Coenzyme $Q_{10}$ Production by Sphingomonas sp. ZUTE03 with Novel Precursors Isolated from Tobacco Waste in a Two-Phase Conversion System

  • Coenzyme $Q_{10}$ ($CoQ_{10}$) is a widely used supplement in heart diseases treatment or antioxidative dietary. The microbial production of $CoQ_{10}$ was enhanced by addition of solanesol and novel precursors recovered from waste tobacco. The novel precursors were separated by silica gel and identified as ${\alpha}$-linolenic acid (LNA) and butylated hydroxytoluene (BHT) based on the effect on $CoQ_{10}$ production and GC-MS. The effects of novel precursors on $CoQ_{10}$ production by Sphingomonas sp. ZUTE03 were further evaluated in a two-phase conversion system. The precursor's combination of solanesol (70 mg/l) with BHT (30 mg/l) showed the best effect on the improvement of $CoQ_{10}$ yield. A maximal $CoQ_{10}$ productivity (9.5 mg $l^{-1}$ $h^{-1}$) was achieved after 8 h conversion, with a molar conversion rate of 92.6% and 92.4% on BHT and solanesol, respectively. The novel precursors, BHT and LNA in crude extracts from waste tobacco leaves, might become potential candidates for application in the industrial production of $CoQ_{10}$ by microbes.

Hysteretic performance of a novel composite wall panel consisted of a light-steel frame and aerated concrete blocks

  • This study aims at investigating the hysteretic performance of a novel composite wall panel fabricated by infilling aerated concrete blocks into a novel light-steel frame used for low-rise residential buildings. The novel light-steel frame is consisted of two thin-wall rectangular hollow section columns and a truss-beam assembled using patented U-shape connectors. Two bare light-steel frames and two composite wall panels have been tested to failure under horizontal cyclic loading. Hysteretic curves, lateral resistance and stiffness of four specimens have been investigated and analyzed. Based on the testing results, it is found that the masonry infill can significantly increase the lateral resistance and stiffness of the novel light-steel frame, about 2.3~3 and 21.2~31.5 times, respectively. Failure mode of the light-steel frame is local yielding of the column. For the composite wall panel, firstly, masonry infill is crushed, subsequently, local yielding may occur at the column if loading continues. Hysteretic curve of the composite wall panel obtained is not plump, implying a poor energy dissipation capacity. However, the light-steel frame of the composite wall panel can dissipate more energy after the masonry infill is crushed. Therefore, the composite wall panel has a much higher energy dissipation capacity compared to the bare light-steel frame.

  • In this paper, we have improved a novel ECB mode using bend structure with high tilt angle in the unique condition by Hot plate equipment. The new control of tilt angle for nematic liquid crystal (NLC) with negative and positive dielectric anisotropy on the rubbed homeotropic polyimide (PI) using baking method by Hot plate equipment was investigated. LC tilt angle decreased with increasing baking temperature and time. We suggest that the development of the novel ECB cell using control of tilt angle on the homeotropic surface is a promising technique for the achievement of fast response time and high contrast ratio.

  • In order to predict the lifetime of InP Heterojunction Bipolar Transistor (HBT) devices and related circuits in the space radiation environment, a novel model including gamma radiation effects is proposed in this paper. Based on the analysis of radiation-induced device degradation effects including both DC and AC characteristics, a set of empirical expressions describing the device degradation trend are presented and incorporated into the Keysight model. To validate the effective of the proposed model, a series of radiation experiments are performed. The correctness of the novel model is validated by comparing experimental and simulated results before and after radiation.

  • Over the past years, many research works have been carried out to investigate the factors which govern the performance of diesel engines. The air pollutant emissions from the diesel engines are still significant concerns environmentally in many countries. In the present study, a novel smoke filtration system of diesel engine is proposed. This novel system is composed of filtering elements and vacuum pump driven by exhaust gases emitted from engine cylinders for capturing smoke. In order to verify the excellency of the novel filtration system, the differential pressure across the filtration system is investigated experimentally comparing with other systems which are used commercially in diesel vehicles. As a result it is founded that the differential pressure of the novel system is the smallest among them.

