V/V, %)이었으며 각각 부재료의 최적 혼합비는 제올라이트 1%, 옥수수강 3%, 탈지강 5%, 백솜 1%, 이분 1%, 왕겨 7%의 순으로 균사생장과 밀도가 양호하였다. 주재료에 부재료 두 종류씩 혼합하였을때 최적 혼합비는 제올라이트:옥수수강의 경우 2:2(V/V, %), 탈지강과 백솜의 경우 3:2(V/V, %), 이분과 왕겨의 경우 1:3(V/V, %)이었다. 이를 다시 주재료와 부재료 모두를 함께 혼합할 경우 면실피 :톱밥 : 볏짚 : 제올라이트 : 옥수수강 : 탈지강 : 백솜 : 이분 : 왕겨 : 피이트를 43.0 : 17.2 : 25.8 : 2.0 : 2.0 : 3.0 : 2.0 : 1.0 : 3.0 1.0(V/V, %)로 혼합하였을 때 균사생장과 밀도가 모두 양호하였다.

조영제 부작용환자의 적절한 병원내 관찰시간에 대한 고찰 (The Optimal Length of Time to Observe Patients with Contrast Media Anaphylaxis in the Emergency Department)

  • Purpose: The purpose of this study was to determine the optimal length of time to observe patients with contrast media anaphylaxis in the Emergency Department. Methods: This study included the patients of all age groups who presented with anaphylaxis due to contrast media to the ED of Pusan National University Hospital from January 2006 to August 2009. The medical records were retrospectively reviewed. We analyzed the 100 patients who met the inclusion criteria. Their median age was 55 (46-62), and 38 out of the 100 patients (38%) were male. Results: The most common symptom of patients was urticaria. Corticosteroid and H1 antihistamine were given to most of the patients in the ED. All were discharged after observation in the ED and none were admitted to the ward. The average time from injection of the contrast media to the onset of symptom was 31 minutes (24-39) and the average time to symptom recovery was 127 minutes (89-188). The mean observation time in the ED was 93 minutes (59-153). Biphasic reactions were reported in only one case (1/100, 1%). Conclusion: These results suggest that patients who present with an anaphylactic reaction after radiologic studies that use intravenous contrast media can be safely discharged early from the ED because the symptom recovery time is not long and a biphasic reaction is rare.

Optimal Configuration of SVC for Satellite Broadcasting Service with Ku/Ka Bands

  • The study investigated the optimal configuration of SVC (Scalable Video Coding) to apply to the satellite broadcasting service, and compared the performance of the SVC with that of the AVC (Advanced Video Coding). To get the optimal configuration, we analyzed the optimal bit rate allocation between the layers and the optimal scalability which requires the least bit rate for the required PSNR for various kinds of contents using JSVM. As a result of investigation, we found that the optimal bit rate allocation occurs when the bit rate of the base layer is minimum, and the spatial scalability shows the best performance. The performance of SVC is similar to that of AVC for spatial scalability, but it depends on contents.

  • Constructed wetland has been widely used for the treatment of sewage, stormwater runoff, industrial wastewater, agricultural runoff, acid mine drainage and landfill leachate. For the removal of nitrogen and phosphorus, uptake by plants and adsorption to media material are the major processes, and, therefore, the selection of media with specific adsorption capacity is the critical factor for the optimal design of wetland along with the selection of appropriate plant species. In this study, two media materials (loess bead and mixed media) possessing different adsorption characteristics for ammonium and phosphate were selected, and their adsorption characteristics were evaluated. In addition, the performance of subsurface-flow wetland systems employing these media was evaluated in both batch and continuous flow systems. With LB medium, beter phosphorus removal was observed, while better ammonia removal was obtained with MM medium. In addition, enhanced removal efficiencies were observed in the wetland systems employing both media and aquatic plants, mainly due to the better environment for microbial growth. As a result, appropriate selection or combination of media with respect to the inflow water quality maybe important factors for the successful design and operation of wetland systems.

  • With increasing social interest in health and health promotion, the government and many organizations are conducting various health campaigns for the public. The public health campaign is aimed at protecting the people from the dangers of disease and contributing to a healthy life and also help establishing a healthy attitude and changing behavior as well. In addition, many researches have been carried out in order to enhance the advertising effect of campaigns aimed at forming preventive attitudes and to verify it by applying many theories. However, as a result, there is a significant lack of research regarding analysis of differences in the effectiveness of the campaign by media. This study is to analyze the effect of health campaign by the type of media which published health campaign advertisements that can affect prevention attitude. To meet the purpose of this study, The 15 print media were to examine the impact of media characteristics and types on participation in campaigns for health campaigns. Through this, we will present the role of the media as an efficient channel to encourage the formation and participation of the health attitude of campaign advertisements, and present significant implications in the selection of optimal media and the execution of campaign budgets.

  • The study was conducted to determine the optimal basal media and the concentration of plant growth regulator for germination of seeds and growth of plantlet from Dendrobium monile. The results obtained were summarized as follows. Germination was similar in light and dark condition, but the growth of plantlet after germination was better under dark than under light condition in several media. Germination was best in Hyponex and Kyoto solution medium among the 9 media tested. The number of roots/shoot was most in the Hyponex medium containing 0,1ppm NAA and 1.Oppm BA.

  • In industrial education which would meet both ends to offer adult-learner (employee) the opportunity to develop himself and to accomplish business aims, it is very important to use instructional media and to prepare the proper educational facilities and equipments. Therefore, this paper would investigate the following aspects. First, the characteristics and optimal training method of industrial education as an adult-training. Secondly, the meaning of instructional media. Thirdly the contents of proper educational facilities and equipments to apply instructional media. Based on the above mentioned investigation, this paper suggests that instructional media and educational facilities and equipments in industrial education should be utilized integratedly and systematically as component factors of total educational program. And then, the basic directions and ways to establish the most desirable educational facilities and equipments are presented in this paper.
