Increases in the Activities of Microsomal ATPases Prepared from the Roots of Lettuce Cultured in Salt-enhanced Nutrient Solutions (양액내 염류농도 증가에 의한 상추뿌리의 마이크로솜 ATPase 활성증가)

  • In order to investigate the mechanism of growth inhibition by salt stress, lettuces were grown hydroponically in three different nutrient solutions, normal and 30 mM or 50 mM $KNO_3$-added nutrient solutions, and the electrical conductivities of these solutions were 1.0, 4.5, and 6.5 dS/m, respectively. The activities of plasma and vacuolar $H^+$-ATPases in the root tissue of lettuce were measured by specific inhibitors, 100 ${\mu}M$ vanadate and 50 mM $NO_3^-$, respectively. Microsomal ATPase activity of lettuce grown in the normal nutrient solution was $356\pm1.5$ nmol/min/mg protein. When lettuces were grown in 30 mM and 50 mM $KNO_3$-added nutrient solutions, total activities of microsomal ATPases were increased by 1.6 and 1.9 times, respectively, and the increases were mainly mediated by vacuolar $H^+$-ATPase. These results show that lettuces adapt themselves to salt-stressed condition by increasing the activities of $H^+$-ATPases. Effects of various heavy metal ions were investigated on the microsomal ATPases and various metal ions at 100 $\mu M$ inhibited the activities by 10$\sim$25%. $Cu^{2+}$ showed the highest inhibitory effect on the vacuolar $H^+$-ATPase. These results suggest that lettuce increases the activities of root ATPases, specially that of vacuolar $H^+$-ATPase, in salt-stressed growth conditions and $Cu^{2+}$ could be a useful tool to control the activity of vacuolar $H^+$-ATPase.

Improvement of K+ and Na+ Ion homeostasis and salt tolerance by Co-inoculation of arbuscular mycorrhizal fungi (AMF) and spore associated bacteria (SAB)

  • Salinity inhibits plant growth and restricts the efficiency of arbuscular mycorrhizal fungi. The selective uptake of nutrients from the soil and their effective transport to host roots make it essential for plant growth and development under salt stress. AMF spore associated bacteria shown to improve mycorrhizal efficiency under stress. Thus, this study aimed to understand the co-inoculation efficiency of AMF and SAB on maize growth and ion homeostasis under salt stress. Two AMF strains and one SAB were inoculated with maize either alone or in combination with one another. The results of our study showed that AMF and SAB co-inoculation significantly improved dry weight and nutrient uptake of maize under salt stress. Co-inoculation significantly reduced proline accumulation in shoots and Na+ accumulation in roots. Co-inoculation treatment also exhibited the high K+/Na+ ratios in roots at 25 mM NaCl. Mycorrhizal colonization showed positive influence for regulation of ZmAKT2, ZmSOS1 and ZmSKOR gene expressions, contributing to K+ and Na+ ion homeostasis. CLSM view showed that SAB were able move and localize into inter and intra cellular spaces of maize roots. In addition, CLSM view of AMF spores showed that gfp-tagged SAB also associated on the spore outer hyaline layer.

Appropriate Pretreatment Method of Coir Bag in Coir Culture (코이어 재배시 적정 전처리 방법 구명)

  • We examined pretreatment methods eliminating potassium and sodium efficiently for coir bag used in hydroponics by analyzing drainage coming from coir bags. In the first experiment we investigated for six coir bags with the high market shares. The three types of pretreatment were washing coir bags with only water for 7 days (W7S0), washing with water for 4 days and further with nutrient solution for 3 days (W4S3), and washing with only nutrient solution for 7 days (W0S7). In the second experiment we tested reproducibility of the experiment results for Bio Grow and coco Mix among six coir bags used in the first experiment to verify the results. As a result, the best pretreatments for the pH stabilization were W4S3 and W0S7. The EC value of the drainage was stabilized to less than 1.0 that is the same as EC of the supply solution on the fourth day in all treatments. The nutrients of the drainage in W7S0 was stabilized in 3~4 days but calcium and magnesium were depleted. We assessed that washing longer than 4 days was waste of water. The stabilization of coir bags in W0S7 was similar to it in W4S3, but washing with the nutrient solution for 7 days seemed to be uneconomical. The reproducibility experiment for two coir bags ensured the results in the first experiment. Therefore, the pretreatment method, which is the most simple to implement and economic, seems to wash with water for 3 days and then with the nutrient solution for 1 day before planting on coir bag.

