Development of fermentation·storage mode for kimchi refrigerator to maintain the best quality of kimchi during storage (김치저장 중 최적의 맛 유지를 위한 김치냉장고 발효·보관 모드의 개발)

  • To maintain the best quality of kimchi during long-term storage, we developed a fermentation storage mode for the kimchi refrigerator. The optimal kimchi fermentation temperature was determined to be $6^{\circ}C$ with fermentation time of 4-7 days in winter and 3-5 days in spring and fall. Based on these results, the fermentation storage mode conditions were programmed to consist of a fermentation temperature of $6^{\circ}C$ and fermentation times of 111 h in winter and 58 h in spring/fall. When kimchi was stored under the developed fermentation storage mode conditions, the total acidity of kimchi was almost the same as that of the control kimchi (stored $-2-\;-1^{\circ}C$ for 12 weeks). However, the number of lactic acid bacteria (LAB) and Leuconostoc sp. in kimchi were higher ($10^1-10^2CFU/mL$) than those in the control kimchi during storage. In addition, kimchi fermented and stored under the fermentation storage mode clearly received higher scores for overall preference than the control kimchi.

Statistical Optimization of Culture Conditions of Probiotic Lactobacillus brevis SBB07 for Enhanced Cell Growth (프로바이오틱 Lactobacillus brevis SBB07의 균체량 증가를 위한 배양 조건 최적화)

  • We recently reported the potential probiotic properties of Lactobacillus brevis SBB07 isolated from blueberries. The present study investigates the effect of culture conditions such as temperature, initial pH, culture time, and medium constituent for industrial application. The ingredients of the medium to improve cell growth were selected by Plackett-Burman design (PBD) and central composite design (CCD) within a desirable range. The PBD was applied with 19 factors: seven carbon sources, six nitrogen sources, and six microelements. Protease peptone, corn steep powder (CSP), and yeast extract were found to be significant factors for the growth of SBB07. The CCD was then applied with three variables found from the PBD at five levels, and the optimum values were decided for the three variables: protease peptone, CSP, and yeast extract. In the case of the growth of SBB07, the proposed optimal media contained 2.0% protease peptone, 2.5% CSP, and 2.0% yeast extract, and the maximum dried-cell weight was predicted to be 2.93963 g/l. By the model verification, it was confirmed that the predicted and actual results are similar. Finally, the study investigated the effects of incubation temperature and initial pH at the optimized medium. It was confirmed that the dried-cell weight increased from $2.2933{\pm}0.0601g/l$ to $3.85{\pm}0.0265g/l$ when compared to the basal medium at $37^{\circ}C$ and initial pH 8.0. Establishing the optimal culture condition for SBB07 provides good potential for applications in probiotics and can serve as the foundation for the industrialization of materials.

An Optimum Harvest Time for Making Grinded Silage of Barley and Wheat for Whole Crop (총체맥류 분쇄 사일리지 조제를 위한 적정 수확시기)

  • This study was carried out to investigate the optimal harvesting time, feed value and fermentation quality of barley and wheat for the making of chopped whole crop silage substitute for formula feed. As a result, the moisture content of barley and wheat decreased with a late harvest, and barley progressed faster than wheat. The plant height was similar with harvesting time, and the number of spikes decreased with prolonged period after heading. The dry matter yield and TDN yield of barley harvested at 35 and 40 days after heading were significantly higher than those at 30 days after heading and wheat was significantly higher at 40 and 45 days than at 35 day after heading(p<0.05). Crude protein content of barley and wheat were increased with later harvesting time, and crude fiber, crude fat and crude ash were slightly decreased, but not statistically significant. NDF and ADF content of barley decreased with later harvesting time, and those showed similar level in wheat. TDN content of barely was slightly increased but there was no difference in wheat. Comparing the effects of fermentation on feed value of chopped whole crop silage, the approximate compositions were slightly increased after fermentation, but the difference was not significant. Fermentations resulted in increasing the pH value of barley silage with late harvesting time, but decreasing the lactic acid content(p<0.05). A pH value of wheat silage showed similar level in different harvest time, and lactic acid content was decreased. Considering the quantity and quality of fermentation, barley and wheat can be used for making chopped silage of whole crop silage when they were harvested at 35 days and 40~45 days after heading, respectively.

