Nutritional Evaluation, Stability of Cereals and Sanitation Status of Processing Utensils and Environments Based on Hygiene Education (위생교육에 따른 선식 제조기구와 작업장의 위생상태변화 및 일부 선식제품의 안정성과 영양적 평가)

  • This study was carried out to investigate the effect of hygiene education on the microbiological changes of processing utensils and the environmental in the manufacture of cereals and to evaluate the Cd, Pb contents and nutrient compositions of 11 cereals. The result of microbiological evaluation was that fungi, coliforms and staphylococcus species were detected in employees, on equipment, utensils and environments in the first inspection. Fungi were detected in most of the cereals, staphylococcus species were detected in soybeans, perilla seeds and sea tangle How, and bacillus cereus was detected in sorghum and black sesame seeds. The water content of rice, barley, glutinous rice, brown rice and carrot flour in packaged products, and in carrot flour, angelica keiskei, carrot, sea mustard and potato in bulk products was in excess of 8.0%. The Pb content of cereals was 0.14-0.51 mg/kg and Cd was not found. The acid value of Job s tears flour and black sesame seed flour was higher than 5.0 mg/g oil. Cereals were manufactured from 41 different cereals and grains, legumes, seeds and nuts, vegetables, potatoes, seaweeds, fruits, glucose and salt. The average content of cereals and grains, legumes, and seeds and nuts in cereals was 75.75%, 16.19% and 4.93%, respectively. The mean nutrient content per 100 g of cereals was calories 365.8kcal, protein 13.3 g, fats 5.9 g, carbohydrates 63.4 g, Ca 91.8 mg, p 269.9 mg, Fe 3.15 mg, Na 76.2 g, K 421.8 mg, Zn 2.33 mg, Vit. A 12.5 R.E., Vit. B$_1$0.23 mg, Vit. $B_2$ 0.16 mg, Vit. $B_6$ 0.46 mg, Niacin 3.5 mg, Vit. C 1.36 mg, folic acid 62.3 $\mu$g and Vit. E 1.24 mg. When nutrients value of 48 g of cereals and 200 $m\ell$l of milk was compared to 1/3 of the RDA, the values were below than 60% of 1/3 of the RDA except Na, K, Vitamin $B_1$ and C contents. The average carbohydrate : protein : fat ratio of energy intake was 54.27 : 17.45 : 28.28, respectively. Therefore, a training program consisting of the education of the staff in surveillance and standard operating procedures, the elimination of dangerous procedures, sanitation checklist, the implementation of Preparation methods and standard recipes for cereals is required.

Effect of Acacia catechu Extract on 3T3-L1 Preadipocyte Differentiation (지방세포의 분화에 미치는 Acacia catechu 추출물의 항비만 효과)

  • The purpose of this study was to investigate the effects of catechu water extract on adipogenesis in 3T3-L1 adipocytes. 3T3-L1 preadipocytes were differentiated with adipogenic regents by incubation for 9 days in the absence or presence of catechu extract ranging from $1{\sim}200{\mu}g/mL$. The effect of catechu extracts on cell proliferation of 3T3-L1 preadipocytes was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The effect of catechu extracts on 3T3-L1 differentiation was examined by measuring intracellular lipid droplet and triglyceride contents. These results were obtained from preadipocyte proliferation and adipocyte differentiation of 3T3-L1. Catechu extracts inhibited lipid accumulation and remarkably decreased triglyceride contents in 3T3-L1 preadipocytes at a concentration showing no cytotoxicity. The anti-adipogenic effects of catechu appeared to be mediated by significant down-regulation of expression of peroxisome proliferator-activated receptor ${\gamma}$, CCAAT/enhancer-binding protein ${\alpha}$, and sterol regulatory element-binding protein 1c proteins apart from expression of hormone-sensitive lipase. We suggest that catechu extracts significantly inhibit adipogenesis and can be used for regulation of obesity.

