Compositions of Fatty Acid, Free Amino Acid and Total Amino Acid of Lespedeza x chiisanensis T. LEE (지리산 싸리의 지질 구성 지방산과 유리 아미노산 및 구성 총 아미노산 조성에 관하여)

  • The compositions of amino acid in the protein and fatty acid in the lipid of Lespedeza x chiisanensis T. LEE were analyzed by HPLC and GC, respectively. The contents of crude oil and protein from the extracts were 11.13% and 5.18%, respectively. The amount of free anino acids in the protein was 443.14mg/100g, and 94.84mg/100g of essential amino acid were contained in the free amino acid. The amount of total amino acids in the protein was 3159.85mg/100g, and 1068.18mg/100g of essential amino acid were contained in the total amino acid. The compositions of fatty acid in the lipid were $C_{18\;:\;2}=45.05%,\;C_{18\;:\;3}=18.71%,\;C_{19}=14.70%,C_{18\;:\;1}=6.81%,\;C_{16}=4.35%,\;C_{16\;:\;1}=1.59%$ in order, respectively. 72.44% of unsaturated fatty acids were contained in the lipid.

Effect of Circadian Rhythms on the Xylene Metabolizing Enzyme Activities in Rats (Xylene 대사 효소 활성에 미치는 주.야 시차의 영향)

  • To evaluate an effect of circadian variation on the xylene metabolizing enzyme activities, 50% m-xylene in olive oil(0.25 $m\ell$/100g body weight) was intraperitoneally administered to the rats every other day for 6 days both in the night; 24:00 and the day; 12:00. Then animals were sacrigiced at 8hr after last injection of m-xylene. Hepatic microsomal cytochrome p450 contents were more increased both in control and xylene treated rats of night phase than those of day phase. But the activity of hepatic alcohol dehydrogenase(ADH) in control of night phase showed the similar value with that in those of day phase and xylene treated rats of day phase showed an increasing tendency of hepatic ADH activity as those of night phase showing similar activity. Furthermore, control rats of night phase than those of day phase. And by xylene treatment, enzyme activities of rats of day phase were higher tendency in rats of control but those of night phase were somewhat inhibited. Besides, xylene-treated animals of night phase showed increasing tendency of urinary methylhippuric acid concentration compared with those of day phase. On the other hand, liver weight per body weight(%), hepatic lipid peroxide content and serum xanthine oxidase activity were higher in night phase. And the activities of hepatic oxygen free radical metabolizing enzymes such as xanthine oxidase, gluthathione S-transferase, and xylene-treated rats of night phase than those of day phase. In conclusion, it can be hypothesized on the basis of the results that the accumulation rate of m-xylene intermediate metabolite, i.e. m-methylbenzaldehyde in liver tissus may be higher in night phase than in day phase and it may be responsible for higher liver toxicity in bight phase than in day phase.

  • Printed mottle of coated paper is one of the most common phenomenon and the most difficult problem in offset printing. Offset printing is processed mainly processed by the attractive properties between water and oil, so all inks for off-printing must work with dampening solution(water). It may cause the emulsion on the printing nip from the printing pressure. This study aimed to investigate the effect of emulsified inks on print mottle from emulsified cyan ink by force with each different IP A contents. We measured the print mottle by densitometer and image analysis method. The emulsified inks also give effect to now properties of inks and it caused more ink transfer rate, however, it caused low density of inks. Moreover the emulsified inks were spreaded to around of dots and cause the thinning density on the non-printing area like print mottle. The results showed that the emulsified inks also cause scumming on the printing result with little mistake of adjusting dampening solution and mostly decreasing dot reproduction. So we could find out the obvious effect of emulsified inks to print mottle.

  • Hydrolysis of biomass for the production of fermentable sugar can be improved by the addition of surfactants. In pulp and paper mills, lignin, which is a by-product of the pulping process, can be utilized as a fine chemical. In the hydrolysis process, lignin is one of the major inhibitors of the enzymatic breakdown cellulose into sugar monomer. Therefore, the conversion of lignin into a biosurfactant offers the opportunity to solve the waste problem and improve hydrolysis efficiency. In this study, lignin derivatives, a biosurfactant, was applied to enzymatic hydrolysis of various lignocellulosic biomass. This Biosurfactant can be prepared by reacting lignin with a hydrophilic polymer such as polyethylene glycol diglycidylethers (PEDGE). In this study, the effect of biosurfactants on the enzymatic hydrolysis of pretreated sweet sorghum bagasse (SSB), oil palm empty fruit bunch, and sugarcane trash with different lignin contents was investigated. The results show that lignin derivatives improve the enzymatic hydrolysis of the pretreated biomass with low lignin content, however, it has less influence on the enzymatic hydrolysis of other pretreated biomass with lignin content higher than 10% (w/w). The use of biosurfactant on SSB kraft pulp can increase the sugar yield from 45.57% to 81.49%.

