포르말린으로 유도된 안면통증에 대한 홍삼추출물의 항염증 효과 (Anti-inflammatory Effects of Korean red ginseng Extract in formalin-induced Orofacial Pain in Rats)

  • 본 연구는 건강보조제 및 약제로서 전신의 염증반응 및 여러 약리적 효능을 가지고 있는 홍삼이 국소적인 안면 통증의 완화에 영향을 끼치는지에 관해 조사하였다. 실험동물은 4그룹으로 대조군(n=6)과 5% 포르말린 주입군(n=6), 5% 포르말린+D.W. 주입군, 홍삼(4.5 ml)(n=6) 복용군으로 나누어 45분간 안면을 긁거나 문지르는 행위를 계수하여 비교하였다. 홍삼 복용군에서 포르말린 주입군에 비해 유의하게 긁거나 문지르는 행위가 줄어들었으며, 15분부터 통증완화효과가 증가되기 시작되어 30분까지 유의한 효과를 보였다. 홍삼투여군에 대한 단백질정량분석 (western blot)으로 통증유발기전에 연관되는 염증과 항산화, NO생성에 관련된 신호전달 단백질 p38 MAPK와 iNOS, Nrf2를 뇌, 연수에서 활성정도를 비교한 결과 감소하는 경향을 확인할 수 있었다. 이상의 결과들은 홍삼이 전신적 약리효과 뿐만 아니라 국소적인 염증, 통증완화에도 효능이 있음 나타낸다.

Hexane fraction from the ethanolic extract of Sargassum serratifolium suppresses cell adhesion molecules via regulation of NF-κB and Nrf2 pathway in human umbilical vein endothelial cells

  • Sargassum serratifolium ethanolic extract has been known for strong antioxidant and anti-inflammatory properties. We prepared hexane fraction from the ethanolic extract of S. serratifolium (HSS) to improve biological activities. In this study, we investigated the effects of HSS on the inhibition of tumor necrosis factor (TNF)-${\alpha}$-induced monocyte adhesion to human umbilical vein endothelial cells (HUVECs). We found that HSS suppressed the production of cell adhesion molecules such as intracellular adhesion molecule-1 and vascular cell adhesion molecule-1 in TNF-${\alpha}$-induced HUVECs. Moreover, TNF-${\alpha}$-induced production of monocyte chemoattractant protein 1 and keratinocyte chemoattractant was inhibited by HSS treatment. HSS suppressed TNF-${\alpha}$-induced nuclear factor kappa B ($NF-{\kappa}B$) activation via preventing proteolytic degradation of inhibitor ${\kappa}B-{\alpha}$. HSS induced the production of heme oxygenase 1 via translocation of Nrf2 into the nucleus in TNF-${\alpha}$-treated HUVECs. Overall, HSS alleviated vascular inflammation through the downregulation of $NF-{\kappa}B$ activation and the upregulation of Nrf2 activation in TNF-${\alpha}$-induced HUVECs. These results indicate that HSS may be used as therapeutic agents for vascular inflammatory disorders.

Protective Role of Corticosterone against Hydrogen Peroxide-Induced Neuronal Cell Death in SH-SY5Y Cells

  • Stress breaks body balance, which can cause diverse physiological disorders and worsen preexisting diseases. However, recent studies have reported that controllable stress and overcoming from stress reinforce resilience to resist against more intense stress afterwards. In this study, we investigated the protective effect of corticosterone (CORT), a representative stress hormone against hydrogen peroxide (H2O2)-induced neuronal cell death and its underlying molecular mechanism in SH-SY5Y cells, a human neuroblastoma cell line. The decreased cell viability by H2O2 was effectively restored by the pretreatment with low concentration of CORT (0.03 μM for 72 h) in the cells. H2O2-increased expression of apoptotic markers such as PUMA and Bim was decreased by CORT pretreatment. Furthermore, pretreatment of CORT attenuated H2O2-mediated oxidative damages by upregulation of antioxidant enzymes via activation of nuclear factor erythroid 2-related factor 2 (Nrf2). These findings suggest that low concentration of CORT with eustressed condition enhances intracellular self-defense against H2O2-mediated oxidative cell death, suggesting a role of low concentration of CORT as one of key molecules for resilience and neuronal cell survival.

