본 연구에서는 FRTL-5 갑상선세포에서 Protein phosphatase type 1(PP-1)과 type 2A(PP-2A) 효소의 억제제인 okadaic acid에 의한 apoptosis 유도 기전에 대하여 이해하고자 하였다. FRTL-5 갑상선세포에 다양한 농도($10{\sim}80nmol$)의 okadaic acid를 처리 한 후 caspase 3와 $PLC-{\gamma}1$ 분절을 확인 한 결과 40nmol 농도에서부터 caspase 3활성과 $PLC-{\gamma}1$ 분절이 나타남을 확인하였다. Okadaic acid에 의한 apoptosis 유도 기전을 조사한 결과 anti-apoptotic 기능이 있는 Bcl-2 단백질 발현은 미약하게 감소하였으나 Bcl-xL은 농도 의존적으로 급격한 감소를 보였다. Caspase 3 효소활성을 저해하는 IAP 단백질 중 cIAP2는 okadaic acid에 영향을 받지 않았으나 cIAP1과 XIAP 단백질 발현은 농도 의존적으로 감소하였다. Okadaic acid에 의한 apoptosis 유도는 caspase 3 의존적인 기전을 보였다. Caspase 3 특이억제제를 okadaic acid와 동시에 처리 시 apoptosis가 억제되었으며 $PLC-{\gamma}1$ 단백질의 절단 현상도 방지되었다. Caspase 3의 활성을 유도하는 cytochrome c의 유리는 okadaic acid처리 농도에 의존적으로 유리하였다. 결론적으로, okadaic acid에 의한 apoptosis 유도는 caspase 3 의존적이며, Bcl-2와 IAP family 단백질 감소현상에 의하여 야기되는 것으로 생각된다.
Phosphatase의 저해제로 알려진 okadaic acid(OA)가 한국산 개구리(북방산개구리 참개구리) 난자의 성숙에 미치는 효과를 조사하였다. 북방산개구리 난자에 약 50 nl의 okadaic acid(0 5-500 mM)를 미세주입한 후 18시간 배양한 결과 0.5 mM의 농도에서 부터 난자의 핵붕괴를 일으키기 시작하였다 동면초기에 처리한 progesterone에 반응하지 않는 난자들도 OA에 의하여 성숙을 일으켰으며. 이 성숙은 배양액에 첨가한 cycloheximide(10 mg/ml)에 영향을 받지 않았다 또한 OA의 처리를 받고 일정시간 배양한 난자의 세포질에는 미성숙 난자의 성숙을 유도하는 maturation promoting factor (MPF)의 활성이 생기었다 참개구리의 난자도 역시 OA에 의해 성숙이 유도되었으며, 주입 후 6시간에서부터 핵붕괴가 일어나기 시작하였다 참개구리에서도 OA의 처리가 MPF의 활성을 촉진하는 것을 세포질내에 Hl histone kinase의 활성도가 증가하는 것으로 확인할 수 있었다 참개구리에서도 OA에 의한 성숙은 cycloheximide나 CAMP에 의해 영향을 받지 않았다 이러한 결과들은 개구리 난자의 MPF 활성화와 성숙과정에 phosphatase가 관여함을 보여주는 것이다.
