Development and Characterization of Novel Rapeseed (Brassica napus L.) Mutant Lines through Mutation Breeding

  • Rapeseed (Brassica napus L.) is one of the most valuable oilseed crop in the world. It is widely used in various industries, such as food, animal feed, energy and chemical industries. In order to improve the industrial requirements for rapeseed, useful agronomic characteristics (higher yields and disease resistance etc.) and modified oil traits (fatty acid composition and fat content) are important in rapeseed. However, Korea has limiting genetic resources of novel traits in rapeseed. In this research, novel rapeseed mutant genotypes by mutation breeding was developed. The mutant lines were generated by the treatment of the seeds of the original cultivar 'Tamra' with 700 Gy of gamma-ray (60Co). Mutants showing varied in flowering time, crude fat content, seed yield and fatty acid content that exhibited stable inheritance of the mutated characteristics from M5 to M7 generations were selected. We investigated genetic variation using SNPs identified from GBS analysis in rapeseed mutant lines derived from the gamma-ray, and interactions between the major agronomic and the oil traits. Significantly associated SNP loci were explored along with candidate genes using SNPs obtained by GBS analysis. As a results of association mapping, a total of 322 SNPs were significantly associated with agronomic traits (155 SNPs) and oil traits (167 SNPs). A total of 70 genes were annotated from agronomic characteristics SNPs; among them 7 genes significantly enriched in developmental process, and a total of 70 genes were annotated from crude fat content and fatty acid compositions SNPs; among them, 11genes were significantly enriched in biosynthetic process. These results could be used for the selection of rapeseed cultivar with enhanced qualities and potential economic benefits.

Characteristics of Purified Horse Oil by Supercritical Fluid Extraction with Different Deodorants Agents

  • This study investigated the impact of activated carbon, palm activated carbon, and zeolite on horse oil (HO) extracted from horse neck fat using supercritical fluid extraction with deodorant-untreated HO (CON) as a comparison. The yield and lipid oxidation of deodorant untreated HO (CON) were not significantly affected by the three deodorants. However, deodorant-treated HOs exhibited significantly elevated levels of α-linolenic acid (C18:3n3) and eicosenoic acid (C20:1n9) compared to CON (p<0.05), while other fatty acids remained consistent. Zeolite-purified HO demonstrated significantly lower levels of volatile organic compounds (VOCs) than other treatments (p<0.05). Remarkably, zeolite decreased the concentration of pentane, 2,3-dimethyl (gasoline odor), by over 90%, from 177.17 A.U. ×106 in CON to 15.91 A.U. ×106. Zeolite also effectively eliminates sec-butylamine (ammonia and fishy odor) as compared to other deodorant-treated HOs (p<0.05). Additionally, zeolite reduced VOCs associated with the fruity citrus flavor, such as nonanal, octanal, and D-limonene in HO (p<0.05). This study suggests that integrating zeolite in supercritical fluid extraction enhances HO purification by effectively eliminating undesirable VOCs, presenting a valuable approach for producing high-quality HO production in the cosmetic and functional food industries.

Drop formation of Carbopol dispersions displaying yield stress, shear thinning and elastic properties in a flow-focusing microfluidic channel

  • The drop formation dynamics of a shear thinning, elastic, yield stress ($\tau_o$) fluid (Carbopol 980 (poly(acrylic acid)) dispersions) in silicone oil has been investigated in a flow-focusing microfluidic channel. The rheological character of each solution investigated varied from Netwonian-like through to highly non-Newtonian and was varied by changing the degree of neutralization along the poly (acrylic acid) backbone. We have observed that the drop size of these non-Newtonian fluids (regardless of the degree of neutralisation) showed bimodal behaviour. At first we observed increases in drop size with increasing viscosity ratio (viscosity ratio=viscosity of dispersed phase (DP)/viscosity of continuous phase (CP)) at low flowrates of the continuous phases, and thereafter, decreasing drop sizes as the flow rate of the CP increases past a critical value. Only at the onset of pinching and during the high extensional deformation during pinch-off of a drop are any differences in the non-Newtonian characteristics of these fluids, that is extents of shear thinning, elasticity and yield stress ($\tau_o$), apparent. Changes in these break-off dynamics resulted in the observed differences in the number and size distribution of secondary drops during pinch-off for both fluid classes, Newtonian-like and non-Newtonian fluids. In the case of the Newtonian-like drops, a secondary drop was generated by the onset of necking and breakup at both ends of the filament, akin to end-pinching behavior. This pinch-off behavior was observed to be unaffected by changes in viscosity ratio, over the range explored. Meanwhile, in the case of the non-Newtonian solutions, discrete differences in behaviour were observed, believed to be attributable to each of the non-Newtonian properties of shear thinning, elasticity and yield stress. The presence of a yield stress ($\tau_o$), when coupled with slow flow rates or low viscosities of the CP, reduced the drop size compared to the Newtonian-like Carbopol dispersions of much lower viscosity. The presence of shear thinning resulted in a rapid necking event post onset, a decrease in primary droplet size and, in some cases, an increase in the rate of drop production. The presence of elasticity during the extensional flow imposed by the necking event allowed for the extended maintenance of the filament, as observed previously for dilute solutions of linear polymers during drop break-up.