Medium Concentration Influencing Growth of the Entomopathogenic Nematode Heterorhabditis bacteriophora and its Symbiotic Bacterium Photorhabdus luminescens

  • The biological control potential of entomopathogenic nematodes (EPN) can be enhanced by improved culture efficiency. Optimization of the media is a key factor for improving in vitro mass production of entomopathogenic nematodes. This study reports the effect of medium concentration. The medium is a combination of carbohydrates, lipids, proteins, sats, and growth factors, on the growth of Heterorhabditis bacteriophora and its symbiotic bacterium Photorhabdus liminescens. The overall optimal medium concentration for nematode recovery, hermaphrodite size, bacterial mass, infective juveniles (IJs) yield, and doubling time was 84 g/l. At this concentration rate, the doubling time of IJs production and the biomass of symbiotic bacteria was 1.6 days and 12.8 g/l, respectively. The maximum yield of $2.4{\times}{10^5}IJs/ml$ was attained within a one-generation cycle (eight days). The yield coefficient was $2.8{\times}{10^6}$ IJs/g medium, and the maximum productivity was $3.1{\times}{10^7}$ IJs per day. Medium concentration affected two independent factors, recovery and hermaphrodite size, which in turn influenced the final yield.

Effects of Potato on the Storage of Kimchi (배추김치의 저장성에 미치는 감자 첨가의 영향)

  • This study was carried out to investigate the properties of Kimchi made with potato as compared to Kimchi made with radish. The pH, acidity, hardness, and number of lactic acid bacteria were examined as properties of the Kimchi to determine the suitability of adding the potato and with focus on the possible duration of storage. First in the case of pH, the storage duration at pH $4.2{\sim}4.3$, which is the state for optimal tasting Kimchi, was approximately 10days in the control group, while that of the experimental group was 10 to 16 days, and thus, longer than the control. Second in the case of acidity, representing the maturity of Kimchi, we examined the time it took to reach 0.6% acidity, in which the experimental group took more longer time than the control. Although the experimental group was slower to mature than the control, the period for keeping at the proper pH was longer than that of the control group. Thirdly, in the case of hardness, which relates to softening, the experimental group had remarkably high values. This suggests that the Kimchi in the experimental group had a more rigid texture and more difficultly softening than the control group. Lastly in the case of the number of lactic acid bacteria, which closely relates to the process of fermentation, there were no significant differences. In conclusion, these results indicate that adding potato to Kimchi can extend its storage period.

A Study on the Recycling of Foodwaste Leachate as External Carbon Sources Using Microbubble (마이크로버블을 이용한 음폐수의 외부탄소원으로서의 재활용 가능성 연구)

  • The purpose of this research was to examine the possibility on the recycling of foodwaste leachate as external carbon sources using microbubble. The following operating conditions were selected: pressurizing tank 3 bar, circulation flow rate 3.65 LPM, and air flow rate 0.3 LPM with batch type. Microbubble contact time of 18 hours is optimal time to satisfy the recycling of foodwaste leachate as external carbon sources with batch type. HRT 18 hours came up to standard for external carbon sources, except for T-P concentration with continuous type. Coagulants need to be used for removal of dissolved phosphorus concentration by more than 88.5% of the total phosphorus concentration. The VFA was influenced by the organic decomposition rate and the concentration in the aerobic condition. It was considered that the VFA was needed for selection the optimal HRT or the addition of acid fermentation process in order to meet recycling standard of foodwaste leachate.

Effect of Pasteurized Substrates in Various Condition on the Mycelial Growth of Oyster Mushroom, Pleurotus spp. (볏짚배지의 살균조건이 느타리버섯균의 균사생장에 미치는 영향)

  • Various conditions for mushroom were investigated to study their effects on the mycelial growth of oyster mushroom and pathogens. Our results show that the optimal pasteurization temperatures for mycelial growth and pathogens are $60^{\circ}C\;and\;121^{\circ}C$ respectively. Both were then decreased by heating over $60^{\circ}C$ and getting down temperature under $121^{\circ}C$, respectively. In the experiment for pasteurization time, it was showed that both 8 and 12 hours pasteurization were greater than 24 hours treatment for the mycelial growth. In contrast, we didn't find any particular effect by different soaking time of substrates. The pH In substrates was leveled well by pasteurization and after-fermentation even though the acidity was lowered by extending the soaking time. Our results also show that the mycelial growth is greatest at $25{\sim}30^{\circ}C$ regardless of pasteurization time for substrates, Through the experiment with paddy straw as a substrate, it was showed that two time pasteurization was greater than one time for mycelium growth and pro-fermentation of paddy straw before pasteurization have resulted in worth mycelial growth compared with that of normal condition.

