The effects of Hemistepta lyrata Bunge (Bunge) fractionated extract on liver X receptor α-dependent lipogenic genes in hepatocyte-derived cells (간 실질세포주에서 니호채(泥胡菜) 분획물이 liver X receptor α 의존적 지방 생성 유전자의 발현에 미치는 효과)

  • Objectives : Hemistepta lyrata Bunge (Bunge) is a wild herb that has been used for managing fever and wound in Korean Traditional Medicine. The present study explored the effects of H. lyrata extract on liver X receptor (LXR) α-dependent lipogenic genes in hepatocyte-derived cells. Methods : After HepG2 cells or Huh7 cells were pre-treated with 1-10 ㎍/mL of H. lyrata extract or its fractionated extract for 0.5 h, the cells were subsequently exposed to LXR ligand for 6-24 h. Cell viability, LXR response element (LXRE)-driven luciferase activity, sterol regulatory element binding protein-response element (SREBP-RE)-driven luciferase activity, SREBP-1c expression, and mRNA levels of LXRα and its-dependent target genes were determined. In addition, LC-MS/MS analysis was conducted to explore major compounds in H. lyrata-chloroform fractionated extract #4 (HL-CF4). Results : Of various H. lyrata extracts tested, chloroform extract and its fractionated extract #4, HL-CF4, significantly decreased T0901317-mediated SREBP-1c expression. In addition, HL-CF4 significantly reduced LXRE atransactivation and LXRα mRNA expression without any cytotoxicity. Moreover, HL-CF4 prevented the SREBP-RE-driven luciferase activity and mRNA levels of fatty acid synthase and stearoyl-CoA desaturase-1 induced by T0901317. Results from LC-MS/MS analysis at positive/negative mode indicated that HL-CF4 contained several compounds showing m/z 197.1176 (C11H17O3), 693.2913/227.1069 (C38H45O12/C15H15O2), 203.1797 (C15H23), 181.1225 (C11H17O2), 591.2957 (C35H43O8), 379.1040 (C18H19O9), 409.1509 (C20H25O9), 309.1348 (C16H21O6), 391.1404 (C20H23O8), and 669.2924/389.1248 (C36H45O12/C20H21O8). Conclusion : Based on its inhibition of the LXRα-dependent signaling pathway, H. lyrata chloroform extract and HL-CF4 have prophylactic potentials for managing non-alcoholic fatty liver.

Antiangiogenic Activity of Coptis chinensis Franch. Water Extract in in vitro and ex vivo Angiogenesis Models (In vitro와 ex vivo 혈관신생 모델에서 황련 냉수추출물의 신생혈관 억제효과)

  • Angiogenesis, the formation of new blood vessels, plays an important role in tumor growth and metastasis; therefore, it has become an important target in cancer therapy. Novel anticancer pharmaceutical products that have relatively few side effects or are non-cytotoxic must be developed, and such products may be obtained from traditional herbal medicines. Coptis chinensis Franch. is an herb used in traditional medicine for the treatment of inflammatory diseases and diabetes. However, potential antiangiogenic effects of C. chinensis water extract (CCFWE) have not yet been studied. The purpose of this study was to determine the antiangiogenic effect of CCFWE in order to evaluate its potential for an anticancer drug. We found that the treatment with CCFWE inhibited the major steps of the angiogenesis process, such as the endothelial cell proliferation, migration, invasion, and capillary-like tube formation in response to vascular endothelial growth factor (VEGF), and also resulted in the growth inhibition of new blood vessels in an ex vivo rat aortic ring assay. We also observed that CCFWE treatment arrested the cell cycle at the G0/G1 phase, preventing the G0/G1 to S phase cell cycle progression in response to VEGF. In addition, the treatment reduced the VEGF-induced activation of matrix metalloproteinases 2 and 9. Taken together, these findings indicate that CCFWE should be considered a potential anticancer therapy against pathological conditions where angiogenesis is stimulated during tumor development.