용매추출 및 화학적 정제법에 의한 고품질의 포도씨유의 제조 (Preparation of High Quality Grape Seed Oil by Solvent Extraction and Chemical Refining Process)

  • 포도씨을 이용한 고부가가치의 포도씨유 제조기술 개발 및 시제품을 생산하기위해 먼저 일반 재배 및 유기농 재배 포도로부터 각각 수거된 포도씨의 수율, 및 기능성성분(지방산, 피토스테롤, 토코페롤, 총카테킨 및 4가지 카테킨 조성)의 차이를 측정하여 비교하였다. 다음, 용매추출법과 화학적 정제방법에의한 포도씨로부터 고품질의 포도씨유의 제조기술을 개발하고 시제품을 생산하한 결과는 다음과 같다. 일반 재배 및 유기농 재배 포도로부터 얻어진 포도씨유 및 추출물의 수율 그리고 기능성성분(지방산, 피토스테롤, 토코페롤, 총카테킨 및 4가지 카테킨류)의 함량을 측정한 후 비교한 결과 대체적으로 일반 재배 및 유기농 재배 포도씨의 기능성성분의 함량에는 큰 차이가 없었다. 건조포도씨(1.0톤)를 볶음처리 후 분쇄하여 노르말-헥산으로 탈지하고 얻은 포도씨 원유를 탈검($0.15\%$ 인산) 및 탈산($20\%$ NaOH 용액) 처리 후 수증기증류 장치($240^{\circ}C$, 2시간, 5 mmHg)를 이용하여 탈색, 탈향 및 탈취공정을 차례로 거치면서 고품질의 포도씨유의 제조기술을 확립하였으며, 아울러 그로부터 최종 포도씨유 제품(50kg, 수율: $5.0\%$/건조 포도씨)을 생산하였다.

목질바이오매스의 급속열분해에 의해 생성된 바이오오일의 특성 분석 (Characterization of Bio-oils Produced by Fluidized Bed Type Fast Pyrolysis of Woody Biomass)

  • 유동형 급속열분해기 (fluidized bed type fast pyrolyzer, 용량 400 g/h)를 이용하여 너도밤나무와 침엽수 혼합재(독일가문비나무/전나무, 50:50) 에서 바이오오일을 생산하였다. 목질바이오매스의 열분해는 약 $470{\pm}5^{\circ}C$에서 1~2초 동안 진행되었다. 목질바이오매스의 열분해 생성물의 조성은 너도밤나무의 경우 바이오오일 60%, 탄 9% 그리고 가스가 31% 정도 생산되었으며, 침엽수 혼합재는 바이오일 49%, 탄 9%, 그리고 42% 가량의 가스가 생성되었다. 두 종류의 목질바이오매스에서 생산된 바이오오일의 수분함량은 약 17~22%이었으며, 밀도는 수종에 관계없이 $1.2kg/{\ell}$이었다. 바이오오일의 원소 조성은 탄소 45%, 산소 47%, 수소 7%, 그리고 질소 1%로 일반적인 목질바이오매스와 큰 차이는 없는 것으로 나타났다. 그러나 화석자원에서 생산되는 오일류와 비교하여 바이오일은 산소함량이 매우 높았고 황은 전혀 포함되어 있지 않았다. 바이오오일의 GC 분석 결과 총 90여종의 방향족(aromatic) 또는 비방향족(non-aromatic) 저분자량 화합물이 검출되었으며 이들의 함량은 바이오오일 전건중량의 31~33%로 분석되었다.

배초향의 수확시기별 생육특성 및 로즈마린산과 정유함량의 변화 (Changes of Growth Characteristics, Rosmarinic Acid and Essential Oil Contents according to Harvest time in Agastache rugosa O. Kuntze)