Effects of Mixed Medicinal Herbs on Adipogenesis and Apoptosis in 3T3-L1 Adipocytes (배합된 한약재의 추출물이 3T3-L1 지방세포의 지방생성과 세포사멸에 미치는 영향)

  • Astragali Radix (AR) has been used in traditional medicinal herb, which is known to tonify the qi and blood. It has been asserted to be a tonic that can improve the function of the lungs, adrenal glands and the gastrointestinal tract, increase metabolism, promote healing and reduce fatigue. Also, Plantago asiatica (PA) has the anti-inflammatory, antiseptic, antitussive, cardiac, diuretic, expectorant, haemostatic effects, though it is considered to be less powerful than the seed. Recent research is showing that it is promising in lowering cholesterol and controlling diabetes. Obesity is characterized at the cellular level by an increase in the number and size of adipocytes differentiated from fibroblastic preadipocytes in adipose tissue. In the present study, we investigated the effects of 3 to 7 extracts of mixed medicinal herbs (water and ethanol extract of Astragali Radix; ARW and ARE, water and ethanol extract of Plantago asitica; PAW and PAE, and those mixed extracts; ARW+PAE, ARW+PAW and ARE+PAE) on adipogenesis and apoptosis in 3T3-L1 adipocytes by MTT assay, Oil-Red-O staining and TUNEL assay. And then, we determined total contents of phenolic compounds and flavonoid compounds in mixed herbal extracts. These result showed that herbal extracts (ARW, PAE and ARW+PAE) affected on adipogenesis and apoptosis in 3T3-L1 adipocytes among 3 to 7 extracts tested. Especially herbal mixed extracts (ARW+PAE) had the highest inhibition on differentiation of preadipocytes and lipoid accumulation of maturing adipocytes. And it did not affect cell viability of mature adipocytes, but herbal mixed extracts (ARW+PAE) increased apoptosis in mature adipocytes, as proved by highest concentration $200\;{\mu}g/m{\ell}$ using TUNEL assay. In addition, contents of total phenoilc compounds and flavonoids showed the highest level in ARE+PAE and PAE than ARW. These results suggest that this main target for adipocyte functions could be effective in improving the metabolic syndrome including obesity.

Effect of Allium Victorialis Var. platyphyllum Leaves on Triton WR-1339-Induced and Poloxamer-407-Induced Hyperlipidemic Rats and on Diet-Induced Obesity Rats (식이성 고지혈 및 비만에 대한 산마늘 추출물의 효과)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var. platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of Is. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks(p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

Study on the Nutritional Composition and Antioxidative Capacity of Mulberry Fruit(Ficus-4x) (4배성 휘커스(Ficus-4x)오디품종의 영양성분 및 항산화능 검색)

  • Nutritional composition and antioxidative capacity of mulberry fruit (Ficus-4x) were investigated for evaluation as new red-colored fruit. Contents of moisture, crude fat, crude protein, and vitamin C were similar, whereas that of crude ash was higher, to those of other berry fruits. Contents of minerals (Ca, 14.33 mg/100 g; P, 39.98 mg/100 g; Fe, 6.01 mg/100 g; Zn, 4.04 mg/100 g; Mn, 2.26 mg/100 g), particularly Fe, were higher than those of other berry fruits. Hardness, springness, cohesiveness, gumminess, and chewiness of mulberry fruit were higher, and color values (L, 36.03; a, 1.80; b, 1.54) were lower than those of strawberry. Relative scavenging activities of mulberry fruit methanol extract and its cyanidine 3-glucoside on 1, 1-diphenyl-2-picrylhydrazy radical (DPPH) were 35.7 and 78.2%, respectively, using butylated hydroxyanisole (BHA) as standard. Antioxidant activities in corn oil (peroxide values and conjugated dienoic acid) were tertiary butylhydroquinone (TBHQ) > mulberry fruit ethanol extract > mulberry fruit water extract > butylated hydroxytoluene (BHT) > tocopherol. Results show mulberry fruit can be very useful red-colored fruit for development of functional foods with beneficial effects on radical scavenging and antioxidative capacities.