Systems Pharmacological Approach to Identification of Schizonepeta teunifolia Extract via Active Ingredients Analysis and Cytotoxicity Effect on A549 Cell Lines (형개 추출물의 시스템 약리학적 분석과 비소세포폐암세포에 대한 증식 억제효과)

  • Objectives : This study aimed to predict the effectiveness and potential of Schizonepeta tenuifolia as an anticancer treatment for non-small cell lung cancer through network-based pharmacology and cellular experiment. Methods : To identify the major bioactive compounds in Schizonepeta tenuifolia, we used the Traditional Chinese Medicine Systems. The target genes for the cancer treatment were selected using the UniProt database and the networked using Cytoscape. We performed functional enrichment analysis based on the Gene Ontology Biological Process and Kyoto Encyclopedia of Genes and Genomes Pathways to predict the mechanisms. To investigate the effect of Schizonepeta tenuifolia on lung cancer cell growth, we treated A549 cells, a lung cancer cell line, with different concentrations of the drug and used the MTT assay for cell viability. Results : Research has shown that the most effective mechanism of active compounds from Schizonepeta tenuifolia is through the pathway of cancer. The results of the network pharmacology analysis indicate that Schizonepeta tenuifolia has potential medicinal value as an adjuvant in anticancer treatment. The concentration-dependent inhibition of cell viability was observed on A549 cells. Furthermore, synergistic anticancer activity with Doxorubicin was also observed. Conclusions : Through a network pharmacological approach, Schizonepeta tenuifolia was predicted to have potential as an anticancer agent, and its efficacy was experimentally demonstrated using A549 cells. These findings suggest that Schizonepeta tenuifolia is a promising candidate for future research.

Hydrolysis of Non-digestible Components of Soybean Meal by α-Galactosidase from Bacillus coagulans NRR1207 (Bacillus coagulans NRR1207이 생산하는 α-galactosidase에 의한 대두박 비소화성분의 가수분해)

  • The fermentation of non-digestible soy meal can convert polysaccharides into many compounds that have a wide variety of biological functions. Bacillus strains are capable of hydrolyzing non-digestible saccharides, such as melibiose, raffinose, and stachyose, found in soy meal components. A highly active ${\alpha}$-galactosidase (${\alpha}$-d-galactoside galactohydrolase, EC 3.2.1.22) was isolated from a bacterium in a traditional Korean fermented medicinal herb preparation. The isolate, T2-16, was identified as Bacillus coagulans based on its 16S rRNA sequence and biochemical properties, and the strain was named Bacillus coagulans NRR-1207. When incubated in 10%(w/v) skim milk, Bacillus coagulans NRR1207 caused a decrease in the pH of the culture medium, as well as an increase in titratable acidity and viable cell counts. This strain also showed higher activities of ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\alpha}$-glucosidase, naphthol-AS-BO-phosphohydrolase, and acid phosphatase when compared to other enzymes. It hydrolyzed oligomeric substrates, such as raffinose and stachyose, and liberated galactose, indicating that the Bacillus coagulans NRR1207 ${\alpha}$-galactosidase hydrolyzed the ${\alpha}$-1,6 glycoside linkage. These results suggest that the decreased stachyose and raffinose contents observed in fermented soy meal are due to this ${\alpha}$-galactosidase activity. Bacillus coagulans NRR1207 therefore has potential probiotic activity and could be utilized in feed manufacturing, as well as for hydrolyzing non-digestible soy meal components.

Review of Clinical Studies on Oriental Medicine Treatment for Obesity in the Korean Literature (비만의 한방 치료에 대한 국내 임상 논문 고찰)

  • The aim of this review is to investigate clinical studies on oriental medicine treatment for obesity in Korean literature and to propose for the better method of clinical studies in order to seek more effective treatment. Electric searches were performed with NDSL, National assembly library, RISS4U, DBPIA, KISS, KMBASE, KoreaMed, Korean traditional knowledge portal and Oasis. Also, 21 major journals associated with oriental medicine in Korea were searched manually. Eighty three studies were included and analyzed in terms of study design, subjects, interventions, results and evaluation of safety. The numbers of controlled, non-controlled and case studies are respectively 26(31%), 43(52%) and 14(17%). Studies using sample size less than 30 accounts for 49% and studies using subjects diagnosed as obese or overweight by specific criteria were 35(42%). Studies which used more than 2 interventions or co-interventions were 76%. Clinical studies which used single herbal medicine were few and clinically studied single herbal medicine were limited to ephedra. More than half of 83 studies were studied for 8 weeks and less. 12% of 69 studies(except case studies) were not statistically analyzed and studies which evaluated safety of intervention were 22(27%) and reported adverse events were 9(11%). To improve the quality of clinical studies on oriental medicine treatment for obesity, studies need to use restricted number of interventions and subjects need to specifically diagnosed as overweight or obese. In addition, various kinds of single herbs need to be clinically studied based on experimental studies and herbalogy.