  • 배초향 국내 수집종의 생육특성 및 수확시기별 로즈마린산과 정유함량을 조사하여 다음과 같은 결과를 얻었다. 1. 수집종들의 개화기는 8월 13일${\sim}$8월25일로 진도 수집종이 가장 늦었고 초장은 $120{\sim}170cm$, 주당 분지수는 $13{\sim}21$개, 화방군(花房群)의 길이는 $10{\sim}15cm$였으며, 화방군(花房群)의 수(數)는 $13{\sim}22$개로 수집종간에 유의성이 없었다. 2. 수집종들의 부위별 건물수량 (kg/10a)은 꽃 $109{\sim}156kg$, 잎 $135{\sim}252kg$, 줄기 $421{\sim}932kg$, 뿌리$172{\sim}394kg$으로 꽃과 뿌리에서는 유의성이 없었으며 건물수량은 개화 중기 (9월 20일)에 수확한 것이 가장 많았다. 3. 수집종들의 로즈마린산 함량은 모든 부위에서 유의성이 인정되었는데 꽃 $0.32{\sim}2.34%$, 잎 $0.97{\sim}1.64%$, 줄기 $0.03{\sim}0.21%$, 뿌리 $0.91{\sim}6.56%$로 뿌리의 함량이 가장 높고 줄기의 함량이 가장 낮았다. 4. 잎의 로즈마린산과 정유함량은 개화전(開花前)이 각각 3.76%, 5.78%로 개화후(開花後)의 1.55%, 2.10%보다 더 높았으며 지상부의 로즈마린산 함량은 개화전에 가장 높았으나 정유 함량은 개화말기에 가장 높았다.

붉나무 순차 용매 추출물의 항산화 효과 비교 (Antioxidative Effect of us javanica Linne Extract by Various Solvents)

  • 75% ethanol 추출물에서 뚜렷한 항산화 효과가 확인된 붉나무 껍질로부터 hexane, ethyl acetate, methanol 그리고 99% ethanol 추출물을 얻고 이들을 팜유와 돈지에 ascorbic acid, citric acid 그리고 ${\delta}-tocopherol$과 함께 첨가하여 Rancimat으로 항산화 효과를 비교하였다. 용매별 추출 수율은 methanol이 14.53%(w/w)로 가장 높았다. hexane, ethyl actate, methanol 그리고 ethanol(99%) 추출물 600ppm을 ascorbic acid 200ppm과 함께 팜유에 첨가하였을 때 antioxidant index(AI : 항산화제첨가 유지의 유도 기간/대상 유지의 유도 기간)는 각각 1.83, 2.25, 2.81 그리고 2.85로 다른 처리구에 비하여 높았고 돈지의 경우는 추출물 600ppm에 ${\delta}-tocopherol$ 200ppm 첨가구가 가장 효과가 높아서 AI는 각각 3.64, 7.83, 7.34 그리고 9.30이었다. ethanol 및 ethyl actate 추출물의 용매별 분획물은 뚜렷한 항산화 증진 효과가 없었고 methanol 추출물을 첨가한 팜유 및 도니지의 $60^{\circ}C$ 저장 시험에서 POV 및 TBA가를 비교한 결과 산화 억제 효과가 뚜렷하였다.

Performance, Digestibility and Carcass Characteristics of Growing/Finishing Pigs Fed Barley-Based Diets Supplemented with an Extruded or Unextruded Blend of Peas and Canola Seed or Meal

  • Seventy-two crossbred pigs weighing an average of 41.5 kg were assigned on the basis of sex, weight and litter to one of four dietary treatments in a factorial (4 treatments${\times}$2 sexes) arrangement. The control diet was based on barley and soybean meal while the experimental treatments consisted of diets in which a portion of the dietary protein was supplied by 20% of a 50:50 blend of extruded ($130^{\circ}C$ for 20 to 25 sec) peas and full-fat canola seed, 20% of a 50:50 blend of unextruded peas and full-fat canola seed or a diet containing 10% peas, 6% canola meal and 4% canola oil (to equal the level of canola oil provided by 10% whole canola seed). Digestibility coefficients for dry matter, crude protein and gross energy were significantly higher (p<0.05) for the control diet than for the other three diets. Extrusion produced no beneficial effects (p>0.05) on nutrient digestibility and there were no differences in digestibility between the diet based on intact canola seed compared with the diet containing canola meal and oil. Choice of protein supplement had no significant effects on gain, feed intake or feed conversion during the grower or finisher phases and over the entire experimental period. Extrusion of the pea-canola blend produced no beneficial effects on pig performance as the performance of pigs fed either the extruded or unextruded blend of peas and canola seed was similar. In addition, the performance of pigs fed diets containing intact canola seed was similar to that of pigs fed canola meal and oil. Castrates gained faster and consumed more feed than gilts (p<0.05). However, their feed conversion was poorer than that of the gilts during the finisher period. There were no significant differences in carcass traits between pigs fed the control and any of the experimental treatments. Extrusion had no effect on carcass traits and the carcasses of pigs fed canola meal and oil did not differ from those of pigs fed whole canola seed. Castrates had a significantly lower dressing percentage, lower estimated lean yield but greater loin fat depth than gilts (p<0.05). The results of this experiment indicate that peas in combination with canola seed or canola meal are an acceptable alternative to soybean meal as a protein supplement for use in growing-finishing swine diets. Extrusion did not appear to have any beneficial effects on the nutritional value of the canola seed-pea blend as nutrient digestibility, growth performance and carcass traits were similar for pigs fed the unextruded blend of peas and canola seed compared with the extruded product. Since the process adds to the cost of the raw products, its use is unlikely to be economical.

