Nutritional Requirements of Actinomyces Isolated from Rumen of Goat

  • The objective of this work was to investigate the nutritional requirements for the growth of Actinomyces sp. 9RCC5 isolated from the rumen of a native goat in Korea. The growth of strain 9RCC5 on the basal medium or the medium minus certain ingredients from the basal medium demonstrated that strain 9RCC5 showed absolute requirement of vitamin B complex mixture, while hemin and volatile fatty acids (VFA) were stimulatory to growth to some extent. The 9RCC5 strain grew well with casein hydrolysate as the sole added nitrogen source. However, neither a complex of 18 amino acids nor ammonium sulfate effectively replaced casein hydrolysate. Vitamins such as riboflavin and pantothenate were essential for growth, while thiamin and biotin were stimulatory. With regard to VFA, the growth was stimulated by acetic acid but inhibited by valeric acid. Relatively large quantities of $Na^+$, $K^+$ and $Ca^{2+}$ were absolutely required for growth. Supplementation of clarified rumen fluid to the basal medium in a range of 0-10% (vol/vol) resulted in an increased rate of growth as well as an increased extent of growth.

  • The nutritional requirements for Prevotella sp. 4PCCNB2 isolated from the rumen of a native goat in Korea and those of the ATCC 19189 strain isolated from the bovine rumen were investigated. The two strains grew well with ammonium sulfate as the sole added nitrogen source. However, neither a complex of amino acids nor casein hydrolysate effectively replaced ammonium sulfate. Biotin, p-aminobenzoic acid, and vitamin $B_12$ were essential to culture the ATCC 19189 strain. Unlike the ATCC 19189 strain, however, $B_12$ was only stimulatory for the growth of the 4PCCNB2 strain. The 4PCCNB2 strain grew well in the basal medium without an individual acid such as acetic acid or valeric acid. In contrast, either acetic or valeric acid was absolutely required for the growth of the ATCC 19189 strain.

A Study of the Nutritional Composition of the Dandelion by Part (Taraxacum officinale) (민들레의 부위별 영양성분 함량 비교)

  • Many studies have presented results about the antioxidative and antimicrobial activities of Dandelions (Taraxacum officinale). There has yet to be a study which makes comparisons of nutrients based on the parts of the Dandelion. To identify the nutrient composition by part of dandelion, nutrient contents were analyzed. Dandelions were taken from Songpa-gu in Seoul and the nutrient composition of the flower, leaf and root were measured. The nutrient content of each part was analysed by using the method developed by the Association of Official Analytical Chemists (AOAC). The proximate components(moisture, protein, fat, fiber, ash, and carbohydrates), minerals and vitamins of the dandelion were analysed. The nutrient composition of the dandelions showed many significant differences among the parts when the differences were determined by using Duncan's multiple range test. The leaf contains more protein, ash, Ca, K, Mg, Zn, vitamin A, B$_1$, B$_2$, and C than the other parts. The root has much more fiber, carbohydrate, P and Fe content. The results demonstrate that dandelions could be used as a food source supplement for fiber, Ca, Fe and vitamin B$_2$ which are common nutritional deficiencies in Korea. It is recommended that more research for other bio-functional factors besides nutrients composition is needed to enhance the utilization of the dandelion.

  • Nutritional components, such as approximate compositions, and amino acid, mineral, vitamin, sugar, and fatty acid contents, of artificially cultivated Pleurotus ferulea were analyzed. Contents of carbohydrates, crude lipids, dietary fibers, crude proteins, total amino acids, particularly essential amino acids, minerals, water-insoluble and-soluble vitamins, glucose, and unsaturated fatty acids such as linoleic acid of P. ferulea were higher than those of P. ostreatus and P. eryngii. Results indicate P. ferulea has abundant essential nutrients and thus is good source of functional healthy food.

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4\;1.0g, \;sucrose\;30.0g, \;MgSo_4{\cdot}7H_2O\;0.5g, \;FeCl_3{\cdot}6H_2O\;1.0mg\;thiamine\;200{\gamma}g$, and distilled water 1000ml, pH6.5.

Growth and Physiological Effects of Immunity Feed Additives on the Juvenile Red Sea Cucumber Stichopus japonicus (홍해삼(Stichopus japonicus) 치삼의 성장 및 면역증강을 위한 면역증강사료첨가제의 개발)

  • The juvenile red variant of the sea cucumber Stichopus japonicus is distributed worldwide. It is a valuable food source in Korea, China, and Japan. Major issues in farming the juvenile red variant sea cucumber include growth rates and disease resistance. In recent years, studies have focused on feed additives to enhance immune system and health. In this study, we used a common diet (CD), a nutritional diet (ND; mixed nutritional supplements), and an immunity diet (ID; mixed probiotics, spirulina, and levan) and compared the growth rates and immunity of juvenile red variant sea cucumbers fed the experimental diets for 12 weeks. The growth and survival rates in the ID group were significantly increased (P>0.05). This suggests that the feed additives positively influenced immunity and growth in the ID group. However, the immune activity was exhibit a stabilizing effect, and further investigation of immune effects is required.