Growth Characteristics and Nutritive Value of Chufa (Cyperus esculentus L.) for Forage Resource (사료자원으로서 Chufa (Gyperus esculentus L.)의 생장특성과 사료가치)

  • Chufa (Cyperus esculentus L.) belongs to one of the sedge family and pows well in summer. The aboveground part of chufa is mostly consisted of leaves and the underground part is mostly composed of a clump of fibrous root with tuber. At the seeding year, it does not reproductive development but produces a lot of tuber. It produced many tillers from the tuber and grows in clumps as a bunch type. The plant height of mature chufa was 73 to 75cm and it grown fully in the middle of July. The number of tillers were increased rapidly until the end of July and still increased slowly after August but it showed very poor growth. The final fresh weight and dry matter yield of aboveground part of chufa were 40.3 tou/ha and 12.1 tou/ha. respectively. The regrowth of aboveground part was vigorous in the early stage of growth after 1st cutting but it was decreased rapidly after the second cutting. In control plot, the number of tubers per a clump were 722 at final stage and their fresh and drymatter yields per m: were 4.2kg and 1.9kg, respectively. In experimental plots, the amount of tubers was decreased steadily according to delay of cutting date, but late cutting date was not affect the tuber formation severely because the tuber produced already early in August. The nutritive value of chufa in vegetative growth stage was good but it was decreased according to growing up. The contents of crude protein(CP), neutral detergent fiber(NDF), dry matter digestibility (DMD) and total digestible nutrients(TDN) of aboveground part of chufa harvested finally were 6.1%, 51.5%, 39.8%, 33.2% and 39.4%, respectively. The contents of crude protein(CP), neutral detergent fiber(NDF), dry matter digestibility(DMD) and total digestible nutrients(TDN) of tubers of chufa were 6.1%, 81.5%, 39.8%, 33.2% and 39.4%, respectively and the content of oil was as high as 16.2%, especially.

Effects of Caffeine and Dietary Fat on Mouse Mammary Development (Caffeine과 지방급여가 생쥐의 유선발달에 미치는 효과)

  • This study was conducted to examine the effect of caffeine and three dietary levels of fat, i.e., 0%, 5% and 20% on mammary gland development. Mice were assigned to three groups (dietary levels 0%, 5%, 20% fat), and treated caffeine of half within the each group. Caffeine-treated mice with 0% or 20% fat levels significantly increased 4$^{th}$ mammary gland development in comparison with that of no caffeine -treated mice (P<0.05). Caffeine-treated mice significantly increased DNA contents of 4$^{th}$ mammary gland in comparison with that of no caffeine-treated mice (P<0.05), and DNA contents of mammary gland increased as fat levels increased within caffeine-treated or no caffeine-treated group. nteraction effect was shown between caffeine and 20% fat diet, [(20% fat+caffeine) - (20% fat + no caffeine) vs (0% fat + caffeine) - (0% fat + no caffeine)](P<0.01). Conclusively, caffeine significantly increased mouse mammary gland development possibly by inhibiting phosphodiesterase activity, and dietary fat supplements increased mammary gland development as the fat content of the diet increased from 0 to 20%. The stimulatory effect of caffeine in mammary development interacted with high level of fat diet.

  • According to the results from the experiments of selecting a suitable variety of forage rape and comparing productivity and nutrient quality between forage rape and oil seed rape, Velox appeared to be the most suitable variety in terms of productivity and nutrient quality at the southern area of Korea among the varieties used in the experiments. Consequently, Velox was grown under two different cultural methods, individual culture and population culture, and productivity and variation of nutrient quality during the growth period were observed and compared between the cultural methods. The results from the experiment are summarized as follows: 1. Although variations were great in plant fresh weight and plant dry matter weight among plants, individual culture showed significantly higher value in these characters at the 1 % level than population culture. Plant fresh weight and plant dry matter weight were largely dependent upon the amount of branches in case of individual culture. However, in population culture, they were equally dependent upon the amount of branches and main stem. There was no significant difference in dry matter percentage between cultural methods, and main stem showed highest dry matter percentage. 2. Content of crude protein was decreasing gradually as plants continued to grow. Individual culture showed higher content of crude protein than population culture from 90 to 120 days after sowing but vice versa from 180 days after sowing to flowering stage. Contents of fiber such as NDF, ADF, cellulose and lignin was low at the early stage of growth. It was increasing gradually as plants grew older and at the latter stage of growth plants under individual culture showed higher values in contents of fiber. 3. In vitro dry matter digestibility (IVDMD) of both stems and leaves was decreasing gradually as plants grew older. Plants under individual culture showed higher IVDMD of stems than plants under population culture, but no significant difference in IVDMD of leaves was observed between cultural methods.

