It was studied in order to improve the yield of serrapeptase production in fermentation that organic nitrogen sources play important roles not only as inducer, repressor and activator, but also nitrogen sources. From the investigation of the effect of Na-caseinate on the induction of serrapeptase production, it was elucidated that real inducer was leucine and strong repressor was cysteine, which were produced through hydrolysis of proteins. Serrapeptase production was strongly induced by Na-caseinate in culture time 12 hrs, but was weakly induced before and after that time. Therefore fed batch culture where partial amount of Na-caseinate is added in 12 hrs, is better than batch culture where total amount of Na-caseinate is added at the beginning. Cysteine, methionine, MgSO$_{4}$, and so on, sulfur-containing materials, repressed the serrapeptase production. In the addition of mineral salts, chlorinated salts is better than sulfated salts because of sulfur repression. The synergic effect of soybean meal with Na-caseinate on the serrapeptase production resulted from Mn$^{2+}$ contained in soybean meal, of which the optimal concentration is 4 mM in enzyme production.

This study was conducted to increase the amount of bio-hydrogen production from microalgae(Chlorella vulgaris) in batch reactors by thermal pre-treatment. The optimization of thermal pre-treatment was conducted using statistic experimental design of response surface methodology. Two experimental parameters of temperature and reaction time were considered. The optimization condition was founded at the coded variables of <0.52, -0.07> corresponding to the experimental of heating temperature of $95.6^{\circ}C$ and reaction time of 57.9 min, respectively. Under the optimal condition, the maximum hydrogen production was predicted to 25.3mL $H_2/g$ dry cell weight (dcw), which was 9.1 times higher value of control(2.8mL $H_2/g$ dcw).
The optimization of fed-batch yeast fermentation process has been performed using genetic algorithm(GA). Three strategies were designed and applied to obtain the optimal feed rate profiles. Genes in the chromosome (input variables for optimization) included feed rates on fixed time intervals (strategy I), or swiching times $t_s1\;and\;t_s2$, and feed rates on singular arc (strategy II), or feed rates and the length of time interval (strategy III). Strategy III showed the best results for all initial conditions due to efficient utilization of genetic information. Simulation results using GA showed similar or better performance compared with previous results by variational caculus and singular control approach.
Whole horse beans (Vicia faba cv. Alfred) (WHB) were pressure toasted at different temperatures of 100, 118 and $136^{\circ}C$ for 3, 7, 15 and 30 minutes in order to determine an optimal heating conditions to increase bypass starch (BPSt) as glucose source which is usually limiting nutrient in highly producing dairy cows in the Netherlands. Starch (St) Ruminal Degradative Kinetics and Fermentation Characteristics of (SRDC) of WHB were determined using in sacco technique in 4 lactating dairy cows fed 47% hay and 53% concentrate according to Dutch dairy cow requirements. Measured characteristics of St were soluble fraction (S), potentially degradable fraction (D) and rate of degradation (Kd) of insoluble but degradable fraction. Based on measured characteristics, percentage bypass starch (BPSt) was calculated according to the Dutch new feed evaluation system: the DVE/OEB system. Pressure toasting temperatures significantly affected starch gelatinization (p<0.01). Degradability of Starch in the rumen was highly reduced by pressure toasting (p<0.01). S varied from 58.2% in the raw WHB (RWHB as a control) to 19.6% in $136^{\circ}C/15min$. S was reduced rapidly with increasing time and temperature (p<0.01). D varied from 41.8% in RWHB to 80.5% in $136^{\circ}C/15min$. D fraction was enormously increased with increasing time and temperature (p<0.01). Kd varied from 4.9%h in RWHB to 3.4%/h in $136^{\circ}C/15min$. All these effects resulted in increasing %BPSt from 29.0% in RWHB to 53.1% in $136^{\circ}C/15min$. Therefore BPSt increased from 93.5 g/kg in RWHB to 173.5 g/kg in $136^{\circ}C/15min$. The effects of pressure toasting on %BPSt and BPSt seemed to be linear up to the highest values tested. Therefore no optimal pressure toasting conditions could be determined at this stage. But among 10 treatments, The treatment of $136^{\circ}C/15min$was the best with the highest BPSt content. It was concluded that pressure toasting was effective in shifting starch degradation from rumen to small intestine to increase bypass starch.
Heating characteristics of red pepper paste packed in report pouches of various thicknesses and their storage stability were investigated to determine the adequate processing conditions that good bacteriological safty and minimal quality changes could be obtained, when sterilized by using a steam-air system retort. A heat penetration into pouch-packed red pepper paste was carried out through by a conductive heat transfer, indicating a simple logarithmic heating curve, and the smaller thickness revealed the higher heat penetration rate, suggesting the possibility of high temperature-short time sterilization of red pepper paste. The processing conditions with Fo-value of 4.5 or higher were sufficient for keeping up bacterial safty, but based on C-value, better quality retention was obtained at pouch thickness of 15mm under the processing temperature of $120^{\circ}C$. Subsequent storage study revealed that the red pepper paste packed in 15mm and processed at $120^{\circ}C$ with Fo=4.5 could be held without any spoilage and overall acceptance change, when stored for 6 months at room temperature under the relative humidity of 70%. After 3 months storage in $38^{\circ}C$ under saturation humidity, overall acceptance of red pepper paste were judged not to be maintainable on the acceptable level, but it may be suggested that above the results could be kept up a desirable quality without any remarkable deterioration.
