Determination and Survey of Fluoroquinolones Residue in Chicken Muscle by HPLC with Fluorescence Detector (액체크로마토그래피-형광검출기를 이용한 닭고기 중 플루오로퀴놀론계 항균물질 정량분석 및 잔류조사)

  • Ofloxacin, norfloxacin, ciprofloxacin, and enrofloxacin in chicken muscle were seperated by liquid extraction and determined with high performance liquid chromatography (HPLC) with fluorescence detector. Analysis was carried out using following conditions; Cl8 column (250${\times}$4.6 mm i.d. 5 ${\mu}{\textrm}{m}$ particle size), mobile phase composed of D.W. (containing 0.4% triethylamine and phospholic acid): methanol : acetonitrile (800:100:100, v/v/v), isocratic pump at a flow rate of 1.0 $m\ell$/min and 50 ${mu}ell$ of injection volume, fluorescence detector with EX278 nm/EM.456 nm. The calibration curves of four fluoroquinolones showed linearity (${\gamma}$$^2$$\geq$0.999) at concenration range of 0.025-0.6 $\mu\textrm{g}$/ml. The recoveries in fortified chicken muscle represented more than 80% with low coefficient of variation (〈10%) for concentration range of four fluoroquinolones. The detection limits for ofloxacin, norfloxacin, ciprofloxacin, and enrofloxacin were 23.5, 3.4, 3.0 and 2.5 ng/g in chicken muscle, respectively. We also monitored fluoroquinolones residue in muscle of chickens (broiler 1:227, Korean native chicken 219, laying chicken 77) using EEC-4-plate screening and HPLC conformation methods. Ten(broiler 5, Korean native chicken 5) out of the fifteen samples which were positively detected by EEC-plate screening method from 1,523 chicken meat were confirmed with ciprofloxacin and enrofloxacin by HPLC. The ranges of residual concentration were 0-0.12 ppm for ciprofloxacin and 0.01-6.79 ppm for enrofloxacin. In conclusion, our method could be applied effectively to determine four fluoroquinolones residues in chicken meat, and further survey for fluoroquinolones residue in chicken meat are needed for more effective control of fluoroquinolones used in livestock.

Comparison of the E-Test with Agar Dilution Susceptibility Test by Using Bacteroides fragilis (Bacteroides fragilis의 E-test와 한천 평판 희석법에 의한 항균제 감수성상의 비교)

  • The susceptibilities of 45 clinical isolates of bacteroides frogilis to cefaclor, ciproflxacin and imipenem were determined by new method, E-test (AB Bidisk, Solna, Sweden) and were compared with those from conventional agar dilution method by using brain heart infusion, Mueller-Hinton and Wilkins Chalgren agar plates. And the susceptibility of 60 clinical isolates of Bacteroides fragilis group (B. fragilis 45 strains, B. distasonis 6 strains, B. ovatus 5 strains, B. thetaiotaomicron 4 strains) to 5 quinolones (ciprofloxacin, enoxacin, norfloxacin, ofloxacin, pefloxacin) were determined by in vitro agar dilution method. Compared with agar dilution MICs for B. fragilis 45 strains, 90.3% of E-test MICs were within ${\pm}$1 dilution of the agar dilutions, and 98.4% were within 2 dilutions. And there were little effect of different medium bases to determine MICs except Mueller-Hinton agar. On Mueller-Hinton agar, B. fragilis showed have or no growth activity. In vitro susceptibility of B. fragjlis group to quinolones, most of the test strains showed resistant patterns to quinolones except ofloxacin and there was little difference of susceptibility patterns between species of B. fragilis group.

Susceptibilities of Quinolones against Erythromycin- and Tetracycline-Resistant Mycoplasma pneumoniae Isolates (Erythromycin과 tetracycline에 저항성인 Myoplasma pneumoniae의 quinolone계 항생물질에 대한 감수성)

  • The in vitro activities of quinolones were determined with 36 strains of tetracycline- resistant, 39 strains of erythromycin-resistant, 21 strains of both resistant to tetracycline and erythromycin, and 27 strains of both susceptible to tetracycline and erythromycin of the M. pneumoniae isolates which have been isolated and classified in previous studies. The $MIC_{90}$ of moxifloxacin, levofloxacin, sparfloxacin, ofloxacin, ciprofloxacin, amikacin, and clarithromycin against 36 strains of tetracycline-resistant M. pneumoniae isolates were 0.125, 0.39, 0.125, 1.0, 1.0, 15.6, and $1.56\;{\mu}g/ml$, respectively. The $MIC_{90}$ against 39 strains of eryhtromycin-resistant M. pneumoniae isolates were 0.06, 0.39, 0.125, 1.0, 1.0, 15.6, and $25.0\;{\mu}g/ml$, respectively. The $MIC_{90}$ against 21 strains of both resistant to tetracycline and eryhtromycin of M. pneumoniae isolates were 0.125, 0.39, 0.125, 1.0, 1.0, 3.9, and $1.56\;{\mu}g/ml$, respectively. The $MIC_{90}$ against 27 strains of both susceptible to tetracycline and eryhtromycin of M. pneumoniae isolates were 0.125, 0.39, 0.125, 1.0, 1.0, 7.8, and $0.09\;{\mu}g/ml$, respectively. These result suggest that moxifloxacin, levofloxacin, and sparfloxacin of the quinolones might be promising antimicrobial agents for the treatment of tetracycline- and erythromycin-resistant M. pneumoniae infection. However, selection of ofloxacin, ciprofloxacin, and clarithromycin for the treatment of M. pneumoniae infection should be cautious decision with antimycoplasmal susceptibility test.

