An experiment was conducted to examine the feasibility of producing multiple calves using embryo transfer in Japanese Black cattle. Milk yield of cows and forage intake of calves were measured for 11 cows with single calves, 14 cows with twins and one cow with triplets. The means of 26 weeks cumulative milk yield were 854, 1028 and 1271 kg for cows having singles, twins and triplets, respectively. Male birth weights for single calves, twins and triplet were 34.9, 26.6 and 19.9 kg, and female ones were 31.7, 24.1 and 22.1 kg, respectively. Weight and daily gain of calves were affected by weeks (W), sex (S), the number of calves (N), parity, birth season, $W{\times}N$, $S{\times}N$ and regression on milk yield. Growth rate was higher for single calves than for twins until about 9 weeks of age, then weights increased at a similar rate. Male calf weaning weights for singles, twins and triplets were 207.0, 177.1 and 162.2 kg, and those for females were 185.4, 151.6 and 180.4 kg, respectively. Average regression coefficients of calf growth on milk yield were significant, and single calf was affected more than twin calves by increment of milk yield. As the number of calves per cow increased, hay intake of calves decreased and concentrate intake tended to increase between 6 and 13 weeks of age.
The use of genomic information in genomic selection programs for dairy and beef cattle breeds has become a reality in recent years. In this investigation, we analyzed single-nucleotide polymorphisms (SNPs) for Hanwoo (n=50) and Holstein (n=50) breeds using the Illumina Bovine SNP50 BeadChip to facilitate genomic selection and utilization of the Hanwoo breed in Korea. Analysis of the entire genomes showed different spectra of SNP frequencies for Hanwoo and Holstein cattle. The study revealed a highly significant (p<0.001) difference between Hanwoo and Holstein cattle in minor allele frequency (MAF). The average MAFs were $0.19{\pm}0.16$ and $0.22{\pm}0.16$ for Hanwoo and Holstein, respectively. From the total of 52,337 SNPs that were successfully identified, about 72% and 79% were polymorphic in Hanwoos and Holsteins, respectively. Polymorphic and fixed SNPs were not distributed uniformly across the chromosomes within breeds or between the two breeds. The number of fixed SNPs on all chromosomes was higher in Hanwoo cattle, reflecting the genetic uniqueness of the Hanwoo breed. In general, the rate of polymorphisms detected in these two breeds suggests that the SNPs can be used for different applications, such as whole-genome association and comparative genetic studies, and are a helpful tool in developing breed identification genetic markers.
Significant SNPs associated with Warner-Bratzler (WB) shear force and sensory traits were confirmed for Hanwoo beef (Korean cattle). A Bonferroni-corrected genome-wide significant association (p< $1.3{\times}10^{-6}$) was detected with only one single nucleotide polymorphism (SNP) on chromosome 5 for WB shear force. A slightly higher number of SNPs was significantly (p<0.001) associated with WB shear force than with other sensory traits. Further, 50, 25, 29, and 34 SNPs were significantly associated with WB shear force, tenderness, juiciness, and flavor likeness, respectively. The SNPs between p = 0.001 and p = 0.0001 thresholds explained 3% to 9% of the phenotypic variance, while the most significant SNPs accounted for 7% to 12% of the phenotypic variance. In conclusion, because WB shear force and sensory evaluation were moderately affected by a few loci and minimally affected by other loci, further studies are required by using a large sample size and high marker density.