A novel laboratory method for measuring the hydraulic conductivity of dredged slurry with high water contents

  • Accurately measuring the hydraulic conductivity of dredged slurry (HCODS) is a difficult task and usually requires highly developed experimental techniques. To resolve such problem, this paper presents a novel laboratory method, where a double drainage sedimentation test (DDST) is proposed to generate a downward seepage after the end of primary consolidation (EOP). Based on the established stress equilibrium equations, it is figured out that the determination of local hydraulic gradients requires the effective stress distribution to be measured. Accordingly, an additional single drainage sedimentation test (SDST) with the same initial water content is performed in the novel laboratory method, which can be utilized to establish the relationship between effective stress and water content for investigated slurry. Thus, HCODS can be determined via a pair of SDST and DDST, with the water contents after the EOP measured. The corresponding calculation procedure is given in details. With a simply-designed settling column, the hydraulic conductivity tests were performed on three types of dredged slurry. The results demonstrated the effectiveness of the novel laboratory method in measuring HCODS.

  • In this paper, we propose a 4-transistor active pixel sensor(APS) with a novel correlated double sampling(CDS) circuit for the purpose of extending dynamic range. Dual sampling techniques can overcome low-sensitivity and temporal disparity problems at low illumination. To accomplish this, two images are obtained at the same time using different sensitivities. The novel CDS circuit proposed in this paper contains MOS switches that make it possible for the capacitance of a conventional CDS circuit to function as a charge pump, so that the proposed APS exhibits an extended dynamic range as well as reduced noise. The designed circuit was fabricated by using $0.35{\mu}m$ 2-poly 4-metal standard CMOS technology and its characteristics have been evaluated.

  • Most researches on monostable LCD and bistable LCD have separately been carried out. We introduce a novel liquid crystal display mode which can be operated as both memory mode and dynamic mode. The novel LCD mode has not only a long term memory time of memory mode but also a fast response time of dynamic mode. We describe switching characteristics of dual mode. Electro-optical characteristics of memory mode and dynamic mode are unique and show the possibility of device application.

A Study on the Novel Rectangular Split Ring Notch Resonators

  • In this paper, a novel notch resonator is proposed based on Rectangular Split Ring Resonator (RRSS) element. We represented the electrical characteristics for R-SRR elements coupled with microstrip line, and measured result is compared with electromagnetic simulation (HFSS) result. We also solved equivalent circuit for R-SRR element, and also discussed influences of change structural parameters of R-SRR. The size of R-SRR is the most important parameters for frequency tuning. Also we can make tunable resonator form the basic structure. This novel resonator can apply to design of tunable bandpass filter and voltage control oscillator.

Novel Vectors for the Convenient Cloning and Expression of In Vivo Biotinylated Proteins in Escherichia coli

  • Biotinylation of recombinant proteins is a powerful tool for the detection and analysis of proteins of interest in a large variety of assay systems. The recent development of in vivo biotinylation techniques in E. coli has opened new possibilities for the production of site-specifically biotinylated proteins without the need for further manipulation after the isolation of the recombinantly expressed proteins. In the present study, a novel vector set was generated which allows the convenient cloning and expression of proteins of interest fused with an N-terminal in vivo biotinylated thioredoxin (TRX) protein. These vectors were derived from the previously reported pBIOTRX vector into which was incorporated part of the pBluescript II+phagemid multiple cloning site (MCS), amplified by PCR using a pair of sophisticated oligonucleotide primers. The functionality of these novel vectors was examined in this system by recombinant expression of rat transforming growth factor-$\beta$. Western-blot analysis using TRX-specific antibodies or peroxidase-conjugated streptavidin confirmed the successful induction of the fusion protein and the in vivo conjugation of biotin molecules, respectively. The convenience of molecular subcloning provided by the MCS and the effective in vivo biotinylation of proteins of interest makes this novel vector set an interesting alternative for the production of biotinylated proteins.