Tribulus terrestris Suppresses the Lipopolysaccharide-Induced Inflammatory Reaction in RAW264.7 Macrophages through Heme Oxygenase-1 Expressions

  • The fruit of Tribulus terrestris L. (Zygophyllaceae) is an important source of traditional Korean and Chinese medicines. In this study, NNMBS223, consisting of the ethanol extract of T. terrestris, showed potent anti-inflammatory activities in RAW264.7 macrophages. We investigated the effect of NNMBS223 in suppressing the protein expression of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 and production of iNOS-derived nitric oxide (NO), COX-2-derived prostaglandin E2 (PGE2) in lipopolysaccharide (LPS)-stimulated macrophages. In addition, NNMBS223 induced expression of heme oxygenase (HO)-1 through nuclear translocation of nuclear factor E2-related factor 2 (Nrf2) in macrophages. The effects of NNMBS223 on LPS-induced production of NO and PGE2 were partially reversed by the HO activity inhibitor tin protoporphyrin (SnPP). These findings suggest that Nrf2-dependent increases in expression of HO-1 induced by NNMBS223 conferred anti-inflammatory activities in LPS stimulated RAW264.7 macrophages.

HaCaT 인간 피부 각질세포에서 과산화수소 유도 산화 손상에 대한 소청자 및 소총2호의 항산화 및 세포보호 효능 (Antioxidant and Cytoprotective Effects of Socheongja and Socheong 2, Korean Black Seed Coat Soybean Varieties, against Hydrogen Peroxide-induced Oxidative Damage in HaCaT Human Skin Keratinocytes)

  • 식품 원료로도 널리 애용되는 검은콩은 풍부한 천연 페놀 화합물을 함유하고 있기 때문에 기능성 소재로서의 개발에도 매우 유용한 자원이다. 본 연구에서는 3가지 검은콩 품종[소청자(SCJ), 소청2호(SC2) 및 청자2호(CJ2)]을 대상으로 TPCs과 항산화 능을 조사하였다. 그 중에서도 TPCs는 CJ2 $H_2O_2$ 처리에 의한 HaCaT 세포의 생존력 감소를 현저히 억제하여 산화적 스트레스에 대한 보호 효과가 있음을 알 수 있었다. SCJ와 SC2 전처리는 또한 HaCaT 세포에서 mitochondrial dysfunction의 차단과 pro-apoptotic Bax의 발현 변화의 정상화를 통해 $H_2O_2$에 의하여 유도된 apoptosis를 효과적으로 억제하였으며, DNA 손상에 대한 보호 효과와 연관성이 있었다. 또한 SCJ와 SC2는 Nrf2와 연관된 TrxR1의 발현을 효과적으로 유도하였으나, 산화적 스트레스에 대한 SCJ와 SC2의 보호 효과는 TrxR 억제제에 의하여 상쇄되었다. 이러한 결과는 SCJ와 SC2가 Nrf2 신호전달 경로 활성을 통하여 산화적 스트레스와 관련된 세포 손상을 차단함으로써 세포 보호 활성을 갖는다는 것을 의미한다. 결론적으로, SCJ와 SC2는 산화스트레스로 인한 피부 질환의 치료와 예방을 위한 응용 가능성이 높음을 보여주었다.

