Development of a Practical and Cost-Effective Medium for Bioethanol Production from the Seaweed Hydrolysate in Surface-Aerated Fermentor by Repeated-Batch Operation

  • To develop a practical and cost-effective medium for bioethanol production from the hydrolysate of seaweed Sargassum sagamianum, we investigated the feasibility and performance of bioethanol production in CSL (corn-steep liquor)-containing medium, where yeast Pichia stipitis was used and the repeated batch was carried out in a surface-aerated fermentor. The optimal medium replacement time during the repeated operation was determined to be 36 h, and the surface aeration rates were 30 and 100 ml/min. Under these conditions, the repeated-batch operation was successfully carried out for 6 runs (216 h), in which the maximum bioethanol concentrations reached about 11-12 g/l at each batch operation. These results demonstrated that bioethanol production could be carried out repeatedly and steadily for 216 h. In these experiments, the total cumulative bioethanol production was 57.9 g and 58.0 g when the surface aeration rates were 30 ml/min and 100 ml/min, respectively. In addition, the bioethanol yields were 0.43 (about 84% of theoretical value) and 0.44 (about 86% of theoretical value) when the surface aeration rates were 30 ml/min and 100 ml/min, respectively. CSL was successfully used as a medium ingredient for the bioethanol production from the hydrolysate of seaweed Sargassum sagamianum, indicating that this medium may be practical and cost-effective for bioethanol production.

Temperature-Dependency Urease Activity in Vibrio parahaemolyticus is Related to Transcriptional Activator UreR

  • Vibrio parahaemolyticus possessing urease-positive property is relatively rare, but such strains consistently exhibit the TDH-related hemolysin (TRH) gene. In this study, we examined the effects of incubation temperature on urease activity expression, using the TH3996 and AQ4673 strains where the enzyme activity is known to be temperature-dependent and -independent, respectively. In the TH3996 strain, $\beta$-galactosidase activity was 4.4-fold lower after $30^{\circ}C$ cultivation than after $37^{\circ}C$ in a ureR-lacZ fusion strain, but temperature dependency was not found in ureD- or nikA-lacZ fusion strains. However, ureR-, ureD-, and nikA-lacZ fusions of the AQ4673 strain was not influenced by incubation temperature. We compared the promoter sequences of ureR between the above two strains. Intriguingly, we detected mismatches of two nucleotides between the two strains located at positions -66 and -108 upstream of the methionine initiation codon for UreR. Additionally, urease activity was not affected by culture temperature at either $30^{\circ}C$ or $37^{\circ}C$ by allelic introduction of the AQ4673 ureR gene into the TH3996 ureR deletion mutant. Taken together, our study demonstrates that the transcriptional factor UreR is involved in the temperature dependency of urease activity, and two nucleotides within the ureR promoter region are of particular importance for the urease activity dependency of V. parahaemolyticus.

Deep Seawater Increases Dendritic Branches of Cultured Rat Hippocampal Neurons (해양심층수에 의한 해마신경세포 가지돌기 수의 증가)

  • Deep seawater (DSW; deep ocean water) is pure, rich in inorganic materials which have attracted attention for various applications. In this study we investigated the effects of the DSW upwelled from the East Sea, offshore Yang Yang (Korea) on the morphological differentiation of cultured rat hippocampal neurons, which were grown in the minimal essential medium containing 10% (v/v) fetal bovine serum and 25% (v/v) DSW with various hardness. DSW had no effect on initial morphological differentiation (17 hr post-plating). When observed on DIV3, 7, 14, and 17, low hardness (0 and 200) DSW reduced dendritic branching. However, dendritic branches within $80\;{\mu}m$ diameter from the center of soma nearly doubled in neurons grown in hardness 1,000 DSW-containing media. DSW with hardness 600 was more or less same as control groups. These results indicate that DSW with appropriate hardness ameliorates neuronal health.

