The ecotoxicological effects of 4-tert-octylphenol were observed on non-biting midge, Chironomus plumosus collected from Anyang stream in Seoul. The survival rate and adult emergence rate on C. plumosus exposed to octylphenol were not significantly affected. However, 4-tert-octylphenol induced developmental delay and disrupted sex ratio in high concentration of octylphenol. The mouth deformity such as tooth deletion or fusion in mentum, and tooth deletion in mandible were observed exposure to 4-tert-octylphenol. The deformity type of the mentum showed deletion (LT, 6.7%), and fusion (LT, 6.3%). Moreover, tooth deletion of mandible was observed in 4-tert-octylphenol treated groups (6.7%, 3 ppm).
The ecotoxicological effects of 4-tert-octylphenol were observed on Harpacticoid copepoda Tigriopus japonicus s.l. gathered and cultured from tidal pool of Korean coast. There were no significant differences in survival rate (except 10 $\mu$g/L; 70.00%) and sex ratio (except 30 $\mu$g/L) on T. japonicus s.l. exposed to 4-tert-octylphenol. However, 4-tert-octylphenol induced developmental delay (copepodite emergence day and adult male mergence day) and retardation of first brooding day of adult female. Moreover the body size and biomass decreased at 4-tert-octylphenol exposure. As a result, detailed life-cycle research of T. japonicus s.l. may yield potential bioindicators for environmental monitoring and assessment.
Recalcitrant 4-t-Octylphenol used as a surfactant was subjected to the biodegradation with wood rot fungi, Basidioradulum molare and Schizopora paradoxa. Two fungi were grown in the culture medium containing various concentrations of 4-t-Octylphenol in order to investigate their resistance against 4-t-octylphenol Schizopora paradoxa was reached to the full growth within 14 incubation days in the concentration of more than 200 ppm of 4-t-Octylphenol, while Basidioradulum molare showed the inhibitory mycelium growth as the concentration was increased Schizopora paradoxa and Basidioradulum molare biodegraded 95% and 36% of initial concentration of 4-t-Octylphenol at first incubation day, respectively. However, the biodegradation capability reached to more than 95% after 3 incubation days. During the biodegradation of 4-t-Octylphenol, the activity of manganese dependent peroxidase was induced by the addition of 4-t-Octylphenol in the culture medium of Schizopora paradoxa, but that of laccase was maximal before the addition. The reduction of estrogenecity was assayed by MCF-7 cell proliferation test and measurement of pS2 mRNA expression. The level of pS2 mRNA was decreased down to the level of baseline at first incubation day. Also, estrogenecity of 4-t-Ocrylphenol completely disappeared after treatment with supernatant by Schizopora paradoxa and Basidioradulum molare from first incubation day of culture down to the levels of vehicle.
A comprehensive analytical method of endocrine disruptors[i.e., nonylphenol(NP), octylphenol(OP), bis(2-ethylhexyl) phthalate(BEHP)] in meat or pork samples was developed. The method employed closed culture tube extraction with dichloromethane and solvent exchange to iso-hexane and SPE(2g) aminopropyl column, followed by determination on gas chromatography linked to mass spectrometry(GC/MS) operated in the single ion monitering(SIM) mode. For the multipoint recovery of nonylphenol, octylphenol and bis(2-ethlhexyl) phthalate OP, NP were showen good recoveries in $0.125-1.25{\mu}g/g$ range of concentration, and BEHP more good recoveries in $0.0125-12.5{\mu}g/g$ wide range of concentration. The present method was applied to beef or pork samples of mart and butcher in Cheonju city and near Cheonju. The range of concentrations was respectively, $0.06-0.24{\mu}g/g$ in nonylphenol(NP) and $0.36-2.35{\mu}g/g$ in bis(2-ethylhexyl) phthalate(BEHP), but octylphenol(OP) was not dected in any samples. This method provides a powerful analytical tool to investigate a wide range of endocrine disruptors in biological samples of limited quantity.
Sewage sludge samples were extracted with dichlomethane by Soxhlet extraction. The extracts were analysed for the endocrine disrupting compounds (EDC), octylphenol(OP), nonylphenol (NP) and bis (2-ethylhexyl)phthalate (DOP) using GC/MS-SIM. In industrial sewage sludge (sample Z-1) the concentration of NP and DOP were respectively $1168{\mu}g/g$ and $1172{\mu}g/g$ whereas in domestic sludge (sample Z-2) the concentration were respectively $10.8{\mu}g/g$ and $80{\mu}g/g$. OP was not detected in Z-2, but detected a small amount in Z-2 of the samples. Environmental EDC have been shown to affect the fertility of range of living organisms, including humans, and the high levels of NP and DOP in industrial sewage sludge could have serious implications for the reproductive health of humans particularly if the compounds entered the human food chain through water or the ingestion of vegetables or animal products.
