Both polar and gravity-induced lateral transport of auxin was markedly reduced in corn coleoptile segments by octanol treatment. Octanol enhance net auxin uptake without affecting that of benzoic acid, suggesting that the effect did not result from a nonspecific action on general membrane permeability. Since naphthylphthalamic acid (NPA) action on both transport and net uptake of auxin was substantially decreased in the presence of octanol, a specific interaction of octanol with the NPA site (efflux carrier) can be postulated. Studies on in vitro binding of NPA to membrane vesicles indicated that octanol did not interfere with NPA binding. When basipetal transport of auxin was impared by plasmolysis, octanol still inhibited auxin transport in the plasmolyzed tissues. The results ruled out the possibility of octanol acting at the plasmodesmata. Kinetic analysis of growth indicated that IAA-sustained growth was rapidly blocked by octanol implicating a common system by which auxin transport is linked to auxin action. Possible mechanisms for octanol action will be discussed.
Triton X-100 계면할성제 존재하에서, $Eu^{3+}$-thenoyltrifluoroacetone(TTA)계의 형광세기의 n-octanol 영향에 관해 연구하였고, n-octanol의 정량을 위한 최적 조건을 규명하였다. 이 착물계에서 들뜨기 파장을 345nm로 주사했을 때, $Eu^{3+}$ 이온의 최대 형광파장은 619nm에서 나타났다. n-octanol의 검정곡선은 $1{\times}10^{-5}M{\sim}1{\times}10^{-7}M$의 범위에서 직선적인 관계를 얻었고, 검출한계는 $1{\times}10^{-9}M$이었다. 이 방법을 이용하여 합성시료를 분석한 결과 분석오차내에서 기지값과 일치하였으며, 상대표준편차는 약 3.5%였다.
A shake-flask method was used to determine the n-octanol/water partition coefficients of sulfamethazine, sulfadimethoxine, sulfamethoxydiazine, sulfamonomethoxine, sulfamethoxazole, sulfaquinoxaline and sulfachloropyrazine from (298.15 to 333.15) K. The results showed that the n-octanol/water partition coefficient of each sulfonamide decreased with the increase of temperature. Based on the fluid phase equilibrium theory, the thermodynamic relationship of n-octanol/water partition coefficient depending on the temperature is proposed, and the changes of enthalpy, entropy, and the Gibbs free energy function for sulfonamides partitioning in n-octanol/ water are determined, respectively. Sulfonamides molecules partitioning in n-octanol/water is mainly an enthalpy driving process, during which the order degrees of system increased. The temperature effect coefficient of n-octanol/water partition coefficient is discussed. The results show that its magnitude is the same as that of values in the literature.
An experimental study was conducted on a 4-stroke direct injection diesel engine to examine the combustion and emission characteristics of 1-octanol/diesel fuel blends. The concentration of 1-octanol in the fuel blends was 10%, 30%, and 50% by volume. Experiments were conducted by varying the engine torque from 6 Nm to 12 Nm at the same engine speed of 2,700 rpm. Results showed that the fuel conversion efficiency increased as the 1-octanol proportion increased under most experimental conditions. However, the brake specific fuel consumption increased due to the relatively low lower heating value of 1-octanol. The smoke opacity and the concentrations of NOx and CO emissions generally decreased with brake mean effective pressure as the 1-octanol proportion increased. On the other hand, the unburned hydrocarbon concentration increased with an ascending ratio of 1-octanol.
Octanol/water partition coefficients of 52 chemicals were calculated using RP-HPLC estimation method and predicted by computer program, PCHEM. The result showed relationship between literature values and RP-HPLC observed values (relative coefficient r$^2$=0.916), but the relationship of PCHEM values with literature values was lower than RP-HPLC value (relative coefficient r$^2$=0.795). The average difference in partition coefficient between the RP-HPLC method and flask-shaking method was log Kow=0.54, while the average difference between the values predicted form the computer program and flask- shaking method was log Kow = 0.36 Compared to octanol/water partition coefficients by 3 methods (Flask-shaking, RP-HPLC, computer prediction), the octanol/water partition coefficient values based on the flask-shaking method were very similar to the literature values, while the octanol/water partition coefficient values by RP-HPLC method without to consider the dead time, and computer prediction values did not significantly differ with the literature values.