Anti-inflammatory effects of Rosa rugosa extracts in RAW264.7 cells exposed to particulate matter (PM10) (미세먼지 PM10에 노출된 RAW264.7 세포에 대한 해당화 추출물의 항염증 활성)

  • Airborne fine dust (FD) particles smaller than 10 ㎛ in diameter (PM10) are one of the major causes of air pollution in East Asia, including Korea, and have become a major contributor to respiratory and skin problems. FD inordinately promotes the production of reactive oxygen species and inflammatory response in macrophages, leading to cell damage and death. Rosa rugosa, a deciduous shrub of the Rosa genus, has been used in traditional East Asian herbal medicine to treat various illnesses. The present study investigated the anti-inflammatory effects of R. rugosa organ extracts on PM10-stimulated RAW264.7 macrophages. Compared to non-treated RAW264.7 cells, treatment with 100 ㎍.ml-1 PM10 resulted in increased nitric oxide (NO) production, similar to lipopolysaccharide treatment. Additionally, 100 ㎍/ml stem extract reduced NO production by more than 45% compared to mock treatment. Furthermore, PM10-induced expression of interleukin (IL)-1β, IL-6, inducible NO synthase, and cyclooxygenase-2 was significantly reduced by stem extract treatment, indicating that the anti-inflammatory effect of the stem extract is mediated by the inhibition of pro-inflammatory mediators in PM10-stimulated RAW 264.7 cells. These results indicate that the R. rugosa stem could be considered a natural remedy with a protective effect against inflammatory responses induced by harmful airborne dust.

The Taste Compounds of Damchi-jeotguk -Concentrated Sea Mussel Extract- (담치젓국의 정미성분(呈味成分))

  • We have examined to evaluate the taste compounds of damchi-jeotguk(concentrated sea mussel extract) which is a kind of traditional processed sea food in Korea. The contents of such compounds as free amino acids, nucleotides their and related compounds, non-volatile organic acid and fatty acid composition were analyzed. The content of total free amino acids was 10520.5mg/100g on dry basis and the major ones were glycine, arginine, aspartic acid and glutamic acid. These amino acids were resulted as 55.7% of total free amino acids in damchi-jeotguk. Hypoxanthine was the highest content($8.77\;{\mu}mole/g$, dry basis) of nucleotides and their related compounds. Free amino acid-N was the most abundant, resulting 53.3% of extractive nitrogen which was 3490.2mg/100g on dry basis and next ammonia-N, nucleotide-N and TMAO-N in order. The major non-volatile organic in damchi-jeotguk were succinic acid(125.5mg/100g, dry basis) and lactic acid 91.9mg/100g, dry basis). In this fatty acid composition of total lipid, polyenoic acid was abundant holding 45.5%. The major fatty acids were 16 : 0, 18 : 2, 22 : 6, 18 : 1 and 20 : 5. It was concluded from the omission test and chemical analysis that the major taste compounds of damchi-jeotguk were free amino acids and non-volatile organic acids.

Cordycepin Inhibits Migration and Invasion of HCT116 Human Colorectal Carcinoma Cells by Tightening of Tight Junctions and Inhibition of Matrix Metalloproteinase Activity (Cordycepin의 치밀결합 강화 및 MMPs의 활성 억제를 통한 HCT116 인체대장암세포의 이동성 및 침윤성의 억제)

