자연해수를 이용한 해수 중 영양염 분석용 표준물질 개발 (Development of Reference Material Using Natural Seawater for Nutrient Analysis in Seawater)

  • 해수 중 용존 영양염은 해양 내 물질순환을 이해하는 가장 기초적인 자료로 전 지구적 환경변화를 감지하는데 필수 생지화학 성분으로 그 중요성이 인식되고 있다. 분석능력이 다양한 연구자들에 의해서 상이한 시공간에서 측정된 용존 영양염 자료의 상호 비교를 위해 영양염 표준물질의 필요성이 증가하고 있다. 본 연구는 경기도 시화호 내측연안, 울진의 인근 연안 표층수와 동해 울릉분지 표층과 1500m에서 채취된 해수를 여과, 멸균하여 제조한 영양염 표준물질의 균질성과 안정성에 대하여 다루었다. 본 연구에서 제작된 영양염 표준물질은 해외에서 제조되는 영양염 표준물질과 유사한 균질성을 가지고 있다. 제조 후 3-13개월 동안 영양염 농도가 일정한 방향으로 감소하거나 증가하는 경향이 관측되지 않았으며 이 보다 더 긴 기간 동안의 안정성은 지속적인 모니터링이 필요하다.

  • To understand the fluctuation of global carbon levels caused by the biogeochemical cycle within the ocean interior, it is essential to achieve comparability of global oceanic nutrient data to a fairly high degree. The Scientific Committee on Ocean Research (SCOR) commissioned a working group (WG147) to establish a system for achieving comparability of oceanic nutrient data within 1% among laboratories around the world. The introduction of international activities for improving nutrient comparability will facilitate the use of nutrient reference material of seawater by researchers within Korea, which will help in meeting international standards of nutrient comparability and promote international cooperation.

  • A certified reference material (CRM) for the analysis of inorganic nutrients in nutritional supplements has been developed. Accurate mass fractions of chromium (Cr), iron (Fe), copper (Cu), and zinc (Zn) were determined by isotope dilution inductively coupled plasma mass spectrometry (ID ICP/MS). The measurement results were used to assign certified values for the CRM, which were metrologically traceable to the definitions of the measurement units in the International System of Units (SI). Production of a candidate reference material (RM) and the certification processes are summarized. Each nutrient in the CRM showed good homogeneity, which was estimated using relative standard deviations of the measurement results of twelve bottles in a batch. This CRM is expected to be an important reference to improve reliability and comparability of nutrient analyses in nutritional supplements and related samples in analytical laboratories.

Development of a Mushroom Powder Certified Reference Material for Element Analysis

  • A certified reference material (CRM) for the analysis of nutrient elements in an edible mushroom (Ganoderma lyceum) powder has been developed (KRISS CRM 108-10-011). The mass fractions of calcium (Ca), iron (Fe), and zinc (Zn) were measured by isotope dilution inductively coupled plasma mass spectrometry (ID ICP/MS). To dissolve the fungi cell wall of mushroom consisted of chitin fibers, sample preparation method by single reaction chamber type microwave-assisted acid digestion with acid mixtures was optimized. The mean measurement results obtained from 12 sample bottles were used to assign as the certified values for the CRM and the between-bottle homogeneities were evaluated from the relative standard deviations. The certified values were metrologically traceable to the definition of the kilogram in the International System of Units (SI). This CRM is expected to be used for validation of analytical methods or quality control of measurement results in analytical laboratories when they determine the mass fractions of elements in mushroom or other similar samples.