To investigate the effects of octylphenol and nonylphenol on the reproduction of the striped field mouse, the mice were subcutaneousely injected with 4-t-octylphenol 800 mg/kg (OP800), nonylphenol 900 mg/kg (NP900) or OP800 + NP900 (OPNP), respectively, in two times a week for 2 months. As the results, there were no differences between the control and the compound-treated groups in the body weight in both sexes, and the GSI (gonadosomatic index) and SVI (seminal vesicles index) in the adult males. But histological abnormalities of the reproductive organs in the mice seems to be related to the compound. This suggestion is supported by the facts that most mice treated with the compounds had only a small number of spermatozoa in the shrunken epididymal tubules. In addition, the shrunken reproductive organ in one mouse treated with the NP, suggests that the abnormalities in the wild striped field mice might be induced by the compound. Furthermore, it is indicated that the compounds must be toxicants to inhibit pregnancy, judging from the fact that all the mice treated with the compounds had failed to deliver.
Determination were performed for the presence of three common environmental endocrine disrupting compounds (EDC), namely nonylphenol (NP), octylphenol (OP), di (2-ethylhexyl)phthalate in sewage sludge from 67 wastes plants, using Soxhlet extraction and GC/MS-SIM. NP and DEHP were detected in almost every sludges but OP was not detected. The range and mean value for NP of all sludge samples were nd~593, $62{\mu}g/g$ dry and the two statistic values for DEHP of those were 13~4733, $1176{\mu}g/g$ dry respectively. DEHP content was 20 times greater than NP in sludges. Concentrations of NP and DEHP in sludge from individual area were increased as following order ; Jeolla South and North Province (Cheju) < Chungcheong South and North Province (Kwangwon) < Gyungsang South and North Province < Wide City < Kyunggi Province.
A comprehensive analytical method of endocrine distruptors[i.e., nonylphenol(NP), octylphenol(OP), bis(2-ethylhexyl)phthalate(BEHP)] is fish samples was developed using two internal standards. This method employed closed culture tube extraction with dichloromethane and solvent exchange to iso-hexane and SPE(2g) aminopropyl column, followed by determination on gas chromatograph linked to mass spectrometer(GC/MS) operated in the single ion monitoring(SIM) mode. The recoveries of nonylphenol and octyphenol in the range of $0.2{\sim}20{\mu}g/g$ using 1-phenyl decanol as one internal standard were over 75%, and recovery of bis(2-ethylhexyl)phthalate in the range of $0.4{\sim}40{\mu}g/g$ using bis(2-ethylbutyl)phthalate(BEBP) as the other internal standards was showed over 102%. The present method was applied to fish samples from Korea and UK. The range of concentrations for nonylphnol(NP) and bis(2-ethylhexyl)phthalate in Korean fish were $0.02{\sim}0.06{\mu}g/g$ in 2 samples and $0.18{\sim}2.03{\mu}g/g$ in 9 samples respectively, but bis(2-ethylhexyl)phthalate(BEHP) in UK samples was found $2.99{\mu}g/g$ in just 1 sample. But octylphenol(OP) was not dected in any samples by this method. This two internal standard method provides a more precise analytical tool to investigate endocrine disruptors in a biological matrices of limited quantity.
Determination were performed for the presence of three common environmental endocrine disrupting compounds (EDC), namely nonylphenol (NP), octylphenol (OP), di(2-ethylhexyl)phthalate (DEHP) in rainwater, distilled water, millipore water, groundwater, tap water, and bottle water in market, using OASIS cartrige extraction and GC/MS-SIM. Octylphenol was not detected in any water samples. The order of nonylphenol concentrations were increased in rainwater < millipore water < distilled water < groundwater < tap water < bottle water. The concentration of nonylphenol in bolltle water was $0.44{\mu}g/L$(ppb) at highest. Di(2-ethylhexyl)phthalates were detected in every water samples, and the concentrations were to be $1.70{\sim}2.92{\mu}g/L$ in rainwater, $8.7{\sim}31.7{\mu}g/L$ in distilled water, $0.6{\sim}5.6{\mu}g/L$(mean: $2.5{\pm}0.3{\mu}g/L$) in millipore water, and that were very stable values than other waters, $1.1{\sim}6.0{\mu}g/L$ in groundwater at near Jeonju, $3.1{\sim}5.8{\mu}g/L$ by means in tap water, $0.5{\sim}67.6{\mu}g/L$ in bottle water from market. The concentration of DEHP in bottle water samples from market were in higher than USEPA regulation at over 40%.
Connexins (Cx) are membrane proteins and monomers for forming gap junction (GJ) channels. Cx46 and Cx50 are also known to function as conductive hemichannels. As part of an ongoing effort to find GJ-specific blocker(s), endocrine disruptors were used to examine their effect on Cx46 hemichannels expressed in Xenopus oocytes. Voltage-dependent gating of Cx46 hemichannels was characterized by slowly activating outward currents and relatively fast inward tail currents. Bisphenol A (BPA, 10 nM) reduced outward currents of Cx46 hemichannels up to ~18% of control, and its effect was reversible (n=5). 4-tert-Octylphenol (OP, $1{\mu}M$) reversibly reduced outward hemichannel currents up to ~28% (n=4). However, overall shapes of Cx46 hemichannel current traces (outward and inward currents) were not changed by these drugs. These results suggest that BPA and OP are likely to occupy the pore of Cx46 hemichannels and thus obstruct the ionic fluxes. This finding provides that BPA and OP are potential candidates for GJ channel blockers.