  • This sudy was conductted for the purpose of determining the optimal amounts of soy tauce, soy bean paste and hot pepper paste, in 3 different Korean soups and comparing the effect of condiments such as salt, soy sauce, black pepper, and Mono Sodium Glutamate, on the taste of two basic Korean Stocks, The results estimated by Palatability Test were as follows: 1. The average NaCl contents of two kinds of commercial soysauce were 15.9%, 25.6%, respectively. 2. The NaCl contents of 2% sardine stock, and 19% beef stock were 0.16~1.17%, 0.17~0.18% respectively. 3. @ According to Saltiness in sardine stock, there was no difference between the soy sauce-added group and the salt-added group. However, for the removing effect of Off-flavor, and Overall Taste, the soy sauce-group had higher score than salt-added group respectively. (p>.0.1) ⓑ In beef stock, there was no difference between the soysauce-added group and the salt-added group in Saltiness test. The soy sauce-added group had higher score in removing effect of Off-flavor (p>.0.1). On other hand, in Overall Taste, the salt- added group was more effective than the soy sauce-added (p>.0.1). 4 @ According to Saltiness test in beef stock, there was no difference between the black pepper-added and non-added group. However, for the removing effect of Off-flavor and, Overall Taste, the added group had higher score than the non-added group, respectively. (p>.10, p>.0.5) ⓑ Also, according to Saltiness test in beef stock, there was no diffetrence between the MSG-added group and non-added group. However for the removing effect of Off-flavor, and Overall taste, the MSG-added group was more effective than the non-added group. (p>.01) 5. The best recipes of soybean sprouts soup for S persons, in this experiment, were soybean sprouts 180 g, sardine 28 g, soysauce 1/2 T.S., salt 7 g, green onion 15 g, choppedgarlic It. s., against 7 C of water, and total cooking time was 20 minutes. 6. The best recipes of clear-beef soup were beef 200 g, radish 200 g, tangle weed 10 g, sesame oil 1/2 t.s., green onion 15 g, chopped garlic 1 t.s., black Pepper 1/8 1.s., and soy sauce 1 T.S., salt 7 g or soy sauce 2 T S., salt 3.5 g. The total cooking time was minutes. 7, The best recipes of soybean pasted Chinese cabbage soup were Chinese cabbage 300 g, sardine 28 g, green onion 15 g, chopped garlic 1 t.s., and bean paste 50 g, hot pepper paste 13 g, salt 3g or bean paste 65 g, hot pepper paste 16 g, salt 1.5 g. And the total cooking time was 25 minutes. 8. Soy sauce, black pepper, M.S.G., bean paste and hot pepper paste were effective on removing Off-flavor of experimental soups.

  • The chemical composition of Undaria pinnatifida Laver (U.P. Laver) were determind. 1. Proximate compositions of U.P. Laver were 22.1% crude protein, 1.3% crude fat, 10.2% crude ash, 66.4% N.F.E, 3 0% crude fiber and 22.8% alginic acid. 2. U.P. Laver protein was chiefly composed of glutamic acid (3.24%) and aspartic acid (2.22%) and considerable quantities of leucine (1.98%), alanine (1.56%), valine (1.32%), lysine (1.22%) and phenylalanine (1.15%). 3. The major fatty acid contents of the lipid extracted from U.P. Laver were linolenic acid (45.2%), palmitic acid (26.14%) and linoleic acid (11.27%) and the minor fatty acid were myristic acid (8.41%), oleic acid (8.11%), and stearic acid (0.87%), respectively. The saturated and unsaturated fatty acid ratio of the oil extracted from U.P. Laver was 35.42/64.58 4. The vitamin contents of U. P. Laver were $28.1{\mu}g/g$ niacin, $25.0{\mu}g/g$ vit. E, $18.8{\mu}g/g$ vit. C. The minerals were composed 1.52% calcium, 0.32% phosphorus, 0.88% magnesium, 0.49% potassium and the others were small quantities.