Analysis of Residual Fluoroquinolones Contents in Fish (어류 중 Fluoroquinolone계 항균제의 분석 및 잔류량 조사)

  • The residual contents of fluoroquinolones in fish and shrimp were analyzed by using HPLC-FLD and LC-MS. The limit of detection (LOD) and limit of quantification (LOQ) for fluoroquinolones by HPLC-FLD were 0.0030-0.0083 and 0.0090-0.0252 mg/kg, respectively. Those by LC-MS were 0.0019-0.0040 and 0.0059-0.0122 mg/kg, respectively. Recoveries regarding ofloxacin, norfloxacin, pefloxacin, ciprofloxacin and enrofloxacin ranged from 76.4-104.6%. The detection rate of fluoroquinolones was 7.5% from a total of 268 samples. In only one Korean bullhead sample, the sum of ciprofloxacin and enrofloxacin contents was detected at a higher level than its maximum residue limits (MRL) of 0.1 mg/kg. Ofloxacin, norfloxacin and pefloxacin were not detected in any samples and detection rates of ciprofloxacin and enrofloxacin were 3.4% and 6.7%, respectively. In detection rate by species eel (2.6%), loach (2.2%), Korean bullhead (1.5%), carp (0.7%) and snakehead (0.4%) were in order.

The Effects of KR-10876, a new Quinolone Antimicrobial Agent, on the Central Nervous System

  • To evaluate KR-10876, a new fluoroquinolone antibacterial agent, its effects on the central nervous system(CNS) were investigated in mmice as part of phamacological study, and the results were compared with those for ciprofloxacin and ofloxacin, two prototypes of quinolone antiabctrial agents. All the parameters indicative of CNS function and acute toxicity were measured by close observation of the animals at regular time intervals after oral treatment of test compounds. KR-10876 did not have any effect on the parameters measured at lower does (100, 300 mg/kg, p.o.), it caused ptosis, suppressed spontaneous locomotor activity, hypothemia, and prolonged hexobarbital-induced sleeping time. KR-10876 also had a slight effect on motor coordination only at high dose. Simialr to ciprofloxacin, KR-10876 did not protect mice from pentylenetetrazol-strychnine-, and electroshock-inducedl convulsions at doses tested. These findings demonstrate that KR-10876 affects CNS functions only at high doses. The rank order for effects is ofloxacin$\le$KR-10876>ciprofloxacin.

Characteristics and Antimicrobial Susceptibility Patterns of Pasteurella multocida Isolated from Pneumonic Lung Lesions of Swine (돼지 폐렴병소에서 분리한 Pasteurella multocida의 특성 및 항생제 감수성 양상)

  • The present study was conducted to investigate the species-specific gene detection and antimicrobial susceptibility of Pasteurella (P.) multocida isolated from pneumonic lung lesions of Youngnam swine herds during the period from July 2006 to September 2007. A total of 91 (36.3%) strains of P. multocida were isolated from 251 pneumonic lung lesions. The species-specific P. multocida gene was detected at 460 bp amplicons by PCR. The P. multocida tested was susceptible to florofenicol (93.4%), amikacin (91.2%), cephalothin (87.9%), cefoxitin (84.6%), ofloxacin (80.2%) and norfloxacin (65.9%) in 27 antimicrobial susceptibility tests. Most of strains were resistant to more than 5 drugs.