능이버섯의 건조특성에 미치는 외부 요인을 조사하기 위하여 온도 3수준$(30^{\circ}C,\;50^{\circ}C,\;70^{\circ}C)$과 풍속 3수준(0.5 m/sec, 1.0 m/sec, 1.5 m/sec)에서 능이버섯의 건조실험을 수행하였으며, 각 조건에서의 향기성분과 건조과정 중 향기 성분의 변화를 조사하였다. 능이버섯의 건조는 상당기간이 감률건조기간으로 나타났으며, 건조속도는 건조온도가 높아질수록 증가했으나, 풍속은 건조에 미치는 영향이 작게 나타났다. 건조온도 $50^{\circ}C$, 풍속 1.5 m/sec의 건조조건에서 1-octen-3-ol, 3-octanone, 1-octanol, 3-octanol등의 peak area가 각각 17976, 11570, 1016, 1077로 다른 건조온도 조건에 비해 높게 나타났다. 건조시간에 따른 능이버섯의 주요 향기성분 함량 변화를 보면 1-octen-3-ol을 비롯한 1-octanol, 2-octen-1-ol, 3-octanol 등의 $C_8$ 화합물 peak area가 크게 감소하였고, C8 화합물 중 1-octen-3-one, 3-octane등의 상대적 함량비는 증가하였다. 또한 건조과정 중 butyric acid, propanoic acid, 3-methyl thiopropanol 등과 같은 화합물이 생성되었다.
Simultaneous distillation-extration method에 의해 4종의 국내에서 인공재배되고 있는 느타리 버섯류 (Pleurotus sp)로부터 휘발성 향기성분을 분리한 다음 GC-MS 및 GC에 의한 표준품과의 머무름 시간 비교에 의해 성분을 확인 하였다. 확인된 54종의 휘발성 화합물중 저온성 느타리 (Pleurotus ostreatus ASI 201)에서는 1-octen-3-ol, 3-octanone, 3-octanol등이, 원형 느타리 (Pleurotus florida--ostreatus-ostreatus)에서는 1-octen-3-ol, n-hexanol, 3-octanone, 3-octanol, phenol등이, 애느타리 (Pleurotus ostreatus)에서는 1-octen-3-ol, n-pentanal, n-hexanol, n-pentanol, 3-octanone, 3-octanol, 1,5-octadien-3-one등이었다. 특히 확인된 성분중에는 탄소수가 8개인 alcohol 및 carbonyl화합물이 많았으며 전체 향기성분에 대한 $C_8$화합물의 비율은 각각 56.60%, 72.46%, 54.84%, 35.85%이었다.
능이버섯의 향기성분을 조사하기 위하여 SDE, SFE, headspace 로 추출하고 GC, GC-MS로 동정하였으며, 능이버섯의 향기특성을 조사하기 위하여 GC-olfactometry 방법중 AEDA법을 이용하였다. 그 결과 SDE 추출물에서는 33개, SFE추출물에서는 26개, headspace 추출물에서는 17개의 화합물이 동정되었다. 확인된 화합물의 주성분은 1-octen-3-ol, 1-octen-3-one, 3-octanone, 2-octen-1-ol, 3-octanol, 1-octanol 등의 $C_8$ 화합물과 benzeneacetaldehyde등이었다. AEDA 결과 능이버섯의 주된 향기특성으로는 생능이버섯내(unknown), 생버섯내(1-octen-3-ol), 곰팡이내(1-octen-3-one), 흙내(3-octanol), 풀내(1-octanol), 강한 풀내(2-octenal), 단 꽃내(benzeneacetaldehyde), 목재내(L-linalool), 치즈내(3-hydroxy-2-butanone), 상큼한 단내(2-decanone), 약간의 불쾌한 고기내(3-methyl thiopropanol), 시큼한 우유내(propanoic acid) 등으로 나타났다.
세포내 유리칼슘(free calcium, $Ca^{2+}$)은 세균에서 고등동물에 이르기까지 거의 모든 세포에서 세포 고유작용을 조절하는 중요한 세포내 신호전달체계(signal transduction system)의 매개체이다. 타액선 세포에서 부교감 신경 자극으로 타액분비가 증가될 때에도 세포내 $Ca^{2+}$ 농도 증가가 가장 중요한 역할을 한다. 그러나 췌장(pancreas)의 경우 세포내 $Ca^{2+}$ 이외에도 인접세포를 전기적 화학적으로 연결해주는 gap junction이 외분비 기능을 직접적으로 조절할 가능설이 제시되었다. 타액선 세포에서도 세포막에 고농도의 gap junction이 존재하고 있으며 gap junction을 통해 인접세포들이 전기적, 화학적으로 연계되어 있어 gap junction이 타액선 세포의 기능을 직접적으로 조절할 가능성을 내포하고 있다. 