2010년 국내 해수 중 영양염 분석 실험실간 상호비교실험 연구 (2010 Inter-laboratory Comparison Study on Nutrient Analysis in Seawater)

  • 2010년에 국내 10개 영양염 분석 실험실을 대상으로 실험실간 상호비교실험이 실시되었다. 균질성과 안정성이 확보된 영양염 농도가 각기 다른 3종의 멸균해수(Bottle 130, 131, 132)가 이용되었으며, 각 실험실은 아질산염(Nitrite), 질산염(Nitrate), 인산염(Phosphate), 규산염(Silicate)의 항목을 5회이상 반복 분석하여 결과를 회신하였다. 각 실험실의 측정정밀도와 실험실간 차이를 평가하기 위해서 분석결과를 통계처리 하였다. 대부분의 실험실이 모든 분석항목에서 10%이내의 측정정밀도를 보였고, 이상치를 제거한 실험실 간 차이는 모든 분석항목에서 4~63%의 상대표준편차를 보였다. 상대백분율차이는 0.04~2217%였다. 각 실험실에서 분석한 영양염 농도의 차이는 우연오차(random error)보다는 계통오차(systematic error)에 기인한다고 볼 수 있다. 균질성과 안정성이 확보된 시료 131을 이용하여 각 실험실의 분석결과를 보정한 결과, 보정 후 실험실 간 차이는 모든 분석항목에서 상대표준편차와 상대백분율차이가 현저히 향상되었다. 본 상호비교실험결과를 종합하면, 본 실험에 참가한 국내 영양염 분석 실험실 내 측정정밀도는 양호한 편이므로 적절한 기준물질을 사용하여 분석자료를 보정할 경우 실험실 간 차이가 줄어들어 다른 실험실에서 생산된 자료와의 자료 공유에 대한 수월성이 높아진다고 볼 수 있다. 따라서 영양염 시료 분석 시 균질성과 안정성이 확보된 표준물질을 사용하여 다른 실험실과의 비교가능성과 소급성을 확립할 것을 제안한다.

The Supplementation of Yam Powder Products Can Give the Nutritional Benefits of the Antioxidant Mineral (Cu, Zn, Mn, Fe and Se) Intakes

  • Yam has been recognized having the beneficial effects for the prevention of various diseases, such as cancer, immunity, infection and obesity etc. There is increasing consideration to supplement the antioxidant nutrients to make up the lack of the antioxidant nutrient intakes. No study has been reported for the analysis of antioxidant mineral contents and comparison to dietary recommended intake for the sense of health promotion. In our study, we analyzed the contents of antioxidant trace elements (Zn, Mn, Fe, Cu and Se) and Cr contents in cultivated Korean yam powders for evaluation of nutrient intake aspects. We collected the commercial yam powders from six different cultivated areas in the South Korea and measured antioxidant minerals (Zn, Mn, Fe, Cu and Se) and Cr contents using trace element-free plasma spectrometer (ICP) or atomic absorption spectrometer (AAS) after dry-ashing and then wet-acid digestion. The accuracy of mineral analysis method was confirmed by the mineral analysis of standard reference material. Each analyzed element contents in yam were compared to dietary reference intakes of Koreans (KDRIs). The average levels of trace elements (Zn, Mn, Fe, Cu, Se and Cr) in yam powders were 18.3, 11.9, 36.0, 3.7, 1.9 and 1.27 ${\mu}g/g$ yam powder, respectively. The intakes of Zn, Fe, Cu and Se of which KDRIs is determined, are accounted as being up to 23.8%, 55.6%, 32.5% and 236% recommended intake (RI) of KDRIs, if daily yam supplementation (50 g) of commercial instruction would be considered. The intake of Mn is about 25% adequate intake (AI) of KDRIs with the daily supplementation of yam powder. Most of mineral intakes from daily yam supplementation were with the range of non-detectable to <10% upper limit (UL) level, which is very much safe. The study results show that daily supplementation of Korean yam power is beneficial to provide the supplemental nutrient intake and also is safe, if the suggested dosage would be considered.