Resistance to fluoroquinolone of Staphylococcus pseudintermedius isolated from dogs and cats in Daegu (대구지역 개와 고양이에서 분리된 Staphylococcus pseudintermedius의 fluoroquinolone 내성)

  • The purpose of this study was to investigated the fluoroquinolone (FQ) resistance and presence of gyrA and grlA gene in 87 Staphylococcus pseudintermedius isolates obtained from clinical samples of dogs and cats. Also, the profiles of FQ resistance compared with methicillin resistant S. pseudintermedius (MRSP) isolates. FQ resistance was observed for enrofloxacin (41.4%), ciprofloxacin (39.1%), norfloxacin (36.8%), ofloxacin and levofloxacin (32.2%, respectively), and moxifloxacin (31.0%). Thirty-eight (43.7%) of 87 S. pseudintermedius isolates were resistant to more than one FQ. Twenty-six (64.5%) of 38 FQ resistant isolates were resistant to all the six FQ tested. Of 38 FQ resistant isolates, gyrA gene was detected in all isolates but grlA gene was not found. Moreover, 19 MRSP isolates were resistant to enrofloxacin (63.2%), ciprofloxacin (57.9%), norfloxacin (52.6%), and ofloxacin, levofloxacin and moxifloxacin (47.4%, respectively). FQ resistance were highly prevalence in S. pseudintermedius isolates from dogs and cats. Our results emphasize the prudent use of antimicrobial agents to companion animals is necessary for prevent antimicrobial resistance.

Polyacrylamide Gel Immobilization of Porcine Liver Esterase for the Enantioselective Production of Levofloxacin

  • Porcine liver esterase was immobilized in polyacrylamide gel for the enantioselective production of levofloxacin from ofloxacin butyl ester. The initial activity of immobilized esterase was found to be significantly affected by the polyacrylamide gel composition. The optimum concentrations of monomer and crosslinker were determined to be 20% and 8.3%, respectively. The activity of immobilized esterase was 55.4% compared to a free enzyme. Enantiomeric excess was maintained at 60%, almost the same level as that of free enzyme. In addition, the immobilized esterase could be used repeatedly up to 10 times without experiencing any severe loss of activity and enantioselectivity.

Effect of Ophthalmic Fluoroquinolones on Bacterial Conjunctivitis: Systematic Review (세균성결막염에 대한 안과용 플루오로퀴놀론계 항균제의 효과: 체계적문헌고찰)

  • This systematic review was conducted to assess the clinical effect of ocular fluoroquinolones used for the treatment of bacterial conjunctivitis. A literature search for randomized controlled clinical trials registered up to January 2010 based on PubMed database, using the following search terms: conjunctivitis and fluoroquinolones (besifloxacin, moxifloxacin, gatifloxacin, levofloxacin, lomefloxacin, ciprofloxacin and ofloxacin) were performed. Pooled data on the clinical resolution and bacterial eradication rates derived from selected 16 studies were reported as the relative risk (RR) and 95% confidence interval (95% CI) compared with placebo. Early clinical resolution and microbiological eradication rates in placebo were 28% and 62% respectively. Fluoroquinolones were significantly effective comparing to placebo: early RR 1.94 (95% CI 1.60~2.34) and late RR 1.30 (1.19~1.43) in clinical resolution rates, and early RR 1.75 (1.58~1.94) and late RR 1.28 (1.18~1.39) in microbiological eradication rates. Besifloxacin, ciprofloaxain and moxifloxacin in clinical resolution, and besifloxacin and levofloxacin in microbiological eradication showed higher RRs than pooled overall fluoroquinolones' RRs. New quinolones had higher antibacterial potencies for all pathogens isolated from bacterial conjunctivitis and resistant isolates than old generation quinolones. In conclusion, ocular 7 fluoroquinolones were all effective than placebo for bacterial conjunctivitis and there were differences between quinolones in early and late clinical resolutions and microbiological eradications, and no differences in safety comparing to placebo.

Isolation and Characterization of Antibiotic and Heavy Metal-Resistant Pseudomonas aeruginosa from Different Polluted Waters in Sohag District, Egypt

  • Different polluted water samples were collected from a wastewater treatment plant, agricultural drainage canals, the River Nile, and irrigation canals. The samples were examined for the enumeration of Pseudomonas aeruginosa in the Sohag area, Egypt over a period of one year. A total of 240 isolates were collected and tested for their resistance to 12 common antibiotics and 6 heavy metals. The isolates were found to be less resistant to norfloxacin(1.7%), ofloxacin(4.6%), amikacin(9.6%), tobramycin (10.4), carbenicillin (15.4), and gentamycin (41.3%), yet more sensitive to rifampicin (75%), kanamycin (89.6%), ampicillin (90.8%), chloramphenicol (91.7%), streptomycin (92.9%), and tetracyclin(96.3%). In contrast, 7.1%, 12.9%, 25.4%, and 53.7% of the isolates were resistant to lead, cadmium, mercury, and zinc, respectively. None of the isolates had developed a resistance to silver or molybdenum. The high frequency of metal-antibiotic double resistance existed between lead and amikacin (56.5%), cadmium and ofloxacin (72.7%), zinc and norfloxacin (100%), and mercury and carbenicillin (94.6%). The high occurrence of antibiotic-resistant bacteria in natural water could be related to the widespread use of antibiotics, with possible public health hazard.