Effect of Growth Temperature and Nutritional Components on the Synthesis of Poly-3-Hydroxybutyric Acid by Filamentation-Suppressed Recombinant Escherichaia coli (Filamentation이 억제된 재조합 대장균에 의한 Poly-3-Hydroxybutyric Acid 합성시 배양온도와 영양분의 영향)

  • The effects of growth temperature and nutritional components on the synthesis of poly-3-hydroxybutyric acid, PHB, by filamentation-suppressed recombinant Escherichia coli XL1-Blue (pSYL107) were studied. After culturing XL1-Blue(pSYL107) for 48 hours in complex medium at 30$\circ$C, 7Al g/l of PHB could be obtained with the PHB content and PHB yield of 82% and 0.371 g PHB/g glucose, respectively. Lower concentration of PHB(3.2 g/l) was obtained when cultu- red at 37$\circ$C, which seemed to be due to the instability of this strain having amplified FtsZ activity. The PHB concentration of 3.75 g/l was obtained after culturing 60 hours in R medium supplemen- ted with 20 g/l glucose at 30$\circ$C, which was more than twice higher than that obtained with XL1-Blue(pSYL105). This suggested that the enhancement of PHB synthesis by suppressing filamenta- tion was more significant in a defined medium than complex medium. PHB synthesis could be further enhanced by supplementing a small amount of various complex nitrogen sources. When 5 g/l of beef extract was added to a defined medium, PHB concentration, PHB content, and PHB yield obtained after 60 hours of cultivation at 30$\circ$C were 7.46 g/l, 86%, and 0.375 g PHB/g glucose,respectively.

Studies on Dietary Habits and Residence Students' Satisfaction with University Dormitory Foodservice in Jeollabuk-do Iksan Area (전라북도 익산 지역 대학교 기숙사 학생의 식습관 및 기숙사 급식 만족도 조사)

  • The purpose of this study was to investigate eating habits and dormitory foodservices' satisfaction in university students using dormitory foodservice in the Jeollabuk-do Iksan area. Self-administered questionnaires were completed by 195 students (86 male, 109 female). Many students (58.5%) ate less than two meals per day and spent around 30 min eating meals. The results show that snack and midnight meals were the main reasons (37.9%) for unhealthy eating habits. Main source of nutritional knowledge and information were TV and the Internet (58.5%), followed by friends and people (25.1%), nutrition books (10.3%), elective courses (4.6%), and newspapers and magazines (1.5%). Men had significantly higher satisfaction scores for nutrition, taste, diversity of menu, as well as hygiene of dormitory food court compared to women (p<0.05). Salty taste was the most important factor in evaluation of taste satisfaction, whereas sour taste was opposite. The reason for taste dissatisfaction in the dormitory food court was not salty enough, and it may be related with their eating habits. The results show that students need education for adequate knowledge and information about the relationship between health and nutrition.

Variations in Reserved Nutrient Consumption and Growth of Pacific Oyster (Crassostra gigas) Larvae during Starvation (참굴 (Crassostrea gigas) 유생의 절식에 따른 성장 및 체내 에너지원의 소비변화)

  • The nutritional demand of oyster larva (Crassostrea gigas) were investigated to determine the optimal culture conditions and improve micro-algae utilization. Changes in nutrients and shell growth were examined in fed and 96-h (48 h in late umbone stage) oysters at four larval stages. Shell growth increased significantly in D shape larvae, regardless of feeding variations. No growth was observed in starved larvae, except in shell length of umbone (to 11.9 ${\mu}m$). Fed larvae showed significant growth in all development stages (P < 0.05). During starvation, lipids were most significantly decreased in all larval stages (by 76.8%, 68.3%, 76.3%, and 40.3%, respectively), followed by protein (41.1%, 31.1%, 33.1%, 16.7%) and nitrogen-free extracts (40.8%, 24.3%, 36.9%, 20.1%), Gross energy (kcal/g) consumption in each larval stage was 49.6%, 35.1%, 39.1%, and 20.4%, respectively. Our results indicate that lipids are the most important energy source during the early larval development stages of C. gigas.

Review of Food Therapy and Development of Diet Therapy Program for Diabetes Mellitus in 「Sikryochanyo」 (「식료찬요」 속 소갈(消渴) 식치방(食治方) 고찰과 이를 활용한 당뇨질환 예방 식단 개발)

  • Century-old nutrition and health concepts can be revived and applied in the modern age in the forms of newly developed menus, recipes, and lifestyle education. Current medical nutrition therapy concepts were first described in the Chosun Dynasty (1392-1897) in Korea based on the philosophy that food and medicine originate from the same source, which is known as 'food as medicine'. Recognizing the importance of culture, tradition, local diet, and lifestyle on health and medical nutrition therapy, we tried to rediscover traditional Korean approaches towards food consumption and nutrition through systematic review of the literature and developed contemporary menus accordingly. The medical nutrition therapy prescriptions described in 'Shikryochanyo' (1460) by the Chosun Dynasty's royal physician Soonyi Jeao cover 45 different diseases. In this project, we developed contemporary menus for those disease models that are most prevalent in modern society. Menus developed with foods that are readily available today were evaluated for their nutritional content and adequacy using a computer-aided nutritional analysis program (CAN pro 3.0, developed by the Korean Nutrition Society for comparison with RDA for Koreans). Therefore, century-old nutrition and health concepts can be revived and applied in modern society as newly developed menus recipes and lifestyle education.
