In vitro-tested ribozyme against synthesized cucumber mosaic virus (CMV) RNA (Agric. Chem. & Biotech. 37:56-63(1994)) was expressed in tobacco plant to develop virus resistant plants. The ribozyme sequence was linked to cauliflower mosaic virus 35S promoter and nopaline synthase(nos) terminator and this chimeric 35S-ribozyme-nos gene was sequenced. The sequenced chimeric gene was transferred to Agrobacterium tumefaciens LBA4404 using tri-parental mating system. The E. coli HB101 containing chimeric gene was incubated with E. coli HB101(pRK2073) as a helper and Agrobacterium tumefaciens LBA4404. Then Agrobacterium cells containing the ribozyme construct was cocultivated with tobacco leaf pieces. Ten different plants were regenerated from kanamycin containing MS medium. The presence of the ribozyme construct in the transgenic tobacco plants was confirmed by polymerase chain reaction (PCR). Seven different transgenic plants in ten different kanamycin resistant plants showed the expected size (570 base pairs) of 35S-ribozyme-nos gene fragment. Total RNAs were isolated from four different transgenic plants and separated on a 1% agarose gel containing formamide. Northern hybridization with 35S-ribozyme-nos gene fragment as a probe indicated that ribozyme transcripts may be degraded tv nuclease. Therefore, nuclease-resistant ribozymes are needed for the development of virus-resistant transgenic plants using ribozymes.

The HCV gene was expressed in potato plants under the control of the constitutive CaMV 355 promoter or tuber-specific patatin promoter. Solanum tuberosum plants carrying a plant expression vector harboring the encoding region of HCV gene were generated by Agrobacterium tumefaciens-mediated in vitro transformation methods. The presence of HCV gene in the plant genome was detected by PCR and DNA hybridization experiments. We obtained the 5 lines of transgenic potato with the pMBPHCV construct and 4 lines of transgenic potato with the pATHCV construct. The HCV transgenic stably integrated into the potato genome, as well as their transcription. HCV mRNA was identified in leaf and tuber tissues of transgenic plants by Northern blot analysis. The transgenic potato plants produced the expected transcript, and the corresponding HCV protein accumulated in individual transgenic plants.
As one way to approach to cold defense mechanism in plants, we previously identified the gene for protein-tyrosine phosphatase (CaTPP1) from hot pepper (Capsicum annuum) using cDNA microarray analysis coupled with Northern blot analysis. We showed that the CaTPP1 gene was strongly induced by cold, drought, salt and ABA stresses. The CaTPP1 gene was engineered under control of CaMV 35S promoter for constitutive expression in transgenic tobacco plants by Agrobacterium-mediated transformation. The resulting CaTPP1 transgenic tobacco plants showed significantly increased cold stress resistance. It also appeared that some of the transgenic tobacco plants showed increased drought tolerance. The CaTPP1 transgenic plants showed no visible phenotypic alteration compared to wild type plants. These results showed the involvement of protein tyrosine phosphatase in tolerance of abiotic stresses including cold and drought stress.
An 1.4 kb of the fad3 cDNA encoding microsomal linoleic acid desaturase catalyzing the conversion of linoleic acid (18:2, ${\omega}-6$) to linolenic acid (18:2, ${\omega}-3$) was introduced into tobacco plants by the Agrobacterium-mediated plant transformation, Among the transgenic tobacco plants conferring kanamycin resistance, five transformants showing increment in unsaturated fatty acid contents were selected and further analyzed for the transgenecity, In genomic Southern blot analyses, copy numbers of the integrated fad3 DNA in chromosomal DNA of the five transgenic tobacco plants were varied among the transgenic lines. By Northern blot analyses, the abundancy of the fad3 mRNA transcript directed by Cauliflower Mosaic Virus 35S promoter was consistent with the relative copy number of the fad3 DNA integrated in the chromosome of transgenic tobacco plants. When compared with the wild type, accumulation of linolenic acid in transgenic tobacco roots was elevated 3.7- to 4.7-fold showing a corresponding decrease in the linoleic acid contents; however, slight increments for linolenic acid were noticed in transgenic leaf tissues. These results indicated that the elevated level of fad3 expression is achieved in transgenic tobacco plants.

This study was conducted to investigate the heavy metal accumulation in wild plants. The samples from 5 plant species on 14 sites were taken on the roadside where urban and industrial regions in northern Gyeonggi do. Artemisia princeps var. orientalis, Chenopodium album L. var. centrobrum, Erigeron canadensis L., Rumex crispus L. and Taraxacum platycapum H. Dahlst were taken from different parts of the plants gathered in surveyed sites for analysing. This study classified which species were suitable to accumulators, excluders, or indicators which were provided as Phytoremediation for heavy metals(Cd, Cr, Ni, Pb and Zn) and considered the applicability of that work. 1. The mean accumulation of 6 heavy metals in plants was as follows; Zn> Cu> Cr> Pb> Ni> Cd in rows except for Ni and Pb in Erigeron. 2. The highest heavy metal accumulators were Taraxacum and Artemisia. 3. The comparison of heavy metal accumulations between the tops and roots in the plants was as follows; Zn> Cd> Pb> Ni> Cu> Cr. 4. Comparison of heavy metal accumulations between tops and roots, root parts were higher than the top parts in most of the plants. Especially, in Taraxacum, a ratio was over 1.0 in Cr, Ni, and Zn. Therefore, Taraxacum was an accumulator for 3 heavy metals. And Erigeron, Chenopodium, Rumex, Artemisia were good for excluders which had lower than 1.0.

