황해 갑각 중형동물플랑크톤의 형태 분석과 DNA 메타바코딩 비교 (Comparison of Morphological Analysis and DNA Metabarcoding of Crustacean Mesozooplankton in the Yellow Sea)

  • Studies on marine zooplankton diversity and ecology are important for understanding marine ecosystem, as well as environmental conservation and fisheries management. DNA metabarcoding is known as a useful tool to reveal and understand diversity among animals, but a comparative evaluation with classical microscopy is still required in order to properly use it for marine zooplankton research. This study compared crustacean mesozooplankton taxa revealed by morphological analysis and metabarcoding of the cytochrome oxidase I (COI). A total of 17 crustacean species were identified by morphological analysis, and 18 species by metabarcoding. Copepods made up the highest proportion of taxa, accounting for more than 50% of the total number of species delineated by both methods. Cladocerans were not found by morphological analysis, whereas amphipods and mysids were not detected by metabarcoding. Unlike morphological analysis, metabarcoding was able to identify decapods down to the species level. There were some discrepancies in copepod species, which could be due to a lack of genetic database, or biases during DNA extraction, amplification, pooling and bioinformatics. Morphological analysis will be useful for ecological studies as it can classify and quantify the life history stages of marine zooplankton that metabarcoding cannot detect. Metabarcoding can be a powerful tool for determining marine zooplankton diversity, if its methods or database are further supplemented.

동태평양 열대해역에서 2009-2010년 침강입자 플럭스의 수직 변화 (Vertical Variation of the Particle Flux in the Eastern Tropical Pacific from 2009 to 2010)

  • A sediment trap had been deployed at 1250 m depth in the Eastern Tropical Pacific (ETP) from September 2009 to July 2010, with the aim of understanding the temporal and vertical variability of particle flux. During the monitoring period, total particle flux varied from 12.4 to 101.0 mg m-2day-1, with the higher fluxes in January-March 2010. Biogenic particle flux varied in phase with the total particle flux. The increase in total particle flux during January-March 2010 was attributed to the enhanced biological production in the surface layer caused by wind-driven mixing in response to the seasonal shifts in the location of the Intertropical convergence zone. The export ratio (e-ratio) was estimated using the particulate organic carbon flux and satellite-derived net primary production data. The estimated e-ratios changed between 0.8% and 2.8% (1.4±0.6% on average). The ratio recorded in the negative phase of Pacific decadal oscillation (PDO) was similar to the previous results obtained from the ETP during the 1992/93 periods in the positive phase of PDO. This suggests that the regime shift of the PDO is not related to the carbon export ratio.

Effects of Recombinant Aquaporin 3 and Seawater Acclimation on the Expression of Aquaporin 3 and 8 mRNAs in the Parr and Smolt Stages of Rainbow Trout, Oncorhynchus mykiss

  • This study aimed to examine the role of two aquaporin isoforms (AQP3 and AQP8) in response to the hyperosmotic challenge of transitioning from freshwater (FW) to seawater (SW) during parr and smoltification (smolt) using the rainbow trout, Oncorhynchus mykiss. We examined the changes in the expression of AQPs mRNAs in the gills and intestine of the parr and smolt stages of rainbow trout transferred from FW to SW using quantitative real-time PCR in an osmotically changing environment [FW, SW, and recombinant AQP3 (rAQP3) injection at two dosage rates]. Correspondingly, AQPs were greater during smoltification than during parr stages in the rainbow trout. Plasma osmolality and gill $Na^+/K^+$-ATPase activity increased when the fish were exposed to SW, but these parameters decreased when the fish were exposed to SW following treatment with rAQP3 during the transition to seawater. Our results suggest that AQPs play an important role in water absorbing mechanisms associated with multiple AQP isoforms in a hyperosmotic environment.

Design of high-speed planing hulls for the improvement of resistance and seakeeping performance

  • High-speed vessels require good resistance and seakeeping performance for safe operations in rough seas. The resistance and seakeeping performance of high-speed vessels varies significantly depending on their hull forms. In this study, three planing hulls that have almost the same displacement and principal dimension are designed and the hydrodynamic characteristics of those hulls are estimated by high-speed model tests. All model ships are deep-V type planing hulls. The bows of no.2 and no.3 model ships are designed to be advantageous for wave-piercing in rough water. No.2 and no.3 model ships have concave and straight forebody cross-sections, respectively. And length-to-beam ratios of no.2 and no.3 models are larger than that of no.1 model. In calm water tests, running attitude and resistance of model ships are measured at various speeds. And motion tests in regular waves are performed to measure the heave and pitch motion responses of the model ships. The required power of no.1 (VPS) model is smallest, but its vertical motion amplitudes in waves are the largest. No.2 (VWC) model shows the smallest motion amplitudes in waves, but needs the greatest power at high speed. The resistance and seakeeping performance of no.3 (VWS) model ship are the middle of three model ships, respectively. And in regular waves, no.1 model ship experiences 'fly over' phenomena around its resonant frequency. Vertical accelerations at specific locations such as F.P., center of gravity of model ships are measured at their resonant frequency. It is necessary to measure accelerations by accelerometers or other devices in model tests for the accurate prediction of vertical accelerations in real ships.