The Development of the HACCP Plan in Korean Rice Cake Manufacturing Facilities (시판 떡류 생산에서 HACCP Plan 개발을 위한 연구)

  • In this study, a Hazard Analysis Critical Control Point (HACCP) plan was developed for the sanitary mass production of commercial Korean rice cake products (Gaepidduk, Injulmi, and Julpyon). The microbiological properties of manufacturing flow were evaluated in order to develop the HACCP Plan. The moisture contents of the rice cakes ranged between 36.2${\sim}$55.3%, whereas the water activity of all samples ranged between 0.954${\sim}$1.0. Microorganisms testing was conducted during various phases of the product flow of Korean rice cake preparation, and included assessments of food equipment, work environment, and cooking employees on a small scale. During the manufacture of Injulmi, Julpyon and Gaepidduk, CCPs were purchasing & storage, steaming and cooling, molding, and holding in the A and B manufactories. At the critical limit of CCPs, storage was conducted below at $5^{\circ}C$ in soybean powder, oil, and paste with redbeans. The steaming process was conducted above at $99^{\circ}C$ for 40 min. Cooling and holding processes were conducted for 2 hours below at $15^{\circ}C$. The molding process included sanitary education for foodhandlers and training for operators. Thus, certain prerequisite programs had to be implemented prior to the implementation of the HACCP system. High levels of bacterial contamination were detected in the aprons worn to work by some employees. Additionally, periodic sanitary education for foodhandlers and training for operators or managers was required. Cross contamination by materials was expected at the place where materials were processed or stored.

Physicochemical Properties of Antioxidant Fractions Extracted from Freeze-Dried Coffee by Various Solvents (동결건조 커피의 순차용매 분획별 특성과 항산화 효과)

  • The relationship between antioxidant activities and physicochemical properties of several fractions obtained from freeze-dried (FD) coffee were investigated. The nine kinds of fraction were consecutively extracted from FD coffee with solvents in increasing order of polarity, and the higher polarity of the solvent the higher extraction yield of the fraction. The antioxidant activities of the fractions were determined by Rancimat and oven test on edible oils. The antioxidant activities of the fractions increased in the order of acetone>ethanol>methanol>50% methanol/water>water fraction, and the antioxidant activities of them were higher on lard than on soybean oil. The antioxidant activity of each fraction was strongly related to the contents of total phenol, total nitrogen content and acidity, whereas color intensity, reducing power, carboxylic acid content showed little contribution to the activity. All fractions had three peaks maxima at 208, 275 and 324 nm on UV-visible spectra, but the only one at 324 nm was linealy proportional to the antioxidant activities of the fractions.

Antiobesity Effect of Baek-Kimchi (Whitish Baechu Kimchl) in Rats Fed High Fat Diet

  • Baek-kimchi (whitish baechu kimchi) was evaluated for anti-obesity properties and effects on triglyceride (TG) and cholesterol in blood and adipose tissues in rats fed a high fat (20 %) diet, and compared to the similar effects of baechu kimchi. Baek-kimchi does not use red pepper powder but contains higher levels of sliced radish and pear than baechu kimchi. SD rats were raised for four weeks on either a normal diet (ND, based on the AIN-93M diet), high fat diet (HFD, supplemented with 16% lard oil in the ND), or HFD containing 5 % baek-kimchi or 5 % baechu kimchi. Feed consumption was not different among the groups, but weight gains were significantly lower in the groups fed either the normal diet or HFD with baek-kimchi or baechu kimchi diets than the group fed HFD alone. The weights of liver and epididymal and perirenal fat pads in baek-kimchi and baechu kimchi diet groups were lower than those of the HFD groups, but the baek-kimchi diet group had lower epididymal and perirenal fat pad weights than the baechu kimchi diet group (p<0.05). The baechu kimchi dietary group also had significantly lower triglyceride and cholesterol contents in liver and epididymal and perirenal fat, reversing the higher levels seen in HFD. Baek-kimchi and baechu kimchi diets were also effective in lowering serum triglyceride and cholesterol levels (p<0.05). These results suggest that baek-kimchi and baechu kimchi consumption can reverse the effects of HFD on weight gain and blood and tissue lipids, and that baek-kimchi is more effective than baechu kimchi. The greater effect is probably due to the higher content of radish and pear used in baek-kimchi.

