추출방법에 따른 산초 종자 정유성분의 이화학적 특성 (Physicochemical properties of Sancho (Zanthoxylum schinifolium) seeds oil base extracts from different method)

  • 추출방법을 달리하여 산초 종자의 정유성분을 추출하였다. 초임계유체추출법은 24, 48 및 72 시간으로 구분하였고, 압착법은 볶음 및 증숙 처리하여 추출한 후 이화학적 특성을 분석하였다. 압착법에 의한 추출수율은 전처리 방법에 관계없이 30%로 동일하였으나 초임계유체 추출시는 추출시간이 길어질수록 수율이 증가하여 72 시간 추출하였을 때 38.5%로 가장 높았다. 각각의 방법에 따라 추출된 산초유의 비중과 굴절율은 각각 0.920~0.297과 1.470~1.473의 범위로서 차이를 나타내지 않았다. 점도는 볶음압착법에서 $115.23{\pm}8.85$ cP로 가장 낮았고, 초임계유체 추출시 $181.88{\pm}4.8{\sim}209.03{\pm}1.8$ cP의 범위로 증가하였다. 추출법에 따른 유지의 색도 중 황색도는 초임계유체 추출시 3.81~8.37 범위였으나 압착추출시는 29.14~29.36으로 유의적으로 높아 추출방법에 따라 큰 차이를 보였다. 산가는 증숙압착법으로 추출하였을 때 $0.93{\pm}0.01mg/g$으로 가장 낮았으며, 초임계유체 추출시 추출시간에 따른 유의차는 없었다. 검화가는 182.96~196.57 mg/KOH g의 범위로서 추출방법에 따른 차이가 없었고, TBA가는 158.96~347.14 mg/kg의 범위로 초임계 추출 시간이 길어질수록 증가하였다. 초임계 유체 및 증숙처리에 의한 정유성분에서는 ${\omega}$-3 계열의 linolenic acid를 포함하였으나 볶음처리 추출에서는 측정되지 않았으며, 포화지방산보다 불포화지방산의 함량(17:83, v/v)이 더 많았다.

  • A dense pellicular solid matrix has been fabricated by coating 4% agarose gel on to dense zironia-silica(ZS) spheres by watr-in-oil emulsification . The agarose evenly laminated the ZS bead to a depth of 30㎛, and the resultin gpellicular assembly was characterised by densities up to 2.39g/mL and a mean particle dimeter of 136 ㎛. In comparative fluidisation tests, the pellicular solid phase exhibited a two-fold greater flow velocity than commercial benchmark ad-sorbents necessary to achieve common values of bed expansion. Furthermore, the perlicular parti-cles were characterised by improved qualities of chromatographic behaviour, particularly with re-spect to a three-fold increase in the apparent effective diffusivity of lysozyme within a pellicular assembly modified with Cibacron Blue 3GA. The properties of rapid protein adsorption/desorp-tion were attributed to the physical design and pellicular deployment of the reactive surface in the solid phase. When combined with enhanced feedstock throughput, such practical advantages recommend the pellicular assembly as a base matrix for the selective recovery of protein products from complex, particulate feedstocks(whole fermentation broths, cell disruptates and biological extracts).

가감목방기탕(加減木防己湯)이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향 (Effects of Kagammokbanggi-Tang on the Adjuvant-Induced Arthritis in Rats)

  • Objectives : This study was to investigate the effect of Kagammokbanggi-tang on the Freund's Complete Adjuvant(FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Arthritic rats were divided into control(n=10) and sample(n=10) group. Control group was taken normal saline for twenty days and sample group was taken extracts of Kagammokbanggi-tang for same duration. Normal group(n=10), non-arthritic group, was injected with mineral oil and was taken normal saline for twenty days. Body weight, paw edema volume and ankle joint thickness were measured at 0, 10, 15, 20 days after treatment. $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$ in synovia were analysed by ELISA at 20 days after treatment. Histochemical investigation of NADPH-d in the PAG and histopathological study on the ankle joint were performed at 20 days after treatment. Results : Paw edema volume, ankle joint thickness, $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$, NADPH-d of sample group were significantly decreased compared with control group. Conclusions : These results indicated that Kagammokbanggi-tang has antiarthritic and analgesic effects, and inhibited expression of NOS on the progression of FCA- induced arthritis in rats.

유기농자재 사용에 따른 고추 병해 발생과 토양 미생물상 구조의 상관관계 (Correlation between Disease Occurrences and Microbial Community Structure by Application of Organic Materials in Pepper)

  • 유기농업은 지속가능한 농업을 위해서 필요하며, 생물 다양성 유지를 위해 필수적인 경작 방법으로 전 세계적으로 성장하고 있는 농업 분야 중 하나이다. 유기농업은 인류의 건강과 환경에 유해한 영향을 끼질 수 있는 화학적 제재의 사용을 줄이고, 친환경적 동·식물성 비료, 식물 추출물, 미생물 제재 등이 사용되기 때문에 소비자에게 안전한 농산물을 제공할 수 있다. 노지 고추를 대상으로 3년간 유기농업 환경과 관행농업 환경에서 주요 병해 발생과 토양 미생물상의 조사하였다. 토양 미생물상의 차이는 차세대 염기서열 분석 기법을 활용하여 조사하였으며, 유기농업에 사용되는 제제의 종류에 따라 토양 미생물상이 다르게 형성되었다. 바이러스, 탄저병에 대해서는 관행농업에서의 병 발생률이 낮았으나, 청고병은 미생물 제재 사용에 의해 병 발생률이 낮게 나타났다. 미생물 제재를 사용한 토양에서 투입된 미생물이 토양 미생물에서 검출되었으며, 이는 미생물이 실제 토양에 정착했음을 시사한다.

활락단(活絡丹)의 Adjuvant 유발 관절염 진행 억제 효과 (Inhibiting Effects of Hwallak-dan(Huoluo-dan) on the Adjuvant-induced Arthritis in Lewis Rats)

  • Objectives : This study was to investigate the antiarthritic effect of Hwallak-dan(Huoluo-dan) on the complete freund's adjuvant(CFA)-induced arthritis in Lewis rats. Methods : Arthritis was induced by intradermal injection of CFA into base of tail. Experimental groups divided into normal(n=10), control(n=10) and treated(n=10) group. Animals of control group received normal saline for twenty days, treated group received extracts Hwallak-dan(Huoluo-dan) for same duration, light mineral oil only injected non-arthritic rats were served as normal group. The incidence of arthritis and arthritic index were observed after treatment. Body weight, paw edema volume and thickness of ankle joint were measured at 0, 11, 14, 17, 20 days after treatment. White blood cell(WBC) counts in blood were analysed at 20 days after treatment and tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta}$), IL-6 contents in paw exudate were analysed by ELISA at 20 days after treatment. Histopathology on the ankle joint were performed at 20 days after treatment. Results : 1. Incidence of arthritis of treated group was 60% and control group was 100% at 20 days after treatment. 2. Arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. 3. Paw edema volume and thickness of ankle joint of treated group was significantly decreased compared with control group at 20 days after treatment. 4. Total WBC of treated group was significantly decreased compared with control group at 20 days after treatment. 5. Neutrophils of treated group was significantly decreased compared with control group at 20 days after treatment. 6. $TNF-{\alpha}$ content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 7. $IL-1{\beta}$ content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 8. IL-6 content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 9. Histopathological arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. Conclusions : These results indicate that Hwallak-dan(Huoluo-dan) has inhibitory effect on the development and progression of CFA-induced arthritis in rats.