Influence of Supplementing Dairy Cows Grazing on Pasture with Feeds Rich in Linoleic Acid on Milk Fat Conjugated Linoleic Acid (CLA) Content

  • Three experiments were conducted to investigate the hypothesis that cows grazing on pasture produce the highest proportion of c-9 t-11 CLA in milk fat and no further increase can be achieved through supplementation of diets rich in linoleic acid, such as full-fat extruded soybeans or soybean oil. In experiment 1, 18 lactating Holstein cows were used in a randomized complete block design with measurements made from wk 4 to 6 of the experiment. In experiment 2, three cannulated lactating Holstein cows were used in a $3{\times}3$ Latin square design. Each period was 4 wk with measurements made in the final wk of each period. Cows in both experiments were assigned at random to treatments: a, conventional total mixed ration (TMR); b, pasture (PS); or c, PS supplemented with 2.5 kg/cow per day of full-fat extruded soybeans (PES). In both experiments, feed intake, milk yield, milk composition, and fatty acid profile of milk and blood serum were measured, along with fatty acid composition of bacteria harvested from rumen digesta in experiment 2. In experiment 3, 10 cows which had continuously grazed a pasture for six weeks were assigned to two groups, with one group (n = 5) on pasture diet alone (PS) and the other group (n = 5) supplemented with 452 g of soy oil/cow per day for 7 d (OIL). In experiment 1, cows in PS treatment produced 350% more c-9, t-11 CLA compared with cows in TMR treatment (1.70 vs. 0.5% of fat), with no further increase for cows in PES treatment (1.50% of fat). Serum c-9, t-11 CLA increased by 233% in PS treatment compared with TMR treatment (0.21 vs. 0.09% of fat) with no further increase for cows in PES treatment (0.18% of fat). In experiment 2, cows in PS treatment produced 300% more c-9 t-11 CLA in their milk fat compared with cows in TMR treatment (1.77 vs. 0.59% of fat), but no further increase for cows in PES treatment (1.84% of fat) was observed. Serum c-9, t-11 CLA increased by 250% for cows in PS treatment compared with cows in TMR treatment (0.27 vs. 0.11% of fat), with no further increase for cows in PES treatment (0.31% of fat). The c-9, t-11 CLA content of ruminal bacteria for cows in PS treatment was 200% or more of TMR treatment, but no further increase in bacterial c-9, t-11 CLA for cows in PES treatment was observed. Supplementation of soy oil in experiment 3 also did not increase the c-9 t-11 CLA content of milk fat compared with cows fed a full pasture diet (1.60 vs. 1.54% of fat). Based on these findings, it was concluded that supplementing with feeds rich in linoleic acid, such as full-fat extruded soybeans or an equivalent amount of soy oil, to cows grazing perennial ryegrass pasture may not increase milk fat c-9 t-11 CLA contents.

Co 및 Mo 기반 촉매에 의한 폴리스티렌의 저온 열분해 특성에 관한 연구 (The Study on Characteristics of Polystyrene by Low Temperature Pyrolysis by using Co and Mo Dispersed Catalysts)

  • 본 연구는 폴리스타렌(PS) 수지의 유화공정 효율성 향상을 위해 저온열분해 회분식 반응기를 이용하여, 단일 PS 수지와 Co 및 Mo 촉매를 각각 첨가한 PS 수지를 반응온도(425, 450, $475^{\circ}C$), 반응시간(20~80분, 15분 간격), 촉매 농도변화에 따른 PS수지의 액화생성물 전환율을 측정하였다. 최적의 열분해 조건은 반응온도 $450^{\circ}C$, 반응시간 35분으로 판단되며, 전환된 액화생성물의 주요 성분은 GC/MS 분석결과 스타이렌 및 벤젠유도체가 대부분으로 나타났다. 생성물은 산업통상자원부에서 고시한 증류성상 온도에 따라 가스, 가솔린, 등유, 경유, 중유로 분류하여 그 수율을 측정하였다. 그리고 $450^{\circ}C$ 반응온도에서 촉매 사용에 따른 전환율은 Co 촉매 > Mo 촉매 > 무촉매 순이었으며, 생성물 중 가스, 등유, 경유수율은 Mo 촉매, 가솔린은 무촉매, 중유는 Co 촉매에서 우수한 것으로 나타났다. Co 및 Mo 촉매 혼합 농도별 전환율 및 열분해 생성물 수율은 Co 촉매 100% 사용 시 가장 우수한 것으로 판단된다.