Effects of recombinant bovine somatotropin (bST) on plasma hormonal concentration, embryo quality, and pregnancy rate were examined during the superovulation and synchronization treatment in donor and recipient cows. Hanwoos (Korean native beef cattle) were treated with controlled internal drug release (CIDR) combined with bST (CIDR+bST) or without bST (CIDR) as donor cows. The embryos recovered from donors were transferred into Holstein recipient heifers treated with bST (CIDR+bST) or without bST (CIDR) for synchronization. The correlation between IGF-I and P4 showed a positive pattern in the CIDR+bST group (r=0.44, p<0.01), but a negative pattern was shown in the CIDR group (r = -0.59, p<0.02) at day 7 of estrous cycles. Although the number of recovered, transferable, and degenerated embryos was not different, quantities of grade 1 (excellent) embryos in CIDR+bST group were significantly higher than those of the CIDR group (p<0.01). The pregnancy rate was higher in the CIDR+bST recipient group compared to CIDR group (p<0.05), when the embryos were recovered from the donors treated with CIDR. However, the pregnancy was maintained highly in both recipient groups, when the embryos were produced by CIDR+bST treated donors. It can be concluded that bST administration combined with CIDR is an effective method for superovulation and synchronization treatment to stabilize plasma hormonal levels, to obtain excellent quality of embryos, and to get higher pregnancy rate.
Intermuscular fat is essential for enhancing the flavor and texture of cultured meat. Mesenchymal stem cells derived from intermuscular adipose tissues are a source of intermuscular fat. Therefore, as a step towards developing a platform to derive intermuscular fat from mesenchymal stem cells (MSCs) for insertion between myofibrils in cultured beef, an advanced protocol of intermuscular adipose tissue dissociation effective to the isolation of MSCs from intermuscular adipose tissues was developed in cattle. To accomplish this, physical steps were added to the enzymatic dissociation of intermuscular adipose tissues, and the MSCs were established from primary cells dissociated with physical step-free and step-added enzymatic dissociation protocols. The application of a physical step (intensive shaking up) at 5 minutes intervals during enzymatic dissociation resulted in the greatest number of primary cells derived from intermuscular adipose tissues, showed effective formation of colony forming units-fibroblasts (CFU-Fs) from the retrieved primary cells, and generated MSCs with no increase in doubling time. Thus, this protocol will contribute to the stable supply of good quality adipose-derived mesenchymal stem cells (ADMSCs) as a fat source for the production of marbled cultured beef.
This study was performed to investigate the FSH levels for superovulation procedure in Korean Native Cattle (Hanwoo). The effectiveness of 200 mg and 400 mg of FSH to initiate superovulation was examined in Hanwoo. Donors, at random stages of the estrous cycle, received a CIDR 7 days later, 200 mg FSH group was treated with 40, 30, 20, 10 mg FSH levels in declining doses twice daily by intramuscular injection fur 4 days. Also, 400 mg FSH group was treated with 80, 60, 40, 20 mg FSH levels. On the 3rd day administration of FSH, 25 mg $PGF_2$${\alpha}$ was administered and CIDR was withdrawn. Donors were artificially inseminated twice at 12 hr intervals. The donor cattle received 250 ${\mu}g$ GnRH at time of 1st insemination and embryos were recovered 8 days after the 1st insemination. As a results, average number of CL treated with FSH 200 mg was higher as $20.9{\pm}1.20$ than $15.8{\pm}0.63$ for donors treated with FSH 400 mg, respectively(p<0.05). Treated group of 200 mg FSH level increased (p<0.05) the number of embryos recovered per procedure compared to 400 mg FSH level ($18.2{\pm}1.18$ vs. $12.38{\pm}0.52$, respectively). When treatment of 200 mg FSH was performed, average transferable embryos/ova increased (p<0.05) to $14.1{\pm}1.12$ from $6.8{\pm}0.33$ of treated of 400 mg FSH. Group of 200 mg FSH increased (p<0.05) to $8.3{\pm}0.76$ from $2.0{\pm}0.26$ in morula stage compare to 400 mg FSH group. Mean of total early blastocyst and expanded blastocyst stage embryos was similar (p<0.05) between the 200 mg and 400 mg FSH levels group ($4.7{\pm}1.19$ vs. $2.9{\pm}0.18$ and $1.2{\pm}0.40$ vs. $1.9{\pm}0.17$). These results suggest that 200 mg FSH level-based superovulation protocol with CIDR may be effectively used fur production of superior embryos in Hanwoo. In other words, the less level of FSH may be effectively applied for Hanwoo (Korean Native Cattle), because Hanwoo was smaller body size than beef or daily cow.
