db/db 마우스에서 상백피탕의 혈당강하 활성 및 기전연구 (Blood Glucose Lowering Activity and Mechanism of Sangbackpitang (SBPT) in db/db Mouse)

  • Antidiabetic activity and mechanism of Sangbackpitang (SBPT) was examined in db/db mice, which is a spontaneously hyperglycemic, hyperinsulinemic and obese animal model. SBPT and acarbose were administered orally for 4 weeks. Fasting and non-fasting serum glucose, glycated hemoglobin and triglyceride were all reduced when compared between db/db control group and SBPT treated group. At 12th week after birth, SBPT increased an insulin secretion although statistic significance was not seen. Total activities of sucrase, maltase and lactase in SBPT treated group were all decreased when compared to db/db control. On the other hand, sucrase and maltase activities in acarbose treated groups were increased. Effect of SBPT on mRNA expression of glucose transporter(GLUT-4) was also examined. Quantitation of glucose transporter was performed by RT-PCR and in vitro transcription with co-amplification of rat-action gene as an internal standard. Muscular GLUT-4 mRNA expression in SBPT treated group was increased significantly. These results may suggest that SBPT lowered blood glucose ascribing to inhibition of glycosidase-catalyzed reaction and upregulation of muscular GLUT-4 mRNA expression.

고지방식이 비만 마우스에 대한 야채 조성물의 항비만 효과 (Anti-obesity Effect of Vegetable Formula on Obese Mice in High Fat Diet)

  • This study was conducted to investigate the cholesterol and Triglyceride (TG) lowering and anti-obesity effects of water extract of vegetable formula (onion 25%, carrot 20%, Cabbage 20%, Sweet Pumpkins 20%, Chinese plum 5%, turmeric 5%, morus leaves 5%, SLS) in mice fed high fat-diet. ICR mice were divided into 3 groups; a normal diet group (ND), a high-fat diet group (HFD), a high-fat diet and SLS with 300 mg/kg treated group (HFD+SLS). Body fat gain was increased by high-fat diet and HFD+SLS group showed a definite weight loss. The total cholesterol level in the HFD+SLS group was 328.4 mg/dl which was 27.4% lower than that in the HFD group and the cholesterol-lowering effect of SLS was confirmed. The HFD + SLS group showed 118.1 mg/dl and the triglyceride level in the serum was decreased by 88.7% compared to the HFD group and SLS significantly decreased blood triglyceride levels. RT-PCR showed that the expression of PPAR-${\gamma}$ and the target gene SCD-1 was inhibited in a dose-dependent manner in the SLS-treated group. These results suggest that the SLS water extract may have a cholesterol and triglyceride-lowering effect and inhibit the expression of PPAR-${\gamma}$ and SCD-1 to have an anti-obesity effect.

오미자(五味子) 헥산 분획 추출물이 고지방 식이에 의한 고지혈증 생쥐의 지질대사 및 간 조직 유전자 발현에 미치는 영향 (Effects of Schisandra Fructus hexane fraction on high fat diet induced hyperlipidemic mice)

  • ■ Objectives The berries of Schisandra chinensis (Schisandra Fructus) are given the name Omiza in Koreane(五味子), and have been used asremedies for many ailments: to resist infections, increase skin health, and combat insomnia, coughing, and thirst. This study was designed to investigate the effects of Schisandra Fructus hexane fraction (SFH) on serum lipid levels in Hyperlipidemic mice. ■ Methods In this experiment, effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured. And in addition, histopathological and gene expression changes in liver tissue was also observed. ■ Results SFH did not affects weight gain, serum AST and ALT in hyperlipidemic mice. Oral administration of SFH lowered levels of total cholesterol and triglyceride, which were elevated by induction of hyperlipidemia. Finally, administration of SFH lowered fasting blood glucose significantly. And SFH also ameliorates anti-oxidative stress systems in internal organs which play key role in disease prevention. ■ Conclusion Results in our study suggest that SFH can prevent obese through regulation of dyslipidemia and hyperglycaemia.