This study was carried out to investigate the flora of Baengnyeongdo (Isl.). The vascular plants which were collected in 5 times (from Aug. 2011 and Nov. 2012) were identified as 732 taxa in total, including 124 families, 411 genera, 647 species, 8 subspecies, 68 varieties, 7 forms and 2 hybrids. 193 of these taxa were first records for the region. In the flora of this area, the endangered plants designated and protected by the wildlife protection law, the Ministry of Environment, Korea were counted to two taxa. Korean endemics plants numbered 8 taxa, and 78 taxa of specially designated floristic regional indicator plants by the Ministry of Environment. And rare plants were identified as 20 taxa. Based on the list of southern and northern limit of distribution plants, 30 taxa and 43 taxa of halophytes were recorded in the investigated area. The naturalized plants were identified as 92 taxa, and their naturalization ratio were found to be 12.5%, respectively.
Growth and ecological differences of some important medicinal plants (Carthamus tinctorius, Astragalus membranaceus, Digitalis purpurea, Melissa officinalis, Anethum graveolens, Angelica gigas and Saururus chinensis) were observed at the different attitudinal sites of Mt. Halla in Cheju from 1997 to 1998. Growth of all medicinal plants was generally higher in the northern slope parts than in the southern slope parts. But at 50m above the sea level in the northern slope regions, the germination, growth and yield of Astragalus membranaceus and Digitalis purpurea were reduced by the wind. As the altitude was higher, the temperature was more slowly down in the northern slope regions than in the southern slope regions, while, the moisture was relatively high. The growth of Carthamus tinctorius, Melissa officinalis, Anethum graveolens and Angelica gigas were generally reduced by the higher altitude. On the other hand, growth of Saururus chinensis was good at the 700m above the sea level.
Long-term observation of the life cycle of plants allows the identification of critical signals of the effects of climate change on plants. Indeed, plant phenology is the simplest approach to detect climate change. Observation of seasonal changes in plants using digital repeat imaging helps in overcoming the limitations of both traditional methods and satellite remote sensing. In this study, we demonstrate the utility of camera-based repeat digital imaging in this context. We observed the biological events of plants and quantified their phenophases in the northern temperate type deciduous broadleaf forest of Jeombong Mountain. This study aimed to identify trends in seasonal characteristics of Quercus mongolica (deciduous broadleaf forest) and Pinus densiflora (evergreen coniferous forest). The vegetation index, green chromatic coordinate (GCC), was calculated from the RGB channel image data. The magnitude of the GCC amplitude was smaller in the evergreen coniferous forest than in the deciduous forest. The slope of the GCC (increased in spring and decreased in autumn) was moderate in the evergreen coniferous forest compared with that in the deciduous forest. In the pine forest, the beginning of growth occurred earlier than that in the red oak forest, whereas the end of growth was later. Verification of the accuracy of the phenophases showed high accuracy with root-mean-square error (RMSE) values of 0.008 (region of interest [ROI]1) and 0.006 (ROI3). These results reflect the tendency of the GCC trajectory in a northern temperate type deciduous broadleaf forest. Based on the results, we propose that repeat imaging using digital cameras will be useful for the observation of phenophases.

Human lactoferrin is an iron-binding glycoprotein with many biological activities, including the protection against microbial and virus infection and stimulation of the immune system. We introduced a human lactoferrin (hLf) cDNA under the control of 35S promoter into sweet potato by particle bombardment. Transgenic plants were regenerated via somatic embryogenesis. Transgenic plants were produced typical tuberous roots in soil. PCR, Southern and northern analyses confirmed that the hLf cDNA was incorporated into the plant genome and was properly expressed in plants. Western blot analysis showed that the 80 kDa full length hLf protein was produced in transgenic tuberous roots. Overall results indicated that sweet potato would be an excellent host to produce human therapeutic proteins.
$\textrm{T}_{5}$ progeny of one transgenic tomato line (To9) carrying antisense polygalacturonase (PG) cDNA was generated by selfing. Five $\textrm{T}_{5}$ plants were used to analyse in detail. The PG antisense gene was stably inherited through fifth generations. In all five $\textrm{T}_{5}$ plants, expression of the antisense transcripts were detected. In consequence, it led to a reduction of the PG enzyme activity in ripe fruit to between 37% and 65% that of normal. In two plants the expression of endogenous PG gene was inhibited in ripe fruit.