NIR-TECHNOLOGY FOR RATIONALE SOIL ANALYSIS WITH IMPLICATIONS FOR PRECISION AGRICULTURE

  • The scope of precision agriculture is to reach the put up cultivation goals by adjusting inputs as precise as possible after what is required by the soil and crop potentials, on a high spatial resolution. Consequently, precision agriculture is also often called site specific agriculture. Regulation of field inputs “on the run” has been made possible by the GPS (Geographical Position System)-technology, which gives the farmer his exact real time positioning in the field. The general goal with precision agriculture is to apply inputs where they best fill their purpose. Thus, resources could be saved, and nutrient losses as well as the impact on the environment could be minimized without lowering total yields or putting product quality at risk. As already indicated the technology exists to regulate the input based on beforehand decisions. However, the real challenge is to provide a reliable basis for decision-making. To support high spatial resolution, extensive sampling and analysis is required for many soil and plant characteristics. The potential of the NIR-technology to provide rapid, low cost analyses with a minimum of sample preparation for a multitude of characteristics therefore constitutes a far to irresistible opportunity to be un-scrutinized. In our work we have concentrated on soil-analysis. The instrument we have used is a Bran Lubbe InfraAlyzer 500 (1300-2500 nm). Clay- and organic matter-contents are soil constituents with major implications for most properties and processes in the soil system. For these constituents we had a 3000-sample material provided. High performance models for the agricultural areas in Sweden have been constructed for clay-content, but a rather large reference material is required, probably due to the large variability of Swedish soils. By subdividing Sweden into six areas the total performance was improved. Unfortunately organic matter was not as easy to get at. Reliable models for larger areas could not be constructed. However, through keeping the mineral fraction of the soil at minimal variation good performance could be achieved locally. The influence of a highly variable mineral fraction is probably one of the reasons for the contradictory results found in the literature regarding organic matter content. Tentative studies have also been performed to elucidate the potential performance in contexts with direct operational implications: lime requirement and prediction of plant uptake of soil nitrogen. In both cases there is no definite reference method, but there are numerous indirect, or indicator, methods suggested. In our study, field experiments where used as references and NIR was compared with methods normally used in Sweden. The NIR-models performed equally or slightly better as the standard methods in both situations. However, whether this is good enough is open for evaluation.

해수 중 용존 암모늄 분석방법 비교: 분광광도법과 형광법 (Comparison of Dissolved Ammonium Analytical Method in Seawater: Spetrophotometry and Fluorometry)

  • 해수 중 용존 암모늄 분석에는 Berthlot's 반응의 분광광도법이 일반적으로 사용되었으나, 최근에는 OPA (orthophthaldialdehyde-sulfite) 형광시약을 이용한 형광법이 국제적으로 활발하게 사용되고 있다. 본 연구에서는 2017년도 호주연구선 Investigator에서 수행된 국제 영양염 선상 상호비교 실험에서 두 기관(KIOST(분광광도법, 1차 선형 회귀식 기울기만 사용), 호주CSIRO(형광법, 2차 비선형 회귀식 사용))간의 영양염 표준물질과 현장 해수시료의 용존 암모늄 농도값 차이(약 0.31 𝜇M)의 원인을 이해하기 위해 분석방법 간 검출한계, 분광광도법에 내재된 시약 굴절률, 검량선식 등에 의한 영향을 고찰하였다. 본 연구에서 측정된 분광광도법의 방법검출한계(0.063 𝜇M)와 시약 굴절률 바탕값(0.054 𝜇M)은 두 기관의 용존 암모늄 농도값 차이를 각각 20%, 17% 정도 설명할 수 있었다. 그러나 분광광도법의 검량선을 형광법과 동일한 2차식 또는 선형회기식의 기울기와 절편으로 농도를 계산할 경우, 두 기관 간의 용존 암모늄 농도값 차이는 현장에서 측정된 분광광도법의 방법검출한계 이하로 줄었다. 따라서 국제 영양염 선상 상호비교 실험에서 영양염 표준물질과 해수 현장시료에서 나타난 두 기관 간의 용존 암모늄 농도값 차이는 분광광도법과 형광법의 분석방법 차이에 의한 영향보다는 두 방법에 사용된 상이한 검량선식 영향인 것으로 판단된다. 향후 해수시료의 용존 암모늄 자료를 비교할 때 분석방법의 차이보다는 분석에 사용된 기저선, 표준용액열 개수, 검량선식 등에 대한 정보에 유의할 것을 권장한다.