Mentha canadensis attenuates adiposity and hepatic steatosis in high-fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: Obesity is a major risk factor for metabolic syndrome, a global public health problem. Mentha canadensis (MA), a traditional phytomedicine and dietary herb used for centuries, was the focus of this study to investigate its effects on obesity. MATERIALS/METHODS: Thirty-five male C57BL/6J mice were randomly divided into 2 groups and fed either a normal diet (ND, n = 10) or a high-fat diet (HFD, n = 25) for 4 weeks to induce obesity. After the obesity induction period, the HFD-fed mice were randomly separated into 2 groups: one group continued to be fed HFD (n = 15, HFD group), while the other group was fed HFD with 1.5% (w/w) MA ethanol extract (n = 10, MA group) for 13 weeks. RESULTS: The results showed that body and white adipose tissue (WAT) weights were significantly decreased in the MA-supplemented group compared to the HFD group. Additionally, MA supplementation enhanced energy expenditure, leading to improvements in plasma lipids, cytokines, hepatic steatosis, and fecal lipids. Furthermore, MA supplementation regulated lipid-metabolism-related enzyme activity and gene expression, thereby suppressing lipid accumulation in the WAT and liver. CONCLUSIONS: These findings indicate that MA has the potential to improve diet-induced obesity and its associated complications, including adiposity, dyslipidemia, hepatic steatosis, and inflammation.

Effects of quercetin derivatives from mulberry leaves: Improved gene expression related hepatic lipid and glucose metabolism in short-term high-fat fed mice

  • BACKGROUND/OBJECTIVES: Mulberry leaves contain quercetin derivatives, which have the effects of reducing obesity and improving lipid and glucose metabolism in mice with obesity. It is not clear whether or not mulberry leaves can directly affect metabolic disorders, in the presence of obesity, because of the interaction between obesity and metabolic disorders. The aim of the current study was to assess the direct action of quercetin derivatives on metabolic disorders in non-obese conditions in short-term high-fat diet fed mice. MATERIALS/METHODS: C57BL/6N mice were fed a high-fat diet, supplemented with either 0% (control), 1%, or 3% mulberry leaf powder (Mul) or 1% catechin powder for five days. Anthropometric parameters and blood biochemistry were determined, and hepatic gene expression associated with lipid and glucose metabolism was analyzed. RESULTS: Body and white fat weights did not differ among the four groups. Plasma triglycerides, total cholesterol, and free fatty acids in the 1%, 3% Mul and catechin groups did not differ significantly from those of the controls, however, plasma glucose and 8-isoprostane levels were significantly reduced. Liver gene expression of gp91phox, a main component of NADPH oxidase, was significantly down-regulated, and PPAR-${\alpha}$, related to ${\beta}$-oxidation, was significantly up-regulated. FAS and GPAT, involved in lipid metabolism, were significantly down-regulated, and Ehhadh was significantly up-regulated. Glucose-metabolism related genes, L-PK and G6Pase, were significantly down-regulated, while GK was significantly up-regulated in the two Mul groups compared to the control group. CONCLUSIONS: Our results suggest that the Mul quercetin derivatives can directly improve lipid and glucose metabolism by reducing oxidative stress and enhancing ${\beta}$-oxidation. The 1% Mul and 1% catechin groups had similar levels of polyphenol compound intake ($0.4{\times}10^{-5}$ vs $0.4{\times}10^{-5}$ mole/5 days) and exhibited similar effects, but neither showed dose-dependent effects on lipid and glucose metabolism or oxidative stress.

Hog millet (Panicum miliaceum L.)-supplemented diet ameliorates hyperlipidemia and hepatic lipid accumulation in C57BL/6J-ob/ob mice

  • Dietary intake of whole grains reduces the incidence of chronic diseases such as obesity, diabetes, cardiovascular disease, and cancer. In an earlier study, we showed that Panicum miliaceum L. extract (PME) exhibited the highest anti-lipogenic activity in 3T3-L1 cells among extracts of nine different cereal grains tested. In this study, we hypothesized that PME in the diet would lead to weight loss and augmentation of hyperlipidemia by regulating fatty acid metabolism. PME was fed to ob/ob mice at 0%, 0.5%, or 1% (w/w) for 4 weeks. After the experimental period, body weight changes, blood serum and lipid profiles, hepatic fatty acid metabolism-related gene expression, and white adipose tissue (WAT) fatty acid composition were determined. We found that the 1% PME diet, but not the 0.5%, effectively decreased body weight, liver weight, and blood triglyceride and total cholesterol levels (P < 0.05) compared to obese ob/ob mice on a normal diet. Hepatic lipogenic-related gene ($PPAR{\alpha}$, L-FABP, FAS, and SCD1) expression decreased, whereas lipolysis-related gene (CPT1) expression increased in animals fed the 1% PME diet (P < 0.05). Long chain fatty acid content and the ratio of C18:1/C18:0 fatty acids decreased significantly in adipose tissue of animals fed the 1% PME diet (P < 0.05). Serum inflammatory mediators also decreased significantly in animals fed the 1% PME diet compared to those of the ob/ob control group (P < 0.05). These results suggest that PME is useful in the chemoprevention or treatment of obesity and obesity-related disorders.

