Recently, Electronic Distance Meter(EDM) is widely used for the traverse surveying and trilateration net. In this study, the closed traverse network is precisely adjusted by simultaneous adjustment of the observed angle and distance, using the Electronic Total Stations (Set 3). And also, it is examined that each kind accidental errors appear observed angles according to the observed distance and the number of repeating angle. In addition to the result, of checking between the simultaneous adjustment, the compass rule and transit rule, simultaneous adjustment in combination of the observed angle and distance is more accurate and effective than other adjustment.
Dermal wound healing responses in the skin of the toad, Bombina orientalis, were examined using transmission electron microscopy. At 12 hours after wounding, debridement and collagenolysis occurred in damaged dermis. Histocyte has a large nucleus and long cytoplasm process. Phagocytic vesicle and lysosome were observed in the cytoplasm. Damaged blood cells were transformed spindle to irregular shape. Autolysis was observed in their cytoplasm. Histocytes are found in poly-band. The irregularly shaped nucleus is located peripheral region in cytoplasm. At 2 days after wounding, partial aggregation of blood cells is observed. Phagocytic, activity is observed in histocyte and collagenolytic collagen fibers are scattered. Fibroblast is observed in the dermis at 3 days after wounding. Clusters of ribosomes and some short cisternae of rough-surfaced endoplasmic reticulum are found in the cytoplasm. In histocyte at 7 days post wounding, various size granules composed of moderately dense material are found the cytoplasm. In this period histocyte is round to rod in profile, with slender processes projecting from the surface. At 7 day after injury, it was observed that formation of connective tissue fibers and amorphous ground substance in regenerating skin.
The author examined the clinical and histological changes on the dorsal tongue mucosa of adult Sprague-Dawley rats after lasing by pulsed Nd:YAG laser. The dorsal tongue was lased for 3 seconds by 1.5, 2.0, 2.5, 3.0, 3.5, 4.0W and 10, 15, 20, 20pps. After tissue changes were observed clinically, the excised samples were observed histologically and the width of tissue destruction was analyzed quantitatively under the Confocal laser microscope respectively. The following results were obtained : 1. Whitening of peripheral tissue was observed more as increasing pulsed per second (Hz) below power 2.5W and observed at all parameters above power 3.0W. 2. Charring of mucosal surface was observed at all parameters but mild at parameters below power 2.0W. 3. The destruction of epithelium was observed at all parameters and tissue destruction was extended to lamina propria at higher pulses per second. 4. The width of tissue destruction was more widening as increasing energies per pulse (p<0.001) and the narrowest at 20Hz than 10Hz, 15Hz and 30Hz(p<0.05). As the above results, author suggests power below-3.5W and 20Hz as the lasing parameters for oral soft tissue therapy.
Marine surface winds observed by two microwave sensors, SeaWinds and Advanced Microwave Scanning Radiometer (AMSR), on the Advanced Earth Observing Satellite-II (ADEOS-II) are evaluated by comparison with off-shore moored buoy observations. The wind speed and direction observed by SeaWinds are in good agreement with buoy data with root-mean-squared (rms) differences of approximately 1 m $s^{-1}$ and $20^{\circ}$, respectively. No systematic biases depending on wind speed or cross-track wind vector cell location are discernible. The effects of oceanographic and atmospheric environments on the scatterometry are negligible. The wind speed observed by AMSR also exhibited reasonable agreement with the buoy data in general with rms difference of 1.2 m $s^{-1}$. Systematic bias which was observed in earlier versions of the AMSR winds has been removed by algorithm refinements. Intercomparison of wind speeds globally observed by SeaWinds and AMSR on the same orbits also shows good agreements. Global wind speed histograms of the SeaWinds data and European Centre for Medium-range Weather Forecasts (ECMWF) analyses agree precisely with each other, while that of the AMSR wind shows slight deviation from them.
