남극 호냉성 미세조 Porosira pseudodenticulata의 형태와 분자적 자료 (Morphology and Molecular Data for Antarctic Cryophilic Microalga, Porosira pseudodenticulata)

  • We have cultured more than 100 Arctic and Antarctic cryophilic microalgal strains in KOPRI culture collections of polar microorganisms (KCCPM). Among them, we tried to identify an Antarctic strain, KOPRI AnM0008 by morphological and molecular analysis. Nuclear SSU rDNA and plastid rbcL sequences were used to identify the strain. It was identified as Porosira pseudodenticulata based on SSU sequence data showing 99% identity with GenBank X85398. This result was supported by morphological features like solitary labiate process, external foramina and internal cribra by optical and scanning electron microscope. Morphological identification and molecular analysis on polar cryophilic microalgae will be accomplished to construct the databases for KCCPM.

  • 유해성 적조생물 Cochlodinium polykrikoides/Gyrodinium aurelum을 포함한 43 종류의 와편조류를 대상으로 SSU 부위 유전자를 분석했다. 유전자 염기서열에 의거한 상호 계통수는 parisomny, distance, maxium 방법으로 실행했다. Noctilura scintillans, Gonyaulax spinifera와 Crythecodinium cohnii 종들은 와편모조류 중 가장 유전적으로 먼 것으로 보였다. Alexandrium과 Symbiodinium 종간의 bootstrap는 70% 이상의 상호 단일 계통도를 보인 반면에, Gymnodinium과 Gyrodinium은 근립절약계수와 최대 유사도 방법에서 다형 계통도를 나타내었다. Gyroddinium aurelum과 G. dorsum/G. galathenum의 유전적 분화율은 7.4% (45 bp) 였고, G. aurelum과 G. mikimotoi 상호간에는 0.9% (5bp) 밖에 나타나지 않았다. 또한 C. polykrikoides와 G. aurelum도 5.2% (31bp)로 낮은 유전적 분화율을 보였다. 계통도를 분석한 결과 G. aureolum과 C. polykrikoides는 Gyrodinium 보다 Gymnodinim 속에 훨씬 더 근접하게 나타났다.

  • 해양 같은 지리적 장벽에도 불구하고, 많은 담수조류는 세계의 다른 대륙에 서식하고 있다. 단세포 담수조류, Mallomonas caudata는 북반구의 미국, 유럽, 및 아시아에서 흔하며, 기후변화 감지나 수화현상 감시 같은 인간생활에 밀접하게 관련되어 있다. 본 종의 계통과 국내출현을 설명하기 위해 여섯 군데의 저수지에서 채집된 균주로부터 엽록체 rbcL과 핵 SSU유전자를 염기서열 분석하였다. 또한, 한국산 균주의 전자현미경적 구조를 조사하였다. 한국과 미국산 종의 SSU염기서열은 0.06%가, rbcL은 0.45%의 차이로 거의 동일하였다. 두 유전자를 이용한 계통수에서 본 종은 속의 다른 종들과 분명히 분리되지만, 단계통군은 아니었다. 근형질은 기저체의 다층판에 부착된 가로무늬의 microfibril들로 구성되어 있었으며, microfibril들의 끝은 핵의 표면 위에 배열되어 있었다. 이 근형질은 Synurophyceae에서 전형적으로 보이는 기저체-핵 연결자이다. Mallomonas가 SSU와 rbcL자료에 의해 지지되지 않는 결과는 분류군 추가와 함께 좀더 연구되어야 할 것으로 사료된다.