광센서 배열을 이용한 무인잠수정의 종단유도장치 시스템 (Underwater Guidance System for AUV using Optical Sensor Array)

  • 본 논문에서는 수중에서 광과광센서를 이용하여 AUV의 도킹스테이션에의 도킹에 대한 새로운 연구를 하였다. 이를 위해 LED를 유도광원으로 사용하는 도킹스테이션과렌즈, 광 변위센서, 신호처리기, 연산처리기로 구성된 센서시스템 무인잠수정에 장착한 무인잠수정의 종단유도장치 시스템을 제안하였다. LED 복사광을 이용해 정밀한 상대각도 측정 정밀도를 얻을 수 있도록 집광렌즈 및 광센서 시스템의 원리해석과 이를 검증하기 위해 집광렌즈와 광센서 시스템을 직접 제작하고 기초실험을 수행하였으며 제작한 광학센서를 이용한 AUV와 도킹시스템을 제작하고 수조에서 기본적인 도킹시험을 수행하여 새로운 도킹 방법으로의 가능성을 검증하였다.

Statics variation analysis due to spatially moving of a full ocean depth autonomous underwater vehicle

  • Changes in gravity and buoyancy of a Full Ocean Depth Autonomous Underwater Vehicle (FOD-AUV) during its descending and ascending process must be considered very carefully compared with a Human Occupied Vehicle (HOV) or a Remotely Pperated Vehicle (ROV) whose activities rely on human decision. We firstly designed a two-step weight dropping pattern to achieve a high descending and ascending efficiency and a gravity-buoyancy balance at designed depth. The static equations showed that gravity acceleration, seawater density and displacement are three key aspects affecting the balance. Secondly, we try our best to analysis the gravity and buoyancy changing according to the previous known scientific information, such as anomaly of gravity acceleration, changing of seawater states. Finally, we drew conclusion that gravity changes little (no more than 0.1kgf, it is impossible to give a accurate value). A density-depth relationship at the Challenger Deep was acquired and the displacement changing of the FOD-AUV was calculated preciously.

Fundamental Research on Spring Season Daytime Sea Fog Detection Using MODIS in the Yellow Sea

  • For the safety of sea, it is important to monitor sea fog, one of the dangerous meteorological phenomena which cause marine accidents. To detect and monitor sea fog, Moderate Resolution Imaging Spectroradiometer (MODIS) data which is capable to provide spatial distribution of sea fog has been used. The previous automatic sea fog detection algorithms were focused on detecting sea fog using Terra/MODIS only. The improved algorithm is based on the sea fog detection algorithm by Wu and Li (2014) and it is applicable to both Terra and Aqua MODIS data. We have focused on detecting spring season sea fog events in the Yellow Sea. The algorithm includes application of cloud mask product, the Normalized Difference Snow Index (NDSI), the STandard Deviation test using infrared channel ($STD_{IR}$) with various window size, Temperature Difference Index(TDI) in the algorithm (BTCT - SST) and Normalized Water Vapor Index (NWVI). Through the calculation of the Hanssen-Kuiper Skill Score (KSS) using sea fog manual detection result, we derived more suitable threshold for each index. The adjusted threshold is expected to bring higher accuracy of sea fog detection for spring season daytime sea fog detection using MODIS in the Yellow Sea.

Morphological Features of Marine Dinoflagellates from Jangmok Harbour in Jinhae Bay, Korea: A Case of 30 Species in the Orders Prorocentrales, Dinophysiales, Gonyaulacales and Gymnodiniales

  • Most previous studies on dinoflagellates in Korean coastal areas were conducted without morphological descriptions and illustrations of the observed dinoflagellates. This indicates that the species and diversity of dinoflagellates may have been respectively misidentified and underestimated in the past, probably due to cell shrinkage, distortion and loss caused by sample fixation. This study provides information on the morphological observations of four dinoflagellate orders (Prorocentrales, Dinophysiales, Gonyaulacales and Gymnodiniales) from Jangmok Harbour in Jinhae Bay, Korea. The unfixed samples were collected weekly from December 2013 to February 2015. A total of 13 genera and 30 species were identified using light and scanning electron microscopy, although some samples were not clarified at the species level. Harmful dinoflagellates, Prorocentrum donghaiense, Tripos furca, Alexandrium affine, A. fundyense, Akashiwo sanguinea and Cochlodinium polykrikoides, were identified based on the morphological observations. The results also reflect the occurrence and identification of dinoflagellates that had not been previously recorded in Jangmok Harbour.

5-bromoprotocatechualdehyde suppresses growth of human lung cancer cells through modulation of ROS and the AKT/MAPK signaling pathway

  • Early-stage lung cancer is the deadliest form of the disease. In this study, we investigated the anticancer activity of 5-bromoprotocatechualdehyde (BPCA) extracted from the seaweed Polysiphonia morrowii Harvey (P. morrowii) in lung cancer H460 cells. We extracted P. morrowii powder thrice with 80% aqueous methanol and separated the extract using high-performance liquid chromatography. We then tested BPCA's effects on cell viability, apoptosis, reactive oxygen species (ROS) generation, and protein expression Our results showed that BPCA inhibited tumor cell growth and ROS production and induced apoptosis through mitogen-activated protein kinase (MAPK) and AKT signaling pathways in lung cancer cells. When BPCA was combined with hydrogen peroxide, ROS production and apoptosis increased even further due to the regulation of AKT signaling and JNK-MAPKs pathways. These findings suggest that BPCA induces lung-cancer-cell death through ROS-mediated phosphorylation in AKT/MAPK signaling. This could lead to the development of new and effective treatments for early-stage lung cancer.