The acute and subacute toxicity of water soluble dimethyl dimethoxy biphenylate derivative (new DDB), hepatitis therapeutics, were investigated in SD rats. In the acute toxicity study, body weights and clinical signs were observed for 7 days after the intravenous injection of new DDB at doses of 140, 182, 236, 307 and 400 mg/kg(r=1.3). Death. Severe convulsion, tremor and decrease motor activity were observed in almost treated groups (except the 140 mg/kg treated group). Changes of body weight in treated groups were not significantly different from control group. Autopsy of survived animals revealed no abnormal gross findings related to new DDB. As a results, the $LD_{50}$ values of new DDB were 244.1 mg/kg for male and 232.5 mg/kg for female. In subacute toxicity study, body weights and clinical signs were observed after intravenous injection of new DDB at doses of 57, 75 and 100 mg/kg/day for 28 days. Death, decrease motor activity and tremor were observed above 75 mg/kg treated groups. Statistically significant changes were observed in hematological and biochemical parameters of new DDB-treated groups; however, these changes were within normal range and had no relationship with dosage. Several abnormal findings were observed in microscopic examination of tissue; however, these findings were not caused by new DDB but environmental factor. The no toxic dose level of new DDB were estimated to be 57 mg/kg/day in this study.
This study was performed to evaluate the acute oral toxicity of an oriental medical prescription for obesity treatment, SH21-B, in Sprague-Dawley rats and Beagle dogs. SH21-B was administered in rats at does of 0mg/kg, 2,000mg/kg, and 5,000mg/kg. And also SH21-B was administered in Beagle dogs at does of 150mg/kg, 300mg/kg, and 600mg/kg. The rats and dogs of both sexes were observed daily for 14 days after single oral administration. Two female rats, one administered at 2,000mg/kg and the other administered at 5,000mg/kg, died, but no dead animal was observed among male rats. Therefore LD50 in the female rat is observed to be 8,710mg/kg, and MLD(Minimum Lethal Dose) of the male rat is observed to be more than 5,000mg/kg. Among dogs, no dead animal was observed up to 600mg/kg and MLD is observed to be more than 600mg/kg.
To obtain a basic information on the development of Genus Viola, ultrastructure and electrofusion process between the two protoplasts from wild Viola callus cells and pansy mesophyll cells were observed with a scanning electron microscopy(SEM) and transmission electron microscopy(TEM). In the ultrastructural observation of wild viola callus protoplasts and pansy mesophyll protoplasts using SEM, their cell walls were removed completely. A knob-like formation was observed on the enlarge surface of viola callus protoplasts. On the surface of pansy mesophyll protoplasts net-like chloroplasts were observed. In SEM observation of pansy mesophyll protoplasts, chloroplasts devoid of membrane were observed on the surface the protoplasts. Pearl chain was formed by applying AC field of 200 V/cm at 1.0 MHz for 43 sec. The lysis of plasma membranes and fusion process occurred by applying a 1,600 V/cm DC pulse twice for 1 sec. After 1-2 hours of a DC pulse application, it was observed that the two protoplasts were fused completely into one cell. In TEM observation of the fused cell, many small vacuoles were located in the fusion area of the two protoplasts. Indeed, two distinct regions were observed during fusing process; in one region, a nucleus was found, while in the other region, both nucleus and nucleous were found.
The present study was designed to help elucidate the effect of glass ionomer cements on the exposed dental pulp by means of histologic examination. A total of 40 cavities of class V were prepared on the teeth of 4 dogs with exposure of 1mm in diameter on the bases of them. 20 cavities were filled with glass ionomer cement as the experimental group and the other 20 cavities were filled with zinc oxide eugenol cement as the control group. The dogs were sacrificed at one, two, three, and four weeks after filling, and the specimens were routinely prepared and stained with Hematoxylin-Eosin. The obtained microscopic findings were as follows: Inflammatory cell infiltrations were observed in control in 1 week, which decreased markedly with time. In all control groups, hemorrhage around exposed pulp tissue and coagulation change of pulp were observed. Secondary dentin formation and thickened predentin were observed in 4 week cases, and the recovery of pulp tissue was favorable on the whole. Inflammatory cell infiltration was observed in all GIC groups. Proliferation of blood vessel and congestion were observed with coagulation changes around the exposed pulp tissue. Secondary dentin formation and thickened predentin were observed in 3 weeks. In the experimental 4 week case, secondary dentin formation was evident. On the whole, pulpal irritation of glass ionomer cement was relatively severe. Recovery of pulp tissue in GIC groups was less favorable compared with that of ZOE groups.