Comparison of protein and amino acid content in selected cultivars of non-glutinous rice in Korea

  • The objective of this study was to determine amino acid and protein contents of brown and milled non-glutinous rice of 13 cultivars in Korea. Protein contents of MRs (milled rices) were in order of Haepum (7.27%) > Hanareum No. 4 (7.14%) > Odae (6.84%). Protein contents of BRs (brown rices) were in order of Haepum (7.68%) > Odae (7.63%) > Hanareum No. 4 (7.60%). The amino acid content was the highest in Haepum (MR 5.76%, BR 6.49%), followed by Haedeul (MR 5.71%, BR 6.30%), and Odae (MR 5.63%, BR 6.29%). The essential amino acid contents of non-glutinous rices were in order of Haepum (MR 2.34%, BR 2.57%) > Haedeul (MR 2.31%, BR 2.48%) > Odae (MR 2.20%, BR 2.56%). The contents of amino acid and protein in BRs were considerably higher than those in MRs. Protein and most of amino acid contents were higher in Haepum than the other cultivars. The certificated reference material (CRM) 1849a (infant/adults nutritional formular) from National Institute of Standard and Technology (NIST) was used as the test sample to determine the precision and accuracy of the analytical method. The regression analyses revealed good correlations (correlation coefficient), greater than 0.99. The recovery values of the amino acids ranged from 93.17 to 99.59%. The limit of detection (LODs) ranged from 0.01 - 0.07 mg·100 g-1 and the limit of quantitation (LOQs) ranged from 0.03 - 0.21 mg·100 g-1 for all analytes.

국내 식품 중 구이, 찜, 볶음, 조림에 존재하는 수용성 비타민 B1, B2 그리고 B3 함량 조사 (Investigation of Water-soluble Vitamin (B1, B2, and B3) Contents in Various Roasted, Steamed, Stir-fried, and Braised Foods Produced in Korea)

  • 본 연구는 국내 식생활의 기본적인 구성식단으로 알려진 구이, 찜, 볶음, 조림에 존재하는 다양한 영양성분 중 수용성 비타민인 $B_1$ (thiamin), $B_2$ (riboflavin), 그리고 $B_3$ (niacin)의 함량을 확인하였다. 실험분석 타당성 검증을 통해서 높은 직선성($r^2$>0.997)을 확인하였고, 검출한계의 경우 $0.001-0.067{\mu}g/mL$ 그리고 정량한계의 경우 $0.002-0.203{\mu}g/mL$를 확인하였다. 또한 실험값에 대한 정밀도와 반복성에 대한 검증을 위해서 standard reference materials를 통해 실험값과 표준값과의 오차가 신뢰도 이내에 존재한다는 결과를 확인하였다. 본 연구에서 제시된 구이식품의 경우 thiamin은 0.039-1.057 mg/100 g의 범위에 존재하였고, riboflavin은 0.058-0.686 mg/100 g의 범위에 존재했으며, niacin의 경우 0.021-21.772 mg/100 g 의 범위로 나타났다. 찜 종류의 경우 thiamin은 0.049-1.066 mg/100 g의 범위에 존재하였고, riboflavin은 0.025-0.548 mg/100 g의 범위에 존재했으며, niacin의 경우 0.134-21.509 mg/100 g의 범위로 나타났다. 볶음의 경우 thiamin은 0.114-0.388 mg/100 g의 범위에 존재하였고, riboflavin은 0.014-1.258 mg/100 g의 범위에 존재했으며, niacin의 경우 0.015-2.319 mg/100 g의 범위로 나타났다. 조림의 경우 thiamin은 0.112-1.656 mg/100 g의 범위에 존재하였고, riboflavin은 0.024-0.298 mg/100 g의 범위에 존재했으며, niacin의 경우 0.322-2.157 mg/100 g의 범위로 나타났다. 본 연구에서 제시된 구이, 찜, 볶음, 그리고 조림 식품에 대한 수용성 비타민 함량에 대한 영양성분 데이터베이스 구축 연구가 국민 식생활의 중요한 기초자료로 이용될 것으로 확신한다.