Genetic Diversity and Pathogenicity of Cylindrocarpon destructans Isolates Obtained from Korean Panax ginseng

  • We analyzed the genetic diversity of Cylindrocarpon destructans isolates obtained from Korean ginseng (i.e., Panax ginseng) roots by performing virulence tests and nuclear ribosomal gene internal transcribed spacer (ITS) and mitochondrial small subunit (mt SSU) rDNA sequence analysis. The phylogenetic relationship analysis performed using ITS DNA sequences and isolates from other hosts helped confirm that all the Korean C. destructans isolates belonged to Nectria/Neonectria radicicola complex. The results of in vivo and ex vivo virulence tests showed that the C. destructans isolates could be divided into two groups according to their distinctive difference in virulence and the genetic diversity. The highly virulent Korean isolates in pathogenicity group II (PG II), together with foreign isolates from P. ginseng and P. quinquefolius, formed a single group. The weakly virulent isolates in pathogenicity group I, together with the foreign isolates from other host plants, formed another group and exhibited a greater genetic diversity than the isolates of PG II, as confirmed by the mt SSU rDNA sequence analysis. In addition, as the weakly virulent Korean isolates were genetically very similar to the foreign isolates from other hosts, they were likely to originate from hosts other than the ginseng plants.

Phylogeny of Phellinus and Related Genera Inferred from Combined Data of ITS and Mitochondrial SSU rDNA Sequences

  • To elucidate phylogenetic relationships of Phellinus and its related genera, nuclear internal transcribed spacer and mitochondrial small subunit ribosomal DNA sequences from 65 strains were determined and compared. The combined dataset of two sequences increased informative characters and led to the production of trees with higher levels of resolution. Phylogenetic analysis of the combined dataset revealed thirteen evolutionary lineages and several unresolved species that were together subdivided into two large clusters consisting of oligonucleate species and binucleate species. These results coincided with previous cytological, morphological, and molecular studies. It is newly recognized that the Phellinus linteus complex forms a sister clade to Inonotus, and that Fulvifomes is somehow related to Inocutis. The Phellinus linteus complex of dimitic perennial taxa made an independent clade from Inonotus and suggested that hyphal miticity and fruitbody permanence had enough phylogenetic significance to keep the complex within the traditional genus Phellinus. Taxa lacking setae were clustered into Fulvifomes, Phylloporia, Inocutis, and Fomitiporia, and the first three were closely related sister groups, but Fomitiporia was a genus distantly related to them. Several taxa with branched setae were shown among distantly related genera. Molecular evidence indicated that the ancestral nuclear type could be a binucleate feature, and that there might be parallel gains of branched setae and parallel losses of setae in the Hymenochaetales.

Newly recorded species of the genus Synura (Synurophyceae) from Korea

  • Background: Species in the heterokont genus Synura are colonial and have silica scales whose ultrastructural characteristics are used for classification. We examined the ultrastructure of silica scales and molecular data (nuclear SSU rDNA and LSU rDNA, and plastid rbcL sequences) to better understand the taxonomy and phylogeny within the section Petersenianae of genus Synura. In addition, we report the first finding of newly recorded Synura species from Korea. Results: We identified all species by examination of scale ultrastructure using scanning and transmission electron microscopy (SEM and TEM). Three newly recorded species from Korea, Synura americana, Synura conopea, and Synura truttae were described based on morphological characters, such as cell size, scale shape, scale size, keel shape, number of struts, distance between struts, degree of interconnections between struts, size of base plate pores, keel pores, base plate hole, and posterior rim. The scales of the newly recorded species, which belong to the section Petersenianae, have a well-developed keel and a characteristic number of struts on the base plate. We performed molecular phylogenetic analyses based on sequence data from three genes in 32 strains (including three outgroup species). The results provided strong statistical support that the section Petersenianae was monophyletic, and that all taxa within this section had well-developed keels and a defined number of struts on the base plate. Conclusions: The phylogenetic tree based on sequence data of three genes was congruent with the data on scale ultrastructure. The resulting phylogenetic tree strongly supported the existence of the section Petersenianae. In addition, we propose newly recorded Synura species from Korea based on phylogenetic analyses and morphological characters: S. americana, S. conopea, and S. truttae.