Attempts of these studies were made to investigate the nonspecific congenital focal accumulation of ectodermal glial cells in the brain of normal piglets. The brain samples were taken from 1-,10-,20-,35-,45- and 70-day-old piglets from a SPF-pig farm and three model pig farms. Occurrences of neuroglial cell foci (NCF) on the brain were observed with light microscope. Appearance degrees of the congenital NCF on 10 to 16 cross section slides per a piglets brain were tentatively designed on a scale from degree+ to ⧻by NCF number: +, less than 20 of NCF number; ⧺, 21-40 of NCF number: ⧻, more than 41 of NCF number. The results obtained were as follows: 1. NCF in the brain were observed mainly on the cerebrum. Regions of higher frequencies on the cerebrum were ordered as subependymal layers of the lateral ventricles, peripheral regions of lateral ventricles in the white matter and some neuron layers under the molecular layer of the gray matter. But NCF were not observed in the cerebellum, pons, medulla oblongata and spinal cords. 2. On the subependymal layers of the lateral ventricles, NCF were observed in 100% of 27 piglets, and appearance degree of ⧻ was observed in 10 piglets(37.0%), ⧺ in 10 piglets(37.0%) and + in 7 Piglets(26.0%) of 27 piglets, respectively. 3. On the white matter of the cerebrum, NCF were observed in 25 piglets(92.6%) of 27 piglets, and appearance degree of ⧻ was observed in 3 piglets(11.1%), ⧺ in 13 piglets(48.2%), + in 9 piglets(33.3%) and - in 2 piglets(7.4%) of 27 piglets, respectively. 4. On the gray matter of the cerebrum, NCF were observed in 21 piglets(77.8%) of 27 piglets, and appearance degree of ⧻ was not observed, appearance degree of ⧺ was observed in 6 piglets(22.2%), + in 15 Piglets(55.6%) and - in 6 piglets(22.2%) of 27 Piglets, respectively. 5. NCF tended to be converged appearance on some regions and tended to be decreased markedly from 35th day after birth, and the shapes of NCF were: global or oval forms crowded by analogous shaped and stained cells in the empty spaces of the brain substrate or on one side of the blood vessels.
The experiment was performed to study the morphological responses of the thymus of the mice, to antitumour agents (5-Fluorouracil or mitomycin C). Healthy adult mice weighing 25 gm each were divided into normal and experimental groups. 5-Fluorouracil (60 mg/kg) or Mitomycin-C $(400{\mu}g/kg)$ were injected subcutaneously to the animals every other day. Animals were sacrificed at 4 days and 7 days following the first injection. Pieces of the tissue taken from the thymus were prefixed with 2.5% paraformaldehyde-l.5% glutaraldehyde, followed by post-fixation with 1% osmium tetroxide. Ultrathin sections stained with uranyl acetate and lead citrate were observed with a JEM 100 CX-II electron microscope. The observed results were as follow: 1. Apoptoses of T-lymphocytes were observed more frequently in the thymus of the experimental groups than in those of a normal group. 2. In the experimental group, the plasma cells with distended cisternae of the granular endoplasmic reticulum and the eosinophile leukocytes were observed frequently. 3. In the experimental group, newly forming Hassall's corpurscles were observed frequently. 4. In the mitomycin-treated group, the epithelial reticular cells containing distended perinuclear cisternae, distended the granular endoplasmic reticula and pyknotic nuclei were observed in the cortico-medullary junctional area. 5. In the mitomycin-treated group, nuclear bodies with medium electron dense materials were often observed in the T lymphocyte. 6. In the 5-fluorouracil-treated groups, fused and dissolved tonofilament bundles and apoptotic bodies were observed in the some epithelial reticular cells in the medullary area. 7. In the 5-fluorouracil-treated groups, some elongated and bar-shaped lysosomes with electron lucent gap were often observed in the macrophages. 8. In the 5-fluorouracil-treated group, membrane complex of the smooth endoplasmic reticulum were ofen observed in the macrophage. From the above results, it was suggested that 5-fluorouracil or mitomycin could induce rapid involution of the thymus, and disturb maturation and differentiation of T lymphocytes, and, in turn, supress immunity.