운동 트레이닝을 통한 심폐체력 반응의 차이가 복부비만 여성의 심혈관계 위험요인과 아디포싸이토카인에 미치는 영향 (Effects of different cardiorespiratory fitness response to exercise training on cardiovascular disease and adipocytokine in abdominal obesity women)

  • 본 연구는 복부비만 여성을 대상으로 운동 트레이닝을 통해 심폐체력의 반응의 차이가 심혈관계 위험요인 및 아디포싸이토카인 분비 및 유전자 발현에 미치는 영향을 알아보는 데 그 목적이 있다. 본 연구는 운동 트레이닝 실시 후 심폐체력 향상의 유무를 이용하여 복부비만 여성 총 48명을 운동-반응 집단(n=34)과 운동-비반응 집단(n=14)으로 나누었다. 운동 트레이닝은 주당 1200kcal를 소모하도록 설계되었으며, 중등도-활발한 운동강도, 걷기나 간단한 조깅의 형태로 12주간 실시하였다. 측정 변인으로 실험 참가 전, 후에 신체조성, 혈압, 심폐체력, 혈중지질, 글루코스, 인슐린 그리고 유리지방산을 측정하였으며, 혈중 아디포싸이토카인과 복부지방 아디포싸이토카인 유전자 발현을 분석하였다. 본 연구의 결과 운동 트레이닝 후 두 집단 모두 비만지표에서 통계적으로 유의하게 감소하였지만, 허리둘레(p=0.040), 체지방률(p=0.031) 그리고 중성지방(p=0.023)과 수축기 혈압(0.046)은 운동-반응 집단과 운동-비반응 집단에서 상호작용 효과를 보였다. 혈중 렙틴은 두 집단에서 상호작용 효과(p=0.022)가 나타났으나 복부지방 렙틴 유전자는 두 집단 모두에서 트레이닝 후(p<0.001)에 유의하게 감소하였다. 이상의 결과를 종합해보면 12주간 운동 트레이닝 실시 후 심폐체력의 향상을 보인 운동-반응 집단은 운동-비반응 집단에 비해 심혈관계 질환 위험요소와 아디포싸이토카인 분비 및 유전자 발현에 더 효과적인 것으로 나타났다. 또한 운동 트레이닝 실시 후 심폐체력의 개선이 없었던 운동-비반응 집단에서도 비만지표와 혈중 렙틴 및 렙틴 유전자 발현이 감소되는 것을 살펴봤을 때 규칙적인 운동 자체가 비록 심폐체력의 개선이 없더라도 인간의 건강상의 이점을 주는 것으로 나타났다. 따라서 심폐체력이 향상되지 않더라도 규칙적인 운동 은 비만 관련 위험요인의 감소 및 예방에 좋은 영향을 주는 것으로 사료된다.

식이로 유도한 비만마우스에서 아피오스의 내장지방 감소 효과 (Anti-Visceral Obesity Effect of Apios americana Medikus in Diet-Induced Obese Mice)

  • 본 연구는 고지방식이(열량의 40%를 지방으로 공급)로 유도한 비만마우스에게 아피오스 분말(10%, w/w)을 12주간 급여한 후 내장지방 감소 및 지방간 개선 효과를 살펴보았다. 아피오스 분말 급여로 인한 유의적인 체중 감소는 보이지 않았으나, 부고환지방과 후복막지방 무게가 유의적으로 감소하여 총 내장지방 무게는 대조군에 비해 14.3% 감소하였다. 부고환지방조직의 지방세포 크기 역시 아피오스 분말군에서 현저히 감소되었다. 혈청의 유리지방산, 중성지질과 총 콜레스테롤 함량은 실험군 간의 유의적인 차이가 없었으나 총 콜레스테롤에 대한 HDL-콜레스테롤 비와 동맥경화 지수는 아피오스 분말군이 대조군에 비해 각각 18.9%와 47.2% 유의적으로 개선되었다. 혈청의 렙틴 함량은 아피오스 분말군이 대조군에 비해 유의적으로 낮았다. 고지방식이를 급여한 마우스에 아피오스 분말 급여는 간 조직 중의 지질축적 및 유리지방산과 중성지질 함량을 유의적으로 낮추는 것으로 나타났다. 간 조직의 지질합성 관련 유전자인 Pparg, Fasn과 Dgat2의 발현이 아피오스 분말 급여로 억제되었다. 이상의 결과에서 아피오스 분말은 고지방식이로 인한 내장지방 축적과 지방간을 개선할 수 있을 것으로 평가되었다.