  • Cordycepin is the major functional component of Cordyceps species and is widely used in traditional oriental medicine. Cordycepin has been shown to possess many pharmacological properties, such as enhancement of immune function along with anti-inflammatory, antioxidant, anti-aging, and anti-cancer effects. Here, we investigated the inhibitory effects of cordycepin on cell migration and invasion, which are two critical cellular processes that are often deregulated during metastasis, using HCT116 human colorectal carcinoma cells. According to our data, cordycepin at non-cytotoxic concentrations markedly inhibited the motility and invasiveness of HCT116 cells in a time-dependent manner. RT-PCR and Western blotting results indicated that cordycepin reduced the levels of claudin proteins, which are major components of tight junctions (TJs), and induced tightening of TJs. Cordycepin also attenuated the expression and activities of matrix metalloproteinases (MMPs)-2 and -9, whereas levels of tissue inhibitor of metalloproteinases (TIMPs)-1 and -2 were simultaneously elevated. These findings suggest that cordycepin reduces the migration and invasion of HCT116 cells by modulating the activities of TJs and MMPs.

A Study on the Organization Design of Regional Innovation Councils for Establishing Strategic Regional Innovation Systems: Based on Kang-Won Province Case (전략적 지역혁신체재 구축을 위한 지역혁신협의회 조직에 관한 연구 - 강원도 사례를 중심으로)

  • In 2004, in order to create core competences and major growth drivers for endogenous regional developments, the Korean government adopted the policies for establishing regional innovation systems, The main organization for the policies is the regional innovation council which is composed of diverse groups of scholars, professionals, and government officials. The regional innovation council of Kang-won Province consists of a couple of sub-divisions such as Operation Committee, Administration Office, Planning and Moderation Department, Strategic Industry Department, Culture and Tourism Industry Department, Regional Industry Department, and Regional Human Resource Development Department. The primary roles and jobs are planning, reviewing, evaluating and finalizing regional innovation policies. However, because of conflicts with local government, insufficient budget problems, organization structure problems, and decision making process problems, the regional innovation councils could not perform their original roles and functions. They could not adopt radical or non-traditional strategies. Although it's failure, a lot of people recognize the importance of regional innovation councils' roles and functions, I believe there would be another regional innovation councils coming continuously which would be take more important roles and have more power over budgeting problems. The purpose of this article is to identify the primary sources of strategies and organization structure problems of the previous regional innovation councils and to provide a couple of answers. In this paper, the author primarily analyze the problems of the Kang-won innovation council. A couple of answers to the desirable organization structures and the desirable roles of regional innovation councils will be presented for discussion.

Prospects of Triticale as Fodder and Feed in Farming of Bangladesh (방글라데시 농업에서 트리티게일의 조사료 및 곡물사료이용 전망)

  • This paper reviews the present situation of Triticale cultivation and examines the potentiality of contribution to livestock as well as poultry sector in Bangladesh Agriculture. Triticale is a human-made cross between rye and durum wheat that has the ability to produce quality green fodder, and then re-grow after first and second cutting to produce grain. In Bangladesh, it is a non-traditional cereal that grows well during the cool and dry Rabi season (November-March) when fodder and feed scarcity is a major limiting factor for ruminant livestock. In Bangladesh Triticale was started to grow in the late Ninety's. The scientists of Bangladesh Agricultural Research Institute (BARI) were first introduced triticale in Bangladesh. Still now the situation of Triticale is grown as fooder and feed in Bangladesh within the scientists under trial. High quality grass fodder was obtained by cutting green triticale plants twice, at 35 and 50 days after seeding, while later the ratooning tillers produced grain to a yield of 1.1-2.4 t/ha of grain for poultry feed or human food. Triticale straw was twice as nutritious as rice or wheat straw and its grain contained more protein than other cereals. Researchers and farmers have also successfully made triticale hay and silage from a mixture of triticale green cuttings, rice straw and molasses. A feeding trial at Bangladesh Livestock Research Institute(BLRI), Savar station showed a large(46%) increase in cow live weight gain and a 36% increase in milk yield (but no change in milk quality or dry matter intake) in cows fed triticale silage compared with those fed rice straw over a period of nine weeks. In another feeding trial, it was found that triticale grain was a good replacement for wheat in the feed blend for chickens in Bangladesh. So it will be a good chance to alive our livestock as well as poultry sector if triticale enters to our existing cropping system as fodder cum grain. The challenge in Bangladesh is to identify fodder technologies that match existing small-scale farmer cropping patterns without needing major inputs or increasing risks. Preliminary field experiments revealed that triticale is a crop with good potential to produce quality fodder and grain for small scale farmers in Bangladesh.