The venom of jellyfish, Chrysaora pacifica, induces neurotoxicity via activating Ca2+-mediated ROS signaling in HT-22 cells

  • Stings of jellyfish, which frequently occur in a warm season, cause severe pain, inflammation and sometimes irreversible results such as the death. Harmful venoms from jellyfish, therefore, have been studied for finding the therapeutic agents to relieve pain or to neutralize toxic components. However, it is still unclear if and how jellyfish venom reveal neuronal toxicity even though pain induction seems to result from the activation of nociceptors such as nerve endings. In this study, using HT-22 cell line, we investigated neurotoxic effects of the venom of Chrysaora pacifica (CpV) which appears in South-East ocean of Korea. In 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay, CpV significantly reduced the viability of HT-22 cells in a dose-dependent manner. Additionally, in 2',7'-Dichlorofluorescin diacetate fluorescence test under the culture condition lacking dominant inflammatory factors, CpV remarkably increased the production of intracellular reactive oxygen species (ROS). Reduced responsive fluorescence to Rhodamine123 and increased expression of intracellular cytochrome c were also observed in HT-22 cells treated with CpV. These indicate that CpV-reduced viability of HT-22 cells may be due to the activation of apoptotic signalings mediated with oxidative stress and mitochondrial dysfunction. Furthermore, removing Ca2+ ion or adding N-acetyl-Lcystein remarkably blocked the CpV effect to reduce the viability of HT-22 cells. The findings in this study clearly demonstrate that CpV may activate Ca2+-mediated ROS signalings and mitochondrial dysfunction resulting in neuronal damage or death, and suggest that blocking Ca2+ pathway is a therapeutic approach to possibly block toxic effects of jellyfish venoms.

Establishment and Characterization of Clonal Cell Lines from Zebrafish, Danio rerio (제브라피쉬(Danio rerio) 배아로부터 동형세포주 확립)

  • Three types of clonal cell lines were isolated according to their size and phenotype from the adherent cell populations in long-term liquid cultures from the embryonic fibroblast cells of Zebrafish, Danio rerio. All kind of cell lines were well proliferated. The size and number of clonal cell lines derived colonies from stable embryonic cells were significantly increased in the presence of NAC and A2P conditioned medium from the cell lines. The stable cell lines and clonal cell lines were cap-able of well proliferation in vitro. These cell lines have been maintained in continuous culture without change in characteristics. A majority of the clonal cells (80%) was shown a normal chromosomal complement (50 chromosomes, 2N) in according with FACs analysis. Majority of cells were positive to vimentin staining and none of them were positive for nestin and Oct -4 by immunocytochemistry. These results indicate that the clonal cell lines obtained from cultured cells are fibroblasts and may be extremely useful in genetic manipulation for further nuclear transfer and fish cloning.

Enhancement of Saccharification Yield of Ulva pertusa Kjellman by High Pressure Homogenization Process for Bioethanol Production (구멍갈파래의 고압 균질 전처리 공정을 통한 바이오에탄올 생산용 당화수율 증진)

  • This study was investigated to improve the saccharification yield of Ulva pertusa Kjellman by the high pressure homogenization process. It was found that the high pressure homogenization pretreatment effectively destructed the cell wall structures only by using water. The high pressure homogenization process was operated under various conditions such as 10000, 20000 or 30000 psi with different recycling numbers. The optimal condition was determined as 30000 psi and 2 pass of recycling numbers and the sugar conversion yields were 16.02 (%, w/w) of glucose and 14.70 (%,w/w) of xylose, respectively. In the case of enzymatic treating the hydrolyzates with 5 FPU/glucan of celullase and 100 units/mL of amyloglucosidase, 65.8% of carbohydrates was converted into glucose. Using the hydrolysates of Ulva pertusa Kjellman, 48.7% of ethanol was obtained in the culture S.cerevisiae. These results showed that the high pressure homogenization process could efficiently hydrolyze the marine resource by using only water for bioethanol production.