C2C12 골격근세포에서 황기의 미토콘드리아 조절 작용 (The Effects of Astragali Radix Extracts on Mitochondrial Function in C2C12 Myotubes)

  • Objective: The prevalence of metabolic syndrome and type 2 diabetes is increasing worldwide. Mitochondrial dysfunction is known to be involved in insulin resistance and obesity, researches have been increasing highly. Astragali Radix extract (ARE) or its main components have been shown to perform comparably to insulin by significantly reducing blood glucose levels in animal models however, the influence on mitochondrial dysfunction are not well understood. Methods: ARE (0.2, 0.5 and 1.0 mg/ml) or metformin (2.5 mM) were treated in C2C12 after 6 day-differentiation. The expressions of adenosine monophosphate (AMP)-activated protein kinase (AMPK) and phosphorylation AMPK, peroxisome proliferators-activated receptror ${\gamma}$ coactivator $1{\alpha}$ ($PGC1{\alpha}$), nuclear respiratory factors 1 (NRF1), mitochondrial transcription factor (Tfam) and myosin heavy chain were detected with western blotting or polymerase chain reaction analysis. The morphological changes were also investigated. Results: ARE dose dependently increased phosphorylation of AMPK and respectively activated mRNA expressions of $PGC1{\alpha}$, NRF1 and Tfam which are mitochondrial biogenesis regulators. Furthermore, there were some morphologic differences of differentiated cells between ARE treatment and control. Conclusions: This study suggests that ARE has the potential to increase muscle mitochondrial function by activating AMPK and $PGC1{\alpha}$.

초음파 데이터의 형상 인지 지수를 이용한 확률 격자 지도의 작성 (Grid Map Building through Neighborhood Recognition Factor of Sonar Data)

  • Representing an environment as the probabilistic grids is effective to sense outlines of the environment in the mobile robot area. Outlines of an environment can be expressed factually by using the probabilistic grids especially if sonar sensors would be supposed to build an environment map. However, the difficult problem of a sonar such as a specular reflection phenomenon should be overcome to build a grid map through sonar observations. In this paper, the NRF(Neighborhood Recognition Factor) was developed for building a grid map in which the specular reflection effect is minimized. Also the reproduction rate of the gird map built by using NRF was analyzed with respect to a true map. The experiment was conducted in a home environment to verify the proposed technique.

망막색소상피세포에서 감초 추출물의 산화적 스트레스에 의한 DNA 손상 및 apoptosis 유발의 차단 효과 (Ethanol Extract of Glycyrrhiza uralensis Protects Against Oxidative Stress-induced DNA Damage and Apoptosis in Retinal Pigment Epithelial Cells)

  • 고령층 인구에서 노인성 황반변성(AMD)은 실명의 주요 원인 중 하나이며, 망막 항상성을 유지하는 망막색소상피(RPE) 세포의 산화적 스트레스에 의한 손상은 AMD의 발달에 기여한다. 감초는 한국을 포함한 아시아 국가의 다양한 질병 치료에 가장 널리 사용되는 약초 중 하나이다. 비록 감초는 다양한 실험 모델에서 항산화 효능이 있는 것으로 알려져 있지만, 망막색소상피세포의 손상에 대한 보호 기전은 알려진 바 없다. 본 연구에서는 ARPE-19 인간 망막색소상피에서 과산화수소($H_2O_2$)에 의해 유도된 산화적 손상에 대한 감초 에탄올 추출물의 효능을 평가하였다. 본 결과에 의하면 감초 추출물은 Nrf2 및 HO-1의 발현을 유도하면서 $H_2O_2$에 의한 ROS의 생성을 유의적으로 차단하였다. 또한 감초 추출물은 $H_2O_2$에 의해 유도된 DNA 손상을 억제하였으며, 미토콘드리아막 전위의 소실을 약화시켰다. 그리고 감초 추출물 $H_2O_2$에 의한 caspase-3의 활성을 억제하면서 apoptosis 유도를 차단하였다. 이러한 결과는 감초 추출물이 DNA 손상을 억제하고 apoptosis를 감소시킴으로써 산화적 손상으로부터 망막색소상피세포를 보호할 수 있음을 의미한다. 비록 감초 추출물에 함유된 생리활성 성분들의 분석과 Nrf2-매개 HO-1 발현의 조절에 대한 추가 연구가 수행되어야 하지만, 감초 추출물은 AMD의 위험을 감소시킬 잠재력을 지니고 있음을 시사한다.