Disposable Nitrate-Selective Optical Sensor Based on Fluorescent Dye

  • Purpose: This study was performed to develop a simple, disposable thin-film optical nitrate sensor. Methods: The sensor was fabricated by applying a nitrate-selective polymer membrane on the surface of a thin polyester film. The membrane was composed of polyvinylchloride (PVC), plasticizer, fluorescent dye, and nitrate-selective ionophore. Fluorescence intensity of the sensor increased on contact with a nitrate solution. The fluorescence response of the optical nitrate sensor was measured with a commercial fluorospectrometer. Results: The optical sensor exhibited linear response over four concentration decades. Conclusions: Nitrate ion concentrations in plant nutrient solutions can be determined by direct optical measurements without any conditioning before measurements.

Adaptive Estimation of Hairy Root Mass Using Conductometry

  • An accurate and efficient method for measuring the mass of hairy roots using conductometry is established. A conductivity equation expressed in terms of the concentration of the ion species in the medium is suggested. By using this equation, the effect of the individual ions on the total conductivity can be quantitatively analyzed. An equation for the in situ estimation of the cell growth coefficient for determining the mass of hairy roots is established based on measurements of the nitrogen concentration and conductivity during cultivation. The proposed equation does not require preliminary experiments to determine the cell growth coefficient. Instead, the physiological characteristics of the plant species are reflected by introducing the cellular nitrogen content. Since the cell growth coefficient is determined by measuring the major ionic nutrient concentrations, it is more effective to express the dynamics of an actual culture system. This improved method for determining the mass of hairy roots was successfully utilized in a fed-batch culture system.

Feasibility Study with Several Sorption Materials to Treat Road Runoff Pollutions (흡착 소재별 도로 노면 유출 오염원의 저감성 비교 연구)

  • This research was conducted to figure out the feasibility of several sorption materials to treat various pollutants in road runoff. In advance of the major feasibility test with various sorption materials, the separation process with $1.2{\mu}m$ filter was conducted and showed that slight portion of pollutants was removed(Orgamic pollutant - 20%, Nutrient salt - 50%, Heavy metals - 0~30%). To remove dissolved pollutants in runoff, various materials were tested through an isotherm sorption experiment. As a result, GAC showed most effective material among them to lessen most contaminants such as organic compounds and nutrients. On the other hand, ion-exchange resin and Zeolite showed limited usefulness on the some heavy metals. Freundlich model was most suitable for the current experiment data, and the amount of adsorbent (GAC) could calculated based on this model.

  • Iron is an essential nutrient for most organisms, which supplied to them in a protein-iron complex known as ferritin. Ferritins are multimeric proteins those are consisted of spherical shell of 24 subunits defining a cavity of about 8nm in diameter. Soluble form of ferritin was separated from disrupted cells, followed by silica powder adsorption. Ferritin was recovered from silica-poweder by distiiled water, which was applied to DEAE anion exchage chromatography. Collected fractions from the DEAE column were assayed to gain the amount and the purity of ferritin by using GF-HPLC.