Preparation of Corncob Grits as a Carrier for Immobilizing Yeast Cells for Ethanol Production

  • In this study, DEAE-corncobs [delignified corncob grits derivatized with 2-(diethylamino)ethyl chloride hydrochloride ($DEAE{\cdot}HCl$)] were prepared as a carrier to immobilize yeast (Saccharomyces cerevisiae) for ethanol production. The immobilized yeast cell reactor produced ethanol under optimized $DEAE{\cdot}HCl$ derivatization and adsorption conditions between yeast cells and the DEAE-corncobs. When delignified corncob grit (3.0 g) was derivatized with 0.5M $DEAE{\cdot}HCl$, the yeast cell suspension ($OD_{600}$ = 3.0) was adsorbed at >90% of the initial cell $OD_{600}$. This amount of adsorbed yeast cells was estimated to be 5.36 mg-dry cells/g-DEAE corncobs. The $Q_{max}$ (the maximum cell adsorption by the carrier) of the DEAE-corncobs was estimated to be 25.1 (mg/g), based on a Languir model biosorption isotherm experiment. When we conducted a batch culture with medium recycling using the immobilized yeast cells, the yeast cells on DEAE-corncobs produced ethanol gradually, according to glucose consumption, without cells detaching from the DEAE-corncobs. We observed under electron microscopy that the yeast cells grew on the surface and in the holes of the DEAE-corncobs. In a future study, DEAE-corncobs and the immobilized yeast cell reactor system will contribute to bioethanol production from biomass hydrolysates.

Effect of Gamma Ray on Growth of Juvenile Abalone, Haliotis discus hannai (참전복(Haliotis discus hannai)의 성장에 대한 감마선 영향)

  • Radiation at very low doses frequently has a stimulating or hermetic effect on growth of organism. Effects of growth and survival rate on various dose of gamma irradiation in the farm culture of juvenile abalone (Haliotis discus hannai) were determined in Hallim (Jeju) from February 2004 to January 2005. The initial shell length of abalone juveniles in this study was average $3.45{\pm}0.4cm$. A change of growth after irradiation $(0{\sim}20Gy)$ was observed for 48 weeks. The highest growth rate was observed in 4 Gy-irradiated group and the lowest growth rate was observed in 20 Gy-irradiated group. The additional research about biochemical changes on juvenile abalone after irradiation should be accomplished. Continuous study for gamma radiation-induced hormesis on growth and metabolism of juvenile abalone will further induce the creation of value in ocean industry.

Variation Landscape Enhancement Strategy through Coloring Plan in the Busan North Port Redevelopment (부산북항재개발사업에서 색채계획을 통한 경관조성방안)

  • In the Busan North port redevelopment project, which is the first large scale port redevelopment project in this country, it adopted the national landscape plan for competitiveness of port city. Furthermore, it includes many efforts on landscape formation in the process of improvement of ocean city image and enhancement of its value, which fits for the situation and characteristics of the original nature, history, and culture resources, together with all facilities, structures, symbols, and eco green belt. Firstly this study attempted to analyze the characteristics of Busan landscape, Busan city design plan(2010), especially coloring plan, together with the case study of landscape color plan for foreign countries. From the basis of this analysis, it was proposed a landscape enhancement strategy in the Busan North port redevelopment project through the coloring plan and it is expected that the result would be used for the basic data for other redevelopment project.

An Economic Analysis of the Extruded Pellets on the Olive Flounder Paralichthys olivaceus Farms in the Jeju Region (제주지역 넙치(Paralichthys olivaceus) 양식업의 경제성 분석: 배합사료 공급 양식어가를 중심으로)

  • The government has implemented a policy to promote the use of extruded pellets in sustainable aquaculture by protecting fishery resources and managing the ocean environment. A survey on the production status and the cost of targeting olive flounder Paralichthys olivaceus culture farms in Jeju Island using extruded pellets was conducted. The survey results were used to examine the profitability and economic feasibility of the test farms, as well as the degree of increase in profitability and economic feasibility of the fish farms receiving government subsidies for employing extruded pellets. The economic feasibility was predicted through a sensitivity analysis of prices and production, which are the variable factors when of using the extruded pellets. Using the economic feasibility analysis, the average NPV (Net Present Value) and IRR (Internal Rate of Return) of sample farms were found to be KRW 5.8 billion and 8.9%, respectively. The result of the economic feasibility analysis of the government subsidy showed a maximum average of about 2.3 times higher NPV and a 3.8% increase in IRR in cases where government subsidies were received.