N형 칼슘통로의 비활성화기전에 관하여는 아직까지도 막전압의존성 기전과 칼슘의존성 기전간에 논란이 계속되고 있다. 2003년에 의학물리에 발표한 논문1)에서 본 연구자는 N형 칼슘통로의 비활성화 기전은 2가지 성분 -빠른 성분과 느린 성분을 가지고 있고 빠른 성분은 칼슘의존적이 아니며 오직 느린 성분만이 칼슘의존적일 가능성을 제시하였다. 본 논문에서는 막전압의존성 기전이 옳건 칼슘의존성 기전이 옳건 간에 세포 신호전달 체계로서 비활성화와 연계된 기전이 필요하므로 이러한 맥락에서 인산화 기전을 연구하였다. 흰쥐 경동맥 결절뉴론을 단일 세포로 얻은 후 whole cell patch clamp technique를 사용하여 N형 칼슘전류를 기록하고 대조 세포내액을 사용하였을 때와 phosphatase inhibitor인 okadaic acid를 포함한 세포내액을 사용하였을 때의 차이를 비교하였다. Okadaic acid에 의하여 비활성화정도가 증가되었고 이러한 okadaic acid 효과는 주로 N형 통로를 통하여 영향을 미침을 N형 칼슘통로 억제제인 $\omega$-conotoxin GVIA를 사용함으로써 확인하였다. Okadaic acid에 의한 비활성화 증가 효과는 protein kinase를 비특이적으로 억제하는 staurosporine에 의하여 억제되었고 또한 calmodulin dependent protein kinase의 특이적 억제제인 lavendustin C에 의하여 억제되었으므로 인산화과정이 N형 칼슘통로 비활성화와 관련되어 있고 특히 calmodulin을 통한 인산화과정이 주로 관여함을 확인하였다. 본 연구자가 발표한 선행논문1)에 의해 외부의 2가 양이온에 의해 빠른 비활성화가 진행되며, 본 논문에 의하여 인산화과정에 의해 빠른 비활성화가 촉진된다는 사실이 확인되었다. 그러나 본 연구결과만으로는 인산화과정이 비활성화 자체라고는 볼 수 없으며 단지 인산화과정에 의해 비활성화가 가속되었다고 해석할 수 밖에 없다. 인산화과정이 비활성화자 체인지 여부는 2가 양이온이 칼슘통로에 작용하는 결합부위에 관한 연구 및 인산화 부위가 칼슘통로인지 아니면 다른 조절 부위인지 여부를 확인할 수 있는 연구가 진행되어야 확실히 알 수 있을 것이다.
protein phosphatase 2A는 bovine brain homogenate의 세포질 fraction에서 얻어졌다. 기질로서 인산화된 histione H1을 이용하여 측정한 phosphatase 의 활성은 dipalmitoyIphophatidylcholine(DPPC) 혹은 phosphatidylserine/DPPC의 혼합물로 구성된 liposome의 존재하에서 저해되었다. Protein phosphatase 2A의 lipid membrane에의 결합은 다중층 지질막의 혼합물 계에서 liposome 의 양이 증가함에 따라서 상등액 중의 phosphatase의 활성이 감소하는 것으로 확인할 수 있었다. 또한 [$^{125}I$]protein phosphatase 2A가 liposome과 동시에 용출되는 것으로도 확인되었다. 그러나 liposome에 대한 protein phosphatase의 친화력은 높지 않았다. 한편, okadaic acid와 liposome은 협동으로 phosphatase의 활성을 감소시켰다. 이것은 okadaic acid가 lipid membrane이나 membrane에 결함한 phosphatase에는 결합하지 않는다는 것을 의미한다. 그러므로 lipid membrane에 의한 protein phosphatase 2A의 활성 저해 효과는 phosphatase 2A와 lipid membrane과의 결합에 의한 것이라고 설명될 수있다.

Six renaturable protein kinases that utilize the myelin basic protein (MBP) as a substrate were activated during prolonged exposure of cardiac myocytes to okadaic acid (OA). We characterized the substrate preference and activation of these kinases, with particular emphasis on 3 novel kinases-MBPK-55, MBPK-62 and MBPK-87. The transcription factors c-Jun, Elk, ATF2, and c-Fos that are used to assess mitogen-activated protein kinase activation were all poor substrates for these three kinases. MAPKAPK2 was also not phosphorylated. In contrast, Histone IIIS was phosphorylated by MBPK-55 and MBPK-62. These protein kinases were activated in cultured cardiac fibroblasts, H9c2 cardiac myoblasts, and Cos cells. High concentrations (0.5 to $1\;{\mu}M$) of OA were essential for the activation of the protein kinases in all of the cell types examined, whereas calyculin A [an inhibitor of protein phosphatase 1 (PP1) and PP2A], cyclosporin A (a PP2B inhibitor), and an inactive OA analog all failed to activate these kinases. The high dose of okadaic acid that is required for kinase activation was also required for phosphatase inhibition, as assessed by immunoblotting whole cell lysates with anti-phosphothreonine antibodies. A variety of chemical inhibitors, including PD98059 (MEK-specific), genistein (tyrosine kinase-specific) and Bisindolylmaleimide I (protein kinase C-specific), failed to inhibit the OA activation of these kinases. Thus, MBPK-55 and MBPK-62 are also Histone IIIS kinases that are widely expressed and specifically activated upon exposure to high OA concentrations.