쇠고기 이력추적제는 소 개체마다 이력번호를 부여하여 수입쇠고기가 한우고기로 둔갑하는 사례를 막고, 소비자에게 구입한 한우고기의 세부 이력 정보들을 제공하는 데 도움을 준다. 이번 연구는 2021년-2022년 서울지역 대형 축산물판매업소에서 유통되는 한우고기 344건에서 DNA 동일성 검사를 실시하여 이력번호 등 주요 표시정보들의 진위를 판별하였다. 그 결과 45건(13.1%)에서 이력번호가 불일치한 것으로 확인되었다. 불일치율은 2021년(14.7%)에 비해 2022년(11.3%) 감소하였고, 도심서북권 20.7%, 동북권 14.4%, 서남권 및 동남권 10.2% 순으로, 북부권(16.9%)이 남부권(10.2%)에 비해 높게 나타났다. 또한 6개 브랜드 중 B사 및 D사는 이력관리가 양호한 반면, E사 및 A사는 이력관리가 취약한 것으로 확인되었고, 통계적으로 유의한 결과 차이를 보였다(P<0.001). 이력번호 불일치 시료의 실제 이력번호는 업소 내 입(출)고 거래내역 자료를 근거로 조사하였고, 그 결과 실제 이력번호 확인율은 53.9%에 불과하였으나, 확인된 범위 내에서 시료의 표시정보 진위 판별 결과, 거짓 표시는 이력번호 13.1%, 성별 2.9%, 도축장명 2.2%, 등급 1.6% 순으로 나타났고, 품종(한우) 거짓 표시는 없었다. 거짓 표시 축산물의 유통 차단을 위해 표시정보의 신속한 진위 판별이 중요하므로 업소 내 이력번호가 기재된 날짜별 부분육 소분할 작업내역 작성을 의무화하는 법적 근거 마련이 필요하다. 본 연구 결과는 투명한 축산물 유통 거래 질서 정착을 위한 이력관리 방향 설정에 참고자료로 활용하고자 한다.
Japanese Brown cattle, one of the four domestic beef breeds in Japan, are suffering from numerical reduction due to economic pressure from profitable breeds. In this study, all the reproductive cows in the Kouchi sub-breed of the Japanese Brown cattle that were alive in July 2005 were investigated by pedigree analysis to clarify genetic structure and composition of genetic variability. In addition, genetically important individuals for the maintenance of genetic variability of the sub-breed were also identified through the core set method. The number of cows analyzed was 1,349. Their pedigrees were traced back to ancestors born around 1940, and pedigree records of 13,157 animals were used for the analysis. Principal component analysis was performed on the relationship matrix of the cows, and their factor loadings were plotted on a three-dimensional diagram. According to their spatial positions in the diagram, all the cows were subdivided into five genetically distinctive subpopulations of 131 to 437 animals. Genetic diversity of the whole sub-breed, which is estimated to be 0.901, was decomposed into 0.856 and 0.045 of within-subpopulation and between-subpopulation components. Recalculation of genetic diversity after removal of one or several subpopulations from the five subpopulations suggested that three of them were genetically important for the maintenance of genetic variability of the sub-breed. Applying the core set method to all the cows, maximum attainable genetic diversity was estimated to be 0.949, and optimal genetic contributions assigned to each cow supported the previous results indicating relative importance of the three subpopulations as useful genetic materials.
Fecal samples from beef and dairy calves less than 12-month-old of Chonnam area were examined for coccidial oocysts. The percentages of cattle and coccidian species found were: Eimeria alabamensis. 9.4; E. auburnensis, 12.5; E. bovis, 29.9; E. brasiliensis, 1.5; E. bukidnonensis, 2.1; E. canadensis, 10.4; E. cylindrica, 7.5; E. ellipsoidalis, 11.5; E. pellita, 1.7; E. subspherica, 10.4; E. wyomingensis, 2.9; and E. zuernii, 22.5. Fifty five percent of the fecal samples from 521 calves were positive for oocysts, and clinical coccidiosis was not found in any of these calves. The number of species in positive samples ranged from 1 to 7.
