The Homologous Region 3 from Bombyx mori Nucleopolyhedrovirus Enhancing the Transcriptional Activity of Drosophila hsp70 Promoter

  • Drosophila melanogaster heat shock protein 70 gene promoter (Dhsp70p) is widely used in transgenic insect to drive exogenous gene, and the homologous region 3 from Bombyx mori nucleopolyhedrovirus (BmNPVhr3) functions as an enhancer for several promoters. To test whether BmNPVhr3 can enhance the Dhsp70ps transcriptional activity, the reporter plasmids, which contain the Dhsp70p, the reporter $\beta$-galactosidase gene with SV40 terminator and BmNPVhr3 fragment, are constructed and transfected into the insect cell lines (Bm-N cells and Sf-21 cells) by lipofectin-mediated method. The results from the transient expression assay show that BmNPVhr3 significantly increases transcriptional activity of Dhsp70p both under the normal condition and under the heat-shock treatment, although the effects are significantly different between in Bm-N cells and in sf-21 cells. The enhancing behavior of BmNPVhr3 on the Dhsp70p is in an orientation-independent manner. Meanwhile, the effects of heat-shock treatment on Dhsp70p alone or Dhsp70p/BmNPVhr3 combination present no significant difference, indicating that BmNPVhr3 only enhances the transcriptional activity of Dhsp70p, but cant alter its characteristic of the response to the heat-shock stress. The above results suggest that the Dhsp70p/BmNPVhr3 combination is more effective one to drive exogenous gene for transgene or stable cell expression system in insects.

  • Numerical simulations for fluid flow and solute transport in a fracture rock masses are performed by using a transient flow model, which is based on the three-dimensional stochastic and discrete fracture network model (DFN model) and is coupled hydraulic model with mechanical model. In the numerical simulations of the solute transport, we used to the particle following algorithm which is similar to an advective biased random walk. The purpose of this study is to predict the response of the tracer test between two deep bore holes (GPK1 and GPK2) implanted at Soultz sous Foret in France, in the context of the geothermal researches.l The data sets used are obtained from in situcirculating experiments during 1995. As the result of the transport simulation, the mean transit time for the non reactive particles is about 5 days between two bore holes.

  • A main steam pipe system is a branch pipe that connects a boiler with a turbine. Water hammering analysis is very important for limiting the damage caused to pipe systems by operation conditions. Water hammering created by an unsteady flow in pipeline systems can cause excessive change in pressure, vibration, and noise. The main steam pipe structure should be designed to safely maintain the pressure pulsation and several vibrations under operation environments. This study evaluated the structural integrity of a main steam pipe during suspended and normal operation by using the ASME fatigue life methodology and finite element analysis. In the analysis, water hammering was used for transient analysis. The calculated alternating stress and fatigue stress were compared with the applicable limits of ASME fatigue life. All the evaluation results satisfied the requirements of the ASME fatigue life.

  • The objective of this work is to apply artificial neural networks for solving inverse problems in the structural optimization of a fiber optic pressure sensor. For the sensor under investigation to achieve a desired accuracy, the change in the distance between the tips of the two fibers due to the applied pressure should not interfere with the phase change due to the change in the density of the air between the two fibers. Therefore, accurate dynamic analysis and structural optimization of the sensor is essential to ensure the accuracy of the measurements provided by the sensor. To this end, a normal mode analysis and a transient response analysis of the sensor were performed by combining commercial finite element analysis package, MSC/NASTRAN, and MATLAB. Furthermore, a parametric study on the design of the sensor was performed to minimize the size of the sensor while fulfilling a number of constraints. In performing the parametric study, the need for a relationship between the design parameters and the response of the sensor was fulfilled by using a neural network. The whole process of the dynamic analysis using commercial finite element analysis package and the parameter optimization of the sensor were automated within the MATLAB environment.

Analysis of Influence on Derailment due to Primary Spring Aging (축상스프링 노화에 따른 탈선안전도 영향 분석)

  • In order to analyze the influence on derailment safety according to the aging of primary rubber springs widely applied to railway vehicles, characteristic tests were carried out on aged primary rubber spring samples. To analyze the effect of primary rubber spring aging on derailment safety, a vehicle dynamic analysis was carried out. The results of the vertical characteristics test for the rubber spring specimens with 17 years of service life revealed that the displacement restoration function was degraded due to rubber aging and the spring stiffness significantly increased. The results of the running dynamic analysis simulating the twist track running in accordance with the EN14363 standard, compared with the normal vehicle model (Case 1), showed that the derailment coefficient and the wheel unloading of the vehicle model (Case 2) using the aging primary spring characteristic increased, and the derailment safety was degraded. IN particular, it was found that the derailment safety due to the reduction of the wheel load is weak in the transient section where a steep slope change occurs.