The sphingomyelin cycle and ceramide generation have been recognized as potential growth suppression signals in mammalian cells. Ceramide has been shown to induce differentiation, cell growth arrest, senescence, and apoptosis. Although the intracelluar target for the action of ceramide remains unknown, recent studies have demonstrated the role of cytosolic ceramideactivated protein phosphatase(CAPP). In this study, the cytotoxic effect of C2-ceramide, a synthetic cellpermeable ceramide analog, on HEp-2 cells and the mechanism by which ceramide induces cell death were investigated. The addition of exogenous C2-ceramide resulted in a concentration dependent cell death. Okadaic acid, a potent inhibitor of CAPP, enhanced ceramide-mediated cell death, which suggests that CAPP is not involved in this process. To understand the mechanism of action of ceramide, we studied the relationship between ceramide and c-Myc and pRb which are defined components of cell growth regulation. Western blot analyses revealed that C2-ceramide (10${\mu}M$) induced c-Myc down-regulation, but there were no significant changes in pRb. However, treatment of okadaic acid (10 nM) enhanced c-Myc and pRb down-regulation. Reduction of the amount of c-Myc and pRb occurred during HEp-2 cell death. These results suggest that the cytotoxic effect of ceramide in HEp-2 cells may not be mediated through the action of CAPP and that the downstream target for ceramide is c-Myc and pRb.
Several lines of evidence have revealed that phosphorylation of amyloid precursor protein (APP) at Thr668 is involved in the pathogenesis of Alzheimer's disease (AD). Okadaic acid (OA), a protein phosphatase-2A inhibitor, has been used in AD research models to increase tau phosphorylation and induce neuronal death. We previously showed that OA increased levels of APP and induced accumulation of APP in axonal swellings. In this study, we found that in OA-treated neurons, phosphorylation of APP at Thr668 increased and accumulated in axonal swellings by c-jun N-terminal kinase (JNK), and not by Cdk5 or ERK/MAPK. These results suggest that JNK may be one of therapeutic targets for the treatment of AD.
LC-MS/MS법에 의하여 설사성 패류독소 7종(okadaic acid, dinophysistoxin-1, -3, pectebotoxin-1, -2, -6, yessotoxin)의 동시 분석 조건을 확립하였고, 2006년 3월부터 9월까지 매월 5종의 이매패(굴, 진주담치, 반지락, 가리비, 개조개, 6월부터 9월까지는 가리비 대신 피조개)를 정기적으로 통영시장에서 구입하여 분석하였으며, 기존의 마우스 검사법과 비교하였다. 마우스 검사법으로는 총 35종 시료 중 3월의 굴과 진주담치에서 0.05${\sim}$0.1 MU/g의 독성을 나타내었으나, 이미 알려진 설사성 패류독소는 전혀 검출되지 않아 다른 성분에 의한 것으로 판단되었다. 한편, LC-MS/MS법에 의하여서는 6월 진주담치에서 dinophysistoxin-1(DTX1)이 0.05 ${\mu}g/g$ 검출되었으며, 8월의 모든 시료에서 okadaic acid(OA)가 0.01${\sim}$0.02 ${\mu}g/g$ 검출되었다. 이들은 외국의 기준치[0.05 MU/g(일본), OAs(OA+DTX1) 0.16 ${\mu}g/g$(EU)]보다 매우 낮은 함량으로 식품 위생상 안전하였다.
Okadaic acid (OA) 군 독소는 설사성 패류중독(diarrhetic shellfish poison, DSP)을 유발하는 해양생물독소이다. 설사성패독은 오카다익산(OA)과 그 유사체인 디노피시스톡신(DTX)으로 구성되어 있으며, 주로 와편모조류에서 생성되어 이매패류의 체내에 축적되어 독화된다. 이에 EFSA, WHO에서는 안전관리를 위하여 수산물 내 OA군 독소 함량 기준을 설정하였다. 최근 우리나라 연안에서도 원인조류인 Dinophysis sp.의 출현이 보고된 바 있으며, 국내 수산물 생산과 소비에도 영향을 미치고 있다. 또한 국제적인 움직임에 발맞추어 국내에서도 2022년 설사성 패독 기준 관리 대상 물질에 DTX2를 추가하여 관리하고 있다. 본 연구는 OA군 독소의 이화학적 특성, 분석법, 인체 노출 사례와 국내외 관리 현황 등의 자료를 검토하여 OA군 독소의 체계적인 모니터링과 안전관리의 기반을 마련하고자 수행되었다.