In this study, precipitation process was developed for the recovery of the lactic acid from calcium lactate fermentation broth. Calcium lactate yield was improved by decreasing the solubility of calcium lactate through the addition of ethanol (25%, v/v) as a co-precipitant. The optimal lime type, lime concentration, stirrer speed, precipitation time, temperature, and solvent amount for $Ca(LA)_2$ precipitation were CaO, 0.0175 g/mL, 220 rpm, 24 h, $5^{\circ}C$, ethanol 25% (v/v), respectively. Lactic acid was easily and efficiently recovered from precipitated $Ca(LA)_2$ by adding sulfuric acid ($Ca(LA)_2/H_2SO_4$ molar ratio=1:1). In the model solution of organic acids and fermentation broth, the overall yields of recovered lactic acid were 62% and 55%, respectively, under the aforementioned optimal conditions.
We investigated the optimal strategy for ethanol production using flocculent Sacchromyces cerevisiae ATCC 96581. Considering the characteristic of flocculent yeast, a repeated fed-batch ethanol fermentation was designed, in which non-sterile glucose powder was fed every 12 hours and, after cell flocculation, new feeding medium was exchanged every 24 or 36 hours. We particularly compared this fermentation process with those when cell flocculation was not carried out. Finally, the maximal total ethanol production was 825 g-ethanol during 120 hours, in which the time interval of withdrawal-fill of feeding medium was 24 hours and cell flocculation was carried out.
Trichoderma harzianum ZF-2 producing laccase was isolated from decaying samples from Shandong, China, and showed dye decolorization activities. The objective of this study was to optimize its culture conditions using a statistical analysis of its laccase production. The interactions between different fermentation parameters for laccase production were characterized using a Plackett-Burman design and the response surface methodology. The different media components were initially optimized using the conventional one-factor-at-a-time method and an orthogonal test design, and a Plackett-Burman experiment was then performed to evaluate the effects on laccase production. Wheat straw powder, soybean meal, and $CuSO_4$ were all found to have a significant influence on laccase production, and the optimal concentrations of these three factors were then sequentially investigated using the response surface methodology with a central composite design. The resulting optimal medium components for laccase production were determined as follows: wheat straw powder 7.63 g/l, soybean meal 23.07 g/l, $(NH_4)_2SO_4$ 1 g/l, $CuSO_4$ 0.51 g/l, Tween-20 1 g/l, $MgSO_4$ 1 g/l, and $KH_2PO_4$ 0.6 g/l. Using this optimized fermentation method, the yield of laccase was increased 59.68 times to 67.258 U/ml compared with the laccase production with an unoptimized medium. This is the first report on the statistical optimization of laccase production by Trichoderma harzianum ZF-2.
Our aim was to ferment soymilk using lactic acid bacteria that showed protease activity and to optimize the condition for fermentation. In total, 108 strains of protease-producing lactic acid bacteria were isolated from various fermented foods such as kimchi and jeotgal, and among them, 29 strains displaying the highest protease activity were selected for further study. From these 29 strains, strain MK1, whose protease activity was 126 $mU/mL{\cdot}min$, was selected as the optimal fermentation strain owing to its high ability to digest soymilk protein. It was henceforth labeled as Lactobacillus paracasei MK1. The optimum conditions for the fermentation of soymilk by using L. paracasei MK1 were determined to be as follows: 30 h of fermentation time at a temperature of $30^{\circ}C$, and at a pH of 6.0 in the initial growth medium.
The task of improving a fungal strain is highly time-consuming due to the requirement of a large number of flasks in order to obtain a library with enough diversity. In addition, fermentations (particularly those for fungal cells) are typically performed in high-volume (100-250 ml) shake-flasks. In this study, for large and rapid screening of itaconic acid (IA) high-yielding mutants of Aspergillus terreus, a miniaturized culture method was developed using 12-well and 24-well microtiter plates (MTPs, working volume = 1-2 ml). These miniaturized MTP fermentations were successful, only when highly filamentous forms were induced in the growth cultures. Under these conditions, loose-pelleted morphologies of optimum sizes (less than 0.5 mm in diameter) were casually induced in the MTP production cultures, which turned out to be the prerequisite for the active IA biosynthesis by the mutated strains in the miniaturized fermentations. Another crucial factor for successful MTP fermentation was to supply an optimal amount of dissolved oxygen into the fermentation broth through increasing the agitation speed (240 rpm) and reducing the working volume (1 ml) of each 24-well microtiter plate. Notably, almost identical fermentation physiologies resulted in the 250 ml shake-flasks, as well as in the 12-well and 24-well MTP cultures conducted under the respective optimum conditions, as expressed in terms of the distribution of IA productivity of each mutant. These results reveal that MTP cultures could be considered as viable alternatives for the labor-intensive shake-flask fermentations even for filamentous fungal cells, leading to the rapid development of IA high-yield mutant strains.