  • The thermal-hydraulic characteristics of the 17$\times$17 OFA (Optimized Fuel Assembly) used in the KNU 7&8 are analyzed and compared with that of the 17$\times$17 SFA (Standard Fuel Assembly) loaded in the KNU 5&6. The thermal-hydraulic characteristics analyzed are minimum DNBR, fuel centerline temperature and exit void fraction at normal operation and design over power transient. Additionally, local linear rod power, which will cause fuel centerline melting, is calculated. The DNBR sensitivity calculations are performed with respect to the reactor operating parameters. COBRA-IV-I code is used for these calculations. The modified W-3 correltion and the drift-flux model are applied for the critical heat flux calculation and the void fraction calculation, respectively. From the calculated results, it has been found that the possibility of DNB occurrence is higher in the OFA than in the SFA. The other hand, the local linear power resulting in fuel centerline moiling of the OFA is nearly equal to that of the SFA.

Effects of Zingiberis Rhizoma Pharmacopuncture Injected at GB30 and ST36 on Neuropathic Pain in Rats (환도(GB30) 및 족삼리(ST36) 건강약침이 신경병증성 통증 유발 흰쥐에 미치는 영향)

  • Objectives : The objective of this study was to investigate the effects of Zingiberis Rhizoma Pharmacopuncture(ZP) at GB30 and ST36 in neuropathic pain induced SD rats by the block of Transient Receptor Potential Vanilloid 1(TRPV1). Methods : Neuropathic pain in rats was induced by tibial and common peroneal nerve transection of right leg. The rat subjects were divided into 6 groups : normal(Nor, n=5), control(Con, n=5), neuropathic pain plus 2 mg/kg ZP injection at GB30 and ST36(ZP-A, n=5), 10 mg/kg ZP(ZP-B, n=5), 20 mg/kg ZP(ZP-C, n=5) and 0.45 mg/kg Tramadol(Tra, n=5). Three days after the surgery, injections were administered once a day for 17 days. Withdrawal response of neuropathic rats' legs were measured by stimulating the paw of Right leg with von frey filament, acetone and radient heat on day 3, 7, 11, 15, 19 after surgery. After all treatments were completed, c-Fos in the midbrain central gray and TRPV1 & TRPA1 of DRG(L5) were analyzed. Results : Groups ZP-B and ZP-C showed a meaningful decrease in the withdrawal response of mechanical allodynia, thermal hyperalgesia and cold allodynia compared to the control group(p<0.05, p<0.01, p<0.001). Groups ZP-B and ZP-C showed a meaningful decrease in the expression of c-fos and TRPV1 protein level compared to the control group(p<0.05, p<0.01, p<0.001). Conclusions : These results suggest that Zingiberis Rhizoma Pharmacopuncture at GB30 and ST36 could decrease mechanical & cold allodynia and thermal hyperalgesia by block the TRPV1 on the model of neuropathic pain.

An Inventory Model-based Spectrum Pooling in Cognitive Radio Networks (인벤토리모델을 이용한 인지무선네트워크에서 스펙트럼풀링)

  • Recently, many research efforts have paid attention to spectrum pooling meshanism that provides efficient way to manage transient spectrum requests of secondary users in cognitive radio networks. Spectrum pooling is maintained by WSP (wireless service provider). WSP leases the spectrums stored in her spectrum pool to secondary users with being paid for it. It is natural that WSP tries to get profits as much as possible, which implies the WSP tries to minimize the cost required for maintaining her spectrum pool. In this paper, we model the spectrum pool into a probabilistic inventory model. Assuming secondary users' spectrum requests follow normal distribution, we give a strategic way that minimizes the maintenance cost of the spectrum pool. By a series of simulations, we show that WSP can reduce the total maintenance cost through our inventory model-based spectrum pooling than maintaining fixed inventory level.

Brachial Artery Thrombosis in an 8-year-old Boy with Antiphospholipid Antibodies Induced by Mycoplasma pneumoniae Infection: a Case Report

  • Antiphospholipid antibodies may be produced in cases involving autoimmune diseases and can sometimes be caused by infections, such as Mycoplasma pneumoniae infection. However, antiphospholipid antibodies causing thrombosis associated with M. pneumoniae pneumonia in children have rarely been reported. We report a case of an 8-year-old boy with M. pneumoniae pneumonia with antiphospholipid antibodies, complicated by brachial artery thrombosis. He was found to have antiphospholipid antibodies and low protein S levels. The brachial artery thrombus was removed via thrombectomy. The titers of antiphospholipid antibodies turned normal within 5 months. This is a rare case of M. pneumoniae infection with brachial artery thrombosis associated with transient antiphospholipid antibodies.

Design of AC/DC Combined V2X System for Small Electric Vehicle (소형 전기차 적용을 위한 AC/DC 복합 V2X 시스템 설계)

  • The small electric vehicles equipped with V2X(vehicle to everything) systems may provide more information and function to the existing navigation system of the vehicle. The key components of V2X technology include V2V (vehicle to vehicle), V2N(vehicle to network) and V2I (vehicle to infrastructure). This study is to design and implementation of VI type E-PTO which is interfaced with external equipments, the work designs the components of E-PTO such as DC/DC converter, DC/AC converter, battery bidirectional charging system etc. Also, it implements the devices and control systems for driving. The test results of VI type E-PTO components showed allowable 10% requirements of transient voltage variation rate and recovery time within 100ms for start/stop and normal operation.