Characterisation and Co-pyrolytic Degradation of the Sawdust and Waste Tyre Blends to Study the Effect of Temperature on the Yield of the Products

  • The present study aimed to determine the effect of co-pyrolysis of sawdust biomass and scrap tyre waste employing different blending ratios of sawdust to waste tyre such as 100:0, 75:25, 50:50, 25:75, and 0:100. The thermochemical characterization of feedstocks was carried out by employing the proximate, ultimate analysis, and thermogravimetric (TGA) analyses, calorific values, and scanning electron microscope coupled with energy dispersive x-ray analysis (SEM-EDX) to select the blending ratio having better bioenergy potential amongst the studied ratios. The blending ratio of 25:75 (sawdust to waste tyre) was selected for the co-pyrolysis study in a fixed-bed pyrolysis reactor system based on its solid biofuels properties such as heating value (30.18 MJ/kg), and carbon (71.81 wt%) and volatile matter (63.82 wt%) contents. The pyrolysis temperatures were varied as 500, 600 and 700 ℃ while the other parameters such as heating rate and nitrogen flowrate were maintained at 30 ℃/min and 0.5 L/min respectively. The bio-oil yields as 31.9, 47.1 and 61.2 wt%, bio-char yields as 34.5, 34.2 and 31.4 wt% and gaseous product yields as 33.6, 18.60 and 7.3 wt% at the pyrolysis temperatures of 500, 600 and 700 ℃ respectively were obtained. The blends of sawdust and waste tyres showed the improved energy characteristics which could provide the solution for the beneficial management of sawdust and scrape tyre wastes via co-pyrolysis processing.

Microorganisms Involved in Natural Fermentation of Asparagus cochinchinensis Roots and Changes in Efficacies after Fermentation (천문동 뿌리의 자연발효에 관여하는 미생물 및 발효 후 효능 변화)

  • Objectives: The aim of this study was to examine the effect of Asparagus cochinchinensis (AC) and fermented AC (fAC) on microorganisms and efficacies. Methods: AC was fermented for four weeks without using any bacterial strains. Then we investigated fermentation characteristics including potential of hydrogen (pH), total sugar, microbial profiling and antioxidant compound contents such as total polyphenol and total flavonoid. The anti-obesity effects of AC and fAC were evaluated by using Oil Red O staining in 3T3-L1 adipocyte. Also anti-diabetic effects of them were evaluated by using 2-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-d-glucose (2-NBDG) uptake in C2C12 skeletal muscle cell. Results: Both pH and total sugar of fAC were decreased significantly compared to unfermented AC. And the abundance of total bacteria and lactic acid bacteria increased during fermentation, especially Lactobacillus plantarum. Also fermentation of AC increased the content of total polyphenol. On the metabolic aspects, we found that AC and fAC suppressed fat accumulation. Conclusions: After four weeks of fermentation, AC increased concentrations of active compounds, altered microbial composition, and inhibited fat accumulation such as triglyceride. These results indicate that fermentation of AC might be a beneficial therapeutic approach for obesity.

Effects of a mixture of essential oils and organic acid supplementation on growth performance, blood profiles, leg bone length, and intestinal morphology in broilers

  • This study was done to evaluate the effects of a mixture of essential oils and organic acid supplementation on growth performance, blood profiles, leg bone length and intestinal morphology in Ross broilers. A total of 40 Ross 308 broilers ($1140{\pm}80g$) were randomly allocated to 2 groups, a basal diet (CON) and a basal diet + 0.05% $Avi-protect^{(R)}$ (AVI, Mixture of 25% citric, 16.7 sorbic, 1.7% thymol, and 1.0% vanillin), with 20 replicates for every group and 1 chicken per replicate per cage. The broilers were raised in a temperature-controlled room maintained at $24{\pm}1^{\circ}C$ and $50{\pm}5%$ humidity. The body weight (p < 0.05) and weight gain (p < 0.05) of the broilers were increased in the AVI group compared with the CON group. The triglyceride (p < 0.05) and low density lipoprotein (LDL) (p < 0.05) contents were significantly decreased in the AVI group compared with the CON group. There was no significant difference in the leg bone length between the AVI and CON groups (p > 0.05). The villi height (p < 0.05) and goblet cell count (p < 0.05) were significantly increased in the AVI group compared with the CON group. In conclusion, $Avi-protect^{(R)}$ as a feed additive improved the growth performance and lipid metabolism and promoted the development of the intestinal morphology of broilers.