New Finding and Optimal Production of a Novel Extracellular Alkaline Lipase from Yarrowia lipolytica NRRL Y-2178

  • Lipases are industrially useful versatile enzymes that catalyze numerous different reactions including hydrolysis of triglycerides, transesterification, and chiral synthesis of esters under natural conditions. Although lipases from various sources have been widely used in industrial applications, such as in food, chemical, pharmaceutical, and detergent industries, there are still substantial current interests in developing new microbial lipases, specifically those functioning in abnormal conditions. We screened 17 lipase-producing yeast strains, which were prescreened for substrate specificity of lipase from more than 500 yeast strains from the Agricultural Research Service Culture Collection (Peoria, IL, U.S.A.), and selected Yarrowia lipolytica NRRL Y-2178 as a best lipase producer. This report presents new finding and optimal production of a novel extracellular alkaline lipase from Y. lipolytica NRRL Y-2178. Optimal culture conditions for lipase production by Y. lipolytica NRRL Y-2178 were 72 h incubation time, $27.5^{\circ}C$, pH 9.0. Glycerol and glucose were efficiently used as the most efficient carbon sources, and a combination of yeast extract and peptone was a good nitrogen source for lipase production by Y. lipolytica NRRL Y-2178. These results suggested that Y. lipolytica NRRL Y-2178 shows good industrial potential as a new alkaline lipase producer.

Effect of Citrus Concentrate on the Physicochemical Properties of Kochujang (감귤 농축액 첨가가 고추장의 품질특성에 미치는 영향)

  • In an effort to improve the quality and palatability of kochujang, the physicochemical and microbial characteristics of kochujang were assessed when different quantities of citrus concentrate were added during fermentation. The moisture content of kochujang increased throughout the entire fermentation process. The pH of kochujang was reduced gradually, but the titratable acidity was increased to 10 weeks of fermentation. The amino-nitrogen and reducing sugar contents of kochujang to which citrus concentrate was added were higher than those of the control, and these values increased with increasing fermentation time. As the ratio of added citrus concentrate increased, the color of the kochujang brightened. The total cell and yeast counts were unaffected by the addition of citrus concentrate. Our sensory evaluation test showed that the addition of 6% citrus concentrate was the optimal condition for improving the quality of kochujang. In conclusion, the addition of citrus concentrate was desirable for the improvement of the quality and palatability of kochujang.

Bioethanol Production from Wasted Corn Stalk from Gangwon Province : from Enzymatic Hydrolysis to Fermentation (강원지역 폐옥수수대로부터 바이오에탄올 생산 : 효소 당화부터 발효까지)

  • Among the samples prepared by various pre-treatment methods, the one pretreated by dilute sulfuric acid showed the highest glucose yield in the enzymatic hydrolysis. Statistical analysis of enzymatic hydrolysis revealed that the glucose yield was in proportion to the enzyme dosage, the ratio of the pre-treated sample to the buffer solution, and the reaction time and that the effect of enzyme dosage was predominant in the experiment range. In addition, the glucose yield was estimated to be 76.1% at an optimal enzymatic hydrolysis condition. In a separate hydrolysis and fermentation (SHF), Saccharomyces cerevisiae converted over 80% of glucose from the enzymatic hydrolysis of pre-treated wasted corn stalk by dilute sulfuric acid to bioethanol with 37% of ethanol yield and 0.42 $g/L{\cdot}hr$ of productivity. In the simultaneous saccharification and fermentation (SSF), 59.5% of conversion from glucan to ethanol and 0.20 $g/L{\cdot}hr$ of productivity were achieved. In both SHF and SSF, approximately 88 g of bioethanol could be obtained from 1 kg of wasted corn stalk. The possible amount of bioethanol in Gangwon province were estimated to be 1.9 kiloton with the assumption of the 50% of collection ratio.