따라서 gap junction이 타액선의 타액분비 작용에도 중요한 역할을 하며 이러한 작용이 세포내 $Ca^{2+}$ 농도를 조절하여 이루어질 것이라는 가정하에 이를 확인하는 실험을 시행하였다. 흰쥐 악하선에서 유리되는 타액양을 측정하기 위해서 악하선으로 혈액을 공급하는 동맥에 가는 관을 삽입하여 생리 식염수를 관류하면서 타액선관을 통해 타액을 채취하였다. 세포내 $Ca^{2+}$ 농도는 분리한 악하선 acini 내에 $Ca^{2+}$ 농도 변화에 민감하게 반응하는 형광물질인 fura-2를 축적시키고 형광 분석기를 사용하여 형광강도를 측정하여 다음과 같은 결과를 얻었다. 1. CCh 투여로 타액 분비가 증가하였을 때 gap junction을 봉쇄하는 약물인 octanol(1 mM)을 투여하면 타액분비가 봉쇄되었으며 이는 가역적 반응이었다. 2. CCh투여로 세포내 $Ca^{2+}$ 농도가 증가하였을 때 1mM octanol을 투여하면 세포내 $Ca^{2+}$ 농도가 CCh 투여전의 상태로 감소되었다. 3. Octanol은 CCh에 의하여 유발된 초기 $Ca^{2+}$ 증가를 억제하지는 못한 반면에 후기 $Ca^{2+}$ 농도를 감소시켰다. 4. 세포막 $Ca^{2+}$ 통로를 열어주는 약물인 thapsigargin($1{\mu}M$)을 투여하여 세포내 $Ca^{2+}$ 농도를 증가시킨 후 1mM octanol을 투여하면 세포내 $Ca^{2+}$ 농도가 thapsigargin 투여 전의 상태로 감소하였다. 5. 2,5-di-tert-butyl-1,4-benzohydroquinone(TBQ)의 투여로 세포막을 통한 $Ca^{2+}$ 농도의 주기적 변동인 $Ca^{2+}$의 oscillation이 유발되었는데, 이때 1mM octanol을 투여한 경우에 $Ca^{2+}$농도의 oscillation이 정지하여 역시 gap junction을 봉쇄하면 TBQ에 의해서 유발된 세포내 $Ca^{2+}$ 농도의 주기적 변동이 사라지고 $Ca^{2+}$ 농도의 감소가 나타남을 확인하였다. 6. Gap junction을 봉쇄하는 또 다른 약물인 glycyrrhetinic acid($100{\mu}M$)도 CCh 자극으로 인한 타액분비를 억제하였다. 이상의 결과로 미루어 gap junction은 흰쥐 악하선 세포로부터의 타액분비 조절에 중요한 역할을 하는데, 이는 gap junction이 세포막 $Ca^{2+}$ 통로를 조절함으로써 수용체 자극으로 유발된 세포내 $Ca^{2+}$ 농도 변화에 영향을 미친 결과인 것으로 추측된다.
Sprague-Dawley계 흰쥐의 악하선에서 선세포를 분리하여 fura-2/AM(fura-2)으로 염색한 후 spectrofluorometer로 세포내 $Ca^{2+}$농도를 측정하였다. 악하선 세포를 관류장치(perfusion chamber)에 넣고 표준용액을 관류시키면서 isoproterenol($1{\mu}M$)과 octanol(1mM)을 투여한 후 $Ca^{2+}$농도 변화를 측정하였는데 단독 투여시 $Ca^{2+}$농도는 거의 변화하지 않았으나 함께 투여한 경우 세포 내 $Ca^{2+}$농도가 증가함을 확인할 수 있었다. Adenylate-cyclase를 활성화 시키는 forskolin($10{\mu}M$)과 octanol을 함께 투여하였을 때도 isoproterenol의 경우와 유사한 증가 현상을 보이는 것으로 볼 때 octanol과 isoproterenol 또는 forskolin이 함께 작용할 때 세포 내 $Ca^{2+}$가 증가하는 것을 확인할 수 있었다. $Ca^{2+}$의 증가기전을 확인하고자 표준용액의 $Ca^{2+}$를 제거함은 물론 EGTA를 처리하여 세포외부의 $Ca^{2+}$를 제거한 후 상기한 바와 동일한 실험을 반복한 결과 $Ca^{2+}$농도의 증가를 보이지 않았다. 따라서 세포 내 $Ca^{2+}$의 증가는 세포 외부로부터의 $Ca^{2+}$유입 때문인 것으로 확인할 수 있었다. 이러한 $Ca^{2+}$의 유입 capacitative entry pathway를 이용하는지 확인코자 gadolinium($10{\mu}M$)을 처리하였을 때 $Ca^{2+}$농도의 증가가 완전히 억제되지는 않았지만 $Ca^{2+}$의 증가속도와 증가량이 감소되어 있음을 확인할 수 있었다. 이상의 실험결과들을 정리하면 세포 내 $Ca^{2+}$농도의 증가와 관련 ${\beta}$-아드레날린계 관련 약물과 옥탄올(octanol)을 함께 처리할 경우 세포 내 $Ca^{2+}$는 세포 외부에서 유입되어 증가되고 그 경로는 일부 capacitative entry pathway를 통함을 확인할 수 있었다.