사상인과 혈청 렙틴 농도의 연관성에 관한 임상연구 (A Clinical Study on the Association between Sasangin and Serum Leptin Level)

  • 1. Objectives: Leptin is ob-gene originated hormone that is concerned with energy metabolism, obesity and asthma which features are supposed to related with Sasangin's 4 viscera physiological theory of Sasang constitutional medicine(SCM). This study aims to investigate whether serum leptin concentration is associated with Sasangin. 2. Methods: Two Way ANOVA analysis was used on the constitution and serum leptin concentration corresponding with BMI among 1054 cases drawn from the Constitutional Information Bank, which was a database with clinically confirmed constitution cases collected from 21 oriental medical institutes in Korea starting from November 2007 to May 2009. The measurement of serum leptin concentration is implemented through radioimmunoassay by request to SCL. 3. Results: Male subjects showed notable difference(p=0.033) in the comparison between constitution and serum leptin concentration corresponding to BMI levels. Tae-eumins showed the highest serum leptin concentration levels in the normal and overweight groups, followed by Soyangin and Soeumin. As the groups changed from overweight to obese, there was an interaction phenomenon in the order of serum leptin concentration levels(p=0.040). As a result, Soeumins showed the highest serum leptin concentration levels and Tae-eumins and Soyangins followed in place. Female subjects did not show statistically notable differences in comparison between constitution and serum leptin concentration corresponding to BMI levels(p=0.239). Serum leptin concentration levels among overweight and obese groups, resulted in the order of Soeumins, Tae-eumins, and Soyangins. But in the as groups changed from overweight to normal the order resulted in Tae-eumin, Soyangin, and Soeumin(p=0.660). 4. Conclusion: Due to a number of limitations in this study, the examination of the relation between constitution and leptin, a main hormone responsible for the digestion, appetite, and energy metabolism, resulted to be uncertain. But statistically notable differences were confirmed in male Tae-eumins with the consideration of BMI. Further examination of the relation between constitution and leptin is necessary through prospective studies with the restriction of confusion variables in the future.

경신해지환(輕身解脂丸) (GGT1)이 형질전환 비만모델 hGHTg 수컷 쥐의 비만관련 유전자 발현에 미치는 영향 (Effects of GyeongshinhaeGihwan 1(GGT1) on the Expression of Obesity-related Genes in Obese Male hGHTg Rats)

  • To investigate whether GyeongshinhaeGihwan 1(GGT1), an anti-obesity herbal medicine widely used in oriental medicine, regulates the expression of obesity-related genes, we measured the changes in mRNA levels of these genes by GGT1 in human growth hormone transgenic (hGHTg) obese male rats, and these effects by GGT1 were compared with those of reductil (RD), an anti-obesity drug approved by FDA. Rats received once daily oral administrations of autoclaved water, RD, or GGT1 for 8 weeks. At the end of study, rats were sacrificed and tissues were harvested. Total RNA from adipose tissue, liver and kidney was prepared and the mRNA levels for LPL (lipoprotein lipase), PPAR $\gamma$ (peroxisome proliferator activated receptor-gamma), PPAR$\delta$ (peroxisome proliferator activated receptor-delta), leptin, TNF$\alpha$ (tumor necrosis factor-alpha), and internal standard G3PDH (glyceraldehyde-3- phosphate dehydrogenase) were analyzed by RT-PCR. PPAR$\gamma$ mRNA levels of liver and kidney were decreased in drug-treated groups compared with control group and the decrease of PPAR$\gamma$ expression was more prominent in GGT1 group than in RD group, suggesting that GGT1 is effective in the inhibition of adipogenesis and lipid storage by decreasing the PPAR$\gamma$ expression. In contrast, PPAR$\delta$ mRNA levels of adipose tissue and kidney were increased by RD and GGT1 , and the magnitudes of increase were higher in GGT1 group than in RD group, indicating that GGT1 stimulates fatty acid oxidation and energy metabolism by activating PPAR$\delta$ expression, Compared with control and RD groups, GGT1 group had higher concentrations of serum leptin, a well-known inhibitor of appetite. However, The mRNA levels of leptin, LPL, and TNF$\alpha$ were not changed by GGT1 and RD, compared with DW. These results demonstrate that GGT1 not only decreases PPAR$\gamma$ expression of liver and kidney, but also increases PPAR$\delta$ expression of adipose tissue and kidney, leading to the regulation of obesity and that these effects were more pronounced in GGT1 group compared with RD group. In addition, GGT1 seems to prevent obesity by increasing the serum leptin levels.