Studies on the Chemical composition of Zostera marina ("거머리 말"의 화학성분에 관한 연구)

  • General components, free sugars, and free amino acids about Zostera marina which is much distributing and producing along the coastlines of Korea have been separated and identified through Ion exchange resin and Paper chromatography method. So distrigushed are sugars contained that this was quantitated by Bertrand method. 1) The result of the general component analysis of Zostera marina shows that there are much free sugars. 2) According to Bertrand method sugar containment quantitated from terrestrial stem and subterranean stem on Po-hang and Ko-hung areas is much more in terrestrial stem than subterranean stem. 3) Seven kinds of sugars are used in experiments(Table 2) among there glucose, fructose and sucrose which keep the sweetness are very much contained. 4) Shown in Fig. 3 ten kinds of free amino acids inspected from experimental liquid by Paper chromatography are separated, which are known, and one spot which in unknown is additionally separated, and in which four kinds of essential amino acid to nutrient are contained.

  • 실시간 온라인 계측에 의한 순환식 양액조제 알고리즘을 위해서 이온센서를 사용하였다. 이온센서의 내구성이 약하고 배양액 내에는 이온센서의 정확한 계측의 방해요인인 이온성분들이 포함되어 있기 때문에 매번 정확한 측도 설정을 수행 해줌으로써 계측값의 신뢰도를 높일 수 있었고 이온센서를 이용한 온라인계측의 가능성을 확인할 수 있었다. 그러나 배양액 속에 장시간 넣어둔 상태로 사용하였을 경우 4시간 이상 경과하면 센서에서 나오는 신호가 지속적으로 상승하는 경향을 보였다. 장시간 동안 배양액에 노출되었던 센서는 측도설정이 불가능한 수준으로 일정농도의 용액 내에서 일정한 값을 나타내지 못하고 계측값이 지속적으로 상승하던가 하강하여 한계값 까지 이르렀다. 이러한 이유로 연구용이 아닌 실제 장치에 적용하여 실용화하기 위해서는 내구성이 강하고 계측하고자 하는 이온에 대해 영향을 주는 방해이온 속에서도 정확도를 유지 할 수 있는 센서의 개발이 선행되어야 한다. 또한 정확한 퇴액 성분의 측정을 위해서는 측도 설정이 선행되어야 하는데 계측이 필요할 때마다 측도 설정을 자동으로 해줄 수 있는 자동 측도 설정 모듈의 개발도 수행되어야 할 것으로 사료된다.

Effect of Concentration of Nutrient Solution on Water and Nutrient Uptake of Tomato Cultivars in Hydroponics (배양액 농도가 수경재배 토마토의 품종별 생육과 양수분 흡수특성에 미치는 영향)

  • This study was carried out to acquire basic data for a long-term hydroponic culture through investigating water and inorganic ion uptake characteristics at different EC level of nutrient solution of three tomato varieties. Three different tomato varieties, the European type(cv. Daphnis), the Asian type(cv. Super Doterang) and cherry type(cv. Minichal), were used for the investigation. Also, the deep flow technique(DFT) was applied. The three different electrical conductivity(EC) level(1.0, 2.0, 3.0, and 4.0 dS·m-1) of hydroponic nutrient solution were used as variable. At a high EC level of nutrient solution, the leaf area and fresh weight decreased in the early stage, and its growth(plant height, leaf number, leaf area, fresh-weight) was poor with salt stress. Result showed that the higher the EC level of the nutrient solution, the lesser was water uptake. The water uptake was not significantly different from varieties in the first survey, but In the second survey, the 'Daphnis' did not show a significant decrease in water uptake in the EC level higher than 2.0 dS·m-1., on the other hand, 'Super Doterang' presented very low water uptake. At a low EC level, N, P, and K, were absorbed more than the concentration of the irrigation water, while Ca, Mg, S uptake were low. At a high EC level, almost ions absorbed less than 50% of the initial concentration of irrigation water. Thus, imbalance among ions was severe at low EC level compared to high EC level. 'Daphnis' was a variety that effectively utilize nutrients under nutrient stress, showing high absorption at low concentration condition and low absorption at high concentration condition. However, 'Daphnis' suffered most seriously by absorbing nutrients excessively.
